17Curcumin in inflammatory bowel diseases:Cellular targets and molecular mechanisms显示文摘Curcumin,a natural product,has exhibited promising effects in both animal models and clinical trials,interacting with a multitude of factors linked to Inflammatory Bowel Disease(IBD).These factors encompass cytokines,oxidative stress-associated enzymes,and modulation of the intestinal microbiota.Notably,curcumin has demonstrated therapeutic potential in animal models of colitis,wherein it exerts a negative regulatory influence on pivotal signaling pathways such as PI3/Akt,JAK/STAT,andβ-catenin.Moreover,it inhibits the expression of proinflammatory enzymes and co-stimulatory molecules(including RANKL,ICAM-1,CD205,CD256,TLR4,among others),while curbing immune cell chemotaxis,thereby attenuating the characteristic neutrophil infiltration observed in IBD.Another facet of curcumin’s action involves its modulation of the intestinal microbiota.Notably,the microbiota itself contributes to beneficial biotrans formations of curcumin,thereby enhancing its effectiveness in IBD treatment.On a clinical front,curcumin has demonstrated the ability to induce clinical and/or endoscopic remission without any reported toxic effects.Hence,curcumin warrants consideration as an adjunctive therapy in IBD management.Subsequent clinical investigations should concentrate on meticulously evaluating curcumin’s impact on these precise therapeutic targets.AMYLLY SANUELLY DA PAZ MARTINS MARLA DE CERQUEIRA ALVES ORLANDO ROBERTO PIMENTEL DE ARAÚJO FABIANA OLIVEIRA DOS SANTOS CAMATARI MARÍLIA OLIVEIRA FONSECA GOULART FABIANA ANDRÉA MOURA 2023BIOCELL2023,47,11:0
18Efficacy of oral consumption of curcumin/Curcuma longa for symptom improvement in inflammatory bowel disease:A systematic review of animal models and a meta-analysis of randomized clinical trials显示文摘The roots of the vegetal Curcuma due to its high content of polyphenols,has been used successfully in several clinical situations.This review assessed the effect of curcumin/Curcuma longa on symptoms and metabolic changes in inflammatory bowel disease(IBD).A systematic review of animal models and randomized clinical trials(RCTs)was conducted by searching the following electronic databases:PubMed,CENTRAL,LILACS,Science Direct,and ClinicalTrials.gov.From 997 found records,62 were included.More than 90%of the animal studies reported an improvement in macroscopic,histologic and/or functional activity;80%identified decreased oxidative and/or inflammatory biomarkers in animals treated with curcumin.Among the RCTs,intention-to-treat analysis showed that oral curcumin was effective in inducing clinical remission(n=281,RR:3.15 CI 95%[1.22–8.10]p=0.0017;i2=72.2%,p=0.006)and clinical response(n=259,RR:1.60 CI 95%[1.09–2.35]p=0.0017;i2=59.7%,p=0.042)but not endoscopic remission(n=161,RR:2.91 CI 95%[0.58–14.58]p=0.195;i2=72.7%,p=0.026).These results confirm that oral supplementation with curcumin/Curcuma longa has beneficial actions in animal colitis and,when associated with drug therapy,is effective in the treatment of patients with IBD.MARLA DE CERQUEIRA ALVES MONISE OLIVEIRA SANTOS NASSIB BEZERRA BUENO ORLANDO ROBERTO PIMENTEL DE ARAúJO MARíLIA OLIVEIRA FONSECA GOULART FABIANA ANDRéA MOURA 2022BIOCELL2022,46,9:0
19Prenatal administration of heparin-binding epidermal growth factor-like growth factor in an experimental model of necrotizing enterocolitis decreased both incidence and severity of the disease显示文摘background Necrotizing enterocolitis(NEC)is the leading gastrointestinal cause of death in premature infants and causes long-term disabilities.Previously,enteral heparin-binding epidermal growth factor-like growth factor(HB-EGF)administered after birth demonstrated decreased incidence and severity of NEC in a neonatal animal model of NEC.We investigated the potential prophylactic strategy of preventing NEC using prenatally administered HB-EGF.Methods An HB-EGF(800μg/kg/dose)dose was injected into pregnant rats via tail vein or intraperitoneal route 2 hours prior to delivery.After cesarean section(C-section)at 21 days’gestation,the rat pups were subjected to the NEC protocol by inducing stressors:hypoxia,hypothermia,hypertonic feeds,and orogastric gavage of lipopolysaccharide(2 mg/kg).Postnatally,pups were monitored for 96 hours and assessed for the development of clinical and postmortem histological NEC.results The experimental NEC incidence in untreated,stressed rat pups was 66%.Compared with untreated pups,the maternal administration of HB-EGF correlated with a significant NEC incidence and severity decrease in rat pups.The strongest decrease was seen when HB-EGF was administered via the intraperitoneal route 2 hours prior to C-section(66%vs 31%,*p<0.05).Prenatal HB-EGF administration significantly increased pups’survival after NEC protocol exposure,with the greatest benefit observed in the group that received HB-EGF intraperitoneally 2 hours before delivery.Conclusions Prenatal administration of HB-EGF decreases the incidence and severity of NEC,preserves gut barrier function and increases survival.This may represent a novel prophylactic clinical strategy for NEC offered to mothers at risk of delivering a premature infant.Marla Ashley Sacks Yomara Stephanie Mendez Faraz A Khan Robert Propst Craig W Zuppan Christopher G Wilson Andrei Radulescu 2022World Journal of Pediatric Surgery2022,5,1:0
