| 1 | An enhanced method for nucleic acid detection with CRISPR-Cas12a using phosphorothioate modified primers and optimized gold-nanopaticle strip显示文摘CRISPR-Cas12a system has been shown promising for nucleic acid diagnostics due to its rapid,portable and accurate features.However,cleavage of the amplicons and primers by the cis-and trans-activity of Cas12a hinders the attempts to integrate the amplification and detection into a single reaction.Through phosphorothioate modification of primers,we realized onepot detection with high sensitivity using plasmids of SARS-CoV-2,HPV16 and HPV18.We also identified the activated Cas12a has a much higher affinity to C nucleotide-rich reporter than others.By applying such reporters,the reaction time required for a lateral-flow readout was significantly reduced.Furthermore,to improve the specificity of the strip-based assay,we created a novel reporter and,when combined with a customized gold-nanopaticle strip,the readout was greatly enhanced owing to the elimination of the nonspecific signal.This established system,termed Targeting DNA by Cas12a-based Eye Sight Testing in an Onepot Reaction(TESTOR),was validated using clinical cervical scrape samples for human papillomaviruses(HPVs)detection.Our system represents a general approach to integrating the nucleic acid amplification and detection into a single reaction in CRISPR-Cas systems,highlighting its potential as a rapid,portable and accurate detection platform of nucleic acids. | Jiaojiao Gong Lijuan Kan Xiuming Zhang Ying He Jiaqiang Pan Liping Zhao Qianyun Li Menghao Liu Jie Tian Sili Lin Zhouyu Lu Liang Xue Chaojun Wang Guanghui Tang | 2021 | Bioactive Materials2021，6，12： | 1 |

| 3 | Association between IDEAL-IQ MRI fat fraction quantification and pelvic bone marrow reserve function in concurrent chemoradiotherapy for cervical cancer显示文摘Objective:To analyze the association between iterative decomposition of water and fat with echo asymmetry and least-squares estimation quantification sequence(IDEAL-IQ)magnetic resonance imaging(MRI)of bone marrow fat fraction and bone marrow reserve function during concurrent chemoradiotherapy for cervical cancer.Methods:The study retrospectively analyzed twenty-six patients with stage IB1 to IVA cervical cancer treated between February 2020 and November 2020.All patients received concurrent chemoradiotherapy that included platinum alone or combined paclitaxel and platinum.Pelvic IDEAL-IQ MRI(plain and enhanced)was performed before and after treatment.Regions of interest,including the fifth lumbar vertebra,sacrum,ilium,ischium,and femoral neck,were manually delineated,and the bone marrow fat fraction was measured.Peripheral blood cell counts were recorded during treatment,and the relationship between the fat fraction values and changes in the blood cell counts was explored.Results:IDEAL-IQ MRI bone marrow fat fraction was associated with platelet nadir and platelet decline during treatment.The average pelvic bone marrow fat fraction before chemoradiotherapy was moderately negatively correlated with platelet count nadir during concurrent chemoradiotherapy(r=-0.450,P?0.021).The change in average pelvic bone marrow fat fraction through chemoradiotherapy was moderately positively correlated with the degree of thrombocytopenia(r=0.399,P=0.044).Conclusion:Bone marrow fat content quantified by IDEAL-IQ was associated with platelet count nadir and the degree of thrombocytopenia in patients with cervical cancer undergoing concurrent chemoradiotherapy. | Xiaomin Wen Qinghua Qin Yanling Wu Zhouyu Li Xinglong Yang Jinquan Liu Ling Lin Mingyi Li | 2023 | Radiation Medicine and Protection2023，4，3： | 0 |
