1Selection and development of representative simple sequence repeat primers and multiplex SSR sets for high throughput automated genotyping in maize显示文摘In the current study, 1900 maize simple sequence repeat (SSR) primers published in MaizeGDB were screened utilizing reference literature, 15 representative Chinese maize inbred lines and 15 Chinese maize hybrids from national regional testing. In total, 500 highly polymorphic primers were identified and used to construct a genetic map. 100 evenly distributed primers, 10 primers per chromosome, were further selected as a set of universal SSR core primers, recommended as preferred primers for general studies. These core primers were then redesigned and used to construct a high throughput multiplex PCR system based on a five-color fluorescence capillary detection system. 2First identification of kdr allele F1534S in VGSC gene and its association with resistance to pyrethroid insecticides in Aedes albopictus populations from Haikou City,Hainan Island,China显示文摘Background:Aedes albopictus is distributed widely in China,as a primary vector of Dengue fever and Chikungunya fever in south of China.Chemical insecticide control is one of the integrated programmes to prevent mosquito-borne diseases.Long-term applications of pyrethroids have resulted in the development of resistance in Ae.albopictus populations in China.However,the susceptibility of Ae.albopictus to pyrethroids in Hainan Island was unclear.Knockdown resistance(kdr),caused by point mutations in the VGSC gene,is one of the mechanisms that confer resistance to DDT and pyrethroids.This study was to investigate the resistance level of Ae.albopictus populations in Haikou City to three pyrethroid insecticides,and elucidate the relationship between the resistant phenotype and kdr mutations.Methods:The Aedes albopictus samples were collected in Xinbu Island(XI),Longtang Town(LT),Shishan Town(ST),Baishamen Park(BP),and Flower Market(FM)from Haikou City,Hainan Island,China.The larval susceptibility to deltamethrin,permethrin and beta-cypermethrin was tested by larval bioassays,and adult susceptibility to deltamethrin and DDT was determined by adult bioassays.The degree of resistance was determined by resistance ratio value(RR50>3)for larvae and by mortality for adult.The kdr alleles at codon 1534 of the VGSC gene were genotyped.The relationship between kdr genotypes and resistant phenotypes was analyzed by Chi-square test.Results:Out of five populations,assessed by larval bioassays,XI was susceptible to deltamethrin and permethrin;LT was susceptible to permethrin and beta-cypermethrin;and ST was susceptible to permithrin.FM and BP both were resistant to all of the three pyrethroids,and FM showed the highest degree of resistance,with RR50 values from 65.17 to 436.36.A total of 493 individuals from the larval bioassays were genotyped for kdr alleles.Five alleles were detected,including two wildtype alleles,TTC(F)(67.04%)and TTT(F)(0.41%),and three mutant alleles,TGC(C)(0.30%),TCC(S)(31.54%)and TTG(L)(0.71%).There was a clear correlation between mutant alleles(or F1534S)and resistant phenotypes(P<0.01).Conclusion:Two novel kdr mutant alleles F1534S and F1534L were detected in the pyrethroid resistant populations of Ae.albopictus in Haikou Hainan,China.For the first time,the mutant F1534S was associated with pyrethroid resistance in Ae.albopictus.Huiying Chen Kaili Li Xiaohua Wang Xinyan Yang Yi Lin Fang Cai Wenbin Zhong Chunyan Lin Zhongling Lin Yajun Ma 2016Infectious Diseases of Poverty2016,5,1:19
3Acupuncture at the Taixi(KI3) acupoint activates cerebral neurons in elderly patients with mild cognitive impairment显示文摘Our previous findings have demonstrated that acupuncture at the Taixi(KI3) acupoint in healthy youths can activate neurons in cognitive-related cerebral cortex. Here, we investigated whether acupuncture at this acupoint in elderly patients with mild cognitive impairment can also activate neurons in these regions. Resting state and task-related functional magnetic resonance imaging showed that the pinprick senstation of acupuncture at the Taixi acupoint differed significantly between elderly patients with mild cognitive impairment and healthy elderly controls. Results showed that 20 brain regions were activated in both groups of participants, including the bilateral anterior cingulate gyrus(Brodmann areas [BA] 32, 24), left medial frontal cortex(BA 9, 10, 11), left cuneus(BA 19), left middle frontal gyrus(BA 11), left lingual gyrus(BA 18), right medial frontal gyrus(BA 11), bilateral inferior frontal gyrus(BA 47), left superior frontal gyrus(BA11), right cuneus(BA 19, 18), right superior temporal gyrus(BA 38), left subcallosal gyrus(BA 47), bilateral precuneus(BA 19), right medial frontal gyrus(BA 10), right superior frontal(BA 11), left cingulate gyrus(BA 32), left precentral gyrus(BA 6), and right fusiform gyrus(BA 19). 5Cloning of a gene-edited macaque monkey by somatic cell nuclear transfer显示文摘Cloning of macaque monkeys by somatic cell nucleus transfer(SCNT) allows the generation of monkeys with uniform genetic backgrounds that are useful for the development of non-human primate models of human diseases. Here, we report the feasibility of this approach by SCNT of fibroblasts from a macaque monkey(Macaca fascicularis), in which a core circadian transcription factor BMAL1 was knocked out by clustered regularly interspaced short palindromic repeat/Cas9 gene editing(see accompanying paper).Out of 325 SCNT embryos transferred into 65 surrogate monkeys, we cloned five macaque monkeys with BMAL1 mutations in both alleles without mosaicism, with nuclear genes identical to that of the fibroblast donor monkey. Further peripheral blood m RNA analysis confirmed the complete absence of the wild-type BMAL1 transcript. 8Effect of scraping therapy on weightlifting ability显示文摘OBJECTIVE:To verify the effects of scraping therapy on the weightlifting ability by measuring the subjective sensation,and changes of biomarkers.METHODS:Five students,who have been trained for 3 years in a sport school in China were participated in this study.A course of scraping therapy was applied to intervene during the normal 7-week of weightlifting training programme.The ability of weightlifting,the scale of rating perceived exertion and serum biochemical markers were measured before and after the intervention.RESULTS:Scraping therapy caused a significant increase in weightlifting ability(P<0.05).The level of rating perceived exertion remained stable with the increase in the training volume.Immuno-globulinA was significantly increased(P<0.05),and creatine kinase and blood urea nitrogen were significantly decreased(P<0.05).No significant changes were observed in white blood cell,neutrophil,and testosterone.CONCLUSION:Scraping therapy may facilitate weightlifting ability mainly by decreasing weight sensation and improving serum biochemical parameters.Xingze Wang Wichai Eungpinichpong Jinsheng Yang Uraiwan Chatchawan Saowanee Nakmareong Yingying Wang Xinyan Gao 2014Journal of Traditional Chinese Medicine2014,34,1:11

11NEAT1/miR-140-3p/MAPK1 mediates the viability and survival of coronary endothelial cells and affects coronary atherosclerotic heart disease显示文摘Studies have shown that long non-coding RNAs(lncRNA)play critical roles in coronary atherosclerotic heart disease(CAD).However,the function of lncRNA nuclear enriched abundant transcript 1(NEAT1)in CAD is unclear.In this study,we aimed to investigate the functions of lncRNA NEAT1 in CAD.RTPCR and western blot analysis were carried out to examine the expressions of related RNAs.Colony formation assay,cell proliferation assay,apoptosis assay,and dual-luc讦erase reporter assay were con ducted to investigate the abilities of colony migration,cell proliferatio n,apoptosis,and targeting.The results showed that NEAT1 was up-regulated in CAD blood samples and in human coronary endothelial cells(HCAECs).Transfection of pcNEATI sign讦icantly inhibited the survival rate of HCAECs and induced apoptosis of HCAECs.MiR-140-3p was down-regulated in HCAECs.NEAT1 directly targeted miR-140-3p,and the expression of miR-140-3p was inversely correlated with the expression of NEAT1 in CAD patients.In addition,co-transfection of NEAT1 with miR-140-3p mimic reversed the effect of pcNEATI on cell viability and apoptosis,mitogenactivated protein kinase 1(MAPK1)was proved to be a target gene of miR-140-3p,and the miR-140-3p mimic was shown to reduce the expression of MAPK1 in HCAECs.pcNEATI sigrdficantly increased the expression level of MAPK1,while shNEATI significantly reduced the expression level of MAPK1.Our results revealed that lncRNA NEAT1 increased cell viability and inhibited CAD cell apoptosis possibly by activating the miR-140-3p/MAPK1 pathway,and lncRNA NEAT1 might serve as a potential therapeutic target for CAD.Hui Zhang Ningning Ji Xinyan Gong Shimao Ni Yu Wang 2020Acta Biochimica et Biophysica Sinica2020,52,9:9
12Preparation and characterization of a composite membrane based on the asphaltene component of coal显示文摘Asphaltene-ceramic composite membranes were fabricated from ceramic supports and an asphaltene component, which was obtained from the separation of coal to give a kind of new carbonaceous precursor material. Using SEM and thermogravimetric analysis to measure the microstructure and properties of the asphaltene component allowed the porosity, permeability, and retention ratios to be determined. The results show that the asphaltene component can be regarded as a good carbon membrane precursor material because of its high carbon content and strong bonding capacity. When ceramic supports are impregnated with asphaltene colloid the asphaltene easily combines with the support surface and forms a good carbonaceous film after carbonization. Little of the asphaltene component permeates into the internal pores of the ceramic support. Although the number of coats applied to the substrate had little affect on the porosity of the asphaltene-ceramic composite membranes the permeability varied depending upon the number of times the substrate was treated. The way bubbles escape from the film, and the phenomenon of coalescence, as affected by different film thicknesses also seem closely related to the number of coats. A composite membrane carbonized at a final temperature of 600 °C is relatively dense and the permeability of Fe(OH) 3 colloid through it is very low. 14Referred Somatic Hyperalgesia Mediates Cardiac Regulation by the Activation of Sympathetic Nerves in a Rat Model of Myocardial Ischemia显示文摘Myocardial ischemia(MI)causes somatic referred pain and sympathetic hyperactivity,and the role of sensory inputs from referred areas in cardiac function and sympathetic hyperactivity remain unclear.Here,in a rat model,we showed that MI not only led to referred mechanical hypersensitivity on the forelimbs and upper back,but also elicited sympathetic sprouting in the skin of the referred area and C8–T6 dorsal root ganglia,and increased cardiac sympathetic tone,indicating sympathetic-sensory coupling.Moreover,intensifying referred hyperalgesic inputs with noxious mechanical,thermal,and electro-stimulation(ES)of the forearm augmented sympathetic hyperactivity and regulated cardiac function,whereas deafferentation of the left brachial plexus diminished sympathoexcitation.Intradermal injection of the α_(2) adrenoceptor(α_(2)AR)antagonist yohimbine and agonist dexmedetomidine in the forearm attenuated the cardiac adjustment by ES.Overall,these findings suggest that sensory inputs from the referred pain area contribute to cardiac functional adjustment via peripheral α_(2)AR-mediated sympathetic-sensory coupling.Xiang Cui Guang Sun Honglei Cao Qun Liu Kun Liu Shuya Wang Bing Zhu Xinyan Gao 2022Neuroscience Bulletin2022,38,4:4

16Linking microbial community composition to farming pattern in selenium-enriched region:Potential role of microorganisms on Se geochemistry显示文摘Selenium(Se)is an essential micronutrient for lives.Indigenous microbial communities play an important role on Se geochemistry in soils.In this study,the microbial community composition and functions of 53 soil samples were investigated using high-throughput sequencing.Samples were divided into 3 groups with different farming types based on the measured geochemical parameters and microbial functional structures.Results indicated that putative Se related bacteria Bacillus,Dyella,Paenibacillus,Burkholderia and Brevibacillus were dominant in dryland plantation soils which were characterized with higher available Se and low contents of H_(2)O,total organic carbon(TOC),NH_(4)^(+) and NO_(2)^(-).In contrast,the putative denitrifier Pseudomonas dominated in flooded paddy soils with higher TOC,NO_(3)^(-) and organic Se,whereas genera Rhizobium,Nitrosospira,and Geobacter preferred woodland soils with higher oxidation-reduction potential(ORP),pH,NH_(4)^(+) and Fe.Farming patterns resulted in distinct geochemical parameters including moisture,pH,ORP,TOC,and contents of soluble Fe,NO_(2)^(-) and NH_(4)^(+),shaping the microbial communities,which in turn affected Se forms in soils.This study provides a valuable insight into understanding of Se biogeochemistry in soils and prospective strategy for Se-rich agriculture production.Yanhong Wang Xinyan Shi Xianxin Huang Chunlei Huang Helin Wang Hanqin Yin Yixian Shao Ping Li 2022Journal of Environmental Sciences2022,34,2:3

18Long-term variation of storm surge-associated waves in the Bohai Sea显示文摘When investigating the long-term variation of wave characteristics as associated with storm surges in the Bohai Sea,the Simulating Waves Nearshore(SWAN)model and Advanced CIRCulation(ADCIRC)model were coupled to simulate 32 storm surges between 1985 and 2014.This simulation was validated by reproducing three actual wave processes,showing that the simulated significant wave height(SWH)and mean wave period agreed well with the actual measurements.In addition,the long-term variations in SWH,pattems in SWH extremes along the Bohai Sea coast,the 100-year retum period SWH extreme distribution,and waves conditional probability distribution were calculated and analyzed.We find that the trend of SWH extremes in most of the coastal stations was negative,among which the largest trend was-0.03 m/a in the western part of Liaodong Bay.From the 100-year return period of the SWH distribution calculated in the Gumbel method,we find that the SWH extremes associated with storm surges decreased gradually from the center of the Bohai Sea to the coast.In addition,the joint probability of wave and surge for the entire Bohai Sea in 100-year return period was determined by the Gumbel logistic method.We therefore,assuming a minimum surge of one meter across the entire Bohai Sea,obtained the spatial SWH distribution.The conclusions of this study are significant for offshore and coastal engineering design.WANG Yanping LIU Yongling MAO Xinyan CHI Yutao JIANG Wensheng 2019Journal of Oceanology and Limnology2019,37,6:2
