1Targeting PFKL with penfluridol inhibits glycolysis and suppresses esophageal cancer tumorigenesis in an AMPK/FOXO3a/BIM-dependent manner显示文摘As one of the hallmarks of cancer,metabolic reprogramming leads to cancer progression,and targeting glycolytic enzymes could be useful strategies for cancer therapy.By screening a small molecule library consisting of 1320 FDA-approved drugs,we found that penfluridol,an antipsychotic drug used to treat schizophrenia,could inhibit glycolysis and induce apoptosis in esophageal squamous cell carcinoma(ESCC).Gene profiling and Ingenuity Pathway Analysis suggested the important role of AMPK in action mechanism of penfluridol.By using drug affinity responsive target stability(DARTS)technology and proteomics,we identified phosphofructokinase,liver type(PFKL),a key enzyme in glycolysis,as a direct target of penfluridol.Penfluridol could not exhibit its anticancer property in PFKL-deficient cancer cells,illustrating that PFKL is essential for the bioactivity of penfluridol.High PFKL expression is correlated with advanced stages and poor survival of ESCC patients,and silencing of PFKL significantly suppressed tumor growth.Mechanistically,direct binding of penfluridol and PFKL inhibits glucose consumption,lactate and ATP production,leads to nuclear translocation of FOXO3a and subsequent transcriptional activation of BIM in an AMPK-dependent manner.Taken together,PFKL is a potential prognostic biomarker and therapeutic target in ESCC,and penfluridol may be a new therapeutic option for management of this lethal disease.Cancan Zheng Xiaomei Yu Yiyao Liang Yidong Zhu Yan He Long Liao Dingkang Wang Yanming Yang Xingfeng Yin Ang Li Qingyu He Bin Li 2022Acta Pharmaceutica Sinica B2022,12,3:4
2Standard pancreatoduodenectomy versus extended pancreatoduodenectomy with modified retroperitoneal nerve resection in patients with pancreatic head cancer:a multicenter randomized controlled trial显示文摘Background:The extent of pancreatoduodenectomy for pancreatic head cancer remains controversial,and more high-level clinical evidence is needed.This study aimed to evaluate the outcome of extended pancreatoduodenectomy(EPD)with retroperitoneal nerve resection in pancreatic head cancer.Methods:This multicenter randomized trial was performed at 6 Chinese highvolume hospitals that enrolled patients between October 3,2012,and September 21,2017.Four hundred patients with stage I or II pancreatic head cancer and without specific pancreatic cancer treatments(preoperative chemotherapy or chemoradiation)within three months were randomly assigned to undergo standard pancreatoduodenectomy(SPD)or EPD,with the latter followed by dissection of additional lymph nodes(LNs),nerves and soft tissues 270◦on the right side surrounding the superior mesenteric artery and celiac axis.The primary endpoint was overall survival(OS)by intention-to-treat(ITT).The secondary endpoints were disease-free survival(DFS),mortality,morbidity,and postoperative pain intensity.Results:TheR1 ratewas slightly lower with EPD(8.46%)thanwith SPD(12.56%).The morbidity and mortality rates were similar between the two groups.The median OS was similar in the EPD and SPD groups by ITT in the whole study cohort(23.0 vs.20.2 months,P=0.100),while the median DFS was superior in the EPD group(16.1 vs.13.2 months,P=0.031).Patients with preoperative CA19–9<200.0 U/mL had significantly improved OS and DFS with EPD(EPD vs.SPD,30.8 vs.20.9 months,P=0.009;23.4 vs.13.5 months,P<0.001).The EPD group exhibited significantly lower locoregional(16.48%vs.35.20%,P<0.001)andmesenteric LNrecurrence rates(3.98%vs.10.06%,P=0.022).The EPD group exhibited less back pain 6 months postoperation than the SPD group.Conclusions:EPD for pancreatic head cancer did not significantly improve OS,but patients with EPD treatment had significantly improved DFS.In the subgroup analysis,improvements in bothOS and DFS in the EPD armwere observed in patients with preoperative CA19–9<200.0 U/mL.EPD could be used as an effective surgical procedure for patients with pancreatic head cancer,especially those with preoperative CA19–9<200.0 U/mL.Qing Lin Shangyou Zheng Xianjun Yu Meifu Chen Yu Zhou Quanbo Zhou Chonghui Hu Jing Gu Zhongdong Xu LinWang Yimin Liu Qingyu Liu MinWang Guolin Li He Cheng Dongkai Zhou Guodong Liu Zhiqiang Fu Yu Long Yixiong Li Weilin Wang Renyi Qin Zhihua Li Rufu Chen 2023Cancer Communications2023,43,2:3
3Vanadium-based metal-organic frameworks and their derivatives for electrochemical energy conversion and storage显示文摘With the excessive consumption of nonrenewable resources,the exploration of effective and durable materials is highly sought after in the field of sustainable energy conversion and storage system.In this aspect,metalorganic frameworks(MOFs)are a new class of crystalline porous organicinorganic hybrid materials.MOFs have recently been gaining traction in energy-related fields.Owing to the coordination flexibility and multiple accessible oxidation states of vanadium ions or clusters,vanadium-MOFs(V-MOFs)possess unique structural characteristics and satisfactory electrochemical properties.Furthermore,V-MOFs-derived materials also exhibit superior electrical conductivity and stability when used as electrocatalysts and electrode materials.This review summarizes the research progress of V-MOFs(inclusive of pristine V-MOFs,V/M-MOFs,and POVbased MOFs)and their derivatives(vanadium oxides,carbon-coated vanadium oxide,vanadium phosphate,vanadate,and other vanadium doped nanomaterials)in electrochemical energy conversion(water splitting,oxygen reduction reaction)and energy storage(supercapacitor,rechargeable battery).Future possibilities and challenges for V-MOFs and their derivatives in terms of design and synthesis are discussed.Lastly,their applications in energy-related fields are also highlighted.Jing Zhu Xiaoyu Chen Ai Qin Thang Fei‐Long Li Dong Chen Hongbo Geng Xianhong Rui Qingyu Yan 2022SmartMat2022,3,3:2
4A survey of real-time rendering on Web3D application显示文摘Background In recent years, with the rapid development of mobile Internet and Web3D technologies, a large number of web-based online 3D visualization applications have emerged. Web3D applications, including Web3D online tourism, Web3D online architecture, Web3D online education environment, Web3D online medical care, and Web3D online shopping are examples of these applications that leverage 3D rendering on the web. These applications have pushed the boundaries of traditional web applications that use text, sound, image, video, and 2D animation as their main communication media, and resorted to 3D virtual scenes as the main interaction object, enabling a user experience that delivers a strong sense of immersion. This paper approached the emerging Web3D applications that generate stronger impacts on people's lives through “real-time rendering technology”, which is the core technology of Web3D. This paper discusses all the major 3D graphics APIs of Web3D and the well-known Web3D engines at home and abroad and classify the real-time rendering frameworks of Web3D applications into different categories. Results Finally, this study analyzed the specific demand posed by different fields to Web3D applications by referring to the representative Web3D applications in each particular field. 5Development of a poly(arylene sulfide sulfone)antibacterial electrospun film as a skin wound dressing application显示文摘Tissue engineering has become a hot issue for skin wound healing because it can be used as an alternative treatment to traditional grafts.Nanofibrous films have been widely used due to their excellent properties.In this work,an organic/inorganic composite poly(arylene sulfide sulfone)/ZnO/graphene oxide(PASS/ZnO/GO)nanofibrous film was fabricated with the ZnO nano-particles blending in an electrospun solution and post-treated with the GO deposition.The optimal PASS/ZnO/GO nanofibrous film was prepared by 2%ZnO nanoparticles,3.0g/mL PASS electrospun solution,and 1%GO dispersion solution.The morphology,hydrophilicity,mechanical property,and cytotoxicity of the PASS/ZnO/GO nanofibrous film were character-ized by using scanning electron microscopy,transmission electron microscope,water contact angle,tensile testing,and a Live/Dead cell staining kit.It is founded that the PASS/ZnO/GO nanofibrous film has outstanding mechanical properties and no cytotoxicity.Furthermore,the PASS/ZnO/GO nanofibrous film exhibits excellent antibacterial activity to both Escherichia coli and Staphylococcus aureus.Above all,this high mechanical property in the non-toxic and antibacterial nanofibrous film will have excellent application prospects in skin wound dressing.Xuzhi Lang Zhurong Tang Zhimei Wei Xiaojun Wang Shengru Long Gang Zhang Jie Yang Qingyu Lin 2022Journal of Innovative Optical Health Sciences2022,15,2:0

8Privacy-Preserving Federated Mobility Prediction with Compound Data and Model Perturbation Mechanism显示文摘Human mobility prediction is important for many applications.However,training an accurate mobility prediction model requires a large scale of human trajectories,where privacy issues become an important problem.The rising federated learning provides us with a promising solution to this problem,which enables mobile devices to collaboratively learn a shared prediction model while keeping all the training data on the device,decoupling the ability to do machine learning from the need to store the data in the cloud.However,existing federated learningbased methods either do not provide privacy guarantees or have vulnerability in terms of privacy leakage.In this paper,we combine the techniques of data perturbation and model perturbation mechanisms and propose a privacy-preserving mobility prediction algorithm,where we add noise to the transmitted model and the raw data collaboratively to protect user privacy and keep the mobility prediction performance.Extensive experimental results show that our proposed method significantly outperforms the existing stateof-the-art mobility prediction method in terms of defensive performance against practical attacks while having comparable mobility prediction performance,demonstrating its effectiveness.Long Qingyue Wang Huandong Chen Huiming Jin Depeng Zhu Lin Yu Li Li Yong 2024China Communications2024,21,3:0
9Simultaneous enhancement of mechanical properties and corrosion resistance of as-cast Mg-5Zn via microstructural modification by friction stir processing显示文摘Magnesium alloys are ideal lightweight materials;however,their applications are extremely limited due to their low strength,poor ductility,and weak corrosion resistance.In the present study,a friction stir processing(FSP)treatment was employed to optimize the mechanical properties and corrosion resistance of an as-cast Mg-5Zn alloy.The average grain size of the Mg-5Zn alloy was refined from 133.8μm to1.3μm as a result of FSP.Along different directions,FSP exhibited the enhancement effects on different mechanical properties.Furthermore,according to the potentiodynamic polarization results,the corrosion current density at the free-corrosion potential of the FSPed sample,was 4.1×10^(-6)A/cm^(2)in 3.5 wt.%Na Cl aqueous solution,which was significantly lower than that of the as-cast sample.Electrochemical impedance spectroscopy revealed that the polarization impedance,Rp,of the FSPed sample was 1534Ω/cm^(2)in 3.5 wt.%NaCl aqueous solution,which was 71.4%greater than that of the as-cast sample.The corrosion morphology of the FSPed sample in 3.5 wt.%NaCl aqueous solution exhibited largely uniform corrosion,rather than severe localized corrosion characteristics,which further reduced the corrosion depth on the basis of reducing the corrosion current density.The results presented herein indicate that FSP is a viable technique for simultaneously improving the mechanical properties and corrosion resistance of the as-cast Mg-5Zn alloy.Fei Long Gaoqiang Chen Mengran Zhou Qingyu Shi Qu Liu 2023Journal of Magnesium and Alloys2023,11,6:0
