1IL-10-producing regulatory B cells restrain the T follicular helper cell response in primary Sjögren’s syndrome显示文摘Increased numbers of T follicular helper(Tfh)cells have been implicated in the development of autoimmune diseases including primary Sjögren’s syndrome(pSS),but how the Tfh cell response is regulated during autoimmune pathogenesis remains largely unclear.Here,we first found negative correlations between IL-10^(+)regulatory B(Breg)cell numbers and Tfh cell responses and disease activity in patients with pSS and mice with experimental Sjögren’s syndrome(ESS).Moreover,we detected high expression of IL-10 receptor on Tfh cells and their precursors in both humans and mice.In culture,IL-10 suppressed human and murine Tfh cell differentiation by promoting STAT5 phosphorylation.By using an adoptive transfer approach and two-photon live imaging,we found significantly increased numbers of Tfh cells with enhanced T cell homing into B cell follicles in the draining cervical lymph nodes of RAG-2−/−mice transferred with IL-10-deficient B cells during ESS development compared with those of RAG-2−/−mice transferred with wild-type B cells.In ESS mice,CD19^(+)CD1d^(hi)CD5^(+)Breg cells with decreased IL-10 production exhibited severely impaired suppressive effects on T cell proliferation.Consistently,CD19^(+)CD24^(+)CD38^(hi) Breg cells from pSS patients showed significantly reduced IL-10 production with defective inhibitory function in the suppression of autologous Tfh cell expansion.Furthermore,the adoptive transfer of IL-10-producing Breg cells markedly suppressed the Tfh cell response and ameliorated ESS progression in ESS mice.Together,these findings demonstrate a critical role for IL-10-producing Breg cells in restraining the effector Tfh cell response during pSS development.Xiang Lin Xiaohui Wang Fan Xiao Kongyang Ma Lixiong Liu Xiaoqi Wang Dong Xu Fei Wang Xiaofei Shi Dongzhou Liu Yan Zhao Liwei Lu 2019Cellular & Molecular Immunology2019,16,12:13
2Geochemical characteristics of natural gas from mud volcanoes in the southern Junggar Basin显示文摘A number of mud volcanoes exist in the southern Junggar Basin. To date few systematic studies on natural gas geochemistry of mud volcanoes have been conducted in China. In June 1991 and August 2010, the authors investigated the mud volcanoes in the southern Junggar Basin twice, and the mud volcanism weakened gradually as seen from the variations such as the decrease of gas pressure and output, the downthrow and dry up of the mud pool. The volcanic intensity was significantly weaker than that in Taiwan. The natural gas from the mud volcanoes in the southern Junggar Basin has similar geochemical characteristics, indicating the same source or origin. The main component of the mud volcano gas is alkane gas with contents of 91.15%-97.49%, and the gas is high-quality commercial gas since methane dominates in the alkane. The 13C1 values are 49.1‰--40.6‰, which are in accordance with the peak 13C1 frequency of mud volcano gas around the world, and the alkane gas displays positive carbon isotopic series, i.e., 13C1< 13C2< 13C3, suggesting typical thermogenic origin. The helium in the mud volcano gas is typically crust-derived due to the low R/Ra values of 0.011-0.054. The mud volcano gas is coal-derived since the 13C2 values are all greater than 28‰, and C1/C1-4 and 13C1 values are in accordance with those of natural gas derived from the Lower-Middle Jurassic coal-measures. 3CAS FGOALS-f3-L Model Datasets for CMIP6 Historical Atmospheric Model Intercomparison Project Simulation显示文摘The outputs of the Chinese Academy of Sciences(CAS) Flexible Global Ocean–Atmosphere–Land System(FGOALS-f3-L) model for the baseline experiment of the Atmospheric Model Intercomparison Project simulation in the Diagnostic,Evaluation and Characterization of Klima common experiments of phase 6 of the Coupled Model Intercomparison Project(CMIP6) are described in this paper. The CAS FGOALS-f3-L model, experiment settings, and outputs are all given. In total,there are three ensemble experiments over the period 1979–2014, which are performed with different initial states. The model outputs contain a total of 37 variables and include the required three-hourly mean, six-hourly transient, daily and monthly mean datasets. The baseline performances of the model are validated at different time scales. The preliminary evaluation suggests that the CAS FGOALS-f3-L model can capture the basic patterns of atmospheric circulation and precipitation well, including the propagation of the Madden–Julian Oscillation, activities of tropical cyclones, and the characterization of extreme precipitation. 5Geochemical Characteristics and Genetic Types of Natural Gas in the Xinchang Gas Field, Sichuan Basin, SW China显示文摘The molecular compositions and stable carbon and hydrogen isotopic compositions of natural gas from the Xinchang gas field in the Sichuan Basin were investigated to determine the genetic types. The natural gas is mainly composed of methane (88.99%–98.01%), and the dryness coefficient varies between 0.908 and 0.997. The gas generally displays positive alkane carbon and hydrogen isotopic series. The geochemical characteristics and gas-source correlation indicate that the gases stored in the 5^(th) member of the Upper Triassic Xujiahe Formation are coal-type gases which are derived from source rocks in the stratum itself. The gases reservoired in the 4^(th) member of the Xujiahe Formation and Jurassic strata in the Xinchang gas field are also coal-type gases that are derived from source rocks in the 3^(rd) and 4^(th) members of the Xujiahe Formation. The gases reservoired in the 2^(nd) member of the Upper Triassic Xujiahe Formation are mainly coal-type gases with small amounts of oil-type gas that is derived from source rocks in the stratum itself. This is accompanied by a small amount of contribution brought by source rocks in the Upper Triassic Ma'antang and Xiaotangzi formations. 6The scientific connotation of oil and gas formations under deep fluids and organic-inorganic interaction显示文摘As a relatively stable craton block in the earth system, the petroliferous basin is influenced by the evolution of the earth system from the early development environment of source rocks, hydrocarbon formation, and reservoir dissolution to hydrocarbon accumulation or destruction. As a link between the internal and external factors of the basin, deep fluids run through the whole process of hydrocarbon formation and accumulation through organic-inorganic interaction. The nutrients carried by deep fluids promote the bloom of hydrocarbon-generating organisms and extra addition of carbon and hydrogen source, which are beneficial to the development of high-quality source rock and enhancement of the hydrocarbon generation potential. The energy carried by the deep fluid promotes the early maturation of the source rock and facilitates the hydrocarbon generation by activation and hydrogenation in high-mature hydrocarbon sources. The dissolution alteration of carbonate rocks and clastic reservoirs by CO_2-rich deep fluids improves the deep reservoir space, thus extending the oil and gas reservoir space into greater depth. The extraction of deeply retained crude oil by deep supercritical CO_2 and the displacement of CH_4 in shale have both improved the hydrocarbon fluidity in deep and tight reservoirs. Simultaneously, the energy and material carried by deep fluids(C, H, and catalytic substances) not only induce inorganic CH_4 formation by Fischer-Tropsch(F-T) synthesis and 'hydrothermal petroleum' generation from organic matter by thermal activity but also cause the hydrothermal alteration of crude oil from organic sources. 9Effects of redox potential on soil cadmium solubility:Insight into microbial community显示文摘Understanding the role of microbes in the solubility of cadmium(Cd) is of fundamental importance for remediation of Cd toxicity. The present study aimed to identify the microbes that involved in regulating Cd solubility and to reveal possible mechanisms. Therefore,microbial communities were investigated through high-throughput sequencing approach, the molecular ecological network was constructed and metagenomes were predicted. Our results indicated that redox conditions affected both the solubility of soil Cd and the microbial communities. Anaerobic microbes, such as Anaerolineaceae, did not only play important roles in shaping the microbial community in soils, but might also be involved in regulating the Cd solubility. Two possible mechanisms that how Anaerolineaceae involved in Cd solubility are(1) Anaerolineaceae are important organic matter degraders under anoxic conditions and(2) Anaerolineaceae can co-exist with methane metabolism microbes, while methane metabolism promotes the precipitation of soluble Cd. Thus, application of Anaerolineaceae in bioremediation of soil Cadmium contamination is a potential approach. 10Effects of blocking Le^Y oligosaccharide on cell surface to MMPs secreted by blastocysts and epithelial cells in mouse in vitro显示文摘In order to understand the role of Le+Y oligosaccharide antigen (Le+Y) during implantation, the relationship of Le+Y on the cell surface with matrix metalloproteinase (MMPs) secreted by blastocysts and monolayer epithelial cells during implantation in the mouse %in vitro% was studied by monoclonal antibody (mAb) AH-6, directed to Le+Y[Fuc α1-2 Gal β1-4 (Fuc α1-3) GlcNAc-], and gelatin zymography. The results showed that MMPs secretion was reduced after Le+Y on the cell surface of either epithelial cells or trophoblasts was blocked. It indicated that MMPs expression which played an important function during the process of implantation were regulated by Le+Y. 12Proteasome inhibition suppresses Th17 cell generation and ameliorates autoimmune development in experimental Sjögren’s syndrome显示文摘Immunoproteasome activation in immune cells is involved in the modulation of immune responses.Increasing evidence indicates that proteasome inhibitors show beneficial effects in treating autoimmune diseases,but it remains unclear whether proteasome inhibition is an effective approach for suppressing autoimmune development in Sjögren’s syndrome(SS).Our previous work has demonstrated a critical role for Th17 cells in the development of experimental SS(ESS)in mice.In this study,we detected high levels of low-molecular-weight protein 7(LMP7),a key subunit of the immunoproteasome,in Th17 cells from ESS mice.Moreover,treatment with bortezomib(BTZ),a proteasome inhibitor,markedly suppressed Th17 differentiation in both murine and human naive T cells in culture.Furthermore,ESS mice treated with BTZ displayed significantly higher saliva flow rates and a reduction in tissue destruction in the salivary glands compared with vehicle-treated ESS mice.Notably,BTZ-treated ESS mice showed markedly decreased Th17 cells,germinal center B cells and plasma cells in the peripheral lymphoid organs.In addition,adoptively transferred wild type naive CD4+T cells rapidly differentiated into Th17 cells and induced salivary dysfunction in IL-17-deficient mice immunized for ESS induction.However,BTZ treatment profoundly suppressed the donor T-cell-derived Th17 response and ameliorated the reduction in salivary secretion in IL-17-deficient recipient mice.Taken together,our findings demonstrate that proteasome inhibition can effectively ameliorate ESS by suppressing the Th17 response,which may contribute to the development of a novel therapeutic strategy for the treatment of SS.Fan Xiao Xiang Lin Jie Tian Xiaohui Wang Qian Chen Ke Rui Jie Ma Shengjun Wang Qingwen Wang Xiaoqi Wang Dongzhou Liu Lingyun Sun Liwei Lu 2017Cellular & Molecular Immunology2017,14,11:6
13Controlled growth and photoconductive properties of hexagonal SnS2 nanoflakes with mesa-shaped atomic steps显示文摘We demonstrated the controlled growth of two-dimensional (2D) hexagonal tin disulfide (SnS2) nanoflakes with stacked monolayer atomic steps. The morphology was similar to flat-topped and step-sided mesa plateaus or step pyramids. The SnS2 nanoflakes were grown on mica substrates via an atmospheric-pressure chemical vapor deposition process using tin monosulfide and sulfur powder as precursors. Atomic force microscopy (AFM), electron microscopy, and Raman characterizations were performed to investigate the structural features, and a sequential layer-wise epitaxial growth mechanism was revealed. In addition, systematic Raman characterizations were performed on individual SnS2 nanoflakes with a wide range of thicknesses (1-100 nm), indicating that the A1g peak intensity and Raman shifts were closely related to the thickness of the SnS2 nanoflakes. Moreover, photoconductive AFM was performed on the monolayer-stepped SnS2 nanoflakes, revealing that the flat surface and the edges of the SnS2 atomic steps had different electrical conductive properties and photoconductive behaviors. This is ascribed to the dangling bonds and defects at the atomic step edges, which caused a height difference of the Schottky barriers formed at the interfaces between the PtIr-coated AFM tip and the step edges or the flat surface of the SnS2 nanoflakes. 14NIR-triggered thermo-responsive biodegradable hydrogel with combination of photothermal and thermodynamic therapy for hypoxic tumor显示文摘Hypoxia is a typical feature of solid tumors,which highly limits the application of the oxygen-dependent therapy.Also,the dense and hyperbaric tumor tissues impede the penetration of nanoparticles into the deep tumor.Thereby,we designed a novel localized injectable hydrogel combining the photothermal therapy(PTT)and the thermodynamic therapy(TDT),which is based on the generation of free radicals even in the absence of oxygen for hypoxic tumor therapy.In our study,gold nanorods(Au NRs)and 2,2-Azobis[2-(2-imidazalin-2-yl)propane]dihydrochlaride(AIPH)were incorporated into the hydrogel networks,which were formed by the copolymerization of hydrophobic N-isopropyl acrylamide(NIPAM)and hydrophilic glycidyl methacrylate modified hyaluronic acid(HAGMA)to fabricate an injectable and near-infrared(NIR)responsive hydrogel.The crosslinked in situ forming hydrogel could not only realize PTT upon the NIR laser irradiation,but also generate free radicals even in hypoxic condition.Meanwhile the shrink of hydrogels upon thermal could accelerate the generation of free radicals to further damage the tumors,achieving the controlled drug release on demand.The designed hydrogel with a sufficient loading capacity,excellent biocompatibility and negligible systemic toxicity could serve as a long-acting implant for NIR-triggered thermo-responsive free radical generation.The in vitro cytotoxicity result and the in vivo antitumor activity illustrated the excellent therapeutic effect of hydrogels even in the absence of oxygen.Therefore,this innovative oxygen-independent platform combining the antitumor effects of PTT and TDT would bring a new insight into hypoxic tumor therapy by the application of alkyl free radical.Xiaoqi Sun Di Liu Xiaoyu Xu Yifeng Shen Yanjuan Huang Zishan Zeng Meng Xia Chunshun Zhao 2020Asian Journal of Pharmaceutical Sciences2020,15,6:5
15A static resistance model and the discontinuous pattern of hydrocarbon accumulation in tight oil reservoirs显示文摘In exploration for tight oil, the content and saturation of hydrocarbon in the tight reservoir is a key factor for evaluating the reserve. Therefore, it is necessary to study the geological history of hydrocarbon accumulation and the tight oil charging process. However, kinetic models used for petroleum development are not applicable for petroleum exploration. In this study, a static resistance model is proposed after analyzing resistances in ultra-slow flow in porous media. Using this model, the discontinuous pattern of oil charging is reproduced through incompressible Navier-Stokes equations, the phase field method and the finite element method. This study also explains macroscopic percolation behavior with microscopic flow mechanisms and discusses some issues in ultra-slow flow in a micro/nano pore-throat network. The resistance analysis reveals that capillary resistance and dissipation resistance are dominant factors in the mechanism of oil accumulation in tight reservoirs. Numerical simulations show that pressure thresholds exist and result in discontinuous oil charging. 17Dynamic blood single-cell immune responses in patients with COVID-19显示文摘The 2019 coronavirus disease(COVID-19)outbreak caused by the SARS-CoV-2 virus is an ongoing global health emergency.However,the virus'pathogenesis remains unclear,and there is no cure for the disease.We investigated the dynamic changes of blood immune response in patients with COVID-19 at different stages by using 5'gene expression,T cell receptor(TCR),and B cell receptors(BCR)V(D)J transcriptome analysis at a single-cell resolution.We obtained single-cell mRNA sequencing(scRNA-seq)data of 341,420 peripheral blood mononuclear cells(PBMCs)and 185,430 donotypic T cells and 28,802 donotypic B cells from 25 samples of 16 patients with COVID-19 for dynamic studies.In addition,we used three control samples.We found expansion of dendritic cells(DCs),CD14+monocytes,and megakaryocytes progenitor cells(MP)/platelets and a reduction of naive CD4+T lymphocytes in patients with COVID-19,along with a significant decrease of CD8+T lymphocytes,and natural killer cells(NKs)in patients in critical condition.The type I interferon(IFN-I),mitogen-activated protein kinase(MAPK),and ferroptosis pathways were activated while the disease was active,and recovered gradually after patient conditions improved.Consistent with this finding,the mRNA level of IFN-I signal-induced gene IFI27 was significantly increased in patients with COVID-19 compared with that of the controls in a validation cohort that included 38 patients and 35 controls.The concentration of interferon-a(IFN-a)in the serum of patients with COVID-19 increased significantly compared with that of the controls in an additional cohort of 215 patients with COVID-19 and 106 controls,further suggesting the important role of the IFN-I pathway in the immune response of COVID-19.TCR and BCR sequences analyses indicated that patients with COVID-19 developed specific immune responses against SARS-CoV-2 antigens.Our study reveals a dynamic landscape of human blood immune responses to SARS-CoV-2 infection,providing clues for therapeutic potentials in treating COVID-19.Lulin Huang Yi Shi Bo Gong Li Jiang Zhixin Zhang Xiaoqi Liu Jialiang Yang Yongquan He Zhilin Jiang Ling Zhong Juan Tang Chunfang You Qi Jiang Bo Long Tao Zeng Mei Luo Fanwei Zeng Fanxin Zeng Shuqiang Wang Xingxiang Yang Zhenglin Yang 2021Signal Transduction and Targeted Therapy2021,6,4:4

20Epigenetic regulation of phosphodiesterase 4d in restrictive cardiomyopathy mice with cTnI mutations显示文摘Epigenetic regulations play an important role in disease development.In this study,we have investigated epigenetic regulations in restrictive cardiomyopathy mice with cTnI 193 His mutation.Our results demonstrated that phosphodiesterase(PDEs)4d was down-regulated in the heart of these mice.Further studies showed that the epigenetic modifications were associated with enhanced acetylation of histone 3 lysine 4 and lysines 9,whereas tri-methylation of histone 3 lysine 4,were decreased in histones near the PDE4d gene promoter regions.The binding levels of histone transmethylase SMYD1 and histone deacetylase HDAC1 were increased in the gene promoter regions in cTnI193 His transgenic hearts.Using immune-fluorescent labeling we found an evidence of cTnI existence in the nucleus of cardiomyocytes and Western blotting further confirmed that both wild type and mutated cTnI could be detected in the cell nucleus of the hearts.Furthermore,an interaction between cTnI and SMYD1,or cTnI and HDAC1 was observed.Overexpression of the mutated cTnI in cultured cardiomyocytes reduced the expression of PDE4d.Our data suggest that the decrease of PDE4d expression in RCM mice caused by cTnI mutations may be related to epigenetic regulation,i.e.,histone acetylation and methylation,and cTnI might be involved in this procedure via an interaction with HDAC1 and SMAD1 in the hearts.Weian Zhao Xiaoqi Wu Zhiyuan Wang Bo Pan Lifei Liu Lingjuan Liu Xupei Huang Jie Tian 2020Science China(Life Sciences)2020,63,4:4
