| 2 | Dynamic blood single-cell immune responses in patients with COVID-19显示文摘The 2019 coronavirus disease(COVID-19)outbreak caused by the SARS-CoV-2 virus is an ongoing global health emergency.However,the virus'pathogenesis remains unclear,and there is no cure for the disease.We investigated the dynamic changes of blood immune response in patients with COVID-19 at different stages by using 5'gene expression,T cell receptor(TCR),and B cell receptors(BCR)V(D)J transcriptome analysis at a single-cell resolution.We obtained single-cell mRNA sequencing(scRNA-seq)data of 341,420 peripheral blood mononuclear cells(PBMCs)and 185,430 donotypic T cells and 28,802 donotypic B cells from 25 samples of 16 patients with COVID-19 for dynamic studies.In addition,we used three control samples.We found expansion of dendritic cells(DCs),CD14+monocytes,and megakaryocytes progenitor cells(MP)/platelets and a reduction of naive CD4+T lymphocytes in patients with COVID-19,along with a significant decrease of CD8+T lymphocytes,and natural killer cells(NKs)in patients in critical condition.The type I interferon(IFN-I),mitogen-activated protein kinase(MAPK),and ferroptosis pathways were activated while the disease was active,and recovered gradually after patient conditions improved.Consistent with this finding,the mRNA level of IFN-I signal-induced gene IFI27 was significantly increased in patients with COVID-19 compared with that of the controls in a validation cohort that included 38 patients and 35 controls.The concentration of interferon-a(IFN-a)in the serum of patients with COVID-19 increased significantly compared with that of the controls in an additional cohort of 215 patients with COVID-19 and 106 controls,further suggesting the important role of the IFN-I pathway in the immune response of COVID-19.TCR and BCR sequences analyses indicated that patients with COVID-19 developed specific immune responses against SARS-CoV-2 antigens.Our study reveals a dynamic landscape of human blood immune responses to SARS-CoV-2 infection,providing clues for therapeutic potentials in treating COVID-19. | Lulin Huang Yi Shi Bo Gong Li Jiang Zhixin Zhang Xiaoqi Liu Jialiang Yang Yongquan He Zhilin Jiang Ling Zhong Juan Tang Chunfang You Qi Jiang Bo Long Tao Zeng Mei Luo Fanwei Zeng Fanxin Zeng Shuqiang Wang Xingxiang Yang Zhenglin Yang | 2021 | Signal Transduction and Targeted Therapy2021,6,4: | 4 |

| 11 | Reducing water and nitrogen inputs combined with plastic mulched ridge-furrow irrigation improves soil water and salt status in arid saline areas,China显示文摘Plastic mulched ridge-furrow irrigation is a useful method to improve crop productivity and decrease salt accumulation in arid saline areas.However,inappropriate irrigation and fertilizer practices may result in ecological and environmental problems.In order to improve the resource use efficiency in these areas,we investigated the effects of different irrigation amounts(400(I1),300(I2)and 200(I3)mm)and nitrogen application rates(300(F1)and 150(F2)kg N/hm^(2))on water consumption,salt variation and resource use efficiency of spring maize(Zea mays L.)in the Hetao Irrigation District(HID)of Northwest China in 2017 and 2018.Result showed that soil water contents were 0.2%-8.9%and 13.9%-18.1%lower for I2 and I3 than for I1,respectively,but that was slightly higher for F2 than for F1.Soil salt contents were 7.8%-23.5%and 48.5%-48.9%lower for I2 than for I1 and I3,but that was 1.6%-5.5%higher for F1 than for F2.Less salt leaching at the early growth stage(from sowing to six-leaf stage)and higher salt accumulation at the peak growth stage(from six-leaf to tasseling stage and from grain-filling to maturity stage)resulted in a higher soil salt content for I3 than for I1 and I2.Grain yields for I1 and I2 were significantly higher than that for I3 and irrigation water use efficiency for I2 was 14.7%-34.0%higher than that for I1.Compared with F1,F2 increased the partial factor productivity(PFP)of nitrogen fertilizer by more than 80%.PFP was not significantly different between I1F2 and I2F2,but significantly higher than those of other treatments.Considering the goal of saving water and nitrogen resources,and ensuring food security,we recommended the combination of I2F2 to ensure the sustainable development of agriculture in the HID and other similar arid saline areas. | LI Cheng WANG Qingsong LUO Shuai QUAN Hao WANG Naijiang LUO Xiaoqi ZHANG Tibin DING Dianyuan DONG Qin'ge FENG Hao | 2021 | Journal of Arid Land2021,13,8: | 0 |
| 12 | Evaluation of FGF10 as a candidate gene for high myopia in a Han Chinese population显示文摘Background:Fibroblast growth factor 10(FGF10)is implicated in the growth and development of the eye.Four singles nucleotide polymorphisms(SNPs)in the FGF10 gene(including rs1384449,rs339501,rs12517396 and rs10462070)were found to be associated with extreme myopia(EM,refractive error≤−10.0 diopters)in Japanese and Chinese Taiwan population.This case-control association study was conducted to explore the relationship between these four SNPs and high myopia in a western Chinese population.Methods:A total of 869 high myopia patients(HM,including 485 EM patients)and 899 healthy controls were recruited.These four SNPs were genotyped using the ABI SNaPshot method.Five genetic models(allelic,homozygous,heterozygous,dominant,and recessive)were applied to further evaluate the possible correlation between the SNPs and high myopia.The linkage-disequilibrium block(LD)structure was tested by Haploview Software.Results:In our study,no statistically significant differences were found between HM/EM patients and controls after Bonferroni multiple-correction(P>0.05)in the allele frequencies of these four SNPs in the FGF10 gene.We further found that rs12517396AA and rs10462070GG carriers showed a decreased risk of HM/EM compared with rs12517396AC+CC and rs10462070GA+AA carriers(P=0.045,OR=0.366;P=0.021,OR=0.131;P=0.03,OR=0.341;P=0.015,OR=0.122;respectively).Additionally,rs12517396AA and rs10462070GG carriers showed the same decreased risk of HM/EM compared with rs12517396CC and rs10462070AA carriers(P=0.048,OR=0.370;P=0.023,OR=0.133;P=0.032,OR=0.346;P=0.017,OR=0.126).However,these significant associations between rs12517396/rs10462070 and HM/EM disappeared after Bonferroni multiple-correction(P>0.05).Conclusion:Our findings indicate that rs12517396 and rs10462070 had marginal association with HM and EM.The other two common polymorphisms in FGF10 unlikely have significant effects in the genetic predisposition to HM/EM in western Chinese population.Further replication studies are needed to validate our findings in both animal models and human genetic epidemiologic studies. | Lingxi Jiang Dongyan Luo Tingting Wang Rui Zheng Yaru Zhai Xiaoqi Liu Bo Gong Zhengzheng Wu Yin Yang Zhenglin Yang Yi Shi | 2019 | Eye and Vision2019,6,1: | 0 |

| 19 | The water-soluble bicyclic 2-pyridone-based fluorescent probe for fast and selective detection of hypochlorite显示文摘As a strong oxidizer,hypochlorite(Cl O^(-))are widely employed as bleaching agents and disinfectants.Determination of ClO^(-)is required to ensure bactericidal effects and avoid hazards caused by excessive residual chlorine.Herein,the derivative bicyclic 2-pyridone,namely DHIP-Py,was prepared successfully to establish a new ClO^(-)-quantitative method.The probe exhibits excellent Cl O^(-)selectivity over other ROS and anions/cations,high sensitivity(LOD=1.32μmol/L),fast response(<5 s),and wide-p H tolerance(pH4~10).Benefit from its good water solubility,DHIP-Py is well suited for water sample analysis and has been successfully applied to detect Cl O^(-)in real-world food and environmental samples,including tap water,bottled water and river water.The detection results were essentially identical to that of obtained from traditional DPD method.Moreover,visual detection of Cl O^(-)via filter paper-based solid sensor and imaging of ClO^(-)in Escherichia coli were also achieved by DHIP-Py.These satisfactory results demonstrate that this bicyclic 2-pyridone-based hypochlorite probe is a promising free chlorine chemosensor with great potential for analytical applications. | Qian Zhou Shanqiang Wang Xiaoyun Ran Linzhi Shen Xiaolin Luo Gui Wang Hui Yang Zhouyu Wang Xiaoqi Yu | 2023 | Chinese Chemical Letters2023,34,6: | 0 |
