Toxic tau: structural origins of tau aggregation in Alzheimer’s disease显示文摘Alzheimer’s disease is characterized by the extracellular accumulation of the amyloidβin the form of amyloid plaques and the intracellular deposition of the microtubule-associated protein tau in the form of neurofibrillary tangles. Most of the Alzheimer’s drugs targeting amyloidβhave been failed in clinical trials.Particularly, tau pathology connects greatly in the pathogenesis of Alzheimer’s disease.Tau protein enhances the stabilization of microtubules that leads to the appropriate function of the neuron.Changes in the quantity or the conformation of tau protein could affect its function as a microtubules stabilizer and some of the processes wherein it is involved. The molecular mechanisms leading to the accumulation of tau are principally signified by numerous posttranslational modifications that change its conformation and structural state. Therefore, aberrant phosphorylation,as well as truncation of tau protein, has come into focus as significant mechanisms that make tau protein in a pathological entity.Furthermore, the shape-shifting nature of tau advocates to comprehend the progression of Alzheimer’s disease precisely. In this review,we emphasize the recent studies about the toxic and shape-shifting nature of tau in the pathogenesis of Alzheimer’s disease.Abdullah Al Mamun Md.Sahab Uddin Bijo Mathew Ghulam Md Ashraf 2020Neural Regeneration Research2020,15,8:21
DL-3-n-butylphthalide ameliorates diabetes-associated cognitive decline by enhancing PI3K/Akt signaling and suppressing oxidative stress显示文摘DL3-n-Butylphthalide(DLNBP),a small molecular compound extracted from the seeds of Ap/um graveo/ens Linn(Chinese celery),has been shown to exert neuroprotective effects due to its anti-inflammatory,anti-oxidative and anti-apoptotic activities.DL-NBP not only protects against ischemic cerebral injury, but also ameliorates vascular cognitive impairment in dementia patients including AD and PD.In the current study,we investigated whether and how DL-NBP exerted a neuroprotective effect against diabetes-associated cognitive decline(DACD)in db/db mice,a model of type-2 diabetes,db/db mice were orally administered DL-NBP(20,60,120 mg·kg^(-1)·d^(-1))for 8 weeks. Then the mice were subjected to behavioral test, their brain tissue was collected for morphological and biochemical analyses. We showed that oral administration of DL-NBP significantly ameliorated the cognitive decline with improved learning and memory function in Morris water maze testing.Furthermore,DL-NBP administration attenuated diabetes-induced morphological alterations and increased neuronal survival and restored the levels of synaptic protein PSD95,synaptophysin and synapsin-1 as well as dendritic density in the hippocampus, especially at a dose of 60 mg/kg.Moreover,we revealed that DL-NBP administration suppressed oxidative stress by upregulating Nrf2/HO-1 signaling, and increased brain-derived neurotrophic factor(BDNF)expression by activating PI3K/Akt/CREB signaling in the hippocampus. These beneficial effects of DL-NBP were observed in high glucose-treated PC12 cells. Our results suggest that DL-NBP may be a potential pharmacologic agent for the treatment of DACD.Bei-ni Wang Cheng-biao Wu Zi-miao Chen Pei-pei Zheng Ya-qian Liu Jun Xiong Jing-yu Xu Pei-feng Li Abdullah Al Mamun Li-bing Ye Zhi-long Zheng Yan-qing Wu Jian Xiao Jian Wang 2021Acta Pharmacologica Sinica2021,42,3:11
Urbanization reduces resource use efficiency of phytoplankton community by altering the environment and decreasing biodiversity显示文摘Urbanization often exerts multiple effects on aquatic and terrestrial organisms, including changes in biodiversity, species composition and ecosystem functions. However, the impacts of urbanization on river phytoplankton in subtropical urbanizing watersheds remain largely unknown. Here,we explored the effects of urbanization on phytoplankton community structure(i.e.,biomass,community composition and diversity)and function(i.e.,resource use efficiency)in a subtropical river at watershed scale in southeast China over 6 years.A total of 318 phytoplankton species belonging into 120 genera and 7 phyla were identified from 108 samples.Bacillariophyta biomass showed an increasing trend with increasing urbanization level. The phytoplankton community shifted from Chlorophyta dominance in rural upstream waters to Bacillariophyta dominance in urbanized downstream waters.Furthermore, phytoplankton diversity and resource use efficiency(RUE=phytoplankton biomass/total phosphorus)were significantly decreased with increasing urbanization level from upstream to downstream.Phytoplankton RUE exhibited a significant positive correlation with species richness, but a negative correlation with phytoplankton evenness. The variation in environmental factors(turbidity, total nitrogen,NH_(4)^(+)-N,total phosphorus,PO_(4)^(3-)-P and percentage urbanized area)was significantly correlated with phytoplankton diversity and RUE.Overall, our results revealed the influence of urbanization on phytoplankton community structure and ecosystem function was due to its altering the environmental conditions.Therefore,human-driven urbanization may play crucial roles in shaping the structure and function of phytoplankton communities in subtropical rivers, and the mechanism of this process can provide important information for freshwater sustainable uses, watershed management and conservation.Yigang Yang Huihuang Chen Mamun Abdullah Al Jean Claude Ndayishimiye Jun R Yang Alain Isabwe Anqi Luo Jun Yang 2022Journal of Environmental Sciences2022,34,2:4

Phytochemical analysis and antioxidant profile of methanolic extract of seed, pulp and peel of Baccaurea ramiflora Lour.显示文摘Objective: To analyze the phytochemical constituents responsible for the plausible antioxidant effect of methanolic extract of the seed, pulp and peel of Baccaurea ramiflora Lour. Methods: Fresh seed, pulp, and peel of Baccaurea ramiflora fruits were extracted with methanol(MEBRse, MEBRpu, MEBRpe) and evaluated by phytochemical analysis for their content of innumerable metabolites(primary and secondary) viz. carbohydrates, alkaloids, glycosides, tannins, phenols, terpenoids, flavonoids, proteins, and fixed oils. The antioxidant efficacy was assessed through different assay methods viz. 2,2-diphenyl-1-picrylhydrazyl(DPPH) free radical scavenging activity, total antioxidant capacity(TAC) and reducing power capacity(RPC). Estimation of total phenolic content(TPC), and total flavonoid content(TFC) was also done to confirm the presence of these phytochemicals. Results: It was revealed from the phytochemical analysis of MEBRse that alkaloids, glycosides, carbohydrates, phenols, and flavonoids were present, while that of MEBRpu showed the existence of carbohydrates, proteins, alkaloids, glycosides, phenols, saponins, flavonoids, and fixed oils. Presence of carbohydrates, alkaloids, phenols, tannins, flavonoids, and terpenoids were found in the MEBRpe. A significant antioxidant activity was revealed by the MEBRpu [EC_(50):(27.612 ± 1.375) μg/mL], compared to MEBRpe, and MEBRse in DPPH assay. The ranking order for RPC was MEBRpu > MEBRpe > MEBRse respectively. The EC_(50) value of TAC of the MEBRpu, MEBRpe, and MEBRse were(25.107 ±0.744) μg/mL,(241.127 ± 7.463) μg/mL and(372.364 ± 11.030) μg/mL, respectively. Quantity of TPC and TFC were the highest in the MEBRpu(124.360 ± 2.078 mg GAE/g and 107.527 ±1.900 mg QRE/g extract) rather than MEBRpe and MEBRse extracts. Conclusions: This study suggests that MEBRpu has a significantly higher antioxidant property than MEBRpe and MEBRse. These extracts might be advantageous in prevention or decelerating the progress of different diseases related to oxidative-stress/damage. Moreover, detailed analysis of these extracts is required to identify the presence of promising compound(s) responsible for their antioxidant activity.Md.Sahab Uddin Md.Sarwar Hossain Abdullah Al Mamun Devesh Tewari Md.Asaduzzaman Md.Siddiqul Islam Mohamed M.Abdel-Daim 2018Asian Pacific Journal of Tropical Medicine2018,11,7:3
TBK1-medicated DRP1 phosphorylation orchestrates mitochondrial dynamics and autophagy activation in osteoarthritis显示文摘Mitochondrial dynamics, including mitochondrial fission and fusion, are critical for maintaining mitochondrial functions.Evidence shows that TANK-binding kinase 1(TBK1)regulates mitochondrial fusion and fission and then mitophagy.Since a previous study demonstrates a strong correlation between mitophagy and osteoarthritis(OA),we herein investigated the potential role of TBK1 in OA process and mitochondrial functions. We demonstrated a strong correlation between TBK1 and OA,evidenced by significantly downregulated expression of TBK1 in cartilage tissue samples of OA patients and in the chondrocytes of aged mice,as well as TNF-α-stimulated phosphorylation of TBK1 in primary mouse chondrocytes.TBK1 overexpression significantly attenuated TNF-α-induced apoptosis and abnormal mitochondrial function in primary mouse chondrocytes.Furthermore,TBK1 overexpression induced remodeling of mitochondrial morphology by directly phosphorylating dynamin-related protein 1(DRP1)at Ser637,abolishing the fission of DRP1 and preventing its fragmentation function.Moreover,TBK1 recruitment and DRP1 phosphorylation at Ser637 was necessary for engulfing damaged mitochondria by autophagosomal membranes during mitophagy. Moreover,we demonstrated that APMK/ULK1 signaling contributed to TBK1 activation. In OA mouse models established by surgical destabilization of the medial meniscus, intraarticular injection of lentivirus-TBK1 significantly ameliorated cartilage degradation via regulation of autophagy and alleviation of cell apoptosis. In conclusion, our results suggest that the TBK1/DRP1 pathway is involved in OA and pharmacological targeting of the TBK1-DRP1 cascade provides prospective therapeutic benefits for the treatment of OA.Sun-li Hu Abdullah Al Mamun Jian Shaw Sun-long Li Yi-feng Shi Xue-man Jin Ying-xin Yu Chao-zhi Pang Ze-yang Li Jia-jie Lu Yue-piao Cai Xiang-yang Wang Jian Xiao 2023Acta Pharmacologica Sinica2023,44,3:2
Community ecology and spatial distribution of trees in a tropical wet evergreen forest in Kaptai national park in Chittagong Hill Tracts, Bangladesh显示文摘We investigated the floristic composition, woody species diversity and spatial distribution of trees in a tropical wet evergreen forest in Kaptai national park in Chittagong Hill Tracks, Bangladesh. We recorded 25 families, 37 genera, 40 species and 1771 woody individuals in a 0.09 ha plot. Euphorbiaceae and Moraceae were the most speciesrich families, and Castanopsis, Ficus and Terminalia were the most species-rich genera. Bursera serrata Wall. ex Colebr. was the dominant species in terms of highest importance value(13%). Trema orientalis(L.)Bl was typically a light demanding species as it appeared in the top canopy with only one individual having the seventh highest IV, but had no regeneration. The expected maximum number of species(Smax) was 140,indicating that many species may invade the forest as the Smax is greater than the recorded total number of species. The nature of the disappearance and appearance of species in the present forest reflects instability of floristic composition. The values of Shannon's index H ′ and Pielou's index J ′(evenness) were 3.36 bit and 0.63, respectively. These values show moderately high species diversity as compared to other subtropical forests in the tropics. In addition, a sample area of 200 m2 in this forest would be sufficient for measuring the diversity indices H ′ and J ′,whereas the trend of J ′may indicate the rate of equality of individuals among the different species decreased with increasing area. The distribution pattern for the total stand was completely random. However, the dominant species showed aggregate distribution for small areas, but random distribution for large areas. The spatial association between species showed that the strongest positive interspecific association occurred between Streblus asper Lour. and Castanea indica Roxb.(ω =0.51). As a whole, most species were weakly associated with each other,of which 58% species associations were completely negative. The result of cluster analysis showed that species pairs were spatially independent at all or most small clusters; stands of species from all clusters are mosaics of complete habitat and pioneer habitat. All patches in this forest community have similar habitat and regeneration niches, which could be a phenomenon for a young growth forest. Thus, the existence of habitat and regeneration niches may be an important factor in the maintenance of diversity in this forest.S. M. 8Analysis and modeling of hysteresis of piezoelectric micro-actuator used in high precision dual-stage servo system显示文摘A dual-stage servo system consists of a primary coarse actuator for facilitating large motion and a secondary micro-actuator for small but precise motion to improve tracking performance. Piezoelectric micro-actuator made from lead zirconate titanate(PZT)has been a popular choice for the secondary stage. However, the advantage gained by the resolution of the secondary PZT actuator is reduced by its inherent hysteresis nonlinearity. Model based hysteresis compensation techniques are preferred due to their simplicity and fast response. Identification and modeling are two substantial parts in such model-based techniques. This paper presents a rigorous analysis and modeling of the hysteresis of PZT micro-actuator. Modified Generalized Prandtl-Ishlinskii and Coleman-Hodgdon models are studied. Identification of the model through nonlinear least square and particle swarm optimization are examined and compared. Several analyses are done through tuning of the model parameters and identification techniques.Experimental analysis and simulation results underscore the effectiveness of this modeling approach. Finally as a design example,a dual-stage simulation analysis is done to show the effectiveness of systematic modeling on hysteresis compensation.Md.Arifur RAHMAN Abdullah Al MAMUN Kui YAO 2015Control Theory and Technology2015,13,2:1
Production of Biochar for Soil Application:A Comparative Study of Three Kiln Models显示文摘Biochar has potentials for soil fertility improvement, climate change mitigation and environmental reclamation, and charred biomass can be deliberately incorporated into soil for long-term carbon stabilization and soil amendment. Many different methods have been used for biochar production ranging from laboratory to industrial scales. However, in countryside of developing countries, biomass is generally used for cooking but not charred. Biochar production techniques at farmer scale have remained poorly developed. We developed and tested biochar production kilns for farmers with a dimension of 50.8 cm × 38.1 cm(height × diameter), using three different setups for optimizing oxygen(O2) limitation and syngas circulation: airtight with no syngas circulation(Model I),semi-airtight with external syngas circulation(Model II) and semi-airtight with internal syngas circulation(Model III). A comparative assessment of these biochar production kiln models was made considering biochar pyrolysis time, fuel to biomass ratio, biochar to feedstock ratio and thermogravimetric index(TGI). Among the models, the best quality biochar(TGI = 0.15) was obtained from Model I kiln taking the longest time for pyrolysis(12.5 h) and the highest amount of fuel wood(1.22 kg kg-1biomass). Model III kiln produced comparatively good quality biochar(TGI = 0.11), but with less fuel wood requirement(0.33 kg kg-1biomass) and shorter pyrolysis time(8.5 h). We also tested Model III kiln in a three times larger size under two situations(steel kiln and pit kiln). The biochar to feedstock ratio(0.38) and quality(TGI = 0.14) increased slightly for the larger kilns. Quality of biochar was found to be mainly related to pyrolysis time. The costs for the biochar stove and pit kiln were US$ 65–77, while it was US$ 154 for the large size steel kiln. Model III kiln can potentially be used for both cooking and biochar production at farmer scale.Shamim MIA Nijam UDDIN Shaikh Abdullah Al MAMUN HOSSAIN Ruhul AMIN Fatima Z.METE Tjisse HIEMSTRA 2015Pedosphere2015,25,5:1
Chloride ingress and macro-cell corrosion of steel in concrete made with recycled brick aggregate显示文摘An investigation on chloride ingress and macro-cell corrosion of steel bars in concrete made with recycled brick aggregate(RBA)was carried out.As control cases, virgin brick aggregate(BA)and stone aggregate(SA)were also investigated. Both cylindrical and cracked prism specimens were studied for 16 different cases. The prism specimens were made with a segmented steel bar providing electrical connection from outside of the specimens to measure macro-cell corrosion current continuously under seawater splash exposure for a period of 30 d using a data logger.Cylindrical specimens were submerged in 3%NaCl solution at a temperature of 40℃ to investigate chloride ingress in concrete made with RBA,BA,and SA after 120 and 180 d.Half-cell potential, corrosion area, and depths of corrosion were also investigated. The chloride ingress as well as corrosion of steel bars in concrete made with the different types of aggregate is ordered as RBA>BA>SA.Tarek Uddin MOHAMMED Mahfuzur RAHMAN Ahmed SABBIR Mohammad Mehedi HASAN Abdullah Al MAMUN 2021Frontiers of Structural and Civil Engineering2021,15,6:0
In vivo analysis of toxic effect of hydrose used in food preparations in Bangladesh显示文摘Objective:To evaluate the toxic effect of hydrose used in the molasses preparation in Bangladesh. Methods:Molasses were collected from open markets in different parts of Bangladesh. The presence of hydrose in selected molasses was detected using commercial kit.To evaluate the toxic effect of hydrose.Swiss albino male mice were divided into four groups.Group Ⅰ was used as control, while Groups Ⅱ,Ⅲ and Ⅳ received hydrose mixing food(5.10 and 25 g/kg food),respectively, and these supplementations were continued to the end of the study(16weeks).Blood was collected from thoracic arteries of the mice under ether anesthesia and then organs were taken. To determine the effect of hydrose on host, blood indices related to liver, heart and kidney dysfunctions were measured.Result:Creatinine and urea levels were significantly(P<0.05) increased in a dose dependent manner in hydrose treated mice, whereas calcium level was significantly decreased in hydrose exposed mice compared to control mice.Histological study of kidney showed the glomeruler inflammation, increased diameter of renal glomeruli and enlargement of proximal tubular lumen of kidneys of mice exposed to hydrose compared to that of control animals. Conclusion:The results of this study indicated that use of hydrose in molasses and other food preparations in Bangladesh may cause kidney impairment.Tuhin Reza Sharmin Aktar Hasan Al Amin Mashiur Rahman Afroza Arefin Nayan Chandra Mohanto Shahnur Alam Abdullah Al Mamun Anwar Habib Asafudullah Farjana Nikkon Khaled Hossain Zahangir Alam Saud 2014Asian Pacific Journal of Tropical Biomedicine2014,4,11:0
Annual pattern of zooplankton communities and their environmental response in a subtropical maritime channel system in the northern Bay of Bengal, Bangladesh显示文摘Zooplankton plays an important role in aquatic food webs by fluxing of energy from primary producer to subsequent trophic levels in the food chain. The annual pattern of zooplankton communities and potential environmental drivers were studied in the Kohelia channel, Bangladesh from summer 2014 to spring 2015.Samples were collected using net at a depth of 1 m. A total of 32 species belonged to 18 orders, 27 families and 15 taxonomic groups were identified. Of these species, 22 distributed in all four seasons of which 8 were dominant and highly contributing to the total communities. Species number peaked in summer next to winter and fall in spring while maximum abundance was in summer and minimum in spring. Multivariate analyses showed that there was a clear annual pattern in the zooplankton communities. Species diversity and evenness peaked in spring but fall in autumn while the high value of species richness was found in winter. Biological-environmental best matching(BIO-ENV) analyses conformed that community pattern of zooplankton was mainly driven by transparency salinity, and temperature individually or combined with water nutrients. These results demonstrate that annual pattern of the zooplankton community shaped by channel environmental factors in subtropical channel ecosystems, thus might be used for community-based subtropical coastal water bioassessment.ABDULLAH Al Mamun ALAM Md.Didarul AKHTAR Aysha XU Henglong ISLAM Md.Shafiqul MUSTAFA KAMAL Abu Hena UDDIN M Muslem ALAM Md.Wahidul 2018Acta Oceanologica Sinica2018,37,8:0

FGF21 alleviates microvascular damage following limb ischemia/reperfusion injury by TFEB-mediated autophagy enhancement and anti-oxidative response显示文摘Dear Editor,Microvascular damage is a pivotal pathological factor in lower limb ischemia/reperfusion(I/R)injury.1 Excessive reactive oxygen species(ROS)formation and disrupted autophagic flux have been recognized as the critical mechanism of cellular death,2,3 especially in I/R injury.Recent investigation has displayed that fibroblast growth factor 21(FGF21)exerts a protective effect against I/R injury via transcription factor EB(TFEB)-mediated autophagy and regulation of anti-oxidative response.4 However, the promising role of FGF21 in acute lower limb I/R injury remains elusive.Fanfeng Chen Jiayu Zhan Xiaoqing Yan Abdullah Al Mamun Yu Zhang Yitie Xu Hongyu Zhang Xiaokun Li Kailiang Zhou Jian Xiao 2022Signal Transduction and Targeted Therapy2022,7,11:0
Crop Improvement and Abiotic Stress Tolerance Promoted by Moringa Leaf Extract显示文摘Moringa leaf extract(MLE)has been shown to promote beneficial outcomes in animals and plants. It is rich in amino acids, antioxidants, phytohormones, minerals, and many other bioactive compounds with nutritional and growth-promoting potential.Recent reports indicated that MLE improved abiotic stress tolerance in plants. Our understanding of the mechanisms underlying MLE-mediated abiotic stress tolerance remains limited. This review summarizes the existing literature on the role of MLE in promoting plant abiotic stress acclimation processes.MLE is applied to plants in a variety of ways, including foliar spray, rooting media, and seed priming.Exogenous application of MLE promoted crop plant growth, photosynthesis, and yield under both nonstress and abiotic stress conditions.MLE treatment reduced the severity of osmotic and oxidative stress in plants by regulating osmolyte accumulation, antioxidant synthesis, and secondary metabolites.MLE also improves mineral homeostasis in the presence of abiotic stress.Overall, this review describes the potential mechanisms underpinning MLEmediated stress tolerance.Md.Abir Ul Islam Juthy Abedin Nupur Charles T.Hunter Abdullah Al Mamun Sohag Ashaduzzaman Sagar Md.Sazzad Hossain Mona F.A.Dawood Arafat Abdel Hamed Abdel Latef Marián Brestič Md.Tahjib-UI-Arif 2022Phyton-International Journal of Experimental Botany2022,91,8:0
