1Influence of puerarin,paeoniflorin,and menthol on structure and barrier function of tight junctions in MDCK and MDCK-MDR1 Cells显示文摘Objective:In this study,the influence of puerarin,paeoniflorin,and menthol on the structure and barrier function of tight junctions(TJs)in MadineDarby canine kidney epithelial(MDCK)and MDCK-multi-drug resistance 1(MDR1)cells was evaluated to determine the mechanisms by which the drugs cross the bloodebrain barrier(BBB).Method:Cells were treated with puerarin,paeoniflorin,and menthol followed by immunohistochemical staining with occludin,claudin-1,and F-actin.The cells were then observed using laser-scanning confocal microscopy.Average optical density(AOD)of the immunofluorescence images of the proteins were analyzed using ImageJ software while Transepithelial electrical resistance(TEER)was measured using an epithelial voltohmmeter.Results:Confocal microscopy revealed that puerarin-and paeoniflorin-treated tight junction proteins were conspicuous while menthol suppressed their expression.Correspondingly,AOD values of cells treated with puerarin or paeoniflorin,or both showed no difference compared to the control group(P>.05)while the menthol group value was downregulated.In 3 h,TEER of cells not treated with menthol were similar to the control group,while treatment with menthol significantly decreased TEER value(P<.05).In addition,application of menthol decreased TEER in MDCK cells earlier than in MDCK-MDR1 cells.Conclusion:Menthol but not puerarin and paeoniflorin may enhance paracellular transport and improve drug penetration of the BBB by disrupting the structure and,thereby,weakening the barrier function of TJs.Lin Zhang Shouying Du Yang Lu Chang Liu Huichao Wu Bing Yang Jie Bai Pengyue Li 2015Journal of Traditional Chinese Medical Sciences2015,2,2:4
2Synthesis and characterization of the flower-like La_(x)Ce_(1-x)O_(1.5+δ) catalyst for low-temperature oxidative coupling of methane显示文摘Lanthanum-based oxides are promising candidates for low-temperature oxidative coupling of methane(OCM).To further lower the OCM reaction temperature,the Ce doped flower-like La_(2)O_(2)CO_(3)microsphere catalysts were synthesized,achieving a significantly low reaction temperature (375℃) while maintaining high C_(2) hydrocarbon selectivity (43.0%).Doping Ce into the lattice of La_(2)O_(2)CO_(3)created more surface oxygen vacancies and bulk lattice defects,which was in favor of the transformation and migration of oxygen species at 350–400℃.The designed H_(2) temperature-programmed reduction (H_(2)-TPR) experiments provided strong evidence that the low reaction temperature of La_(x)Ce_(1-x)O_(1.5+δ)can be attributed to the transformation and migration of oxygen species,which dynamically generated surface oxygen vacancies for continuous oxygen activation to selectively convert methane.Moreover,designed temperatureprogrammed surface reaction (TPSR) clarified that two kinds of surface oxygen species in La_(x)Ce_(1-x)O_(1.5+δ)catalysts were concerned with catalytic performance,that is,the surface chemisorbed oxygen species for the activation of CH_(2)and the formation of CH_(2)·intermediates,surface La-Ce-O lattice oxygen species that caused the excessive oxidation of CH_(2)·intermediates.Finally,the factors affecting the transformation and migration of oxygen species were explored.Ru Feng Pengyu Niu Bo Hou Qiang Wang Litao Jia Minggui Lin Debao Li 2022Journal of Energy Chemistry2022,31,4:2
3LIM domain proteins Pinch1/2 regulate chondrogenesis and bone mass in mice显示文摘The LIM domain-containing proteins Pinch1/2 regulate integrin activation and cell–extracellular matrix interaction and adhesion.Here,we report that deleting Pinch1 in limb mesenchymal stem cells(MSCs)and Pinch2 globally(double knockout;dKO)in mice causes severe chondrodysplasia,while single mutant mice do not display marked defects.Pinch deletion decreases chondrocyte proliferation,accelerates cell differentiation and disrupts column formation.Pinch loss drastically reduces Smad2/3 protein expression in proliferative zone(PZ)chondrocytes and increases Runx2 and Col10a1 expression in both PZ and hypertrophic zone(HZ)chondrocytes.Pinch loss increases sclerostin and Rankl expression in HZ chondrocytes,reduces bone formation,and increases bone resorption,leading to low bone mass.In vitro studies revealed that Pinch1 and Smad2/3 colocalize in the nuclei of chondrocytes.Through its C-terminal region,Pinch1 interacts with Smad2/3 proteins.Pinch loss increases Smad2/3 ubiquitination and degradation in primary bone marrow stromal cells(BMSCs).Pinch loss reduces TGF-β-induced Smad2/3 phosphorylation and nuclear localization in primary BMSCs.Interestingly,compared to those from single mutant mice,BMSCs from dKO mice express dramatically lower protein levels ofβ-catenin and Yap1/Taz and display reduced osteogenic but increased adipogenic differentiation capacity.Finally,ablating Pinch1 in chondrocytes and Pinch2 globally causes severe osteopenia with subtle limb shortening.Collectively,our findings demonstrate critical roles for Pinch1/2 and a functional redundancy of both factors in the control of chondrogenesis and bone mass through distinct mechanisms.Yiming Lei Xuekun Fu Pengyu Li Sixiong Lin Qinnan Yan Yumei Lai Xin Liu Yishu Wang Xiaochun Bai Chuanju Liu Di Chen Xuenong Zou Xu Cao Huiling Cao Guozhi Xiao 2020Bone Research2020,8,4:2

11Composition,dispersion,and health risks of bioaerosols in wastewater treatment plants:A review显示文摘Bioaerosols are defined as airbome particles(0.05-100 um in size)of biological origin.They are considered potentially harmful to human health as they can contain pathogens such as bacteria,fungi,and viruses.This review summarizes the most recent research on the health risks of bioaerosols emitted from wastewater treatment plants(WWTPs)in order to improve the control of such bioaerosols.The concentration and size distribution of WWTP bioaerosols;their major emission sources,composition,and health risks;and considerations for future research are discussed.The major themes and findings in the literature are as follows:the major emission sources of WWTP bioaerosols include screen rooms,sludge-dewatering rooms,and acration tanks;the bioaerosol concentrations in screen and sludge-dewatering rooms are higher than those outdoors.WWTP bioacrosols contain a variety of potentially pathogenic bacteria,fungi,antibiotic resistance genes,viruses,endotoxins,and toxic metal(loid)s.These potentially,pathogenic substances spread with the bioaerosols,thereby posing health risks to workers and residents in and around the WWTP.Inhalation has been identified as the main exposure route,and children are at a higher risk of this than adults.Future studies should identify emerging contaminants,establish health risk assessments,and develop prevention and control systems.Yunping Han Lin Li Ying Wang Jiawei Ma Pengyu Li Chao Han Junxin Liu 2021Frontiers of Environmental Science & Engineering2021,15,3:0
12Study on Relationship Between Arc Behavior and Joint Forming in Cold Metal Transfer Welding Technology with Polarity⁃Exchanging显示文摘Cold metal transfer with polarity⁃exchanging is a new integrated welding technology based on MIG.Due to the alternation of the positive and negative polarities of the wire,favorable control upon the deposition rate and the welding shape coefficient was obtained in order to meet the desired joint design,and the related controlling principles and joint characteristics were reported.Droplet transfer physical behavior exhibited strong dependability on the studied welding parameters,such as welding voltage,welding current,wire feeding speed,and polarity⁃exchanging.This welding technology provides a new way for the welding of body⁃in⁃white(BIW)thin sheet with special demands.Moreover,the typical quality defects of MIG were greatly improved.Our study provides important technical information from the perspective of industrial application of MIG and sheds light on the higher application level of MIG in BIW welding.Lijun Han Pengyu Lin Gengwei Zhang Lihui Zhong Changhua Liu 2020Journal of Harbin Institute of Technology(New Series)2020,27,5:0
