| 1 | IL-10-producing regulatory B cells restrain the T follicular helper cell response in primary Sjögren’s syndrome显示文摘Increased numbers of T follicular helper(Tfh)cells have been implicated in the development of autoimmune diseases including primary Sjögren’s syndrome(pSS),but how the Tfh cell response is regulated during autoimmune pathogenesis remains largely unclear.Here,we first found negative correlations between IL-10^(+)regulatory B(Breg)cell numbers and Tfh cell responses and disease activity in patients with pSS and mice with experimental Sjögren’s syndrome(ESS).Moreover,we detected high expression of IL-10 receptor on Tfh cells and their precursors in both humans and mice.In culture,IL-10 suppressed human and murine Tfh cell differentiation by promoting STAT5 phosphorylation.By using an adoptive transfer approach and two-photon live imaging,we found significantly increased numbers of Tfh cells with enhanced T cell homing into B cell follicles in the draining cervical lymph nodes of RAG-2−/−mice transferred with IL-10-deficient B cells during ESS development compared with those of RAG-2−/−mice transferred with wild-type B cells.In ESS mice,CD19^(+)CD1d^(hi)CD5^(+)Breg cells with decreased IL-10 production exhibited severely impaired suppressive effects on T cell proliferation.Consistently,CD19^(+)CD24^(+)CD38^(hi) Breg cells from pSS patients showed significantly reduced IL-10 production with defective inhibitory function in the suppression of autologous Tfh cell expansion.Furthermore,the adoptive transfer of IL-10-producing Breg cells markedly suppressed the Tfh cell response and ameliorated ESS progression in ESS mice.Together,these findings demonstrate a critical role for IL-10-producing Breg cells in restraining the effector Tfh cell response during pSS development. | Xiang Lin Xiaohui Wang Fan Xiao Kongyang Ma Lixiong Liu Xiaoqi Wang Dong Xu Fei Wang Xiaofei Shi Dongzhou Liu Yan Zhao Liwei Lu | 2019 | Cellular & Molecular Immunology2019,16,12: | 13 |
| 2 | Preparation of mesoporous silica microspheres with multi-hollow cores and their application in sustained drug release显示文摘Mesoporous silica microspheres with multi-hollow cores(MSMMCs)were prepared by an O1/W/O2 double emulsion technique.When tetraethyl orthosilicate(TEOS)was hydrolyzed in the O1/W/O2 double emulsion with P123(a triblock copolymer with a formula of HO(CH2CH2O)20(CH2CH(CH3)O)70(CH2CH2O)20H)dissolved in the aqueous phase,the internal oil droplets would serve as templates for the empty hollow cores,P123 function as templates for the mesopores in the shell,and the aqueous phase act as space-limiting micro-reactors for the hydrolysis process,thus forming MSMMCs with controllable size and hollow cores.After further modification by a high-temperature aging process,the prepared MSMMCs had a diameter of 2-10 m,with macro-multi-hollow cores with a size of 100-1000 nm,mesopores in the shell with a size of 3.8-4.4 nm,and a surface area of 383-735 m2/g.In addition,MSMMCs were used as carriers to load avermectin through an immersing and evaporating process,and the loaded avermectin showed a well sustained release behavior from the MSMMCs matrix.This study demonstrated a promising and simple method for preparing silica micro-particles with porous multi-hollow core structure for sustained release applications. | Chen Zhang Tian Hou Jianfeng Chen Lixiong Wen | 2010 | Particuology2010,8,5: | 7 |
| 3 | IL-17 sustains the plasma cell response via p38-mediated Bcl-xL RNA stability in lupus pathogenesis显示文摘Recent studies have demonstrated a central role for plasma cells in the development of autoimmune diseases,such as systemic lupus erythematosus(SLE).Currently,both the phenotypic features and functional regulation of autoreactive plasma cells during SLE pathogenesis remain largely unclear.In this study,we first found that a major subset of IL-17 receptor-expressing plasma cells potently produced anti-dsDNA IgG upon IL-17A(IL-17)stimulation in SLE patients and lupus mice.Using a humanized lupus mouse model,we showed that the transfer of Th17 cell-depleted PBMCs from lupus patients resulted in a significantly reduced plasma cell response and attenuated renal damage in recipient mice compared to the transfer of total SLE PBMCs.Moreover,long-term BrdU incorporation in lupus mice detected highly enriched long-lived BrdU+subsets among IL-17 receptor-expressing plasma cells.Lupus mice deficient in IL-17 or IL-17 receptor C(IL-17RC)exhibited a diminished plasma cell response and reduced autoantibody production with attenuated renal damage,while the adoptive transfer of Th17 cells triggered the plasma cell response and renal damage in IL-17-deficient lupus mice.In reconstituted chimeric mice,IL-17RC deficiency resulted in severely impaired plasma cell generation but showed no obvious effect on germinal center B cells.Further mechanistic studies revealed that IL-17 significantly promoted plasma cell survival via p38-mediated Bcl-xL transcript stabilization.Together,our findings identified a novel function of IL-17 in enhancing plasma cell survival for autoantibody production in lupus pathogenesis,which may provide new therapeutic strategies for the treatment of SLE. | Kongyang Ma Wenhan Du Fan Xiao Man Han Enyu Huang Na Peng Yuan Tang Chong Deng Lixiong Liu Yulan Chen Jingjing Li Shiwen Yuan Qin Huang Xiaoping Hong Dajun Hu Xiaoyan Cai Quan Jiang Dongzhou Liu Liwei Lu | 2021 | Cellular & Molecular Immunology2021,18,7: | 6 |
| 4 | Numerical and experimental investigations of liquid mixing in two-stage micro-impinging stream reactors显示文摘A two-stage micro-impinging stream reactor(TS-MISR) that combined a first pre-mixing stage with a second micro-impinging stream reacting stage for continuous multi-component reacting systems has been built from commercial T-junctions and steel micro-capillaries. Both of operating parameters and reactor configurations,such as jet Reynolds number(Rej), volumetric flow ratio(R), the first-stage junction angle(φ), the connecting capillary length(Lc) and connecting capillary diameter(dc), had significant effects on the micromixing efficiency of the reactor. Such effects were investigated for both of the two stage structures, respectively, by experimental and CFD methods and were optimized for the best micromixing performance. | 5 | Enhanced permeation performance of polyether-polyamide block copolymer membranes through incorporating ZIF-8 nanocrystals显示文摘Membrane-based CO_2 separation is a promising alternative in terms of energy and environmental issues to other conventional techniques. Polyether-polyamide block copolymer(Pebax) membranes are promising for CO_2 separation because of their excellent selectivity, but limited by their moderate gas permeability. In this study,fresh-prepared zeolitic imidazolate framework-8(ZIF-8) nanocrystals were integrated into the Pebax?1657matrices to form mixed matrix membranes. The resulting membrane exhibits significantly improved CO_2permeability(as high as 300% increase), without the sacrifice of the selectivity, to the pristine polymer membrane. Several physical characterization techniques were employed to confirm the good interfacial interaction between ZIF-8 fillers and Pebax matrices. The effect of added ZIF-8 fillers on the transport mechanism through MMMs is also explored. Mixed-gas permeation for both CO_2/N_2 and CO_2/CH_4 was also evaluated. | 6 | Two-layer networked learning control using self-learning fuzzy control algorithms显示文摘Since the existing single-layer networked control systems have some inherent limitations and cannot effectively handle the problems associated with unreliable networks, a novel two-layer networked learning control system (NLCS) is proposed in this paper. Its lower layer has a number of local controllers that are operated independently, and its upper layer has a learning agent that communicates with the independent local controllers in the lower layer. To implement such a system, a packet-discard strategy is firstly developed to deal with network-induced delay and data packet loss. A cubic spline interpolator is then employed to compensate the lost data. Finally, the output of the learning agent based on a novel radial basis function neural network (RBFNN) is used to update the parameters of fuzzy controllers. | 8 | TiO_2-modified nano-egg-shell Pd catalyst for selective hydrogenation of acetylene显示文摘Pd-based egg-shell nano-catalysts were prepared using porous hollow silica nanoparticles (PHSNs) as support, and the as-prepared catalysts were modified with TiO 2 to promote their selectivity for hydro-genation of acetylene. Pd nanoparticles were loaded evenly on PHSNs and TiO 2 was loaded on the active Pd particles. The effects of reduction time and temperature and the amount of TiO 2 added on catalytic performances were investigated by using a fixed-bed micro-reactor. It was found that the catalysts showed better performance when reduced at 300°C than at 500 °C, and if reduced for 1 h than 3 h. | 9 | Integrated Building Envelope Design Process Combining Parametric Modelling and Multi-Objective Optimization显示文摘As an important element in sustainable building design, the building envelope has been witnessing a constant shift in the design approach. Integrating multi-objective optimization(MOO) into the building envelope design process is very promising, but not easy to realize in an actual project due to several factors, including the complexity of optimization model construction, lack of a dynamic-visualization capacity in the simulation tools and consideration of how to match the optimization with the actual design process. To overcome these difficulties, this study constructed an integrated building envelope design process(IBEDP) based on parametric modelling, which was implemented using Grasshopper platform and interfaces to control the simulation software and optimization algorithm. A railway station was selected as a case study for applying the proposed IBEDP, which also utilized a grid-based variable design approach to achieve flexible optimum fenestrations. To facilitate the stepwise design process, a novel strategy was proposed with a two-step optimization, which optimized various categories of variables separately. | 10 | CFD and experimental investigations on the micromixing performance of single countercurrent-flow microchannel reactor显示文摘Microchannel reactors are widely used in different fields due to their intensive micromixing and, thus, high masstransfer efficiency. In this work, a single countercurrent-flow microchannel reactor(S-CFMCR) at the size of ~1 mm was developed by steel micro-capillary and laser drilling technology. Utilizing the Villermaux/Dushman parallel competing reaction, numerical and experimental studies were carried out to investigate the micromixing performance(expressed as the segregation index XS) of liquids inside S-CFMCR at the low flow velocity regime.The effects of various operating conditions and design parameters of S-CFMCR, e.g., inlet Reynolds number(Re),volumetric flow ratio(R), inlet diameter(d) and outlet length(L), on the quality of micromixing were studied qualitatively. It was found that the micromixing efficiency was enhanced with increasing Re, but weakened with the increase of R. Moreover, d and L also have a significant influence on micromixing. CFD results were in good agreement with experimental data. In addition, the visualization of velocity magnitude, turbulent kinetic energy and concentration distributions of various ions inside S-CFMCR was illustrated as well. | 11 | Changes of gut microbiome and metabolome in the AOM/DSS mouse model of colorectal cancer with FLASH radiation显示文摘Objective:To investigate the differences in small intestinal toxicity and taxonomic composition,diversity,and functional pathways of gut microbiome and metabolome after different radiotherapies in mouse colorectal cancer(CRC)model.Methods:Azoxymethane/dextran sodium sulfate(AOM/DSS)-induced mouse CRC model was treated with single pulse FLASH-RT(dose rate 100 Gy/s)or CONV-RT(dose rate 2 Gy/min)at whole abdomen.At 12 d after radiotherapy,sections of small intestinal tract tissue were dissected for hematoxylin and eosin(HE)staining and the fresh feces were collected for 16S ribosomal RNA(rRNA)microbiome sequencing and liquid chromatography and mass spectrometry(LC-MS)metabolomics sequencing to assess changes in the gut microbiota and metabolites.Microbial high-throughput 16S rRNA data was analyzed with QIIME2 and LEfSe softwares.ProteoWizard,XCMS and Ropls softwares were used for LC-MS analysis.Results:HE staining showed that FLASH-RT maintained small intestinal integrity and reduced the radiotherapyinduced injury.Sequencing analysis of gut fecal microbiome showed that phylum Bacteroidetes and genera Prevotella and Lactobacillus of microbial community were increased after FLASH-RT.Metabolomics sequencing analysis revealed that the metabolites after FLASH-RT were enriched in amino acid metabolism,while cholesterol metabolism was top enriched after CONV-RT.Conclusions:FLASH-RT significantly mitigates the small intestine tissue damage compared with CONV-RT.FLASHRT and CONV-RT have different impact on gut microbiota and its metabolites.Our results provide a theoretical basis for the early evaluation,prediction and individualized treatment of the irradiation effect after novel FLASHRT on tumors through the evaluation of intestinal microbiota and metabolites. | Mengmeng Xu Xinyu Qiu Qiu Chen Tianyu Yang Jingze Xu Liang Chen Lixiong Shuai Zhiming Xu Xinyang Cheng Yongsheng Zhang Zhifei Cao | 2023 | Radiation Medicine and Protection2023,4,1: | 2 |

| 13 | Combined treatment using acupuncture and music therapy on children with cerebral palsy Gross motor function measure comparison in 60 cases显示文摘BACKGROUND: The use of acupuncture has received recognition to effectively treat cerebral palsy. Moreover, music therapy can be used to modify treatment of cerebral palsy. OBJECTIVE: To study the effects of combined treatment using acupuncture and music therapy on gross motor function measure (GMFM) of children with cerebral palsy, compared with acupuncture treatment alone. DESIGN, TIME AND SETTING: Randomized, controlled, clinical study. The experiment was conducted in Shenzhen Hospital of Traditional Chinese Medicine between January 2007 and September 2007. PARTICIPANTS: All children with cerebral palsy in the trial were from the outpatient department of Shenzhen Hospital of Traditional Chinese Medicine. The children were randomly divided into two groups: 30 children in Group B received acupuncture and music therapy, while 30 children in Group A received only acupuncture therapy. METHODS: Subjects in Groups A and B received acupuncture based on syndrome differentiation. The main acupoints were necessary for all participants. At first, flash needling was applied to the acupoints. For the remaining acupoints, the technique of transverse needling was applied to the head acupoints, and perpendicular needling was used for the other points. The inserted needles were twirled and then maintained for 30 minutes. The needle was twirled for one second every other 10 minutes, without reinforcing-reducing techniques. The therapy was performed every other day. The trial consisted of three periods each, and lasted for 84 days. Subjects in Group B received music therapy. They listened to music that they preferred while acupuncture was being performed. Following acupuncture, they were allowed to perform musical activities, such as percussion, singing, and dancing. The music therapy was scheduled for one hour, including listening to music for 30 minutes and music activities for 30 minutes. MAIN OUTCOME MEASURES: The comprehensive functional evaluation scale of cerebral palsy and the GMFM were applied to test symptoms before and after respective treatment. RESULTS: All 60 children were included in final analysis. The general function of all children with cerebral palsy improved, but the general efficiency in Group B was obviously higher than in Group A (93% and 73%, χ2 = 4.32, P < 0.05). GMFM evaluation showed that the function of crawling and kneeling, standing, and walking improved more significantly in Group B than in Group A (P < 0.05), while the function of lying down, rolling, and sitting remained similar between the two groups (P > 0.05). CONCLUSION: The combination of acupuncture and music therapy on the treatment of cerebral palsy is more effective than acupuncture alone.
