| 1 | Gene expression profile induced by oral administration of baicalin and gardenin after focal brain ischemia in rats显示文摘Aim: To investigate differential gene expression and the pharmacological mechanism of baicalin and gardenin in focal cerebral ischemia in rats with high-density cDNA microarray. Methods: Rat left middle cerebral arteries were occluded and treated with either baicalin or gardenin. The pharmacological effects were investigated using the difference in infarction areas before and after treatment, which were determined by 2,3,5-triphenyltetrazolium chloride (TTC) staining. Gene ex-pression was demonstrated using a “Biostar40S” gene microarray. Semiquantitative reverse transcription-polymerase chain reaction (RT-PCR) was used to verify the result of the selected genes. Results: Both baicalin and gardenin reduced the infarction areas in focal cerebral ischemia rats (P<0.05). The differential genes were 211,177, and 70 (upregulated or downregulated) in the model group, baicalin, and gardenin treatment groups compared with the sham-operated group, respectively. Gene expression of RpL19 and Csnk2 underwent an approximately 1.9 and 2.1-fold increase, respectively, verified by semiquantitative RT-PCR, which was the same trend as the cDNA microarray. Conclusion: Differential gene expression with respect to the pharmacological effects of baicalin and gardenin on focal cerebral ischemia by cDNA microarray revealed a number of clues with respect to the therapeutic mechanisms of Chinese traditional medicine. | 2 | Effects of paclitaxel on proliferation and apoptosis in human acute myeloid leukemia HL-60 cells显示文摘AIM: To investigate the regulatory effect of paclitaxel on proliferation and apoptosis in human acute leukemia HL- 60 cells. METHODS: HL-60 cell growth was measured by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tertrazolium bromide (MTT) colorimetric assay. Cell cycle kinetics and apoptosis were analyzed by flow cytometry and micro- scopic examination. In addition, DNA microarrays containing 14400 EST elements were used to investigate the gene expression pattern of HL-60 cells exposed to paclitaxel 1 μmol/L. RESULTS: Paclitaxel inhibited HL-60 cell growth significantly in a dose-dependent and time-dependent manner (P<0.01). Marked cell accumulation in G2/M phase and multinucleated cells were also observed after treatment with paclitaxel 0.1 and 1 μmol/L. Among 14400 EST elements, 277 genes were found to be markedly up- or down-expressed in the HL-60 cells treated with paclitaxel 1 μmol/L for 0.5 h, comprising 210 known genes and 67 unknown genes. CONCLUSION: Paclitaxel suppresses the growth of HL-60 cells in vitro by causing cell-cycle arrest and apoptosis. | 3 | Effect of substitution of Si by Al on the microstructure and mechanical properties of bainitic transformation-induced plasticity steels显示文摘The effect of partial or full substitution of Si by Al on the microstructure and mechanical properties has been extensively studied in multi-phase transformation-induced plasticity(TRIP) steels with polygonal ferrite matrix, but rarely studied in bainitic TRIP steels. The aim of the present study is to properly investigate the effect of Al and Si on bainite transformation, microstructure and mechanical properties in bainitic steels in order to provide guidelines for the alloying design as a function of process parameters for the 3 rd generation advanced high strength steels(AHSS). It is shown from the dilatometry study,microstructural investigations and tensile properties measurements that the Al addition results in an acceleration whereas Si addition leads to a retardation in bainite transformation kinetics. The addition of Al retards the decomposition of austenite into pearlite and carbides at holding temperatures higher than450℃ whereas Si retards the decomposition of austenite into carbides at temperatures lower than 450℃.Consequently, the Al-added bainitic steel has a better strength-elongation combination at bainitic holding temperatures higher than 450℃ while Si-added steel has a better strength-elongation combination at temperatures lower than 450℃. The higher yield strength of Al-added steel is mainly attributed to its finer bainitic lath. | 4 | Controllable synthesis of well-ordered TiO_2 nanotubes in a mixed organic electrolyte for high-efficiency photocatalysis显示文摘Well-ordered TiO 2 nanotube arrays (TNAs) were fabricated by electrochemical anodization in a mixed organic electrolyte consisting of ethylene glycol and glycerol. The morphology, structure, crystalline phase, and photocatalytic properties of TNAs were characterized by using TEM, SEM, XRD and photodegradation of methylene blue. It was found that the morphology and structure of TNAs could be significantly influenced by the anodization time and applied voltage. The obtained tube length was found to be proportional to anodization time, and the calculated growth rate of nanotubes was 0.6 m/h. The microstructure analysis demonstrated that the diameter and thickness of the nanotubes increased with the increase of anodization voltage. The growth mechanism of TNAs was also proposed according to the observed relationship between current density and time during anodization. | 5 | Flocculation behaviors of a novel papermaking sludge-based flocculant in practical printing and dyeing wastewater treatment显示文摘In our previous studies,several papermaking sludge-based flocculants(PSBFs)were synthesized from wood pulp papermaking sludge.The structure-activity relationships of the PSBFs have been investigated in simulated dye wastewater treatment,but their efficiencies in practical printing and dyeing wastewater(PPDW)treatment are unknown.Herein,an PSBF was prepared,and its performance is discussed in comparison to polyaluminium chloride(PAC)and polyacrylamide(PAM)in PPDW treatment.The PSBF was used in three ways:as an independent flocculant,as a PAC aid,or used to treat the effluent of the PAC system.The results indicated that adding PSBF alone produced similar color and chemical oxygen demand(COD_(Cr))removals as the PAC system alone,but PSBF performed better than PAC when the pH of PPDW was higher than 7.0.Adding PSBF as a PAC aid improved the color,COD_(Cr)and turbidity removals,but the elimination efficiencies were slightly lower than those of the PAC+PAM system.However,when PSBF was used as a flocculant to treat the effluent of the PAC system(PAC→PSBF),the effluent qualities were enhanced.Compared with the PAC system,the color and COD_(Cr)removals of PAC→PSBF system increased by 16.21%and 13.26%,respectively.The excitation and emission matrix fluorescence results indicated that PSBF removed tryptophan-like pollutants more efficiently than PAC.Considering the pH requirements of the subsequent bioreactor treatment in practice,the PAC→PSBF system were also investigated at the PPDW pH level of 7.0.Its maximum removal efficiencies of color,COD_(Cr)and turbidity were 90.17%,32.60%and 82.50%,respectively. | Kangying Guo Baoyu Gao Jie Wang Jingwen Pan Qinyan Yue Xing Xu | 2021 | Frontiers of Environmental Science & Engineering2021,15,5: | 3 |
| 6 | cDNA Array Analysis of the Differential Expression Change in Virulence-related Genes During the Development of Resistance in Candida albicans显示文摘Candida albicans is the most frequently isolated fungus in immunocompromised patients associated with mucosal and deep-tissue infections, To investigate the correlation between virulence and resistance on a gene expression profile in C. albicans, we examined the changes in virulence-related genes during the development of resistance in C, albicans from bone marrow transplant patients using a constructed cDNA array representing 3096 unigenes. In addition to the genes known to be associated with azole resistance,16 virulence-related genes were identified, whose differential expressions were newly found to be associated with the resistant phenotype. Differential expressions for these genes were confirmed by RT-PCR independently. Furthermore, the up-regulation of EFG1, CPH2, TEC1, CDC24, SAP10, ALS9, SNF1, SP072 and BDF1, and the down-regulation of RAD32, IPF3636 and UB14 resulted in stronger virulence and invasiveness in the resistant isolates compared with susceptible ones.
