2CONSTRUCTION OF HU-PBL/SCID CHIMERAS AND DEVELOPMENTOF EBV-RELATED LYMPHOMAS显示文摘Objective To construct hu-PBL/SCID chimeras and to investigate the development of lymphoma and oncogenicity of the Epstein-Barr virus (EBV). Methods Human peripheral blood lymphocytes (PBLs) were isolated from healthy adult donors and transplanted intraperitoneally into severe combined immunodeficient(SCID) mice. Mice with hu-PBL engraftment from healthy EBV seronegative donors were injected intraperitoneally with EBV-containing supernatant from suspension culture of B95-8 cell line (active infection), whereas mice receiving lymphocytes from healthy EBV seropositive donors were not re-infected with B95-8 derived EBV (latent infection). Pathological examination and molecular analysis were performed on experime-ntal animals and induced neoplasms. Results In the early stage of this experiment, 12 mice died of acute graft-versus-host disease, mortality was 34.3% (12/35 mice) with an average life span of 17.5 days. In 19 survival hu-PBL/SCID chimeric recipients from 12 healthy donors, tumor incidence was 84.2% (16/19 mice). The average survival time of tumor-bearing mice was 65.5 days. EBV-related neoplasms in SCID mice were nodular tumors with aggressive and fatal features. Histological morphology of tumors exhibited diffuse large cell lymphomas. Immunohistochemistry revealed that LCA (CD45) and L26 (CD20) were positive, but both PS1 (CD3) and UCHL-1 (CD45RO) were negative, and EBV products ZEBRA, LMP1, and EBNA2 were expressed in a small number of tumor cells. EB virus particles were seen in the nuclei of some tumor cells by electron microscopy, and EBV DNA could be amplified in the tumor tissues by PCR. In situ hybridization indicated that the nuclei of tumor cells contained human-specific Alu sequence. Conclusions EBV-induced tumors were human B-cell malignant lymphomas. 3Smoke suppressant in flame retarded thermoplastic polyurethane composites: Synergistic effect and mechanism study显示文摘Considerable smoke and toxic volatiles generation has compromised the application of thermoplastic polyurethane (TPU) and caused a great threat to human life. Here, nano-MgFe layered double hydroxide (MgFe-LDH) with uniform particle size was synthesized to reduce smoke density and toxic gases of TPU composites using ammonium polyphosphate (APP) as a flame retardant agent. The results show that the combination of 16 wt.% APP and 4 wt.% MgFe-LDH greatly decreased the smoke density (D20min and Ds, max), smoke production rate (SPR) and heat release rate (HRR) of TPU composites. Furthermore, the MgFe-LDH synergist demonstrated high efficiency in decreasing total volatiled products and toxic volatiles evolved, such as the CO, HCN and isocyanates. The reason was mainly attributed to the chemical reaction between MgFe-LDH and APP, which can promote the compactness of char layers with fine microstructure formed in the decomposition process of MgFe-LDH/APP/TPU composites. 10Critical Role of Cys168 in Noggin Protein's Biological Function显示文摘Previous studies have indicated that noggin exerts its neural inducing effect by binding and antagonizing bone morphogenetic protein 4 (BMP4). In order to further clarify the relationship between the structure and the function of noggin, and elucidate the possible mechanism responsible for noggin-BMP4 interaction, we generated three noggin mutants, C168S, C174S and C197S, by using a site-directed mutagenesis method. Ectopic expression of wild-type (WT) noggin, C174S or C197S, in Xenopus animal caps (ACs) by mRNA injection converted the explants (prospective ectoderm) into neural tissue, as indicated by the neural-like morphology and expression of the neural cell adhesion molecule (NCAM) in the ACs. In contrast, ACs expressing C168S suffered an epidermal fate similar to the control caps. Similarly, among the three mutants, only C168S lost the dorsalizing function. These studies highlight the critical role played by Cys168 in noggin's biological activities. 12Hyperosmolarity promotes macrophage pyroptosis by driving the glycolytic reprogramming of corneal epithelial cells in dry eye disease显示文摘Tear film hyperosmolarity plays a core role in the development of dry eye disease(DED)by mediating the disruption of ocular surface homeostasis and triggering inflammation in ocular surface epithelium.In this study,the mechanisms involving the hyperosmolar microenvironment,glycolysis mediating metabolic reprogramming,and pyroptosis were explored clinically,in vitro,and in vivo.Data from DED clinical samples indicated that the expression of glycolysis and pyroptosis-related genes,including PKM2 and GSDMD,was significantly upregulated and that the secretion of IL-1βsignificantly increased.In vitro,the indirect coculture of macrophages derived from THP-1 and human corneal epithelial cells(HCECs)was used to discuss the interaction among cells.The hyperosmolar environment was found to greatly induce HCECs’metabolic reprogramming,which may be the primary cause of the subsequent inflammation in macrophages upon the activation of the related gene and protein expression.2-Deoxy-d-glucose(2-DG)could inhibit the glycolysis of HCECs and subsequently suppress the pyroptosis of macrophages.In vivo,2-DG showed potential efficacy in relieving DED activity and could significantly reduce the overexpression of genes and proteins related to glycolysis and pyroptosis.In summary,our findings suggested that hyperosmolar-induced glycolytic reprogramming played an active role in promoting DED inflammation by mediating pyroptosis.Yu Han Yu Zhang Kelan Yuan Yaying Wu Xiuming Jin Xiaodan Huang 2023Frontiers of Medicine2023,17,4:0
13Magnesium Demethylcantharidate induces apoptosis in hepatocellular carcinoma cells via ER stress显示文摘Cantharidin(CTD)is a bioactive ingredient isolated from Cantharis vesicatoria(blister beetles),which has potential therapeutic value as an anticancer agent.Magnesium Demethylcantharidate(MDC)is a recently developed derivative of Cantharidin(CTD),and previous studies have illustrated its excellent anticancer activity on HCC cells.However,the effect and mechanism of MDC remains unclear and need to be further studied.In particular,whether MDC can cause ER stress in HCC is still unknown.In this study,we demonstrated that endoplasmic reticulum stress(ERS)-related proteins were changed in SMMC-7721 and Bel-7402 cells after being exposed to MDC.Moreover,we found that MDC could significantly inhibit the growth of xenograft tumor in nude mice.In summary,we confirmed that MDC could induce ERS in HCC cells and thus induce apoptosis.XINTING ZHU MENG YE KELAN FANG FANG LIU JING HUI MEICHEN LIU XIAOFEI LI RONG YAN YUN LIU 2022BIOCELL2022,46,12:0
14A study on the dynamic evaluation of urbanization development quality:An example of 9 counties in Lishui City, Zhejiang Province显示文摘As an important indicator to measure the economic development and modern civilization of a country or region, the evaluation of urbanization development quality is a complex systematic project, which is helpful to clarify the strengths and weaknesses and regulate and guide the development strategies. Based on hierarchical analysis, geographically weighted regression analysis and cluster analysis, this paper takes Lishui City, Zhejiang Province as an example and selects 1 primary indicator, 4 secondary indicators and 17 tertiary indicators to construct an urbanization development evaluation index system and make an objective evaluation of the development status. The results show that: the level of urbanization development in Lishui City is relatively backward in the province, but the development trend is good;in terms of spatial correlation, the development of 9 counties and districts is relatively close, among which Qingtian County plays a key role in the development of urbanization in Lishui City;in terms of cluster analysis, the city’s 9 counties and districts can be divided into 'agglomeration enhancement type' and 'coordination In terms of cluster analysis, the city’s 9 counties and districts can be divided into three major types.' Cluster Enhancement Type', 'Coordination Promotion Type' and'Weak Development Type'.
