| 1 | The histone H3 lysine-27 demethylase Jmjd3 plays a critical role in specific regulation of Th17 cell differentiation显示文摘Interleukin(IL)17-producing T helper(Th17)cells play critical roles in the clearance of extracellular bacteria and fungi as well as the pathogenesis of various autoimmune diseases,such as multiple sclerosis,psoriasis,and ulcerative colitis.Although a global transcriptional regulatory network of Th17 cell differentiation has been mapped recently,the participation of epigenetic modifications in the differentiation process has yet to be elucidated.We demonstrated here that histone H3 lysine-27(H3K27)demethylation,predominantly mediated by the H3K27 demethylase Jmjd3,crucially regulated Th17 cell differentiation.Activation of naı¨ve CD41 T cells immediately induced high expression of Jmjd3.Genetic depletion of Jmjd3 in CD41 T cells specifically impaired Th17 cell differentiation both in vitro and in vivo.Ectopic expression of Jmjd3 largely rescued the impaired differentiation of Th17 cells in vitro in Jmjd3-deficientCD41 T cells.Importantly,Jmjd3-deficient mice were resistant to the induction of experimental autoimmune encephalomyelitis(EAE).Furthermore,inhibition of the H3K27 demethylase activity with the specific inhibitor GSK-J4 dramatically suppressed Th17 cell differentiation in vitro.At the molecular level,Jmjd3 directly bound to and reduced the level of H3K27 trimethylation(me3)at the genomic sites ofRorc,which encodes the masterTh17 transcription factorRorgt,and Th17 cytokine genes such as Il17,Il17f,and Il22.Therefore,our studies established acritical role of Jmjd3-mediatedH3K27demethylation inTh17 cell differentiation andsuggest that Jmjd3 can be a novel therapeutic target for suppressing autoimmune responses. | Zhi Liu Wei Cao Longxia Xu Xi Chen Yu Zhan Qian Yang Sanhong Liu Pengfei Chen Yuhang Jiang Xiaohua Sun Yu Tao Yiming Hu Cuifeng Li Qi Wang Ying Wang Charlie Degui Chen Yufang Shi Xiaoren Zhang | 2015 | Journal of Molecular Cell Biology2015,7,6: | 6 |
| 2 | Uncertainty analysis of hydrological processes based on ARMA-GARCH model显示文摘Uncertainty analysis and risk analysis are two important areas of modern water resource management,in which accurate variance estimation is required.The traditional runoff model is established under the assumption that the variance is a constant or it changes with the seasons.However,hydrological processes in the real world are often heteroscedastic,which can be tested by McLeod-Li test and Engle Lagrange multiplier test.In such cases,the GARCH model of hydrological processes is established in this article.First,the seasonal factors in the sequence are removed.Second,the traditional ARMA model is established.Then,the GARCH model is used to correct the residual.At last,the daily runoff data in 1949-2001 of Yichang Hydrological Station is taken to be an example.The result shows that compared to the traditional ARMA model,the GARCH model has the ability to predict more accurate confidence intervals under the same confidence level. | WANG HongRui GAO Xiong QIAN LongXia YU Song | 2012 | Science China(Technological Sciences)2012,55,8: | 6 |

| 13 | K-Means Clustering with Local Distance Privacy显示文摘With the development of information technology,a mass of data are generated every day.Collecting and analysing these data help service providers improve their services and gain an advantage in the fierce market competition.K-means clustering has been widely used for cluster analysis in real life.However,these analyses are based on users’data,which disclose users’privacy.Local differential privacy has attracted lots of attention recently due to its strong privacy guarantee and has been applied for clustering analysis.However,existing K-means clustering methods with local differential privacy protection cannot get an ideal clustering result due to the large amount of noise introduced to the whole dataset to ensure the privacy guarantee.To solve this problem,we propose a novel method that provides local distance privacy for users who participate in the clustering analysis.Instead of making the users’records in-distinguish from each other in high-dimensional space,we map the user’s record into a one-dimensional distance space and make the records in such a distance space not be distinguished from each other.To be specific,we generate a noisy distance first and then synthesize the high-dimensional data record.We propose a Bounded Laplace Method(BLM)and a Cluster Indistinguishable Method(CIM)to sample such a noisy distance,which satisfies the local differential privacy guarantee and local dE-privacy guarantee,respectively.Furthermore,we introduce a way to generate synthetic data records in high-dimensional space.Our experimental evaluation results show that our methods outperform the traditional methods significantly. | Mengmeng Yang Longxia Huang Chenghua Tang | 2023 | Big Data Mining and Analytics2023,6,4: | 0 |

| 15 | Electrophysiological Characteristics and Radiofrequency Ablation of the Focal Atrial Fibrillation Originating From the Superior Vena Cava(摘要)显示文摘Introduction: The superior vena cava (SVC) has cardiac musculature extending from the right atrium and could be one potential origin of focal atrial fibrillation (AF). Paroxysmal AF originated from the SVC was identified in three patients. Methods and Results: It consisted of 2 female aged 55, 58 and 60 years respectively. Two of them had brady-tachy syndrome and a pacemaker implantation. Daily paroxysmal palpitation due to paroxysmal AF and runs of atrial tachycardia initiated by atrial ectopic beats was observed through 12 leads ECG and Holter recordings in all patients. The morphology of P` wave was positive with a high amplitude in leads II, III, aVF, and negative in aVR. The SVC was mapped with a circumferential 10-electrode catheter. The atrial electrogram was preceded by SVC muscular potential (spike-like) during the ectopic activity and a temporal reversal was exhibited during sinus rhythm. Spontaneous AF initiated by a burst of rapid ectopic beats from the SVC was consistently observed. Ostial radiofrequency ablation was performed at segments characterized by fusion of the atrial electrogram and the SVC muscular potential and SVC electrical disconnection was achieved. During 12, 9 and 1 months’ follow-up, all patients were free from recurrences of AF despite difficult arrhythmia control pre-procedure.Conclusion:Focal AF may originate from SVC with a characteristic morphology of P` wave.
