1Isolation and Identification of Cancer Stem-Like Cells from Murine Melanoma Cell Lines显示文摘In current study,cancer stem-like cells in the murine melanoma B16F10 cells were investigated.CD phenotypes of the B16F10 cells were analyzed by flow cytometry,and the specific CD phenotype cells from the B16F10 cells were isolated by MACS.Then we used colony formation assay in soft agar media,the cell growth assay in serum-free culture media as well as the tumorigenicity investigation of the specific CD phenotype cells in C57BL/6 mice, respectively,to identify cancer stem-like cells in the B16F10 cells.The results showed that the B16F10 cells could form spherical clones in serum-free culture media,and the rate of clonegenesis of CD133 + ,CD44 + and CD44 + CD133 + cells was higher than that of CD133 - ,CD44 - and CD44 + CD133 - cells in soft agar media,respectively. The tumorigenic potential of CD133 + ,CD44 + ,CD44 + CD133 + cells and CD44 + CD133 + CD24 + cells was stronger than that of CD133 - ,CD44 - ,CD44 + CD133 - cells and CD44 + CD133 + CD24 - cells in mice,respectively.In conclusion,the CD44 + CD133 + CD24 + cells have some biological properties of cancer stem-like cells or are highly similar to the characteristics of cancer stem cells(CSC).These results provide an important method for identifying cancer stem-like cells in B16F10 cells and for further cancer target therapy.Jun Dou Meng Pan Ping Wen Yating Li Quan Tang Lili Chu Fengshu Zhao Chuilian Jiang Weihua Hu Kai Hu Ning Gu 2007Cellular & Molecular Immunology2007,4,6:25
2Left posterior fascicular block: a new endpoint of ablation for verapamil-sensitive idiopathic ventricular tachycardia显示文摘Background Verapamil-sensitive, idiopathic left ventricular tachycardia (ILVT) with right bundle branch block configuration and left-axis deviation is known to be due to re-entry mechanism but the exact nature of reentrant circuit in ILVT is not fully elucidated. Radiofrequency (RF) ablation was applied during ventricular tachycardia (VT) and termination of the VT or abolishing the inducibility of the tachycardia was used as an endpoint for successful RF. In this study, the left posterior fascicular block in surface electrocardiogram (ECG) was used as a new endpoint of ablation to cure ILVT. Methods Electrophysiological studies and radiofrequency ablation were performed in 39 consecutive patients [30 men, 9 women; age ranging from 10 to 64 years, mean (29±16) years] with verapamil-sensitive ILVT and structurally normal hearts. VT could be terminated by the intravenous administration of verapamil in all patients. The target site was the midseptum of LV where the earliest Purkinje potentials were recorded during VT. RF current was applied to the target site with or without late diastolic potential (LDP) during sinus rhythm in 37 patients and during VT in 2 patients to meet the ablation endpoint: the left posterior fascicular block in the surface ECG. Results Thirty-seven patients with ILVT had been treated by RF ablation during sinus rhythm and two had been treated during VT. All of them met the endpoint of the left posterior fascicular block. Thirty-eight cases were symptom-free without medications during the follow-up period (range from 3 to 95 months, median 17 months). One patient developed a clinical recurrence and the left posterior fascicular block in surface ECG disappeared. The patient received another treatment. The endpoint was met and the procedure was successful. Conclusions The left posterior fascicular block in surface ECG used as an endpoint of RF ablation to treat ILVT is effective. It is important especially in those patients whose VT can not be induced or the inducible condition is unstable. 4How we can improve patients’comfort after Milligan- Morgan open haemorrhoidectomy显示文摘AIM:To demonstrate the value of Diosmin(flavonidic fraction)in the management of post-haemorhoidectomic symptoms.METHODS:Eighty-six consecutive patients with grades ⅢandⅣacute mixed hemorrhoids admitted to the Anorectal Surgical Department of First Affiliated Hospital,Xinjiang Medical University from April 2009 to April 2010,were enrolled in this study.An observerblinded,randomized trial was conducted to compare post-haemorhoidectomic symptoms with use of Diosmin flavonidic fraction vs placebo.Eighty-six patients were randomly allocated to receive Diosmin flavonidic fraction 500 mg for 1 wk(n=43)or placebo(n=43).The Milligan-Morgan open haemorrhoidectomy was performed by a standardized diathermy excision method.Pain,bleeding,heaviness,pruritus,wound edema and mucosal discharge were observed after surgery.The postoperative symptoms and hospitalization time were recorded.RESULTS:The mean age of the Diosmin group and controls was 53.2 and 51.3 years,respectively.In Diosmin group,haemorrhoid piles were of the third degree in 33 patients and the fourth degree in 10;and in the control group,29 were of the third degree and 14 were of the fourth degree.There was no statistically significance in age,gender distribution,degree and number of excised haemorrhoid piles,and the mean duration of haemorrhoidal disease between the two groups.There was a statistically significant improvement in pain,heaviness,bleeding,pruritus from baseline to the 8th week after operation(P<0.05).Patients taking Diosmin had a shorter hospitalization stay after surgery(P< 0.05).There was also a significant improvement on the proctoscopic appearance(P<0.001).However,there was no statistical difference between the two groups in terms of wound mucosal discharge.Two patients experienced minor bleeding at the 8th week in Diosmin group,and underwent surgery.CONCLUSION:Diosmin is effective in alleviating postoperational symptoms of haemorrhoids.Therefore,it should be considered for the initial treatment after haemorrhoid surgery.However,further prospective randomized trials are needed to confirm the findings of this study.Ma-Mu-Ti-Jiang A ba-bai-ke-re Hong-Guo Huang Wen-Ni Re Kai Fan Hui Chu Er-Ha-Ti Ai Mai-Mai-Ti-Tu-Er-Xun KE Li-Mu Yi-Rui Wang Hao Wen 2011World Journal of Gastroenterology2011,17,11:13
5ζ-Carotene Isomerase Suppresses Tillering in Rice through the Coordinated Biosynthesis of Strigolactone and Abscisic Acid显示文摘Rice tillering is an important agronomic trait affecting grain yield.Here,we identified a high-tillering mutant tillering20(t20),which could be restored to the wild type by treatment with the strigolactone(SL)analog rac-GR24.T20 encodes a chloroplast ζ-carotene isomerase(Z-ISO),which is involved in the biosynthesis of ca-rotenoids and their metabolites,SL and abscisic acid(ABA).The t20 mutant has reduced SL and ABA,raising the question of how SL and ABA biosynthesis is coordinated,and whether they have overlapping functions in tillering.We discovered that rac-GR24 stimulated T20 expression and enhanced all-trans-p-carotene biosynthesis.Importantly,rac-GR24 also stimulated expression of Oryza sativa 9-CIS-EPOXY-CAROTENOID DIOXYGENASE 1(OsNCED1)through induction of Oryza sativa HOMEOBOX12(0sHOX12),promoting ABA biosynthesis in shoot base.On the other hand,ABA treatment significantly repressed SL biosynthesis and the ABA biosynthetic mutants displayed elevated SL biosynthesis.ABA treatment reduced the number of basal tillers in both t20 and wild-type plants.Furthermore,while ABA-deficient mu-tants aba1 and aba2 had the same number of basal tillers as wild type,they had more unproductive upper tillers at maturity.This work demonstrates complex interactions in the biosynthesis of carotenoid,SLs and ABA,and reveals a role for ABA in the regulation of rice tillering.Xue Liu Qingliang Hu Jijun Yan Kai Sun Yan Liang Meiru Jia Xiangbing Meng Shuang Fang Yiqin Wang Yanhui Jing Guifu Liu Dianxing Wu Chengcai Chu Steven M.Smith Jinfang Chu Yonghong Wang Jiayang Li Bing Wang 2020Molecular Plant2020,13,12:11
6Immunogenicity and safety of a severe acute respiratory syndrome coronavirus 2 inactivated vaccine in healthy adults: randomized, double-blind, and placebo-controlled phase 1 and phase 2 clinical trials显示文摘Background:The significant morbidity and mortality resulted from the infection of a severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)call for urgent development of effective and safe vaccines.We report the immunogenicity and safety of an inactivated SARS-CoV-2 vaccine,KCONVAC,in healthy adults.Methods:Phase 1 and phase 2 randomized,double-blind,and placebo-controlled trials of KCONVAC were conducted in healthy Chinese adults aged 18 to 59 years.The participants in the phase 1 trial were randomized to receive two doses,one each on Days 0 and 14,of either KCONVAC(5 or 10 mg/dose)or placebo.The participants in the phase 2 trial were randomized to receive either KCONVAC(at 5 or 10 mg/dose)or placebo on Days 0 and 14(0/14 regimen)or Days 0 and 28(0/28 regimen).In the phase 1 trial,the primary safety endpoint was the proportion of participants experiencing adverse reactions/events within 28 days following the administration of each dose.In the phase 2 trial,the primary immunogenicity endpoints were neutralization antibody seroconversion and titer and anti-receptor-binding domain immunoglobulin G seroconversion at 28 days after the second dose.Results:Inthe phase1 trial,60 participantswere enrolled andreceived at least one dose of 5-mgvaccine(n=24),10-mgvaccine(n=24),or placebo(n=12).In the phase 2 trial,500 participantswere enrolled and received at least one dose of 5-mg vaccine(n=100 for 0/14 or 0/28 regimens),10-mg vaccine(n=100 for each regimen),or placebo(n=50 for each regimen).In the phase 1 trial,13(54%),11(46%),and seven(7/12)participants reported at least one adverse event(AE)after receiving 5-,10-mg vaccine,or placebo,respectively.In the phase 2 trial,16(16%),19(19%),and nine(18%)0/14-regimen participants reported at least oneAEafter receiving 5-,10-mg vaccine,or placebo,respectively.Similar AE incidences were observed in the three 0/28-regimen treatment groups.No AEs with an intensity of grade 3+were reported,expect for one vaccine-unrelated seriousAE(foot fracture)reported in the phase 1 trial.KCONVACinduced significant antibody responses;0/28 regimen showed a higher immune responses than that did 0/14 regimen after receiving two vaccine doses.Conclusions:Both doses of KCONVAC are well tolerated and able to induce robust immune responses in healthy adults.These results support testing 5-mg vaccine in the 0/28 regimen in an upcoming phase 3 efficacy trial.Trial Registration:http://www.chictr.org.cn/index.aspx(No.ChiCTR2000038804,http://www.chictr.org.cn/showproj.aspx?proj=62350;No.ChiCTR2000039462,http://www.chictr.org.cn/showproj.aspx?proj=63353).Hong-Xing Pan Jian-Kai Liu Bao-Ying Huang Gui-Fan Li Xian-Yun Chang Ya-Fei Liu Wen-Ling Wang Kai Chu Jia-Lei Hu Jing-Xin Li Dan-Dan Zhu Jing-Liang Wu Xiao-Yu Xu Li Zhang Meng Wang Wen-Jie Tan Wei-Jin Huang Feng-Cai Zhu 2021Chinese Medical Journal2021,,11:11
7A buried Neolithic paddy soil reveals loss of microbial functional diversity after modern rice cultivation显示文摘It has been documented that human activities are causing the rapid loss of taxonomic, phylogenetic, genetic and functional diversity in soils. However, it remains unclear how modern intensive rice cultivation impacts the soil microbiome and its functionality. Here we examined the microbial composition and function differences between a buried Neolithic paddy soil and an adjacent,currently-cultivated paddy soil using high throughput metagenomics technologies. Our results showed that the currently cultivated soil contained about 10-fold more microbial biomass than the buried one. Analyses based on both 16S rRNA genes and functional gene array showed that the currently cultivated soil had significantly higher phylogenetic diversity, but less functional diversity than the buried Neolithic one. The community structures were significantly different between modern and ancient soils,with functional structure shifting towards accelerated organic carbon(C) degradation and nitrogen(N) transformation in the modern soils. 8Mycorrhizal symbiosis modulates the rhizosphere microbiota to promote rhizobia–legume symbiosis显示文摘Plants establish symbioses with mutualistic fungi,such as arbuscular mycorrhizal(AM)fungi,and bacteria,such as rhizobia,to exchange key nutrients and thrive.Plants and symbionts have coevolved and represent vital components of terrestrial ecosystems.Plants employ an ancestral AM signaling pathway to establish intracellular symbioses,including the legume–rhizobia symbiosis,in their roots.Nevertheless,the relationship between the AM and rhizobial symbioses in native soil is poorly understood.Here,we examined how these distinct symbioses affect root-associated bacterial communities in Medicago truncatula by performing quantitative microbiota profiling(QMP)of 16S rRNA genes.We found that M.truncatula mutants that cannot establish AM or rhizobia symbiosis have an altered microbial load(quantitative abundance)in the rhizosphere and roots,and in particular that AM symbiosis is required to assemble a normal quantitative root-associated microbiota in native soil.Moreover,quantitative microbial co-abundance network analyses revealed that AM symbiosis affects Rhizobiales hubs among plant microbiota and benefits the plant holobiont.Through QMP of rhizobial rpoB and AM fungal SSU rRNA genes,we revealed a new layer of interaction whereby AM symbiosis promotes rhizobia accumulation in the rhizosphere of M.truncatula.We further showed that AM symbiosis-conditioned microbial communities within the M.truncatula rhizosphere could promote nodulation in different legume plants in native soil.Given that the AM and rhizobial symbioses are critical for crop growth,our findings might inform strategies to improve agricultural management.Moreover,our work sheds light on the co-evolution of these intracellular symbioses during plant adaptation to native soil conditions.Xiaolin Wang Huan Feng Yayu Wang Mingxing Wang Xingguang Xie Huizhong Chang Like Wang Jicheng Qu Kai Sun Wei He Chunyan Wang Chuanchao Dai Zhaohui Chu Changfu Tian Nan Yu Xuebin Zhang Huan Liu Ertao Wang 2021Molecular Plant2021,14,3:6

10Equivalent efficacy study of QL1101 and bevacizumab on untreated advanced non-squamous non-small cell lung cancer patients: a phase 3 randomized, double-blind clinical trial显示文摘Objective:This phase 3 study aimed to test equivalence in efficacy and safety for QL1101,a bevacizumab analogue in Chinese patients with untreated locally advanced non-squamous non-small cell lung cancer(NSCLC).Methods:Eligible patients were randomly assigned 1:1 to receive carboplatin and paclitaxel in combination with either QL1101 or bevacizumab,15 mg/kg every 3-week for 6 cycles.This was followed by maintenance treatment with single agent QL1101 every 3-week.The primary end-point was objective response rate(ORR),with secondary end-points being progression-free survival(PFS),overall survival(OS),disease control rate(DCR),and adverse events(AEs).Results:Of 675 patients,535 eligible patients were randomized to the QL1101 group(n=269)and bevacizumab group(n=266).ORRs were 52.8%and 56.8%,respectively,for the QL1101 and bevacizumab groups,with an ORR hazard ratio 0.93(95%confidence interval:0.8-0131.1).The PFS,OS,DCR,and AEs were comparable between the 2 groups,which remained the same after stratification according to epidermal growth factor receptor mutation or smoking history.Conclusions:QL1101 showed similar efficacy and safety profiles as compared to bevacizumab among Chinese patients with untreated locally advanced non-squamous NSCLC.Tianqing Chu Jun Lu Minghong Bi Helong Zhang Wu Zhuang Yan Yu Jianhua Shi Zhendong Chen Xiaochun Zhang Qisen Guo Quan Liu Huijuan Wu Jian Fang Yi Hu Xiuwen Wang Cuicui Han Kai Li Baohui Han 2021Cancer Biology & Medicine2021,18,3:4
11Critical function of DNA methyltransferase 1 in tomato development and regulation of the DNA methylome and transcriptome显示文摘DNA methylation confers epigenetic regulation on gene expression and thereby on various biological processes.Tomato has emerged as an excellent system to study the function of DNA methylation in plant development.To date,regulation and function of DNA methylation maintenance remains unclear in tomato plants.Here,we report the critical function of tomato(Solanum lycopersicum)Methyltransferase 1(Sl MET1)in plant development and DNA methylome and transcriptome regulation.Using CRISPR-Cas9 gene editing,we generated slmet1 mutants and observed severe developmental defects with a frame-shift mutation,including small and curly leaves,defective inflorescence,and parthenocarpy.In leaf tissues,mutations in Sl MET1 caused CG hypomethylation and CHH hypermethylationon a whole-genome scale,leading to a disturbed transcriptome including ectopic expression of many RIN target genes such as ACC2 in leaf tissues,which are normally expressed in fruits.Neither the CG hypomethylation nor CHH hypermethylation in the slmet1 mutants is related to tissue culture.Meanwhile,tissue culture induces non-CG hypomethylation,which occurs more frequently at gene regions than at TE regions.Our results depict Sl MET1-and tissue culture-dependent tomato DNA methylomes,and that Sl MET1 is required for maintaining a normal transcriptome and normal development of tomato.Yu Yang Kai Tang Tatsiana U Datsenka Wenshan Liu Suhui Lv Zhaobo Lang Xingang Wang Jinghui Gao Wei Wang Wenfeng Nie Zhaoqing Chu Heng Zhang Avtar K HANDa Jian-Kang Zhu Huiming Zhang 2019Journal of Integrative Plant Biology2019,61,12:3
12Preparation, microstructure and degradation performance of biomedical magnesium alloy fine wires显示文摘With the development of new biodegradable Mg alloy implant devices, the potential applications of biomedical Mg alloy fine wires are realized and explored gradually. In this study, we prepared three kinds of Mg alloy fine wires containing 4 wt% RE(Gd/Y/Nd) and 0.4 wt% Zn with the diameter less than 0.4 μm through casting, hot extruding and multi-pass cold drawing combined with intermediated annealing process. Their microstructures, mechanical and degradation properties were investigated. In comparison with the corresponding as-extruded alloy, the final fine wire has significantly refined grain with an average size of 3–4 μm, and meanwhile shows higher yield strength but lower ductility at room temperature. The degradation tests results and surface morphologies observations indicate that Mg–4Gd–0.4Zn and Mg–4Nd–0.4Zn fine wires have similar good corrosion resistance and the uniform corrosion behavior in SBF solution. 13Seabed scene simulation and its realization in extending Vega显示文摘Realistic simulation of underwater scene is always difficult because of the special and complex vision effects in underwater space. Seabed is an important part of underwater environment. This paper describes the methods for seabed scene simulation based on OpenGL. It includes construction of fluctuant terrain based on the random sinusoidal algorithm, simulation of seabed flicker effect by means of circular texture mapping and generation of turbidity effect by using fog techniques. For the application based on the leading high level 3D development environment-Vega, underwater scene simulation is still a difficulty since there is no module for it. Based on the analysis of Vega software and the research on seabed scene simulation methods, a Vega extending module named 'Underwater Space' was created through developing module class and extending lynx interface. The module class was designed through developing DLL written in C++. The Lynx was extended through developing keyword configure file, GUI configure file and lynx plug-in DLL. The problem that Vega can't simulate underwater space, is elementarily resolved. 16Pyrolysis of single large biomass particle: Simulation and experiments显示文摘Pyrolysis and heat transfer characteristics of single large biomass particle were investigated using threedimensional unsteady heat transfer model coupled with chemical reactions.The consumption of biomass and the production of products were simulated.Some experiments were designed to provide model parameters for simulation calculations.The simulation was verified by pyrolysis experiments of large biomass particle in a vertical tube furnace.The simulation results show the internal heat and mass transfer law during the pyrolysis of large biomass particle.When the biomass particle diameter is between 10 and 30 mm,for every 5 mm increase in particle diameter,the time required for complete pyrolysis will increase on average by about 50 s.When the pyrolysis temperature is between 673 K and 873 K,a slight decrease in the pyrolysis temperature will cause the time required for the biomass to fully pyrolyze to rise significantly.And the phenomenon is more obvious in the low temperature range.The results indicate that the numerical simulation agrees well with the experimental results.Kai Wang Huiyan Zhang Sheng Chu Zhenting Zha 2021Chinese Journal of Chemical Engineering2021,34,1:2
17Discovery of novel double pyrazole Schiff base derivatives as anti-tobacco mosaic virus(TMV) agents显示文摘Many pyrazole derivatives were reported to exhibit highly activity towards tobacco mosaic virus(TMV).In this work,an optimized pyrazole Schiff base scaffold was designed and introduced to derive novel potential TMV inhibitors.Thirty-six compounds were synthesized,characterized by elemental analysis,mass spectra and nuclear magnetic resonance(NMR) spectroscopy and evaluated by biological experiments.The bioassay results showed that some of the synthesized compounds exhibited excellent anti-TMV activities.Especially,5-chloro-3-methyl-1H-pyrazole contained compound 4j showed ningnanmycin comparable inhibitory activity and can be considered as potential anti-TMV candidate agent.With molecular docking,compound 4j insert into nucleotide sequence(GAAGUU) of OriRNA stably which revealed nucleotide could be a target of these compounds.Xian-Hai Lv Zi-Li Ren Dong-Dong Li Ban-Feng Ruan Qing-Shan Li Ming-Jie Chu Cheng-Ying Ai Dao-Hong Liu Kai Mo Hai-Qun Cao 2017Chinese Chemical Letters2017,28,2:2

20Characteristics of Ce-Mn film on 6061 alloys and its improved performance显示文摘HF2- was applied to accelerate the Ce-Mn film formation on 6061A1 alloy in the Ce3+-MnO4-solution.The process of film formation,the composition and structure of the film were analyzed by scanning electron microscopy(SEM) equipped with energy-dispersive spectroscopy(EDS),X-ray photoelectron spectroscopy(XPS)and X-ray diffractometer(XRD).The film formation process includes three stages.At the initial stage,a threedimensional(3D) skeleton was formed quickly,and then the skeleton was fully filled with cerium oxide and manganese oxide,resulting in a dense structure.Subsequently,a new skeleton was formed and also filled.Al,Ce,O and Mn were detected in the film.Ce existed mainly in the form of Ce4+(89%).The film existed in an amorphous form and was composed of ceria(cerium hydroxide),manganese dioxide and aluminum oxide.After electrostatically spraying fluorocarbon powder,the resultant products satisfied the required mechanical performance and exhibited almost non-filament corrosion compared with commercially available chromium-free conversion film.Its corrosion resistant time to acetate spray can reach 2000 h,which is consistent with that of fluorocarbon paint.The results showed that Ce-Mn film can offer an attractive prospect to eliminate volatile organic compounds(VOC) problem arisen by using fluorocarbon paint in the process of industrial production.Xue-Long Hao Wen Ma Chu Luo Shi-Chao Yang Hong Ji Kai Zhang 2019Rare Metals2019,38,10:1
