Nitrogen addition to 1Cr12NiMo steel can effectively hinder the austenite grain growth, refine the martensite lath, and increase the strength and hardness. | 8 | Unraveling a genetic roadmap for improved taste in the domesticated apple显示文摘Although taste is an important aspect of fruit quality, an understanding of its genetic control remains elusive in apple and other fruit crops. In this study, we conducted genomic sequence analysis of 497 Malus accessions and revealed erosion of genetic diversity caused by apple breeding and possible independent domestication events of dessert and cider apples. Signatures of selection for fruit acidity and size, but not for fruit sugar content, were detected during the processes of both domestication and improvement. Furthermore, we found that single mutations in major genes affecting fruit taste, including Ma1, MdTDT, and MdSOT2, dramatically decrease malate, citrate, and sorbitol accumulation, respectively, and correspond to important domestication events. Interestingly, Ma1 was identified to have pleiotropic effects on both organic acid content and sugar:acid ratio, suggesting that it plays a vital role in determining fruit taste. Fruit taste is unlikely to have been negatively affected by linkage drag associated with selection for larger fruit that resulted from the pyramiding of multiple genes with minor effects on fruit size. | 10 | GSH-sensitive polymeric prodrug: Synthesis and loading with photosensitizers as nanoscale chemo-photodynamic anti-cancer nanomedicine显示文摘Precisely delivering combinational therapeutic agents has become a crucial challenge for anti-tumor treatment. In this study, a novel redox-responsive polymeric prodrug(molecular weight,MW: 93.5 k Da) was produced by reversible addition-fragmentation chain transfer(RAFT) polymerization. The amphiphilic block polymer-doxorubicin(DOX) prodrug was employed to deliver a hydrophobic photosensitizer(PS), chlorin e6(Ce6), and the as-prepared nanoscale system [NPs(Ce6)] was investigated as a chemo-photodynamic anti-cancer agent. The glutathione(GSH)-cleavable disulfide bond was inserted into the backbone of the polymer for biodegradation inside tumor cells, and DOX conjugated onto the polymer with a disulfide bond was successfully released intracellularly. NPs(Ce6) released DOX and Ce6 with their original molecular structures and degraded into segments with low MWs of 41.2 k Da in the presence of GSH. NPs(Ce6) showed a chemo-photodynamic therapeutic effect to kill 4 T1 murine breast cancer cells, which was confirmed from a collapsed cell morphology, a lifted level in the intracellular reactive oxygen species, a reduced viability and induced apoptosis. Moreover, ex vivo fluorescence images indicated that NPs(Ce6) retained in the tumor, and exhibited a remarkable in vivo anticancer efficacy. The combinational therapy showed a significantly increased tumor growth inhibition(TGI,58.53%). Test samples in this study were subjected to different solution treatments and the same aging treatment(at 760?C for 1 h).When solution annealing temperature was elevated from 1020?C to 1150?C, the stress-rupture life at750?C/320 MPa was increased from 60 h to 300 h, the stress-rupture elongation was enhanced from12% to 17%, and the elongation of the tensile at 750?C was improved from 11% to 24%. All tensile and stress-rupture samples displayed an intergranular dimple mixed fracture. Intergranular micro-cracks had a great relationship with the morphology of grain boundary carbides. Most carbides retained the morphology of globular shape and continuous thin plate. After tensile and stress-rupture tests, a few carbides were converted into lamellar. The results showed that intergranular micro-cracks were easier to form at continuous thin plate carbides than at globular shape carbides. Lamellar carbides hardly caused the nucleation of micro-cracks. As the decrease of tem perature and stress, the stress rupture life gradually increased. A Larson-Miller Parameter (LMP) method was used for analyzing the stress rupture life under different conditions. The linear fitting formula between stress (σ) and LMP was derived as σ= 3166.455-119.969 ×LMP and the fitting coefficient w as 0.98. After testing, the dislocation configurations of all stress rupture samples w ere investigated by transm ission electron microscopy (TEM). The tem perature and stress had a significant impact on the deform ation m echanism, thereby affected the stress rupture life of K4750 alloy. As the increasing stress at a given tem perature, the deform ation m echanism gradually transform ed from Orowan looping to stacking fault shearing. Based on experim ental results, the threshold stress at 650 ℃, 700 ℃, 750 ℃ and 800 ℃ for the transition of deformation mechanism was estimated to be about 650 MPa, 530 MPa, 430 MPa and 350 MPa, respectively. Below the threshold stress,γ phase effectively hindered dislocation motion by Orowan looping mechanism, K4750 alloy had along stress rupture life. Slightly above the threshold stress, Orowan looping combining stacking fault shearing was the dom inant mechanism ,the stress rupture life decreased. Experimental results indicate that S-nitrides precipitated in the interdendritic region of the ESR structure at temperatures higher than 1100 ℃. S-nitrides could spread throughout the entire interdendritic area after sufficient exposure, and they were more numerous and much finer than primary nitrides.
