3Development and characterization of a clinical strain of Coxsackievirus A16 and an eGFP infectious clone显示文摘Coxsackievirus A16(CA16) is one of the major causes of hand, foot, and mouth disease(HFMD) worldwide, which is a common illness that affects children. The frequent occurrence of HFMD outbreaks has become a serious public health problem in Asia. Therefore, it is important to understand the pathogenesis and replication of CA16. In this study, a stable infectious c DNA clone of an epidemic strain of Coxsackievirus A16(CA16) was assembled, and subsequently a reporter virus(e GFP-CA16) was constructed by inserting the e GFP gene between the 5'-UTR and the N-terminus of VP4, with the addition of a 2A protease cleavage site(ITTLG) at its C-terminus. This was transfected into Vero cells to generate infectious recombinant viruses. The growth characteristics and plaque morphology, in vitro, in mammalian cells were found to be indistinguishable between the parental and recombinant viruses. Although the e GFP-CA16 showed smaller plaque size as compared to recombinant CA16, both were found to exhibit similar growth trends and EC50 of NITD008. In summary, this stable infectious c DNA clone should provide a valuable experimental system to study CA16 infection and host response. 4Effect of seed electron injection on chorus-driven acceleration of radiation belt electrons显示文摘Using the hybrid finite difference method,we solve the Fokker-Planck equation to study the effect of seed electron injection on acceleration of radiation belt electrons driven by chorus waves.Numerical results show that in the absence of injection chorus waves can accelerate electrons at large pitch angles(e >60°),producing enhancements in the phase space density(PSD) of(1-2 MeV) electrons by a factor of 100-1000 within 1-2 days.In the presence of injection,chorus waves yield increase in PSD of electrons by accelerating the injected seed electrons.Meanwhile,the PSD evolution increases as the pitch angle increases but decreases as electron energy increases.Moreover,the PSD evolution can extend to higher energies with a time scale of 1-2 days for 1-2 MeV energies.When the injection increases by a factor of 10 higher than the initial value and remains for about two days,maximum values of PSD for 1 or 2 MeV increase to 6 or 3 times respectively higher than those without injection in two days.The current results suggest that the injected seed electrons play an important role in the evolution of the radiation belt electrons.YAN Qi SHI LiQing LIU SiQing 2013Science China(Technological Sciences)2013,56,2:4
5A regional ionospheric TEC mapping technique over China and adjacent areas: GNSS data processing and DINEOF analysis显示文摘A technique is developed to derive two-dimensional maps of total electron content(TEC) over China and adjacent areas using Global Navigation Satellite System(GNSS) data from the Crustal Movement Observation Network of China(CMONOC) and the International Global Navigation Satellite System Service(IGS). A revised self-calibration of pseudo-range errors(SCORE) algorithm is used to derive the TEC and to determine the Differential Code Biases(DCBs) simultaneously. The accuracy and validity of this technique is verified in two ways. Firstly, the estimated TEC is compared with the results derived using DCBs from Center for Orbit Determination of Europe(CODE) under different solar activity conditions and seasons; secondly, sample TEC along the receiver-to-satellite ray paths are simulated by Ne Quick model and are reprocessed by this TEC derivation technique to make the accuracy test. Two-dimensional maps of vertical TEC of ionospheric pierce points(IPPs) are obtained accordingly with a time resolution of 30 s. The data interpolating empirical orthogonal functions(DINEOF) technique is then used to make the extrapolation for the unknown or missing data points. The optimal EOF modes for data reconstruction are specified via cross-validation method. 7Genome-wide analysis reveals selection for Chinese Rongchang pigs显示文摘Livestock have undergone domestication and consequently strong selective pressure on genes or genomic regions that control desirable traits. To identify selection signatures in the genome of Chinese Rongchang pigs, we generated a total of about 170 Gb of DNA sequence data with about 6.4-fold coverage for each of six female individuals. By combining these data with the publically available genome data of 10 Asian wild boars,we identified 449 protein-coding genes with selection signatures in Rongchang pigs, which are mainly involved in growth and hormone binding, nervous system development, and drug metabolism. The accelerated evolution of these genes may contribute to the dramatic phenotypic differences between Rongchang pigs and Chinese wild boars. 8Passively Q-switched erbium doped fiber laser using a gold nanostars based saturable absorber显示文摘In this paper, we propose and demonstrate an all-fiber passively Q-switched erbium doped fiber laser(EDFL) by using gold nanostars(GNSs) as a saturable absorber(SA) for the first time, to the best of our knowledge. In comparison with other gold nanomorphologies, GNSs have multiple localized surface plasmon resonances, which means that they can be used to construct wideband ultrafast pulse lasers. By inserting the GNS SA into an EDFL cavity pumped by a 980 nm laser diode, a stable passively Q-switched laser at 1564.5 nm was achieved for a threshold pump power of 40 mW. By gradually increasing the pump power from 40 to 120 mW, the pulse duration decreases from 12.8 to 5.3 μs and the repetition rate increases from 10 to 17 kHz. 9Extensive evolution analysis of the global chikungunya virus strains revealed the origination of CHIKV epidemics in Pakistan in 2016显示文摘Chikungunya virus(CHIKV) is a mosquito-borne virus that causes epidemics widely in the world especially in the tropical and subtropical regions. Phylogenetic analysis has found that the CHIKV lineages were associated with the spatial and temporal distributions, which were related to the virus adaption to the major mosquito species and their distributions. In this study, we reported the complete genome sequences of eight CHIKV isolates from the outbreak in Pakistan last year. Then we reviewed the evolutionary history using extensive phylogenetic analysis, analyzed lineagespecific substitutions in viral proteins, and characterized the spreading pathway of CHIKV strains including the Pakistani strains. The results showed that the Pakistani stains belonged to the ECSA.IOL sub-lineage and derived from India. The genetic properties of the Pakistani strains including the adaptive substitution to vectors were further characterized, and the potential risks from the occurrence of CHIKV infection in Pakistan were discussed. 10IL-33 drives the antitumor effects of dendritic cells via the induction of Tc9 cells显示文摘Dendritic cell(DC)tumor vaccines exert their antitumor effects through the induction of effector T cells.We recently identified Tc9 cells as a new potent antitumor effector T cell subset.However,approaches to direct DCs to preferably prime antitumor Tc9 cells should be further exploited.Here,we demonstrate that the addition of interleukin(IL)-33 potently promotes the induction of Tc9 cells by DCs in vitro and in vivo.IL-33 treatment also drives the cytotoxic activities of DC-induced Tc9 cells.Notably,IL-33 treatment enhances cell survival and proliferation of DC-primed CD8+T cells.More importantly,the addition of IL-33 during in vitro priming of tumor-specific Tc9 cells by DCs increases the antitumor capability of Tc9 cells.Mechanistic studies demonstrated that IL-33 treatment inhibits exhaustive CD8+T cell differentiation by inhibiting PD-1 and 2B4 expression and increasing IL-2 and CD127(IL-7 receptor-α,IL-7Rα)expression in CD8+T cells.Finally,the addition of IL-33 further promotes the therapeutic efficacy of DC-based tumor vaccines in the OT-I mouse model.Our study demonstrates the important role of IL-33 in DC-induced Tc9 cell differentiation and antitumor immunity and may have important clinical implications.Ning Liu Yuxue Jiang Jintong Chen He Nan Yinghua Zhao Xiao Chu Alison Wang Dongjiao Wang Tianxue Qin Sujun Gao Ying Yue Siqing Wang 2019Cellular & Molecular Immunology2019,16,7:2
11Highly efficient acousto-optic modulation using nonsuspended thin-fil1m lithium niobate-chalcogenide hybrid waveguides显示文摘A highly efficient on-chip acousto-optic modulator is as a key component and occupies an exceptional position in microwave-to-optical conversion.Homogeneous thin-film lithium niobate is preferentially employed to build the suspended configuration for the acoustic resonant cavity,with the aim of improving the modulation efficiency of the device.However,the limited cavity length and complex fabrication recipe of the suspended prototype restrain further breakthroughs in modulation efficiency and impose challenges for waveguide fabrication.In this work,based on a nonsuspended thin-film lithium niobate-chalcogenide glass hybrid Mach-Zehnder interferometer waveguide platform,we propose and demonstrate a built-in push-pull acousto-optic modulator with a half-wave-voltage-length product VnL as low as 0.03 V cm that presents a modulation efficiency comparable to that of a state-of-the-art suspended counterpart.A microwave modulation link is demonstrated using our developed built-in push-pull acousto-optic modulator,which has the advantage of low power consumption.The nontrivial acousto-optic modulation performance benefits from the superior photoelastic property of the chalcogenide membrane and the completely bidirectional participation of the antisymmetric Rayleigh surface acoustic wave mode excited by the impedance-matched interdigital transducer,overcoming the issue of low modulation efficiency induced by the incoordinate energy attenuation of acoustic waves applied to the Mach-Zehnder interferometer with two arms in traditional push-pull acousto-optic modulators.Lei Wan Zhiqiang Yang Wenfeng Zhou Meixun Wen Tianhua Feng Siqing Zeng Dong Liu Huan Li Jingshun Ppan Ning Zhu Weiping Liu Zhaohui Li 2022Light(Science & Applications)2022,11,6:2
12Augmented cellular uptake and homologous targeting of exosome-based drug loaded IOL for posterior capsular opacification prevention and biosafety improvement显示文摘Posterior capsular opacification(PCO),the most common complication after cataract surgery,is caused by the proliferation,migration and differentiation of residual lens epithelial cells(LECs)on the surface of the intraocular lens(IOL).Although drug-loaded IOLs have been successfully developed,the PCO prevention efficacy is still limited due to the lack of targeting and low bioavailability.In this investigation,an exosome-functionalized drug-loaded IOL was successfully developed for effective PCO prevention utilizing the homologous targeting and high biocompatibility of exosome.The exosomes derived from LECs were collected to load the anti-proliferative drug doxorubicin(Dox)through electroporation and then immobilized on the aminated IOLs surface through electrostatic interaction.In vitro experiments showed that significantly improved cellular uptake of Dox@Exos by LECs was achieved due to the targeting ability of exosome,compared with free Dox,thus resulting in superior anti-proliferation effect.In vivo animal investigations indicated that Dox@Exos-IOLs effectively inhibited the development of PCO and showed excellent intraocular biocompatibility.We believe that this work will provide a targeting strategy for PCO prevention through exosome-functionalized IOL.Siqing Zhu Huiying Huang Dong Liu Shimin Wen Liangliang Shen Quankui Lin 2022Bioactive Materials2022,7,9:1

17A 3D analysis of the occurrence of fractures in hot dry rock reservoirs based on the spatial distribution of natural fractures显示文摘Hot dry rock(HDR)geothermal energy is a kind of widely distributed clean energy with huge reserves.However,its commercial development has been constrained by reservoir stimulation.In the early stage of HDR geothermal energy development,properly determining spatial distribution patterns of natural fractures in HDR reservoirs can effectively guide reservoir stimulation.This study analyzes the spatial distribution of natural fractures by using FracMan software based on the actual geological data and log data of well M-2 in the Matouying Uplift area,Hebei Province.The fracture parameters are counted and Monte Carlo simulation technique is introduced to optimize the parameters,which makes the natural fracture model more accurate and reliable.Furthermore,this study simulates hydraulic fracturing using the model combined with the actual in-situ stress parameters and the construction scheme.As verified by fitting the changes in simulated wellhead pressure during hydraulic fracturing with the actual wellhead pressure data detected during construction,the methods for natural fracture modeling used in this study are scientific and reasonable.The preliminary prediction results show that the displacement design scheme with a pump displacement of 2.0-3.0 m^(3)/min,4.0-5.5 m^(3)/min and 6-7 m^(3)/min in the early,middle and late stages,respectively,has good fracturing effect.The results of this study can be utilized as a reference for preparing development schemes for HDR reservoirs.Siqing He Bo Feng Jinshou Zhu Xiyao Liu Shuantong ShangGuan Xiaofei Qi Jiulong Liu 2023Energy Geoscience2023,4,4:1
18Comparison of a new model with previous models for the low-latitude magnetopause size and shape显示文摘In this study, the advantages and the limitations of previous low-latitude magnetopause empirical models are discussed. In order to overcome their limitations and inherit their advantages, a new continuous function for the influence of the interplanetary magnetic field (IMF) Bz on the magnetopause, the Shue model function and the 613 low-latitude magnetopause crossings are used to construct a new low-latitude magnetopause model parameterized by the solar wind dynamic pressure (Dp) and IMF Bz. In comparison with the previous low-latitude magnetopause models, it is found that the new model improves the prediction capability and has a large range of validity for the low-latitude magnetopause. 20Rabies surveillance and control in China over the last twenty years显示文摘Globally,rabies remains a serious threat to public health.In particular,large number of cases continue to be detected in Africa and Asia.China has experienced three major outbreaks with the current epizootic starting in 1990s,peaking in 2007 with 3,300 cases,and consistently decreasing since this point.A key factor of the success in keeping the rabies outbreak under control is the National Public Health Reporting Program that was introduced to coordinate surveillance at provincial and regional levels.Through this program,it is possible to obtain an integrated overview of how rabies was spreading and how various factors were contributing to the outbreak.At the same time,samples have been collected from suspected rabies cases and a subset of positive samples selected for sequencing.In this way,it is possible to investigate the outbreak from a phylogenetic perspective,providing the comprehensive description of a rabies outbreak to date.In thiswork,we review rabies control efforts over the last 20 years,and showhowthis integrated approach has led to a deeper characterization of the outbreak and a better understanding of the factors driving the spread of the disease.Xiaoyan Tao Siqing Liu Wuyang Zhu Simon Rayner 2021Biosafety and Health2021,3,3:1
