| 1 | LncRNAs-directed PTEN enzymatic switch governs epithelial–mesenchymal transition显示文摘Despite the structural conservation of PTEN with dual-specificity phosphatases,there have been no reports regarding the regulatory mechanisms that underlie this potential dual-phosphatase activity.Here,we report that K27-linked polyubiquitination of PTEN at lysines 66 and 80 switches its phosphoinositide/protein tyrosine phosphatase activity to protein serine/threonine phosphatase activity.Mechanistically,high glucose,TGF-β,CTGF,SHH,and IL-6 induce the expression of a long non-coding RNA,GAEA(Glucose Aroused for EMT Activation),which associates with an RNA-binding E3 ligase,MEX3C,and enhances its enzymatic activity,leading to the K27-linked polyubiquitination of PTEN.The MEX3C-catalyzed PTEN^K27-polyUb activates its protein serine/threonine phosphatase activity and inhibits its phosphatidylinositol/protein tyrosine phosphatase activity.With this altered enzymatic activity,PTEN^K27-polyUb dephosphorylates the phosphoserine/threonine residues of TWIST1,SNAI1,and YAP1,leading to accumulation of these master regulators of EMT.Animals with genetic inhibition of PTEN^K27-polyUb,by a single nucleotide mutation generated using CRISPR/Cas9(Pten^K80R/K80R),exhibit inhibition of EMT markers during mammary gland morphogenesis in pregnancy/lactation and during cutaneous wound healing processes.Our findings illustrate an unexpected paradigm in which the lncRNA-dependent switch in PTEN protein serine/threonine phosphatase activity is important for physiological homeostasis and disease development. | Qingsong Hu Chunlai Li Shouyu Wang Yajuan Li Bo Wen Yanyan Zhang Ke Liang Jun Yao Youqiong Ye Heidi Hsiao Tina K.Nguyen Peter K.Park Sergey D.Egranov David H.Hawke Jeffrey R.Marks Leng Han Mien-Chie Hung Bing Zhang Chunru Lin Liuqing Yang | 2019 | Cell Research2019,29,4: | 8 |
| 2 | Photoluminescence enhancement of red-emitting GdVO_4:Eu and YVO_4:Eu phosphors by adding zinc显示文摘We synthesized the rare-earth activated R0.94-xEu0.06ZnxVO4 (R: Gd and Y; 0≤x≤0.08) phosphors with a spherical morphology and a smooth surface by the ultrasonic spray pyrolysis. The annealed R0.94-xEu0.06ZnxVO4 crystallized in the tetragonal zircon type structure, belonging to the space group of I41/amd. The incorporation of a small amount of Zn to R0.94Eu0.06VO4 improved the emission characteristics. The emission intensities of the Gd0.88Eu0.06Zn0.06VO4 and Y0.9Eu0.06Zn0.04VO4 phosphors at 619 nm were 72% and 21% stronger than those of the Gd0.94Eu0.06VO4 and Y0.94Eu0.06VO4 phosphors, respectively. | 17 | Behavioral evidence for contextual olfactory-mediated avoidance of the ubiquitous phytopathogen Botrytis cinerea by Drosophila suzukii显示文摘Herbivorous insects may benefit from avoiding the smell produced by phytopathogens infecting plant host tissue if the infected tissue reduces insect fitness.However,in many cases the same species of phytopathogen can also infect host plant tissues that do not directly affect herbivore fitness.Thus,insects may benefit from differentiating between pathogen odors emanating from food and nonfood tissues.This is based on the hypothesis that unecessarily staying attentive to pathogen odor from nonfood tissue may incur opportunity costs associated with not responding to other important survival functions.In this study adults of Drosophila suzukii Matsumura,an invasive larval frugivore,showed reduced attraction to the odor of raspberry fruit,a food tissue,when infected with Botrytis cinerea Pers.,a ubiquitous phytopathogen,in favor of odors of uninfected raspberry fruit.Moreover,D.suzukii oviposited fewer eggs on infected raspberry fruit relative to uninfected raspberry fruit.Larval survival and adult size after eclosion were significantly reduced when reared on B.cinerea-infected raspberry relative to uninfected fruit.Interestingly,when the behavioral choice experiment was repeated using Botrytis-infected vsuninfected strawberry leaves,a nonfood tissue,in combination with fresh raspberry fruit,odor from B.cinerea-infected leaves did not reduce D.suzukii attraction to raspberries rel-ative to raspberries with uninfected leaves.These behavioral results illustrate the important role context can play in odor-mediated interactions between insects,plants and microbes.We discuss implications of our findings for developing a repellent that can be useful for the management of D.suzukii. | Dong H.Cha Stephen P.Hesler Gabrielle Brind'Amour Karen S.Wentworth Sara Villani Kerik D.Cox Matthew T.Boucher Anna Wallingford Shinyoung K.Park Jan Nyrop Gregory M.Loeb | 2020 | Insect Science2020,27,4: | 0 |
