| 1 | Inhibition of SARS-CoV-2 (previously 2019-nCoV) infection by a highly potent pan-coronavirus fusion inhibitor targeting its spike protein that harbors a high capacity to mediate membrane fusion显示文摘The recent outbreak of coronavirus disease(COVID-19)caused by SARS-CoV-2 infection in Wuhan,China has posed a serious threat to global public health.To develop specific anti-coronavirus therapeutics and prophylactics,the molecular mechanism that underlies viral infection must first be defined.Therefore,we herein established a SARS-CoV-2 spike(S)protein-mediated cell-cell fusion assay and found that SARS-CoV-2 showed a superior plasma membrane fusion capacity compared to that of SARS-CoV.We solved the X-ray crystal structure of six-helical bundle(6-HB)core of the HR1 and HR2 domains in the SARS-CoV-2 S protein S2 subunit revealing that several mutated amino acid residues in the HR1 domain may be associated with enhanced interactions with the HR2 domain.We previously developed a pan-coronavirus fusion inhibitor,EK1,which targeted the HR!domain and could inhibit infection by divergent human coronaviruses tested,including SARS-CoV and MERS-CoV.Here we generated a series of lipopeptides derived from EK1 and found that EK1C4 was the most potent fusion inhibitor against SARS-CoV-2 S protein-mediated membrane fusion and pseudovirus infection with IC50s of 1.3 and 15.8 nM,about 241-and 149-fold more potent than the original EK1 peptide,respectively.EK1C4 was also highly effective against membrane fusion and infection of other human coronavirus pseudoviruses tested,including SARS-CoV and MERS-CoV,as well as SARSr-CoVs,and potently inhibited the replication of 5 live human coronaviruses examined,including SARS-CoV-2.Intranasal application of EK1C4 before or after challenge with HCoV-OC43 protected mice from infection,suggesting that EK1C4 could be used for prevention and treatment of infection by the currently circulating SARS-CoV-2 and other emerging SARSr-CoVs. | Shuai Xia Meiqin Liu Chao Wang Wei Xu Qiaoshuai Lan Siliang Feng Feifei Qi Linlin Bao Lanying Du Shuwen Liu Chuan Qin Fei Sun Zhengli Shi Yun Zhu Shibo Jiang Lu Lu | 2020 | Cell Research2020,30,4: | 81 |

| 6 | Improving Plant Genome Editing with High-Fidelity xCas9 and Non-canonical PAM-Targeting Cas9-NG显示文摘Two recently engineered SpCas9 variants, namely xCas9 and Cas9-NG, show promising potential in improving targeting specificity and broadening the targeting range. In this study, we evaluated these Cas9 variants in the model and crop plant, rice. We first tested xCas9-3.7, the most effective xCas9 variant in mammalian cells, for targeted mutagenesis at 16 possible NGN PAM (protospacer adjacent motif) combinations in duplicates. xCas9 exhibited nearly equivalent editing efficiency to wild-type Cas9 (Cas9-WT) at most canonical NGG PAM sites tested, whereas it showed limited activity at non-canonical NGH (H = A, C, T) PAM sites. High editing efficiency of xCas9 at NGG PAMs was further demonstrated with C to T base editing by both rAPOBECI and PmCDAI cytidine deaminases. With mismatched sgRNAs, we found that xCas9 had improved targeting specificity over the Cas9-WT. Furthermore, we tested two Cas9-NG variants, Cas9-NGv1 and Cas9-NG, for targeting NGN PAMs. Both Cas9-NG variants showed higher editing efficiency at most non-canonical NG PAM sites tested, and enabled much more efficient editing than xCas9 at AT-rich PAM sites such as GAT, GAA, and CAA. Nevertheless, we found that Cas9-NG variants showed significant reduced activity at the canonical NGG PAM sites. In stable transgenic rice lines, we demonstrated that Cas9-NG had much higher editing efficiency than Cas9-NGv1 and xCas9 at NG PAM sites. To expand the base-editing scope, we developed an efficient C to T base-editing system by making fusion of Cas9-NG nickase (D10A version), PmCDAI, and UGI. | 7 | Target classification with low-resolution radar based on dispersion situations of eigenvalue spectra显示文摘Most low-resolution radar systems, especially ground surveillance radar systems, work at relatively low pulse repeat frequency (PRF) and with short time-on-target (TOT) (duration in scanning). Low PRF leads to Doppler ambiguity and short TOT results in low Doppler resolution, which poses a problem to target classification with low-resolution radar based on the jet engine modulation (JEM) characteristic of radar echo. From the pattern classification viewpoint, we propose a method of using dispersion situations of JEM eigenvalue spectra to categorize aeroplanes into three kinds, namely turbojet aircraft, prop aircraft and helicopter. We analyze the mathematical model of JEM echoes consisting of a series of line spectra and regard them as a sum of several series of harmonious waves. Classification features can be extracted based on the harmonious wave sum model. Some schemes for extracting features from echoes within or between pulses are proposed. Low-dimensional features are extracted to reduce computation burden. Our methods do not compensate for the fuselage echoes and are insensitive to the variation of fuselage Doppler. | 8 | Influence of up-regulation of Notch ligand DLL4 on biological behaviors of human gastric cancer cells显示文摘AIM: To investigate the potential roles of Delta-like ligand 4 (DLL4) on the biological behavior of gastric cancer cells and its molecular mechanisms. METHODS: A recombinant eukaryotic expression vector containing human DLL4 gene was constructed and transfected into the human gastric cancer cell line SGC7901. Clones with up-regulated DLL4 were selected and amplified. The effect of DLL4 up-regulation on gastric cancer cell growth was assessed using cell growth assay. The migration and invasion were assessed using a transwell migration assay and matrigel invasion assay. Matrix metalloproteinases were detected using the zymogram technique. Cells were implanted subcutaneously into male BALB/c nu/nu mice. Tumor volumes were then calculated and compared. DLL4 staining in the implanted tumor was performed using immunohistochemistry technique. RESULTS: Growth curves over a six-day time course showed significantly promoted cell proliferation of SGC7901 cells with up-regulated DLL4. DLL4 up-regulation in SGC7901 cells promoted the migration (205.4 ± 15.2 vs 22.3 ± 12.1, P < 0.05) and invasion (68.8 ± 5.3 vs 18.2 ± 6.0, P < 0.05) in vitro and tumorigenicity in vivo (2640.5 ± 923.6 mm 3 vs 1115.1 ± 223.8 mm 3 , P < 0.05). Furthermore, significantly increased mRNA level and increased secretion of matrix metalloproteinase-2 (MMP-2) proenzyme were observed in SGC7901 cells with up-regulated DLL4. However, increased MMP-9 mRNA level but decreased extracellular MMP-9 proenzyme level was observed. It is generally acknowledged that Tm is in an approximate linear relationship with surface temperature Ts, and the relationship presents regional variation. This paper employed sliding average method to calculate correlation coefficients and linear regression coefficients between Tm and Ts at every 2°× 2.5° grid point using Ts data from European Centre for Medium-Range Weather Forecasts(ECMWF) and Tm data from 'GGOS Atmosphere', yielding the grid and bilinear interpolation-based Tm Grid model. Tested by Tm and Ts grid data, Constellation Observation System of Meteorology, Ionosphere, and Climate(COSMIC) data and radiosonde data, the Tm Grid model shows a higher accuracy relative to the Bevis Tm-Ts relationship which is widely used nowadays. | 18 | An adjustment method for active reflector of large high-frequency antennas considering gain and boresight显示文摘The design of the Qitai 110 m Radio Telescope(QTT) with large aperture and very high working frequency(115 GHz) was investigated in Xinjiang, China. The results lead to a main reflector with high surface precision and high pointing precision. In this paper, the properties of active surface adjustment in a deformed parabolic reflector antenna are analyzed. To assure the performance of large reflector antennas such as gain and boresight, which can be obtained by utilizing an electromechanical coupling model, and satisfy them simultaneously, research on active surface adjustment applied to a new parabolic reflector as target surface has been done. Based on the initial position of actuators and the relationship between adjustment points and target points, a novel mathematical model and a program thatdirectly calculates the movements of actuators have been developed for guiding the active surface adjustment of large reflector antennas. This adjustment method is applied to an 8 m reflector antenna,in which we only consider gravity deformation. | 20 | Simulation of groundwater table dynamics based on Feflow in the Minqin Basin,China显示文摘As groundwater table declination is an important factor resulting in degradation of eco-environment in the Minqin Basin,China,it is significant to investigate and understand the groundwater table dynamics in this area. According to the physical and geographical conditions of the Minqin Basin,a hydrogeological conceptual model and a mathematical model were established,and the mathematical model was figured out by using Finite Element subsurface Flow system(Feflow).Accurate hydrogeological parameters were acquired,and the spatio-temporal distribution dynamics of groundwater table for 1983–2001 were also simulated.The model performed well with a correlation coefficient of 0.977 and a mean error of 0.9768 m.The inflow and outflow of the groundwater system were predicted by time series analysis,and the groundwater table dynamics for 2011 were further acquired.Generally the groundwater table in the Minqin Basin would continue to decline.The groundwater table would decline during spring and summer irrigation,while it would rise during autumn-winter irrigation.The groundwater depression cones would expand with the increase of center depths.Therefore,regulatory measures should be taken to prevent the declination of groundwater table and improve the eco-environment of this area. | Lan MA XiaoMei WEI AnMing BAO ShiFei WANG | 2012 | Journal of Arid Land2012,4,2: | 5 |
