3rtcisE2F promotes the self-renewal and metastasis of liver tumorinitiating cells via N^(6)-methyladenosine-dependent E2F3/E2F6 mRNA stability显示文摘Liver cancer is highly heterogeneous,and the tumor tissue harbors a variety of cell types.Liver tumor initiating cells(TICs)well contribute to tumor heterogeneity and account for tumor initiation and metastasis,but the molecular mechanisms of liver TIC self-renewal are elusive.Here,we identified a functional read-through rt-circRNA,termed rtcisE2F,that is highly expressed in liver cancer and liver TICs.rtcisE2F plays essential roles in the self-renewal and activities of liver TICs.rtcisE2F targets E2F6 and E2F3 mRNAs,attenuates mRNA turnover,and increases E2F6/E2F3 expression.Mechanistically,rtcisE2F functions as a scaffold of N^(6)-methyladenosine(m^(6)A)reader IGF2BP2 and E2F6/E2F3 mRNA.rtcisE2F promotes the association of E2F6/E2F3 mRNAs with IGF2BP2,and inhibits their association with another m^(6)A reader,YTHDF2.IGF2BP2 inhibits E2F6/E2F3 mRNA decay,whereas YTHDF2 promotes E2F6/E2F3 mRNA decay.By switching m^(6)A readers,rtcisE2F enhances E2F6/E2F3 mRNA stability.E2F6 and E2F3 are both required for liver TIC self-renewal and Wnt/β-catenin activation,and inhibition of these pathways is a potential strategy for preventing liver tumorigenesis and metastasis.In conclusion,the rtcisE2F-IGF2BP2/YTHDF2-E2F6/E2F3-Wnt/β-catenin axis drives liver TIC self-renewal and initiates liver tumorigenesis and metastasis,and may provide a strategy to eliminate liver TICs.Zhenzhen Chen Lan Huang Kaili Wang Lulu Zhang Xiang Zhong Zhongyi Yan Benyu Liu Pingping Zhu 2022Science China(Life Sciences)2022,65,9:2
4High temperature performance of coaxial h-BN/CNT wires above 1,000 ℃:Thermionic electron emission and thermally activated conductivity显示文摘The development of wires and cables that can tolerate extremely high temperatures will be very important for probing extreme environments, such as in solar exploration, fire disasters, high-temperature materials processing, aeronautics and astronautics. In this paper, a lightweight high-temperature coaxial h-boron nitride (BN)/carbon nanotube (CNT) wire is synthesized by the chemical vapor deposition (CVD) epitaxial growth of h-BN on CNT yarn. The epitaxially grown h-BN acts as both an insulating material and a jacket that protects against oxidation. It has been shown that the thermionic electron emission (1,200 K) and thermally activated conductivity (1,000 K) are two principal mechanisms for insulation failure of h-BN at high temperatures. The thermionic emission of h-BN can provide the work function of h-BN, which ranges from 4.22 to 4.61 eV in the temperature range of 1,306-1,787 K. The change in the resistivity of h-BN with temperature follows the ohmic conduction model of an insulator, and it can provide the “electron activation energy”(the energy from the Fermi level to the conduction band of h-BN), which ranges from 2.79 to 3.08 eV, corresponding to a band gap for h-BN ranging from 5.6 to 6.2 eV. However, since the leakage current is very small, both phenomena have no obvious influence on the signal transmission at the working temperature. This lightweight coaxial h-BN/CNT wire can tolerate 1,200 ℃ in air and can transmit electrical signals as normal. 7Rapid and sensitive analysis of trace b-blockers by magnetic solidphase extraction coupled with Fourier transform ion cyclotron resonance mass spectrometry显示文摘A rapid and sensitive method for analyzing trace b-blockers in complex biological samples,which involved magnetic solid-phase extraction(MSPE)coupled with Fourier transform ion cyclotron resonance mass spectrometry(FTICR-MS),was developed.Novel nanosilver-functionalized magnetic nanoparticles with an interlayer of poly(3,4-dihydroxyphenylalanine)(polyDOPA@Ag-MNPs)were synthesized and used as MSPE adsorbents to extract trace b-blockers from biological samples.After extraction,the analytes loaded on the polyDOPA@Ag-MNPs were desorbed using an organic solvent and analyzed by FTICR-MS.The method was rapid and sensitive,with a total detection procedure of less than 10 min as well as limits of detection and quantification in the ranges of 3.5-6.8 pg/mL and 11.7-22.8 pg/mL,respectively.The accuracy of the method was also desirable,with recoveries ranging from 80.9%to 91.0%following the detection of analytes in human blood samples.All the experimental results demonstrated that the developed MSPE-FTICR-MS method was suitable for the rapid and sensitive analysis of trace b-blockers in complex biological samples.Xue Xiao Kaili He Ya-Jun Hou Zhangmin Xiang Yunyun Yang 2022Journal of Pharmaceutical Analysis2022,12,2:0
8Bidirectional micro-actuators based on eccentric coaxial composite oxide nanofiber显示文摘It is of great importance to develop new micro-actuators with high performance by optimizing the structures and materials.Here we develop a VO2/AI2O3/CNT eccentric coaxial nanofiber,which can be potentially applied as a micro-actuator.The specific eccentric coaxial structure was efficiently fabricated by conventional thin film deposition methodology with individual CNT templet.Activated by thermal and photothermal stimuli,the as-developed actuator delivers a bidirectional actuation behavior with large amplitudes and an ultra-fast response,〜2.5 mS.A tweezer can be further made by assembling two such nanofibers symmetrically onto a tungsten probe.Clamping and unclamping can be realized by laser stimulus.More experimental and simulation investigations indicated that the actuation behaviors could be attributed to the nanostructured eccentric coaxial geometry,the thermal coefficient mismatch between layers and the fast phase transition of V02.The micro-actuators will have potentials in micro manipulators,nanoscaled switches,remote controls and other autonomous systems.Furthermore,a large variety of coaxial and eccentric coaxial nanofibers with various functions can also be developed,giving the as-developed methodology more opportunities.Guang Wang He Ma Xiang Jin Hua Yuan Yang Wei Qunqing Li Kaili Jiang Shoushan Fan 2020Nano Research2020,13,9:0
9Neurturin gene cloning and expression in Vero cells显示文摘BACKGROUND:Transplantation of cell lines expressing neurturin(NTN) has been used to treat animal models of Parkinson's disease.However,gene homology between humans and rats varies greatly,so experimental results are not entirely suitable for understanding cellular transplantation in humans. OBJECTIVE:To explore expression of NTN in African green monkey kidney cells(Vero cells);to obtain a stably NTN expressing cell line. DESIGN,TIME AND SETTING:An observation study of gene engineering and cellular biology was performed at the Institute of Medical Biology,Chinese Academy of Medical Science and Peking Union Medical College between 2005 and 2008. MATERIALS:Human embryonic hepatic tissues,expression vector pcDNA3,and Vero cells were prepared in this laboratory.Primers were synthesized by TaKaRa Biotechnology,Dalian,China; RNA extraction kit and plasmid extraction kit were purchased from Shanghai Watson Bioengineering,China;G418 and MTT were purchased through Sigma,USA;Lipofectamine2000 was a product of Invitrogen,USA;mice anti-human NTN antibody and fluorescent labeling goat anti-mouse IgG antibody were provided by Jingmei Biotech,China. METHODS:Total RNA was harvested from human embryonic hepatic tissues,and NTN cDNA was cloned by RT-PCR method,followed by subcloning into the pcDNA3 eukaryotic expression vector.The obtained pcDNA3/hNTN was stably transfected into Vero cells using Lipofectamine 2000,and stably expressing clones were selected using G418. MAIN OUTCOME MEASURES:NTN mRNA and protein expressions were respectively identified by RT-PCR and immunofluorescence.The morphology of transfected cells was observed under inverted microscopy,and the growth characteristics of those cells were determined using MTT method. RESULTS:A clonal cell line,stably expressing human NTN mRNA and protein,was obtained through stable transfection of pcDNA3/hNTN into Vero cells.The transfected Vero cells exhibited irregular morphology,rather than a spindle shape.The growth retardation phase was prolonged, but the number of cells was identical to non-transfected cells. 10Notoginsenoside R1 Protects against Diabetic Nephropathy through TXNIP-NLRP3 Signaling Pathway显示文摘Background:Diabetic nephropathy(DN)is a microvascular complication of diabetes mellitus(DM).DN results from many factors,including changes in glomerular hemodynamics,oxidative stress and inflammation,and in-terstitial fibrosis and tubular atrophy.Panax notoginseng,a commonly used Chinese medicine,has been used in the treatment of kidney disease.Notoginsenoside R1(NGR1),the main compound isolated from P.notoginseng,has been reported to have a renoprotective role in DN.However,the therapeutic effect and mechanism of NGR1 in DN remain unclear.Objective:The present study aimed to investigate the therapeutic effect and mechanism of NGR1 in DN.Methods:In this study,mouse podocytes(MPC-5 cells)and db/db mice were used to investigate the effect of NGR1 on DN in vitro and in vivo,respectively.Blood glucose,renal function,inflammatory factors,and PI3K/AKT-Nrf2-NLRP3 signaling pathway proteins were assessed.Results:The study results indicated that NGR1 reversed cell viability induced by high glucose(HG,30 mM).The related mechanism results showed that NGR1 decreased oxidative stress by inhibiting reactive oxygen species(ROS)level and upregulating the expression of Nrf2,NQO1,and HO-1 via TXNIP targeting.In addition,NLRP3 inflammasome and PI3K/AKT were engaged in NGR1-based protection against HG-stimulated podocytes.In db/db mice,NGR1 significantly decreased blood glucose,urine protein,urine output,blood urea nitrogen,and other parameters as well as reversed kidney injury by inhibiting oxidative stress and proinflammatory response.Conclusion:Taken together,this study revealed that NGR1 exerted a significant therapeutic effect on DN both in vitro and in vivo via a mechanism related to the TXNIP-Nrf2 pathway and NLRP3 inflammasome,suggesting that NGR1 is a potential therapeutic option for DN.Chunting Zhang Renyikun Yuan Siyuan Li Guodong Huang Kaili Sun Jiaping Pan Qiuxia Liu Xiang Gao Zhijing Wang Tongyu Li Shilong Lu Jianzhen Lv Liting Huang Hongwei Gao 2023Clinical Complementary Medicine and Pharmacology2023,3,4:0
11HIGH-ACCURACY EXPLICIT TWO-STEP METHODS WITH MINIMAL PHASE-LAG FOR y″=f(t,y)显示文摘In this paper, two families of high accuracy explicit two-step methods with minimal phase-lag are developed for the numerical integration of special second-order periodic initial-value problems. In comparison with some methods in [1-4,6], the advantage of these methods has a higher accuracy and minimal phase-lag. The methods proposed in this paper can be considered as a generalization of some methods in [1,3,4].
