| 2 | Characteristics of secondary inorganic aerosol and sulfate species in size-fractionated aerosol particles in Shanghai显示文摘Sulfate, nitrate and ammonium(SNA) are the dominant species in secondary inorganic aerosol, and are considered an important factor in regional haze formation. Size-fractionated aerosol particles for a whole year were collected to study the size distribution of SNA as well as their chemical species in Shanghai. SNA mainly accumulated in fine particles and the highest average ratio of SNA to particulate matter(PM) was observed to be 47% in the fine size fraction(0.49–0.95 μm). Higher sulfur oxidation ratio and nitrogen oxidation ratio values were observed in PM of fine size less than 0.95 μm. Ion balance calculations indicated that more secondary sulfate and nitrate would be generated in PM of fine size(0.49–0.95 μm). Sulfur K-edge X-ray absorption near-edge structure(XANES) spectra of typical samples were analyzed. Results revealed that sulfur mainly existed as sulfate with a proportion(atomic basis) more than 73% in all size of PM and even higher at 90% in fine particles. Sulfate mainly existed as(NH4)2SO4 and gypsum in PM of Shanghai. Compared to non-haze days, a dramatic increase of(NH4)2SO4 content was found in fine particles on haze days only, which suggested the promoting impact of(NH4)2SO4 on haze formation. According to the result of air mass backward trajectory analysis, more(NH4)2SO4 would be generated during the periods of air mass stagnation. | 3 | Ectopic expression of VRT-A2 underlies the origin of Triticum polonicum and Triticum petropavlovskyi with long outer glumes and grains显示文摘Polish wheat (Triticum polonicum) is a unique tetraploid wheat species characterized by an elongated outer glume. The genetic control of the long-glume trait by a single semi-dominant locus, P1 (from Polish wheat), was established more than 100 years ago, but the underlying causal gene and molecular nature remain elusive. Here, we report the isolation of VRT-A2, encoding an SVP-clade MADS-box transcription factor, as the P1 candidate gene. Genetic evidence suggests that in T. polonicum, a naturally occurring sequence rearrangement in the intron-1 region of VRT-A2 leads to ectopic expression of VRT-A2 in floral organs where the long-glume phenotype appears. Interestingly, we found that the intron-1 region is a key ON/OFF molecular switch for VRT-A2 expression, not only because it recruits transcriptional repressors, but also because it confers intron-mediated transcriptional enhancement. Genotypic analyses using wheat accessions indicated that the P1 locus is likely derived from a single natural mutation in tetraploid wheat, which was subsequently inherited by hexaploid T. petropavlovskyi. | 4 | Effects of PP4 suppression on the proliferation of MCF7 cells显示文摘PP4, one of the few protein phosphata- ses associated with centrosome in cells of many species such as Drosophila, C. elegans and mam- mals, plays an essential role in the regulation of cen- trosome functions in Drosophila and C. elegans. In order to explore the role of PP4 in mammalian cells, full-length PP4 gene was obtained by RT-PCR from MCF7 cell total RNA and inserted into eukaryotic expression vector pEGFP-C1. The resultant con- struct pEGFP-C1-PP4 was transfected into MCF7 cells and immunostaining was carried out to confirm the centrosome localization of PP4. Then we re- versely subcloned a non-conserved domain of PP4 into pXJ41 to construct an anti-sense vector pXJ41- as-PP4. By transfecting pXJ41-as-PP4 into MCF7 cells and screening with G418, we obtained a stable cell line in which PP4 expression was stably sup- pressed. The cell line was analyzed on cell mor- phology, cytoskeleton structure, growth characteris- tics and the mitosis process. It was found that the proliferation rate decreased and serum-dependence increased in PP4-suppressed cells. Furthermore, flow cytometry and mitotic index analysis showed that G2/M transition was prolonged. PP4 suppression resulted in abnormal interphase microtubule, forma- tion of multipolar spindles and an increase in per- centage of multinuclear cells. | 12 | Potential existence of two independent centrosome-targeting domains in PP4显示文摘Protein phosphatase 4 (PP4) is an important member in the PPP family of protein Ser/Thr phosphatases. It has been proven to regulate a variety of cellular processes such as centrosome maturation, micro- tubule nucleation, splicesome assembly, and JNK pathway activation. Compared to the crystallized and structurally well defined phosphatase PP1 and PP2B, little is known about the structure of PP4. Besides the conserved motifs characteristic of the PPP family, no information is available on the other domains of PP4. PP4 is reported to localize to the centrosome in many species such as Drosophila, Caenor- habditis elegans and mammalian cells, which suggests a conserved role of PP4 in the regulation of centrosome function. Unlike several other centrosomal proteins, no sequence has been identified for PP4 that can target it to specific centrosomal localization. In this study, we used a combination of PCR mutagenesis and transient expression of GFP-tagged proteins in mammalian cells, and identified two PP4 centrosome-targeting domains of 68―136 and 134―220 aa. These two domains may be associated for appropriate localization to the centrosome.
