| 1 | Identification of a novel coronavirus causing severe pneumonia in human:a descriptive study显示文摘Background:Human infections with zoonotic coronaviruses(CoVs),including severe acute respiratory syndrome(SARS)-CoV and Middle East respiratory syndrome(MERS)-CoV,have raised great public health concern globally.Here,we report a novel batorigin CoV causing severe and fatal pneumonia in humans.Methods:We collected clinical data and bronchoalveolar lavage(BAL)specimens from five patients with severe pneumonia from Wuhan Jinyintan Hospital,Hubei province,China.Nucleic acids of the BAL were extracted and subjected to next-generation sequencing.Virus isolation was carried out,and maximum-likelihood phylogenetic trees were constructed.Results:Five patients hospitalized from December 18 to December 29,2019 presented with fever,cough,and dyspnea accompanied by complications of acute respiratory distress syndrome.Chest radiography revealed diffuse opacities and consolidation.One of these patients died.Sequence results revealed the presence of a previously unknownβ-CoV strain in all five patients,with 99.8%to 99.9%nucleotide identities among the isolates.These isolates showed 79.0%nucleotide identity with the sequence of SARS-CoV(GenBank NC_004718)and 51.8%identity with the sequence of MERS-CoV(GenBank NC_019843).The virus is phylogenetically closest to a bat SARS-like CoV(SL-ZC45,GenBank MG772933)with 87.6%to 87.7%nucleotide identity,but is in a separate clade.Moreover,these viruses have a single intact open reading frame gene 8,as a further indicator of bat-origin CoVs.However,the amino acid sequence of the tentative receptor-binding domain resembles that of SARS-CoV,indicating that these viruses might use the same receptor.Conclusion:A novel bat-borne CoV was identified that is associated with severe and fatal respiratory disease in humans. | Li-Li Ren Ye-Ming Wang Zhi-Qiang Wu Zi-Chun Xiang Li Guo Teng Xu Yong-Zhong Jiang Yan Xiong Yong-Jun Li Xing-Wang Li Hui Li Guo-Hui Fan Xiao-Ying Gu Yan Xiao Hong Gao Jiu-Yang Xu Fan Yang Xin-Ming Wang Chao Wu Lan Chen Yi-Wei Liu Bo Liu Jian Yang Xiao-Rui Wang Jie Dong Li Li Chao-Lin Huang Jian-Ping Zhao Yi Hu Zhen-Shun Cheng Un-Lin Liu Zhao-Hui Qian Chuan Qin Qi Jin Bin Cao Jian-Wei Wang | 2020 | Chinese Medical Journal2020,,9: | 99 |
| 2 | Extract of Naotaifang, a compound Chinese herbal medicine, protects neuron ferroptosis induced by acute cerebral ischemia in rats显示文摘Objective:Our previous research showed that Naotaifang(a compound traditional Chinese herbal medicine)extract(NTE)has clinically beneficial effects on neurological improvement of patients with acute cerebral ischemia.In this study,we investigated whether NTE protected acute brain injury in rats and whether its effects on ferroptosis could be linked to the dysfunction of glutathione peroxidase 4(GPX4)and iron metabolism.Methods:We established an acute brain injury model of middle cerebral artery occlusion(MCAO)in rats,in which we could observe the accumulation of iron in neurons,as detected by Perl’s staining.Using assay kits,we measured expression levels of ferroptosis biomarkers,such as iron,glutathione(GSH),reactive oxygen species(ROS)and malonaldehyde(MDA);further the expression levels of transferrin receptor1(TFR1),divalent metal transporter 1(DMT1),solute carrier family 7 member 11(SLC7 A11)and GPX4 were determined using immunohistochemical analysis,real-time quantitative polymerase chain reaction and Western blot assays.Results:We found that treatment with NTE reduced the expression levels of TFR1 and DMT1,reduced ROS,MDA and iron accumulation and reduced neurobehavioral scores,relative to untreated MCAO rats.Treatment with NTE increased the expression levels of SLC7 A11,GPX4 and GSH,and the number of Nissl bodies in the MCAO rats.Conclusion:Taken together,our data suggest that acute cerebral ischemia induces neuronal ferroptosis and the effects of treating MCAO rats with NTE involved inhibition of ferroptosis through the TFR1/DMT1 and SCL7 A11/GPX4 pathways. | Bin Lan Jin-wen Ge Shao-wu Cheng Xi-long Zheng Jun Liao Chao He Zheng-qing Rao Guo-zuo Wang | 2020 | Journal of Integrative Medicine2020,18,4: | 37 |
| 3 | A nomogramfor predicting overall survival in patients with low-grade endometrial stromal sarcoma: A population-based analysis显示文摘Background:Low-grade endometrial stromal sarcoma(LG-ESS)is a rare tumor that lacks a prognostic prediction model.Our study aimed to develop a nomogram to predict overall survival of LG-ESS patients.Methods:A total of 1172 patients confirmed to have LG-ESS between 1988 and 2015 were selected from the Surveillance,Epidemiology and End Results(SEER)database.They were further divided into a training cohort and a validation cohort.The Akaike information criterion was used to select variables for the nomogram.The discrimination and calibration of the nomogram were evaluated using concordance index(C-index),area under time-dependent receiver operating characteristic curve(time-dependent AUC),and calibration plots.The net benefits of the nomogram at different threshold probabilities were quantified and compared with those of the International Federation of Gynecology and Obstetrics(FIGO)criteria-based tumor staging using decision curve analysis(DCA).Net reclassification index(NRI)and integrated discrimination improvement(IDI)were also used to compare the nomogram’s clinical utilitywith that of the FIGO criteria-based tumor staging.The risk stratifications of the nomogram and the FIGO criteria-based tumor staging were compared.Results:Seven variables were selected to establish the nomogram for LG-ESS.The C-index(0.814 for the training cohort and 0.837 for the validation cohort)and the time-dependent AUC(>0.7)indicated satisfactory discriminative ability of the nomogram.The calibration plots showed favorable consistency between the prediction of the nomogram and actual observations in both the training and validation cohorts.The NRI values(training cohort:0.271 for 5-year and 0.433 for 10-year OS prediction;validation cohort:0.310 for 5-year and 0.383 for 10-year OS prediction)and IDI(training cohort:0.146 for 5-year and 0.185 for 10-year OS prediction;validation cohort:0.177 for 5-year and 0.191 for 10-year OS prediction)indicated that the established nomogram performed significantly better than the FIGO criteria-based tumor staging alone(P<0.05).Furthermore,DCA showed that the nomogram was clinically useful and had better discriminative ability to recognize patients at high risk than the FIGO criteria-based tumor staging.Conclusions:A prognostic nomogram was developed and validated to assist clinicians in evaluating prognosis of LG-ESS patients. | Jie Wu Huibo Zhang Lan Li Mengxue Hu Liang Chen Bin Xu Qibin Song | 2020 | Cancer Communications2020,40,7: | 35 |

| 5 | Field released transgenic papaya effect on soil microbial communities and enzyme activities显示文摘Soil properties, microbial communities and enzyme activities were stud ied in soil amended with replicase (RP)-transgenic or non-transgenic papaya unde r field conditions. Compared with non-transgenic papaya, significant differences (P<0.05) were observed in total nitrogen in soils grown with transgenic papaya. There were also significant differences (P<0.05) in the total number of colony forming units (CFUs) of bacteria, actinomycetes and fungi between soils amended with RP-transgenic plants and non-transgenic plants. Compared with non-transgen ic papaya, the total CFUs of bacteria, actinomycetes and fungi in soil with tran sgenic papaya increased by 0.43—1.1, 0.21—0.80 and 0.46—0.73 times respective ly. Significantly higher (P<0.05) CFUs of bacteria, actinomycetes and fungi res istant to kanamycin (Km) were obtained in soils with RP-transgenic papaya than t hose with non-transgenic papaya in all concentrations of Km. Higher resistance q uotients for Kmr (kanamycin resistant) bacteria, actinomycetes and fungi were fo und in soil planted with RP-transgenic papaya, and the resistance quotients for Kmr bacteria, actinomycetes and fungi in soils with transgenic papaya increased 1.6—4.46, 0.63—2.5 and 0.75—2.30 times. RP-transgenic papaya and non-transg enic papaya produced significantly different enzyme activities in arylsulfatase (5.4—5.9x), polyphenol oxidase (0.7—1.4x), invertase (0.5—0.79x), cellulase ( 0.23—0.35x) and phosphodiesterase (0.16—0.2x). The former three soil enzymes appeared to be more sensitive to the transgenic papaya than the others, and cou ld be useful parameters in assessing the effects of transgenic papaya. | 6 | Dihydromyricetin alleviates carbon tetrachloride-induced acute liver injury via JNK-dependent mechanism in mice显示文摘AIM: To assess the effects of dihydromyricetin(DHM) as a hepatoprotective candidate in reducing hepatic injury and accelerating hepatocyte proliferation after carbon tetrachloride(CCl4) treatment.METHODS: C57 BL/6 mice were used in this study. Mice were orally administered with DHM(150 mg/kg) for 4 d after CCl4 treatment. Serum and liver tissue samples were collected on days 1, 2, 3, 5 and 7 after CCl4 treatment. The anti-inflammatory effect of DHM was assessed directly by hepatic histology detection and indirectly by serum levels of aspartate aminotransferase(AST), alanine aminotransferase(ALT), albumin, and superoxide dismutase(SOD). Inflammatory cytokines, such as interleukin(IL)-1β, IL-6 and tumor necrosis factor-α(TNF-α), were detected using ELISA kits. Proliferating cell nuclear antigen(PCNA) staining was used to evaluate the role of DHM in promoting hepatocyte proliferation. Hepatocyte apoptosis wasmeasured by TUNEL assay.Furthermore,apoptosis proteins Caspases-3,6,8,and 9 were detected by Western blot.SP600125 were used to confirm whether DHM regulated liver regeneration through JNK/TNF-αpathways.RESULTS:DHM showed a strong anti-inflammatory effect on CCl4-induced liver injury in mice.DHM could significantly decrease serum ALT,AST,IL-1β,IL-6 and TNF-αand increase serum albumin,SOD and liver SOD compared to the control group after CCl4 treatment(P<0.05).PCNA results indicated that DHM could significantly increase the number of PCNA positive cells compared to the control(348.9±56.0 vs 107.1±31.4,P<0.01).TUNEL assay showed that DHM dramatically reduced the number of apoptotic cells after CCl4 treatment compared to the control(365.4±99.4 vs 90.5±13.8,P<0.01).Caspase activity detection showed that DHM could reduce the activities of Caspases-8,3,6 and 9 compared to the control(P<0.05).The results of Western blot showed that DHM increased the expression of JNK and decreased TNF-αexpression.However,DHM could not affect TNF-αexpression after SP600125 treatment.Furthermore,DHM could significantly improve the survival rate of acute liver failure(ALF)mice(73.3%vs 20.0%,P<0.0001),and SP600125 could inhibit the effect of DHM.CONCLUSION:These findings demonstrate that DHM alleviates CCl4-induced liver injury,suggesting that DHM is a promising candidate for reversing liver injury and ALF. | Jun Xie Jie Liu Tu-Ming Chen Qing Lan Qing-Yu Zhang Bin Liu Dong Dai Wei-Dong Zhang Li-Ping Hu Run-Zhi Zhu | 2015 | World Journal of Gastroenterology2015,21,18: | 17 |

| 9 | Management of granulomatous lobular mastitis: an international multidisciplinary consensus(2021 edition)显示文摘Granulomatous lobular mastitis(GLM) is a rare and chronic benign inflammatory disease of the breast. Difficulties exist in the management of GLM for many front-line surgeons and medical specialists who care for patients with inflammatory disorders of the breast. This consensus is summarized to establish evidence-based recommendations for the management of GLM. Literature was reviewed using PubMed from January 1, 1971 to July 31, 2020. Sixty-six international experienced multidisciplinary experts from 11 countries or regions were invited to review the evidence.Levels of evidence were determined using the American College of Physicians grading system, and recommendations were discussed until consensus. Experts discussed and concluded 30 recommendations on historical definitions,etiology and predisposing factors, diagnosis criteria, treatment, clinical stages, relapse and recurrence of GLM. GLM was recommended as a widely accepted definition. In addition, this consensus introduced a new clinical stages and management algorithm for GLM to provide individual treatment strategies. In conclusion, diagnosis of GLM depends on a combination of history, clinical manifestations, imaging examinations, laboratory examinations and pathology.The approach to treatment of GLM should be applied according to the different clinical stage of GLM. | 12 | Mechanisms and pharmacokinetic/pharmacodynamic profiles underlying the low nephrotoxicity and ototoxicity of etimicin显示文摘Etimicin(ETM),a fourth-generation aminoglycosides(AGs),is now widely clinically used in China due to its high efficacy and low toxicity.However,the mechanisms underlying its low nephrotoxicity and ototoxicity remain unclear.In the present study we compared the antibacterial and toxicity profiles of etimicin,gentamicin(GM,a second-generation AG),and amikacin(AMK,a third-generation AG),and investigated their pharmacokinetic properties in the toxicity target organs(kidney and inner ear)and subcellular compartments.We first demonstrated that ETM exhibited superior antibacterial activities against clinical isolates to GM and AMK,and it exerted minimal nephrotoxicity and ototoxicity in rats following multi-dose administration.Then,we conducted pharmacokinetic studies in rats,showed that the three AGs accumulated in the kidney and inner ear with ETM being distributed to a lesser degree in the two toxicity target organs as compared with GM and AMK high-dose groups.Furthermore,we conducted in vitro experiments in NRK-52E rat renal tubular epithelial cells and HEI-OC1 cochlear hair cells,and revealed that all the three AGs were distributed predominantly in the mitochondria with ETM showing minimal accumulation;they not only directly inhibited the activity of mitochondrial complexesⅣandⅤbut also inhibited mitochondrial function and its related PGC-1α-NRF1-TFAM pathway;ETM caused minimal damage to the mitochondrial complex and mitochondrial biogenesis.Our results demonstrate that the minimal otonephrotoxicity of ETM results from its lesser accumulation in mitochondria of target cells and subsequently lesser inhibition of mitochondrial function.These results provide a new strategy for discovering novel AGs with high efficacy and low toxicity. | Lan Yao Jing-wei Zhang Bin Chen Ming-min Cai Dong Feng Qi-zhi Wang Xin-yu Wang Jian-guo Sun Yi-wen Zheng Guang-ji Wang Fang Zhou | 2020 | Acta Pharmacologica Sinica2020,41,6: | 10 |
| 13 | A large carbon pool in lake sediments over the arid/semiarid region, NW China显示文摘Carbon burial in lake sediments is an important component of the global carbon cycle. However, little is known about the magnitude of carbon sequestered in lake sediments over the arid/semiarid region of China(ASAC).In this study, we estimate both organic and inorganic carbon burial since *AD 1800 based on nine lakes in ASAC,and discuss the most plausible factors controlling carbon burial. Our estimates show that the annual organic carbon burial rate(OCBR) ranges from 5.3 to 129.8 g cm-2year-1(weighted mean of 49.9 g cm-2year-1), leading to a standing stock of 1.1–24.0 kg cm-2(weighted mean of 8.6 kg cm-2)and a regional sum of *108 Tg organic carbon sequestered since *AD 1800. The annual inorganic carbon burial rate(ICBR) ranges from 11.4 to 124.0 g cm-2year-1(weighted mean of 48.3 g cm-2year-1), which is slightly lower than OCBR. The inorganic carbon standing stock ranges from2.4 to 26.0 kg cm-2(weighted mean of 8.1 kg cm-2),resulting in a sum of *101 Tg regional inorganic carbon burial since *AD 1800, which is slightly lower than the organic carbon sequestration. OCBR in ASAC shows a continuously increasing trend since *AD 1950, which is possibly due to the high autochthonous and allochthonous primary production and subsequently high sedimentation rate in the lakes. This increasing carbon burial is possibly related to both climatic changes and enhanced anthropogenic activities, such as land use change, deforestation, and eutrophication in the lake. Based on the observation that two mechanisms such as mushrooming and cavitation are involved in cavity expansion by a long rod penetrator, the model is constructed by using the laws of conservation of mass, momentum, energy, together with the u-v relationship of the newly suggested 1D theory of long rod penetration (see Lan and Wen, Sci China Tech Sci, 2010, 53(5): 1364–1373). | 17 | The molecular characterization of maize B chromosome specific AFLPs显示文摘The origin and evolution of B chromosomes could be explained by the specific DNA sequence on them. But the specific sequences known were quite limited. To investigate maize B chromosome sqicific DNA sequeces, maize genomes with and without B chromosomes were analyzed by AFLP. Only 5 markers were found specific to genomes with B chromosomes among about 2000 AFLP markers. Southern hybridization and sequence analysis revealed that only the sequence of M8-2D was a B chromosome specific sequence. This sequence contained the telomeric repeat unit AGGGTTT conserved in plant chromosome telomeres. In addition, the sequence of M8-2D shared low homology to clones from maize chromosome 4 centromere as well. Researches were also conducted to explore the function of the nano-LDHs in the composite fire-retardant agents in air and nitrogen atmosphere, respectively. The results indicated that the nano-LDHs species were responsible for the catalytic oxidation of the rich-carbon compound in oxygen atmosphere. | 19 | Source apportionment of PM_(2.5)light extinction in an urban atmosphere in China显示文摘Haze in China is primarily caused by high pollution of atmospheric fine particulates(PM2.5).However, the detailed source structures of PM2.5 light extinction have not been well established, especially for the roles of various organic aerosols, which makes haze management lack specified targets. This study obtained the mass concentrations of the chemical compositions and the light extinction coefficients of fine particles in the winter in Dongguan, Guangdong Province, using high time resolution aerosol observation instruments. We combined the positive matrix factor(PMF) analysis model of organic aerosols and the multiple linear regression method to establish a quantitative relationship model between the main chemical components, in particular the different sources of organic aerosols and the extinction coefficients of fine particles with a high goodness of fit(R^2= 0.953). The results show that the contribution rates of ammonium sulphate,ammonium nitrate, biomass burning organic aerosol(BBOA), secondary organic aerosol(SOA) and black carbon(BC) were 48.1%, 20.7%, 15.0%, 10.6%, and 5.6%, respectively. It can be seen that the contribution of the secondary aerosols is much higher than that of the primary aerosols(79.4% versus 20.6%) and are a major factor in the visibility decline. BBOA is found to have a high visibility destroying potential, with a high mass extinction coefficient, and was the largest contributor during some high pollution periods.
