| 1 | Test and simulation of full-scale self-centering beam-to-column connection显示文摘A new type of beam-to-column connection for steel moment frames,designated as a'self-centering connection,'is studied.In this connection,bolted top-and-seat angles,and post-tensioned(PT)high-strength steel strands running along the beam are used.The PT strands tie the beam flanges on the column flange to resist moment and provide self-centering force.After an earthquake,the connections have zero deformation,and can be restored to their original status by simply replacing the angles.Four full-scale connections were tested under cyclic loading.The strength,energy-dissipation capacity,hysteresis curve,as well as angles and PT strands behavior of the connections are investigated.A general FEM analysis program called ABAQUS 6.9 is adopted to model the four test specimens.The numerical and test results match very well.Both the test and analysis results suggest that:(1)the columns and beams remain elastic while the angles sustain plastic deformations for energy dissipation when the rotation of the beam related to the column equals 0.05 rad,(2)the energy dissipation capacity is enhanced when the thickness of the angle is increased,and(3)the number of PT strands has a significant influence on the behavior of the connections,whereas the distance between the strands is not as important to the performance of the connection. | Deng Kailai Pan Peng Alexandre Lam Pan Zhenhua Ye Lieping | 2013 | Earthquake Engineering and Engineering Vibration2013,12,4: | 8 |
| 2 | Using knowledge inference to suppress the lamp disturbance for fire detection显示文摘Fire detection in buildings is crucial for people’s lives and property.Conventional temperature and smoke sen-sors have many disadvantages:the limited cover range;detection delays;the difficulty in distinguishing smoke and fire.Recently,research on convolutional neural networks(CNN)for fire image detection has become a hot topic.However,existing fire classification and object detection methods are often interfered with by flash-lights,red objects and the high-brightness background,resulting in a high false alarm rate.Besides,light and lamps often exist in buildings.To address this issue,this paper focuses on introducing scene prior knowledge and causal inference mechanisms to suppress the lamp disturbance.Firstly,we train the YoloV3 network to detect and recognize lamps.Secondly,to reduce the dataset bias,we mask the lamp regions with the pro-posed Local Grabcut segmentation method.Last,compared with direct fire classification methods,our proposed methods reduce about 34.6%false alarm rate based on InceptionV4 networks.The experimental results verify the effectiveness among different CNN architectures(Resnet101,Firenet,Densenet121).The code is online at https://github.com/kailaisun/fire-detection-without-lamp. | Kailai Sun Qianchuan Zhao Xinwei Wang | 2021 | Journal of Safety Science and Resilience2021,2,3: | 3 |
| 3 | S100A8 mediates the activation of P65/HLA-B/S100A8/BCL-2/Caspase-9 (-3) pathway in laryngeal carcinogenesis显示文摘S100 calcium binding protein A8 (S100A8),a possible novel member of NF-kappa B signal pathway in laryngeal squamous cell carcinoma (LSCC),interacts with human leukocyte antigen B (HLA-B) which carries an NF-kappa B binding site within the enhancer A. The objective of this study was to explore the molecular mechanism of S100A8 in laryngeal carcinogenesis. RT-PCR,Western blotting and immuno-histochemistry staining were applied to evaluate the expression levels of IKKα,P65,REL-B,S100A8,APAF-1 and BCL-2 genes. The signal transduction passway in which S100A8 might participate was explored by RNA interference. Flow cytometry,TUNEL assay and cell invasion in vitro were used to detect the biological behavior of Hep2 cells induced by S100A8 gene. Our results showed that high expression of S100A8 was related to tumorigenesis in LSCC and negatively correlated with the degree of differentiation,indicating that S100A8 gene could inhibit apoptosis and promote metastasis in LSCC. Additionally,the suppression of S100A8 by RNA interference down-regulated BCL-2 but not APAF-1,P65 and IKKα,while,the suppression of P65 could significantly down-regulate the expression of S100A8 gene. | 8 | Expression for peroxisome proliferator-activated receptor gamma in pituitary adenomas 38 cases for semi-quantitative immunohistochemical analysis显示文摘BACKGROUND: It has been reported that peroxisome proliferator-activated receptor γ(PPARγ) is highly expressed in lung cancer, colon cancer, and gastric cancer, as well as other tumors. OBJECTIVE: To study expression of PPARγin pituitary adenomas and analyze the role of PPARγin hormonal typing of pituitary adenomas. DESIGN, TIME AND SETTING: Semi-quantitative immunohistochemistry of pathological specimens. The experiment was conducted at the Department of Neurosurgery, Wuxi Second Hospital Affiliated to Nanjing Medical University between January 2002 and May 2005. MATERIALS: Surgical resection samples of pituitary adenomas from 38 cases (18 male and 20 female) were analyzed. Eight cases were determined to be invasive pituitary adenomas and 30 cases were non-invasive pituitary adenomas. Hormonal classification of the types of pituitary adenomas revealed somatotrophic adenomas in six cases, corticotrophic adenoma in five cases, prolactinomas in 13 cases, multi-hormone secreting adenomas in six cases, and eight cases of adenoma without altered endocrine function. Five autopsy specimens were collected during the same period from patients of matching age that died from unrelated diseases and were included as normal anterior pituitary controls. METHODS: Cell counts for positive immunohistochemical signals were recorded from histopathological sections. The percentage of positive cells was reported as a semi-quantitative analysis. MAIN OUTCOME MEASURES: The rate of PPARγpositive cells in different types of adenoma was based on hormonal levels and invasiveness of pituitary tumor cells. RESULTS: All tumor biopsies were determined to express PPARγ The rate of PPARγ-positive cells ranged between 8%-65% in the pituitary adenomas. According to hormonal type, PPARγexpression did not vary between the groups. In addition, there was no significant difference in PPARγexpression between the non-invasive and invasive pituitary adenomas. | 16 | Bottlebrush inspired injectable hydrogel for rapid prevention of postoperative and recurrent adhesion显示文摘Postsurgical adhesion is a common clinic disease induced by surgical trauma,accompanying serious subsequent complications.Current non-surgical approaches of drugs treatment and biomaterial barrier administration only show limited prevention effects and couldn’t effectively promote peritoneum repair.Herein,inspired by bottlebrush,a novel self-fused,antifouling,and injectable hydrogel is fabricated by the free-radical polymerization in aqueous solution between the methacrylate hyaluronic acid(HA-GMA)and N-(2-hydroxypropyl)methacrylamide(HPMA)monomer without any chemical crosslinkers,termed as H-HPMA hydrogel.The H-HPMA hydrogel can be tuned to perform excellent self-fused properties and suitable abdominal metabolism time.Intriguingly,the introduction of the ultra-hydrophilic HPMA chains to the H-HPMA hydrogel affords an unprecedented antifouling capability.The HPMA chains establish a dense hydrated layer that rapidly prevents the postsurgical adhesions and recurrent adhesions after adhesiolysis in vivo.The H-HPMA hydrogel can repair the peritoneal wound of the rat model within 5 days.Furthermore,an underlying mechanism study reveals that the H-HPMA hydrogel significantly regulated the mesothelial-to-mesenchymal transition(MMT)process dominated by the TGF-β-Smad2/3 signal pathway.Thus,we developed a simple,effective,and available approach to rapidly promote peritoneum regeneration and prevent peritoneal adhesion and adhesion recurrence after adhesiolysis,offering novel design ideas for developing biomaterials to prevent peritoneal adhesion. | Jushan Gao Jinpeng Wen Datao Hu Kailai Liu Yuchen Zhang Xinxin Zhao Ke Wang | 2022 | Bioactive Materials2022,7,10: | 0 |

| 18 | miR-208a Promotes Apoptosis in H9c2 Cardiomyocytes by Targeting GATA4显示文摘Background:microRNAs are crucial for cardiovascular development and are associated with congenital heart disease(CHD).Recent studies have shown that microRNAs play a role in heart development and is closely related to CHD.The present study investigated the underlying mechanism of microRNA-208a(miR-208a)in“simple”CHD.Material and Methods:Reverse transcription-quantitative PCR(RT-qPCR)demonstrated miR-208a expression levels in children with CHD(n=27)compared with normal controls(n=29),in cardiomyocytes from embryo 10(E10)to post-birth(P7)and organs in adult rats in healthy rats.Apoptosis of H9c2 cells after transfection with miR-208a detected by TUNEL assay.B-cell lymphoma(Bcl)-2,an anti-apoptotic gene,was detected by RT-qPCR,as well as Gata4.After 48h overexpression of miR-208a,GATA4 was detected via western blotting.Dual luciferase reporting system was used to identify the binding sites of miR-208a to Gata4.Results:Expression of miR-208a was upregulated in the CHD group via the control group(p<0.01).At P7,miR-208a had the highest expression(p<0.01),and which was highest in myocardiocytes via other organs or tissues(p<0.01)in adult rats.The number of apoptotic cells increased significantly post-transfection with miR-208a(p<0.01),while decreased with the miR-208a inhibitor via the control group(p<0.01).Compared with the control group,there was no significant difference in the expression level of Bcl-2 after miR-208a overexpression(p>0.05).The present study proved that miR-208a binds directly to the 3´-UTR of Gata4 at site 1,363-1,369 bp.Expression of GATA4 decreased after miR-208a overexpression(p<0.01),but increased following transfection with a miR-208a inhibitor via the control group(p<0.05).Conclusions:Our study demonstrated that miR-208a downregulates Bcl-2 by directly targeting GATA4,which may cause CHD.miR-208a may become a new biomarker or therapeutic target for CHD in the future. | Liying Gong Hongkun Jiang Guangrong Qiu Kailai Sun | 2021 | Congenital Heart Disease2021,16,5: | 0 |
| 19 | Overexpression of inhibinα(1-32)fusion protein promotes apoptosis and cell cycle arrest in a cervical cancer cell model(Hela cells)显示文摘Inhibins play important roles in the reproductive system. To evaluate whether inhibin α (1-32) fusion protein plays a role in cervical cancer growth, the plasmid pVAX-inhα was constructed and its effect on proliferation and apoptosis of the human cervical cancer cell line (Hela) was checked by flow cytometry and real-time PCR. The expression and localization of inhibin α protein were detected by RT-PCR and confocal mi- croscopy which showed that inhibin α protein was expressed and localized in the nucleus of Hela cells. Over ex- pression of inhibin α gene significantly induced cell apoptosis and ceased S phase of cell cycle. Furthermore, cell proliferation was significantly suppressed 96 h post-transfection and mRNA level of anti-apoptosis genes (Bcl-2, NFκB) were decreased but pro-apoptosis genes (Bax, wild type p53) and inhibin co receptor (TGFβR3) were increased, indicating that inhibin, through its co-receptor, might activate apoptotic and cell growth cascades which regulate proliferation and apoptosis in Hela cells. These results suggest that inhibin α (1-32) fusion pro- tein, located in the cell nucleus, can regulate Hela cells growth and apoptosis by induction of apoptotic pathways such as NFκB, Bcl-2 and p53 families.
