7One feature of the activated southern Ordos block:the Ziwuling small earthquake cluster显示文摘Small earthquakes( Ms > 2.0) have been recorded from 1970 to the present day and reveal a significant difference in seismicity between the stable Ordos block and its active surrounding area.The southern Ordos block is a conspicuous small earthquake belt clustered and isolated along the NNW direction and extends to the inner stable Ordos block;no active fault can match this small earthquake cluster.In this paper,we analyze the dynamic mechanism of this small earthquake cluster based on the GPS velocity field( from 1999 to2007),which are mainly from Crustal Movement Observation Network of China( CMONOC) with respect to the north and south China blocks.The principal direction of strain rate field,the expansion ratefield,the maximum shear strain rate,and the rotation rate were constrained using the GPS velocity field.The results show that the velocity field,which is bounded by the small earthquake cluster from Tongchuan to Weinan,differs from the strain rate field,and the crustal deformation is left-lateral shear.This left-lateral shear belt not only spatially coincides with the Neo-tectonic belt in the Weihe Basin but also with the NNW small earthquake cluster( the Ziwuling small earthquake cluster).Based on these studies,we speculate that the NNW small earthquake cluster is caused by left-lateral shear slip,which is prone to strain accumulation.When the strain releases along the weak zone of structure,small earthquakes diffuse within its upper crust.The maximum principal compression stress direction changed from NE-SW to NEE-SWW,and the former reverse faults in the southwestern margin of the Ordos block became a left-lateral strike slip due to readjustment of the tectonic stress field after the middle Pleistocene.The NNW Neo-tectonic belt in the Weihe Basin,the different movement character of the inner Weihe Basin( which was demonstrated through GPS measurements) and the small earthquake cluster belt reflect the activated southern margin of the Ordos block,which was generated through readjustment of the tectonic stress field after the middle Pleistocene.Li Yuhang Wang Qingliang Cui Duxin Hao Ming Ji Lingyun Qin Shanlan 2014Geodesy and Geodynamics2014,5,3:1

9Structure-tissue exposure/selectivity relationship(STR) correlates with clinical efficacy/safety显示文摘Drug optimization, which improves drug potency/specificity by structure-activity relationship(SAR) and drug-like properties, is rigorously performed to select drug candidates for clinical trials. However,the current drug optimization may overlook the structure-tissue exposure/selectivity-relationship(STR) in disease-targeted tissues vs. normal tissues, which may mislead the drug candidate selection and impact the balance of clinical efficacy/toxicity. In this study, we investigated the STR in correlation with observed clinical efficacy/toxicity using seven selective estrogen receptor modulators(SERMs) that have similar structures, same molecular target, and similar/different pharmacokinetics. The results showed that drug’s plasma exposure was not correlated with drug’s exposures in the target tissues(tumor, fat pad, bone, uterus),while tissue exposure/selectivity of SERMs was correlated with clinical efficacy/safety. Slight structure modifications of four SERMs did not change drug’s plasma exposure but altered drug’s tissue exposure/selectivity.Seven SERMs with high protein binding showed higher accumulation in tumors compared to surrounding normal tissues, which is likely due to tumor EPR effect of protein-bound drugs. These suggest that STR alters drug’s tissue exposure/selectivity in disease-targeted tissues vs. normal tissues impacting clinical efficacy/toxicity. 13Seco-cyclic phorbol derivatives and their anti-HIV-1 activities显示文摘Phorbol esters are recognized for their dual role as anti-HIV-1 agents and as activators of protein kinase C(PKC).The efficacy of phorbol esters in binding with PKC is attributed to the presence of oxygen groups at positions C20,C3/C4,and C9 of phorbol.Concurrently,the lipids located at positions C12/C13 are essential for both the anti-HIV-1 activity and the formation of the PKC-ligand complex.The influence of the cyclopropane ring at positions C13 and C14 in phorbol derivatives on their anti-HIV-1 activity requires further exploration.This research entailed the hydrolysis of phorbol,producing seco-cyclic phorbol derivatives.The anti-HIV-1 efficacy of these derivatives was assessed,and the affinity constant(Kd)for PKC-δprotein of selected seco-cyclic phorbol derivatives was determined through isothermal titration calorimetry.The findings suggest that the chemical modification of cyclopropanols could affect both the anti-HIV-1 activity and the PKC binding affinity.Remarkably,compound S11,with an EC50 of 0.27μmol·L^(−1) and a CC50 of 153.92μmol·L^(−1),demonstrated a potent inhibitory effect on the intermediate products of HIV-1 reverse transcription(ssDNA and 2LTR),likely acting at the viral entry stage,yet showed no affinity for the PKC-δprotein.These results position compound S11 as a potential candidate for further preclinical investigation and for studies aimed at elucidating the pharmacological mechanism underlying its anti-HIV-1 activity.HUANG Xiaolei HUANG Xusheng LI Qirun MA Mengdi CUI Yadong YANG Liumeng WANG Haibo LUO Ronghua CHEN Jinglei YANG Jingxuan LIN Jinrong LI Duxin ZHENG Yongtang ZHANG Jian 2024Chinese Journal of Natural Medicines2024,22,4:0
14Analysis of the diagnosis and treatment of 78 gastric cancer cases in young people显示文摘Objective: To summarize the causes of difficulty in gastric cancer diagnosis in young people and explore potential methods of improving diagnostic accuracy. Methods: We retrospectively analyzed 78 cases of gastric cancer in young people (14–35 years). Results: The clinical manifestations of gastric carcinoma in young people show no specificity, and the initial symptoms were diverse, with discomfort in the upper abdomen as the main symptom. There are four causes of difficulty in early detection: (1) The previous physician relied on antacids and did not use or analyze the results of gastroscopic examination and barium meal examination of the upper digestive tract; (2) The physician performing the endoscopic examination did not strictly follow the standards in the biopsy of fine pathological changes in suspected early cancer; (3) The physician lacked a good understanding of the clinical progress of ulcerating-healing-ulcerating in the ulcerating type of early gastric cancer; (4) Lacked a good understanding of precancerous lesions in the gastric mucosa. Conclusion: Early gastroscopy and pathological examina- tion must be conducted for people who above 20 years of age with one or more of the following symptoms: dull pain in the upper abdomen, anorexia, fatigue, tarry stool and vomiting of unknown origin. The results of gastroscopic examination should be taken into consideration for patients with recurrent ulcer, and biopsy should be standardized. Biopsy specimens should be taken from several loci in the suspected lesions, and should be repeated if necessary. 15The insight of why:Causal inference in Earth system science显示文摘The utilization of big Earth data has provided insights into the planet we inhabit in unprecedented dimensions and scales.Unraveling the concealed causal connections within intricate data holds paramount importance for attaining a profound comprehension of the Earth system.Statistical methods founded on correlation have predominated in Earth system science(ESS)for a long time.Nevertheless,correlation does not imply causation,especially when confronted with spurious correlations resulting from big data.Consequently,traditional correlation and regression methods are inadequate for addressing causation related problems in the Earth system.In recent years,propelled by advancements in causal theory and inference methods,particularly the maturity of causal discovery and causal graphical models,causal inference has demonstrated vigorous vitality in various research directions in the Earth system,such as regularities revealing,processes understanding,hypothesis testing,and physical models improving.This paper commences by delving into the origins,connotations,and development of causality,subsequently outlining the principal frameworks of causal inference and the commonly used methods in ESS.Additionally,it reviews the applications of causal inference in the main branches of the Earth system and summarizes the challenges and development directions of causal inference in ESS.In the big Earth data era,as an important method of big data analysis,causal inference,along with physical model and machine learning,can assist the paradigm transformation of ESS from a model-driven paradigm to a paradigm of integration of both mechanism and data.Looking forward,the establishment of a meticulously structured and normalized causal theory can act as a foundational cornerstone for fostering causal cognition in ESS and propel the leap from fragmented research towards a comprehensive understanding of the Earth system.Jianbin SU Duxin CHEN Donghai ZHENG Yang SU Xin LI 2023Science China Earth Sciences2023,66,10:0
16Geodetic constraints on contemporary three-dimensional crustal deformation in the Laji Shan—Jishi Shan tectonic belt显示文摘The Laji Shan—Jishi Shan tectonic belt(LJTB),located in the southern part of the northeastern Tibetan Plateau(NETP),is a tectonic window to reveal regional tectonic deformation in the NETP.However,its kinematics in the Holocene remains controversial.We obtain the latest and dense horizontal velocity field based on data collected from our newly constructed and existing GNSS stations.Combined with fault kinematics from geologic observations,we analyze the crustal deformation characteristics along the LJTB.The results show that:(1)The Laji Shan fault(LJF)is inactive,and the northwest-oriented Jishi Shan fault(JSF)exhibits a significant dextral and thrust slip.(2)The transpression along the arc-shaped LJTB accommodates deformation transformation between the dextral Riyue Shan fault and the sinistral west Qinling fault.(3)With the continuous pushing of the Indian plate,internal strains in the Tibetan Plateau are continuously transferred in the northeast via the LJTB as they are gradually dissipated near the LJTB and translated into significant crustal uplift in these regions.Wenquan Zhuang Duxin Cui Ming Hao Shangwu Song Zhangjun Li 2023Geodesy and Geodynamics2023,14,6:0
17N-glycans released from glycoproteins using a commercial kit and comprehensively analyzed with a hypothetical database显示文摘The glycosylation of proteins is responsible for their structural and functional roles in many cellular activities.This work describes a strategy that combines an efficient release, labeling and liquid chromatography–mass spectral analysis with the use of a comprehensive database to analyze N-glycans. The analytical method described relies on a recently commercialized kit in which quick deglycosylation is followed by rapid labeling and cleanup of labeled glycans. This greatly improves the separation, mass spectrometry(MS) analysis and fluorescence detection of N-glycans. A hypothetical database, constructed using Glyc Resoft, provides all compositional possibilities of N-glycans based on the common sugar residues found in N-glycans. In the initial version this database contains > 8,700 N-glycans, and is compatible with MS instrument software and expandable. N-glycans from four different well-studied glycoproteins were analyzed by this strategy. The results provided much more accurate and comprehensive data than that had been previously reported. This strategy was then used to analyze the N-glycans present on the membrane glycoproteins of gastric carcinoma cells with different degrees of differentiation. Accurate and comprehensive N-glycan data from those cells was obtained efficiently and their differences were compared corresponding to their differentiation states.
