| 1 | Multi-modal radiomics model to predict treatment response to neoadjuvant chemotherapy for locally advanced rectal cancer显示文摘BACKGROUND Neoadjuvant chemotherapy is currently recommended as preoperative treatment for locally advanced rectal cancer(LARC);however,evaluation of treatment response to neoadjuvant chemotherapy is still challenging.AIM To create a multi-modal radiomics model to assess therapeutic response after neoadjuvant chemotherapy for LARC.METHODS This retrospective study consecutively included 118 patients with LARC who underwent both computed tomography(CT)and magnetic resonance imaging(MRI)before neoadjuvant chemotherapy between October 2016 and June 2019.Histopathological findings were used as the reference standard for pathological response.Patients were randomly divided into a training set(n=70)and a validation set(n=48).The performance of different models based on CT and MRI,including apparent diffusion coefficient(ADC),dynamic contrast enhanced T1 images(DCE-T1),high resolution T2-weighted imaging(HR-T2WI),and imaging features,was assessed by using the receiver operating characteristic curve analysis.This was demonstrated as area under the curve(AUC)and accuracy(ACC).Calibration plots with Hosmer-Lemeshow tests were used to investigate the agreement and performance characteristics of the nomogram.RESULTS Eighty out of 118 patients(68%)achieved a pathological response.For an individual radiomics model,HR-T2WI performed better(AUC=0.859,ACC=0.896)than CT(AUC=0.766,ACC=0.792),DCE-T1(AUC=0.812,ACC=0.854),and ADC(AUC=0.828,ACC=0.833)in the validation set.The imaging performance for extramural venous invasion detection was relatively low in both the training(AUC=0.73,ACC=0.714)and validation(AUC=0.578,ACC=0.583)sets.The multi-modal radiomics model reached an AUC of 0.925 and ACC of 0.886 in the training set,and an AUC of 0.93 and ACC of 0.875 in the validation set.For the clinical radiomics nomogram,good agreement was found between the nomogram prediction and actual observation.CONCLUSION A multi-modal nomogram using traditional imaging features and radiomics of preoperative CT and MRI adds accuracy to the prediction of treatment outcome,and thus contributes to the personalized selection of neoadjuvant chemotherapy for LARC. | Zheng-Yan Li Xiao-Dong Wang Mou Li Xi-Jiao Liu Zheng Ye Bin Song Fang Yuan Yuan Yuan Chun-Chao Xia Xin Zhang Qian Li | 2020 | World Journal of Gastroenterology2020,26,19: | 7 |

| 4 | Grain refinement and macrosegregation behavior of direct chill cast Al-Zn-Mg-Cu alloy under combined electromagnetic fields显示文摘The combined electromagnetic fields were achieved by the application of an alternating magnetic field and a stationary magnetic field and were used during direct chill(DC) casting process to control the microstructure and macrosegregation of an Al-Zn-Mg-Cu alloy. Ingot microstructures were analyzed under an optical microscope(Leica DMR). The composition at different locations in the ingots was measured with inductively coupled plasma mass spectrometry(ICP) method. The results showed that the grain structure is transformed from dendrite to equiaxed structure and significantly refined with the application of combined electromagnetic fields. The uniformity of microstructure is also greatly improved. The combined electromagnetic fields show a significant effect on the distribution of elements. | 8 | Rapid Detection of Rifampin-resistant Clinical Isolates of Mycobacterium tuberculosis by Reverse Dot Blot Hybridization显示文摘Objective A PCR-reverse dot blot hybridization(RDBH) assay was developed for rapid detection of rpo B gene mutations in ‘hot mutation region' of Mycobacterium tuberculosis(M. tuberculosis). Methods 12 oligonucleotide probes based on the wild-type and mutant genotype rpoB sequences of M. tuberculosis were designed to screen the most frequent wild-type and mutant genotypes for diagnosing RIF resistance. 300 M. tuberculosis clinical isolates were detected by RDBH, conventional drug-susceptibility testing(DST) and DNA sequencing to evaluate the RDBH assay. Results The sensitivity and specificity of the RDBH assay were 91.2%(165/181) and 98.3%(117/119), respectively, as compared to DST. When compared with DNA sequencing, the accuracy, positive predictive value(PPV) and negative predictive value(NPV) of the RDBH assay were 97.7%(293/300), 98.2%(164/167), and 97.0%(129/133), respectively. Furthermore, the results indicated that the most common mutations were in codons 531(48.6%), 526(25.4%), 516(8.8%), and 511(6.6%), and the combinative mutation rate was 15(8.3%). One and two strains of insertion and deletion were found among all strains, respectively. | 9 | A simple AuNPs-based colorimetric aptasensor for chlorpyrifos detection显示文摘In this work, a simple gold nanoparticles(Au NPs) based colorimetric biosensor was developed for chlorpyrifos(Chl) detection using an aptamer as the capture probe. The Chl-aptamer with low dissociation constant(Kd) of 58.59 ± 6.08 nmol/L was selected by ss DNA library immobilized systematic evolution of ligands by enrichment(SELEX). In the absence of Chl, the Chl-aptamer acted as the stabilizer for Au NPs in salt solution. In the presence of Chl, the highly specific Chl-aptamer bound with Chl targets immediately,thus a self-aggregation of Au NPs induced by salt was displayed. The fabricated colorimetric aptasensor exhibited an excellent sensitivity for Chl detection with the LOD as low as 14.46 nmol/L. | 15 | High-static-low-dynamic stiffness isolator based on an electromagnetic negative stiffness spring with long linear stroke显示文摘Negative stiffness mechanisms can improve low-frequency vibration isolation performance and have been widely used in the vibration isolation of precision equipment. However, the negative stiffness mechanism usually introduces a nonlinear stiffness,resulting in a nonlinear response and worsening the vibration isolation performance, especially under large amplitude vibration.In this paper, an electromagnetic spring with linear negative stiffness(ESLNS) is proposed, in which the antagonistic ampere forces of the energized coils are used to generate negative stiffness within a long linear stroke. The magnetic field distribution is improved through the design of the magnetic circuit, thereby increasing the stiffness generation efficiency. The stiffness can be adjusted bidirectionally by current within the range of positive and negative stiffness. An electromagnetic stiffness model was established based on the equivalent magnetic circuit method. Experimental measurements verified the accuracy of the model and proved the linearity of the electromagnetic spring. A vibration isolator with high static and low dynamic stiffness(HSLDS) based on the ESLNS is designed and tested. The experimental results prove that the introduction of the ESLNS can effectively expand the isolation frequency band without changing the equilibrium position. Moreover, the vibration isolator with ESLNS does not produce nonlinear response. | 18 | Using endogenous glycogen as relaxation agent for imaging liver metabolism by MRI显示文摘Glycogen plays essential roles in glucose metabolism.Imaging glycogen in the liver,the major glycogen reservoir in the body,may shed new light on many metabolic disorders.^(13)C magnetic resonance spectroscopy(MRS)has become the mainstream method for monitoring glycogen in the body.However,the equipment of special hard-ware to standard clinical magnetic resonance imaging(MRI)scanners limits its clinical applications.Herein,we utilized endogenous glycogen as a T_(2)-based relaxation contrast agent for imaging glycogen metabolism in the liver in vivo.The in vitro results demonstrated that the transverse relaxation rate of glycogen strongly correlates with the concentration,pH,and field strength.Based on the Swift-Connick theory,we characterized the exchange property of glycogen and measured the exchange rate of glycogen as 31,847 Hz at 37°C.Besides,the viscosity and echo spacing showed no apparent effect on the transverse relaxation rate.This unique feature enables vi-sualization of glycogen signaling in vivo through T_(2)-weighted MRI.Two hours-post intraperitoneal injection of glucagon,a clinical drug to promote glycogenolysis and gluconeogenesis,the signal intensity of the mice’s liver increased by 1.8 times from the T_(2)-weighted imaging experiment due to the decomposition of glycogen.This study provides a convenient imaging strategy to non-invasively investigate glycogen metabolism in the liver,which may find clinical applications in metabolic diseases. | Shizhen Chen Mou Jiang Yaping Yuan Baolong Wang Yu Li Lei Zhang Zhong-Xing Jiang Chaohui Ye Xin Zhou | 2023 | Fundamental Research2023,3,4: | 0 |
