| 3 | Autophagic flux response and glioblastoma sensitivity to radiation显示文摘Objective:Glioblastoma is the most common primary brain tumor in adults and one of the most lethal human tumors.It constitutes a unique non-metastasizing human tumor model with high resistance to radiotherapy and chemotherapy.The current study investigates the association between autophagic flux and glioblastoma cell resistance.Methods:The expression kinetics of autophagy-and lysosome-related proteins following exposure of two glioblastoma cell lines(T98 and U87)to clinically relevant radiation doses was examined.Then,the response of cells resistant to radiotherapy and chemotherapy was investigated after silencing of LC3A,LC3B,and TFEB genes in vitro and in vivo.Results:Following irradiation with 4 Gy,the relatively radioresistant T98 cells exhibited enhanced autophagic flux.The more radiosensitive U87 cell line suffered a blockage of autophagic flux.Silencing of LC3A,LC3B,and TFEB genes in vitro,significantly sensitized cells to radiotherapy and temozolomide(U87:P<0.01 and<0.05,respectively;T98:P<0.01 and<0.01,respectively).Silencing of the LC3A gene sensitized mouse xenografts to radiation.Conclusions:Autophagy in cancer cells may be a key factor of radio-resistance and chemo-resistance in glioblastoma cells.Blocking autophagy may improve the efficacy of radiochemotherapy for glioblastoma patients. | Achilleas G.Mitrakas Dimitra Kalamida Alexandra Giatromanolaki Stamatia Pouliliou Avgi Tsolou Rafail Kyranas Michael I.Koukourakis | 2018 | Cancer Biology & Medicine2018,15,3: | 1 |

| 7 | Application of hurdle technology for the shelf life extension of European eel(Anguilla anguilla)fillets显示文摘Fresh fish,and especially fatty species,are highly perishable due to oxidative deterioration of fish flesh and the elevated microbial load on fish surface.The implementation of a variety of“mild hurdles”may significantly decrease the rates of fish chemical degradation and microbiological spoilage,by better retaining the initial quality,compared to more intense preservation techniques.The aim of this work is the comparative study of different,single or combined,treatments at 15℃on the quality and shelf life of chilled eel fillets.Fish fillets were treated using osmotic solutions consisting of glycerol(30%–40%–45%)and 5%NaCl with and without former antioxidant impregnation by using Rosemary Serum.In all cases,water activity decreased to approximately 0.90 after 420 min of osmotic treatment.Untreated and osmotically treated fish fillets were subsequently stored at 4℃and their stability under chill conditions was assessed based on microbial growth and oxidative deterioration.Microbial growth of treated samples was significantly delayed,especially due to the osmotic step(OS)and the derived water activity decrease.Lipid oxidation,a major cause of rejection for fatty fish such as eel,was greatly inhibited in treated fillets,owing to both‘hurdles’,aw lowering(OS)and antioxidant impregnation with rosemary serum(RS/OS),showing the synergistic effect of these successive procedures.Shelf life of treated eel fillets exhibited a more than 10-fold increase,as compared to the untreated samples,based on chemical composition and a 2 to 3-fold shelf life improvement,in terms of microbial growth. | Maria C.Giannakourou Natalia Stavropoulou Theofania Tsironi Vladimiros Lougovois Vassiliki Kyrana Spyros J.Konteles Vassilia J.Sinanoglou | 2023 | Aquaculture and Fisheries2023,8,4: | 1 |

| 16 | Long-term outcomes of pediatric liver transplantation in acute liver failure vs end-stage chronic liver disease:A retrospective observational study显示文摘BACKGROUND Children with acute liver failure(ALF)who meet the criteria are eligible for super-urgent transplantation,whereas children with end-stage chronic liver disease(ESCLD)are usually transplanted electively.Pediatric liver transplantation(PLT)in ALF and ESCLD settings has been well described in the literature,but there are no studies comparing the outcomes in these two groups.AIM To determine if there is a difference in post-operative complications and survival outcomes between ALF and ESCLD in PLT.METHODS This was a retrospective observational study of all primary PLTs performed at a single center between 2000 and 2019.ALF and ESCLD groups were compared for pretransplant recipient,donor and operative parameters,and post-operative outcomes including graft and patient survival.RESULTS Over a 20-year study period,232 primary PLTs were performed at our center;195 were transplanted for ESCLD and 37 were transplanted for ALF.The ALF recipients were significantly older(median 8 years vs 5.4 years;P=0.031)and heavier(31 kg vs 21 kg;P=0.011).Living donor grafts were used more in the ESCLD group(34 vs 0;P=0.006).There was no difference between the two groups concerning vascular complications and rejection,but there were more bile leaks in the ESCLD group.Post-transplant patient survival was significantly higher in the ESCLD group:1-,5-,and 10-year survival rates were 97.9%,93.9%,and 89.4%,respectively,compared to 78.3%,78.3%,and 78.3%in the ALF group(P=0.007).However,there was no difference in 1-,5-,and 10-year graft survival between the ESCLD and ALF groups(90.7%,82.9%,77.3%vs 75.6%,72.4%,and 66.9%;P=0.119).CONCLUSION Patient survival is inferior in ALF compared to ESCLD recipients;the main reason is death in the 1st year post-PLT in ALF group.Once the ALF children overcome the 1st year after transplant,their survival stabilizes,and they have good long-term outcomes. | Amr M Alnagar Abdul R Hakeem Khaled Daradka Eirini Kyrana Marumbo Methga KarthikeyanPalaniswamy Sanjay Rajwal Jamila Mulla Moira O'meara Vivek Upasani Dhakshinamoorthy Vijayanand Raj Prasad Magdy S Attia | 2023 | World Journal of Transplantation2023,13,3: | 1 |
