1hsa-miR-29c and hsa-miR-135b differential expression aspotential biomarker of gastric carcinogenesis显示文摘AIM: To investigate the expression profiles of hsa-mi R-29 c and hsa-mi R-135 b in gastric mucosal samples and their values as gastric carcinogenesis biomarkers. METHODS: The expression levels of hsa-mi R-29 c and hsa-mi R-135 b in normal gastric mucosa, non-atrophic chronic gastritis, intestinal metaplasia and intestinaltype gastric adenocarcinoma were analysed using quantitative real-time PCR. The difference between hsa-mi R-29 c and hsa-mi R-135 b expression profiles in the grouped samples was evaluated by ANOVA and Student's t-test tests. The results were adjusted for multiple testing by using Bonferroni's correction. P values ≤ 0.05 were considered statistically significant. To evaluate hsa-mi R-29 c and hsa-mi R-135 b expressions as potential biomarkers of gastric carcinogenesis, we performed a receiver operating characteristic curve analysis and the derived area under the curve, and a Categorical Principal Components Analysis. In silico identification of the genetic targets of hsa-mi R-29 c and hsa-mi R-135 b was performed using different prediction tools, in order to identify possible genes involved in gastric carcinogenesis.RESULTS: The expression levels of hsa-mi R-29 c were higher in normal gastric mucosal samples, and decreased progressively in non-atrophic chronic gastritis samples, intestinal metaplasia samples and intestinal-type gastric adenocarcinoma samples. The expression of hsa-mi R-29 c in the gastric lesions showed that non-atrophic gastritis have an intermediate profile to gastric normal mucosa and intestinal-type gastric adenocarcinoma, and that intestinal metaplasia samples presented an expression pattern similar to that in intestinal-type gastric adenocarcinoma. This micro RNA(mi RNA) has a good discriminatory accuracy between normal gastric samples and(1) intestinal-type gastric adenocarcinoma; and(2) intestinal metaplasia, and regulates the DMNT3 A oncogene. hsa-mi R-135 b is up-regulated in non-atrophic chronic gastritis and intestinal metaplasia samples and down-regulated in normal gastric mucosa and intestinal-type gastric adenocarcinoma samples. Non-atrophic chronic gastritis and intestinal metaplasia are significantly different from normal gastric mucosa samples. hsa-mi R-135 b expression presented a greater discriminatory accuracy between normal samples and gastric lesions. 2Genetic diversity,population structure,and relationships in a collection of pepper(Capsicum spp.)landraces from the Spanish centre of diversity revealed by genotypingby-sequencing(GBS)显示文摘Pepper(Capsicum spp.)is one of the most important vegetable crops;however,pepper genomic studies lag behind those of other important Solanaceae.Here we present the results of a high-throughput genotyping-by-sequencing(GBS)study of a collection of 190 Capsicum spp.accessions,including 183 of five cultivated species(C.annuum,C.chinense,C.frutescens,C.baccatum,and C.pubescens)and seven of the wild form C.annuum var.glabriusculum.Sequencing generated 6,766,231 high-quality read tags,of which 40.7%were successfully aligned to the reference genome.SNP calling yielded 4083 highly informative segregating SNPs.Genetic diversity and relationships of a subset of 148 accessions,of which a complete passport information was available,was studied using principal components analysis(PCA),discriminant analysis of principal components(DAPC),and phylogeny approaches.C.annuum,C.baccatum,and C.chinense were successfully separated by all methods.Our population was divided into seven clusters by DAPC,where C.frutescens accessions were clustered together with C.chinense.C.annuum var.glabriusculum accessions were spread into two distinct genetic pools,while European accessions were admixed and closely related.Separation of accessions was mainly associated to differences in fruit characteristics and origin.Phylogeny studies showed a close relation between Spanish and Mexican accessions,supporting the hypothesis that the first arose from a main genetic flow from the latter.Tajima’s D statistic values were consistent with positive selection in the C.annuum clusters,possibly related to domestication or selection towards traits of interest.This work provides comprehensive and relevant information on the origin and relationships of Spanish landraces and for future association mapping studies in pepper.Leandro Pereira-Dias Santiago Vilanova Ana Fita Jaime Prohens Adrián Rodríguez-Burruezo 2019Horticulture Research2019,6,1:4
3In vitro adherence of conjunctival bacteria to different oculoplastic materials显示文摘AIM: To investigate the resistance to bacterial adhesion of materials used in oculoplastic surgery, particularly materials used in the manufacture of orbital implants.METHODS: Seven organisms of conjunctival flora(two strains of Staphylococcus epidermidis and one strain each of Staphylococcus aureus, Staphylococcus hominis, Corynebacterium amycolatum, Acinetobacter calcoaceticus, and Serratia marcescens) were selected. A lactic acid bacterium(Lactobacillus rhamnosus) was also included as positive control because of its well-known adhesion ability. Eight materials used to make oculoplastic prostheses were selected(glass, steel, polytetrafluoroethylene, polymethylmethacrylate, silicone from orbital implants, commercial silicone, porous polyethylene, and semismooth polyethylene). Materials surfaces and biofilms developed by strains were observed by scanning electron microscopy. Kinetics of growth and adhesion of bacterial strains were determined by spectrophotometry. Each strain was incubated in contact with plates of the different materials. After growth, attached bacteria were re-suspended and colony-forming units(CFUs) were counted. The number of CFUs per square millimetre of material was statistically analyzed.RESULTS: A mature biofilm was observed in studied strains except Staphylococcus hominis, which simply produced a microcolony. Materials showed a smooth surface on the microbial scale, although steel exhibited 1.0-μm-diameter grooves. Most organisms showed significant differences in adhesion according to the material. There were also significant differences in thetotal number of CFUs per square millimetre from each material(P=0.044). 4Discrimination of ventricular tachycardia and localization of its exit site using surface electrocardiography显示文摘Differential diagnosis of supraventricular tachycardia (SVT) and ventricular tachycardia (VT) is of paramount importance for appropriate patient management. Several diagnostic algorithms for discrimination of VT and SVT based on surface electrocardiogram (ECG) analysis have been proposed. Following established diagnosis of VT,a specific origination tachycardia site can be supposed according to QRS complex characteristics.
