1Exosome and Exosomal MicroRNA: Trafficking, Sorting, and Function显示文摘Exosomes are 40–100 nm nano-sized vesicles that are released from many cell types into the extracellular space. Such vesicles are widely distributed in various body fluids. Recently,m RNAs and micro RNAs(mi RNAs) have been identified in exosomes, which can be taken up by neighboring or distant cells and subsequently modulate recipient cells. This suggests an active sorting mechanism of exosomal mi RNAs, since the mi RNA profiles of exosomes may differ from those of the parent cells. Exosomal mi RNAs play an important role in disease progression, and can stimulate angiogenesis and facilitate metastasis in cancers. 2Prevalence of minimal hepatic encephalopathy and quality of life evaluations in hospitalized cirrhotic patients in China显示文摘AIM:To investigate the prevalence of minimal hepatic encephalopathy(MHE)and to assess corresponding health-related quality of life(HRQoL)in hospitalized cirrhotic patients in China.METHODS:This multi-center cross-sectional study included 16 teaching hospitals,which were members of 'Hepatobiliary Cooperation Group,Society of Gastroenterology,Chinese Medical Association',from different areas of China carried out between June and October in 2011.All the eligible hospitalized cirrhotic patients(n = 538)were required to complete triplicate number connection tests combined with one digit symbol test for diagnosing MHE.Patients' clinical examination data were complemented by a modified questionnaire assessing HRQoL.Written informed consent was obtained from each patient.RESULTS:Male was predominant(68.6%)in 519 patients who met the criteria of the study,with a mean age of 49.17 ± 11.02 years.The most common cause of liver cirrhosis was chronic hepatitis B(55.9%).The prevalence of MHE was 39.9% and varied by ChildPugh-Classification score(CPC-A:24.8%,CPC-B:39.4% and CPC-C:56.1%,P < 0.01).MHE(P < 0.01)and higher CPC scores(P < 0.01)were associated with a high HRQoL scores(reflecting poorer quality of life).The prevalence of MHE was proportionate to CPC(P = 0.01)and high quality of life scores(P = 0.01).CONCLUSION:Hospitalized cirrhotic patients have a high prevalence of MHE that is proportionate to the degree of liver function and HRQoL impairment.Ji-Yao Wang Ning-Ping Zhang Bao-Rong Chi Yu-Qing Mi Li-Na Meng Ying-Di Liu Jiang-Bin Wang Hai-Xing Jiang Jin-Hui Yang Yun Xu Xiao Li Jian-Ming Xu Guo Zhang Xin-Min Zhou Yu-Zheng Zhuge De-An Tian Jin Ye Yu-Lan Liu 2013World Journal of Gastroenterology2013,19,30:64
3High-Strength and High-Plasticity TWIP Steel for Modern Vehicle显示文摘In this paper new high-strength and high-plasticity twinning induced plasticity (TWIP) steel for modern automobile body was investigated. Some basic experimental results were given. The results indicate the TWlP steel has excellent properties. It exhibits high ultimate tensile strength (600~1100 MPa) and extremely large elongation of 60% to 90%. 4Prevalence of Thyroid Nodules and Its Relationship with Iodine Status in Shanghai:a Population-based Study显示文摘Objective This study was designed to evaluate the prevalence of thyroid nodules(TNs) and its relationship with urine iodine concentrations(UICs) after the regional rapid economic growth and lifestyle changes. Methods A cross-sectional survey was conducted in the general population aged 15-69 years. A questionnaire regarding general and personal characteristics and relevant information was administered. Ultrasonography of the thyroid was performed, and serum triiodothyronine(T3), tetraiodothyronine(T4), serum thyroid stimulating hormone(TSH), free triiodothyronine(FT3), free tetraiodothyronine(FT4), thyroglobulin antibody(Tg Ab), thyroid peroxidase antibody(TPOAb), and TSH receptor antibody(TRAb) levels were measured for each individual subject. Results The prevalence rates of TNs in the whole population, females and males were 27.76%, 34.04%, and 21.60%, respectively. The prevalence of multiple nodules increased with age, whereas the prevalence peaks differed between males and females. The median UICs in the whole population and females with non-TNs were higher than those of subjects with TNs(P=0.0035, P=0.0068). The median UICs in subjects with a single TN were higher than those in subjects with multiple TNs(P=0.0164, P=0.0127). The result showed a U-shaped curve relationship between UIC and prevalence of TNs. The prevalence of TNs was the lowest when the UIC was 140-400 μg/L. Conclusion The prevalence of TNs was nearly 30% and increased with age. The relationship between UIC and prevalence of TNs is U-shaped, with an increase in risk when the UIC was <140 μg/L and >400 μg/L. 6Antigenic variation of the human influenza A(H3N2) virus during the 2014–2015 winter season显示文摘The human influenza A(H3N2) virus dominated the 2014–2015 winter season in many countries and caused massive morbidity and mortality because of its antigenic variation. So far, very little is known about the antigenic patterns of the recent H3N2 virus. By systematically mapping the antigenic relationships of H3N2 strains isolat ed since 2010, we discov ered that two groups with obvious antigenic divergence, named SW13(A/Switzerland/9715293/2013-like strains) and HK14(A/Hong Kong/5738/2014-like strains), co-circulated during the 2014–2015 winter season. HK14 group co-circulated with SW13 in Europe and the United States during this season, while there were few strains of HK14 in China's Mainland, where SW13 has dominated since 2012. Furthermore, we found that substitutions near the receptor-binding site on hemagglutinin played an important role in the antigenic variation of both the groups. 8The Intervention Effect of Rosiglitozone in Ovarian Fibrosis of PCOS Rats显示文摘Objective To explore the Intervention effect of Rosiglitozone in ovarian fibrosis of PCOS rats.Methods 60 female SD rats were randomly divided into 3 groups:control group,model group and treatment group.The model and treatment groups were established by subcutaneous injection of DHEA,while the treatment group was given RGZ.The serum hormone values,pathohistology of ovarian structure of rats,ovarian ultrastructure and the expressions of TGF-β 1 and CTGF were detected.Results The PCOS model was established successfully.The expression intensity of TGF-β 1 and CTGF in Oocytes of the PCOS groups was 9.545±2.954 and 9.665±2.400,respectively and was significantly higher than that of the control group 6.636±2.264 and 7.036±2.133;after treatment with rosiglitazone,the expression was significantly decreased 6.980±2.421 and 6.642±2.721 as compared with that of the model group (P<0.05,P<0.001).The values in serum of the PCOS groups were 3.749±2.054 and 0.265±0.129,and 1.914±1.801 and 0.096±0.088 in the control group which had statistically significant difference (P<0.05,P<0.001).After treatment with rosiglitazone,the values were 2.3100±1.825 and 0.112±0.187 and were significantly different with those of the model group (P<0.05,P<0.001).Conclusion TGF-β 1 and CTGF play an important role in the development of ovary fibrosis in PCOS.However,RGZ may postpone the development of fibrosis by decreasing the levels of TGF-β 1 and CTGF.MIAO Zhu Lin GUO Liang WANG Yong Xia CUI Rong YANG Ning HUANG Mi Qiong QIN Wei Bing CHEN Jin LI Hong Mei WANG Zi Neng WEI Xiang Cai 2012Biomedical and Environmental Sciences2012,25,1:14
9Uptake of HIV Self-testing among Men Who have Sex with Men in Beijing, China: a Cross-sectional Study显示文摘Objective To examine HIV self-testing uptake and its determinates among men who have sex with men(MSM) in Beijing, China. Methods A cross-sectional online survey was conducted in Beijing, China in 2016. Participants were users of a popular Chinese gay networking application and had an unknown or negative HIV status. Univariate and multivariate logistic regression analyses were conducted to examine factors associated with HIV self-testing based on adjusted odds ratio(AOR) and 95% confidence interval(CI). Results Among the 5,996 MSM included in the study, 2,383(39.7%) reported to have used HIV self-testing kits. Willingness to use an HIV self-test kit in the future was expressed by 92% of the participants. High monthly income(AOR = 1.49; CI = 1.10-2.02; P = 0.010), large number of male sex partners(≥ 2: AOR = 1.24; CI = 1.09-1.43; P = 0.002), sexual activity with commercial male sex partners(≥ 2: AOR = 1.94; CI = 1.34-2.82; P = 0.001), long-term drug use(AOR = 1.42; CI = 1.23-1.62; P < 0.001), and long-term HIV voluntary counseling and testing(VCT) attendance(AOR = 3.62; CI = 3.11-4.22; P < 0.001) were all associated with increased odds of HIV self-testing uptake. Conclusion The nearly 40% rate of HIV self-testing uptake among MSM in our sample was high. In addition, an over 90% willingness to use kits in the future was encouraging. 10Bioactive biodegradable polycitrate nanoclusters enhances the myoblast differentiation and in vivo skeletal muscle regeneration via p38 MAPK signaling pathway显示文摘Complete skeletal muscle repair and regeneration due to severe large injury or disease is still a challenge.Biochemical cues are critical to control myoblast cell function and can be utilized to develop smart biomaterials for skeletal muscle engineering.Citric acid-based biodegradable polymers have received much attention on tissue engineering,however,their regulation on myoblast cell differentiation and mechanism was few investigated.Here,we find that citrate-based polycitrate-polyethylene glycol-polyethylenimine(POCG-PEI600)nanoclusters can significantly enhance the in vitro myoblast proliferation by probably reinforcing the mitochondrial number,promote the myotube formation and full-thickness skeletal muscle regeneration in vivo by activating the myogenic biomarker genes expression of Myod and Mhc.POCG-PEI600 nanoclusters could also promote the phosphorylation of p38 in MAP kinases(MAPK)signaling pathway,which led to the promotion of the myoblast differentiation.The in vivo skeletal muscle loss rat model also confirmed that POCG-PEI600 nanoclusters could significantly improve the angiogenesis,myofibers formation and complete skeletal muscle regeneration.POCG-PEI600 nanocluster could be also biodegraded into small molecules and eliminated in vivo,suggesting their high biocompatibility and biosafety.This study could provide a bioactive biomaterial-based strategy to repair and regenerate skeletal muscle tissue.Yi Guo Min Wang Juan Ge Wen Niu Mi Chen Wei Cheng Bo Lei 2020Bioactive Materials2020,5,3:12

12Effects of recombinant retroviral vector mediated human insulin like growth factor-1 gene transfection on skeletal muscle growth in rat显示文摘Background This study transferred a recombinant gene encoding human insulin like growth factor-1 (hIGF-1) into modified primary skeletal myoblasts with a retroviral vector (pLgXSN ) and determined whether the hIGF-1 promoted growth of skeletal muscle in rat. Methods hIGF-1cDNA was amplified in vitro from normal human liver cells by using RT-PCR and cloned into plasmid vector pLgXSN. The recombinant vector pLghIGF-1SN and control vector pLgGFPSN were transfected into packaging cell PT67 and G418 was used to select positive colony. Myoblasts were infected with a high titre viral supernatant and transduction efficiency was evaluated as GFP expression. The expression of hIGF-1 mRNA in myoblasts was investigated by immunocytochemistry and RT-PCR. MTT assays detected the growth of myoblasts in vitro. Myoblasts transduced with pLghIGF-1SN were injected into hind limb muscles of 10-12 week male SD rats. Formed tissues were harvested 4 weeks later. Myocyte diameter, mean weight of hind limb and body were measured to evaluate the skeletal muscle growth. Results Recombinant retroviral plasmid vector pLghIGF-1SN was constructed successfully. The titre of the packaged recombinant retrovirus was 1×106 cfu/ml. The transfection rate of PT67 cells reached 100% after G418 screening. hIGF-1 expression was positive in myoblast-IGF-1. The proliferation rate of myoblast-IGF-1 in vitro was higher than GFP-myoblast or myoblast (P< 0.05). The mean weights of hind limb and body of rats injected myoblast-IGF-1 were higher than those of the rats injected with myoblast-GFP or myoblast (P< 0.05). Myocyte diameter had a significant increase in IGF-1 group compared to GFP group and myoblast group (P< 0.05). Conclusions The transfection of the human IGF-1 gene mediated by a retroviral vector can promote the growth of skeletal muscle in rats. 13Microstructure and tensile properties of low cost titanium alloys at different cooling rate显示文摘Titanium and titanium alloys have several advantages, but the cost of titanium alloys is very expensive compared with the traditional metal materials. This article introduces two new low-cost titanium alloys Ti-2.1Cr-1.3Fe (TCF alloy) and Ti-3Al-2.1Cr-1.3Fe (TACF alloy). In this study, we used Cr-Fe master alloy as one of the raw materials to develop the two new alloys. We introduce the microstructure and tensile properties of the two new alloys from β solution treated with different cooling methods. Optical microscopy (OM), X-ray diffractometry (XRD), and transmission electron microscopy (TEM) were employed to analyze the phase constitution, and scanning electron microscopy (SEM) was used to observe the fracture surfaces. The results indicate that the microstructures consist of β grain boundary and α′ martensite after water quenching (WQ), β matrix and α phase after air cooling (AC) and furnace cooling (FC), respectively. Also, the microstructure is the typical basketweave structures after FC. Of course, athermal ω is also observed by TEM after WQ. The strength increases with decreasing cooling rates and the plasticity is reversed. Because of the athermal ω, the strength and ductility are highest and lowest when the cooling method is WQ. The strength of TACF alloy is higher than the TCF alloy, but the plasticity is lower. The fracture surfaces are almost entirely covered with dimples under the cooling methods of AC and FC. 15Influence of Inclusion on Crack Initiation in Wheel Rim显示文摘Speed increase and heavy haul of the railway freight car are the effective measures to raise the transport ability of railway,and they have been the new trend for railway freight car development.The increase of speed and axle load will lead to the increase of wheel-rail contact stress,which will make a great change of stress distribution in the whole wheel rim.Goodier equation was employed to analyze the stress state around inclusion and cavity.In the operation of wheel-rail contact stress,stress concentration occurred in the pole of Al2O3 spherical inclusion,while it appeared on the equator of cavity.The critical inclusion sizes of 250 kN axle load freight car wheel at different velocities,a certain distance away from the tread,were calculated by Murakami equation.Comparing the inclusion in cast steel wheel with that in rolled steel wheel,it is found that the inclusion in cast steel wheel has much more excellent crack resistance than that in rolled steel wheel.In the meantime,in order to reduce the probability of rim crack,some suggestions were put forward.MI Guo fa NAN Hong yan LIU Yan-lei ZHANG Bin ZHANG Hong SONG Guo xiang 2011Journal of Iron and Steel Research(International)2011,18,1:8
16Efficacy of a Chinese herbal formula on hepatitis B e antigenpositive chronic hepatitis B patients显示文摘BACKGROUND No guideline recommends antiviral therapy for hepatitis B e antigen(HBeAg)-positive chronic hepatitis B patients with persistently normal alanine aminotransferase levels and a high hepatitis B virus(HBV)DNA viral load.AIM To evaluate the feasibility and safety of a Chinese herbal formula as a therapeutic option for chronic HBV infection.METHODS In total,395 patients(30–65 years old)with confirmed HBeAg-positive chronic hepatitis B infection and persistently normal alanine aminotransferase were randomized to receive either Chinese herbal formula or placebo for 96 wk.Endpoints to evaluate therapeutic efficacy included:(1)HBV DNA levels decreased to less than 4 log10 IU/mL at weeks 48 and 96;and(2)HBeAg clearance and seroconversion rates at weeks 48 and 96.RESULTS HBV DNA levels≤4 log10 IU/mL were 10.05%at week 48 and 18.59%at week 96 in the treatment group.The HBeAg clearance and conversion rates were 8.54%and 8.04%at week 48 and 16.08%and 14.57%at week 96,respectively.However,HBV DNA levels≤4 log10 IU/mL were 2.55%and 2.55%at weeks 48 and 96,respectively,and the HBeAg clearance rates were 3.06%and 5.61%at weeks 48 and 96,respectively,in the control group.The quantitative hepatitis B surface antigen and HBeAg levels at baseline and changes during the treatment period as well as the alanine aminotransferase elevation at weeks 12 and 24 were strong predictors of HBeAg clearance.CONCLUSION High rates of HBV DNA reduction,HBeAg clearance and seroconversion could be achieved with Chinese herbal formula treatments,and the treatments were relatively safe for HBeAg-positive chronic hepatitis B-infected patients with persistently normal alanine aminotransferase.The ability of the compound to modulate host immune function probably contributed to this effect.Yu-Feng Xing Chun-Shan Wei Tian-Ran Zhou Dan-Ping Huang Wei-Chao Zhong Bin Chen Hua Jin Xiao-Yu Hu Zhi-Yun Yang Qing He Kai-Ping Jiang Jun-Min Jiang Zhen-Bin Hu Xin Deng Fan Yang Feng-Yi Li Gang Zhao Li-Chun Wang Yu-Qiang Mi Zuo-Jiong Gong Peng Guo Jian-Hua Wu Wei-Qun Shi Hong-Zhi Yang Da-Qiao Zhou Guang-Dong Tong 2020World Journal of Gastroenterology2020,26,30:7

18Protective effect of total flavonoid C-glycosides from Abrus mollis extract on lipopolysaccharide-induced lipotoxicity in mice显示文摘Abrus mollis is a widely used traditional Chinese medicine for treating acute and chronic hepatitis, steatosis, and fibrosis. It was found that the total flavonoid C-glycosides from Abrus mollis extract(AME) showed potent antioxidant, anti-inflammatory, and hepatoprotective activities. To further investigate the hepatoprotective effect of AME and its possible mechanisms, lipopolysaccharide(LPS)-induced liver injury models were applied in the current study. The results indicated that AME significantly attenuated LPS-induced lipid accumulation in mouse primary hepatocytes as measured by triglyceride(TG) and total cholesterol(TC) assays and Oil Red O staining. Meanwhile, AME exerted a protective effect on LPS-induced liver injury as shown by decreased liver index, serum aminotransferase levels, and hepatic lipid accumulation. Real-time PCR and immunoblot data suggested that AME reversed the LPS-mediated lipid metabolism gene expression, such as sterol regulatory element-binding protein-1(SREBP-1), fatty acid synthase(FAS), and acetyl-CoA carboxylase 1(ACC1). In addition, LPS-induced overexpression of activating transcription factor 4(ATF4), X-box-binding protein-1(XBP-1), and C/EBP homologous protein(CHOP) were dramatically reversed by AME. Furthermore, AME also decreased the expression of LPS-enhanced interleukin-6(IL-6) and cyclooxygenase-2(COX-2). Here, it is demonstrated for the first time that AME ameliorated LPS-induced hepatic lipid accumulation and that this effect of AME can be attributed to its modulation of hepatic de novo fatty acid synthesis. 19Agglutinin isolated from Arisema heterophyllum Blume induces apoptosis and autophagy in A549 cells through inhibiting PI3K/Akt pathway and inducing ER stress显示文摘Arisaema heterophyllum Blume is one of the three medicinal plants known as traditional Chinese medicine Rhizoma Arisaematis(RA). RA has been popularly used to treat patients with convulsions, inflammation, and cancer for a long time. However, the underlying mechanisms for RA effects are still unclear. The present study was designed to determine the cytotoxicity of agglutinin isolated from Arisema heterophyllum Blume(AHA) and explore the possible mechanisms in human non-small-cell lung cancer A549 cells. AHA with purity up to 95% was isolated and purified from Arisaema heterophyllum Blume using hydrophobic interaction chromatography. AHA dose-dependently inhibited the proliferation of A549 cells and induced G_1 phase cell cycle arrest. AHA induced apoptosis by up-regulating pro-apoptotic Bax, decreasing anti-apoptotic Bcl-2, and activating caspase-9 and caspase-3. In A549 cells treated with AHA, the PI3K/Akt pathway was inhibited. Furthermore, AHA induced increase in the levels of ER stress markers such as phosphorylated eukaryotic initiation factor 2α(p-eIF2α), C/EBP-homologous protein(CHOP), inositol-requiring enzyme 1α(IRE1α), and phosphorylated c-Jun NH_2-terminal kinase(p-JNK). AHA also induced autophagy in A549 cells. Staining of acidic vesicular organelles(AVOs) and increase in the levels of LC3II and ATG7 were observed in AHA-treated cells. These findings suggested that AHA might be one of the active components with anti-cancer effects in Arisaema heterophyllum Blume. 20Stable carbon and nitrogen isotope evidence of human and pig diets at the Qinglongquan site,China显示文摘Previous studies on the Rice-Millet(foxtail millet and common millet) Blended Zone in Chinese Neolithic have not clearly addressed such questions as the importance of primitive rice-millet mixed agriculture to human lifestyle and livestock managements within this region,the relationship among the development of the agriculture,paleoenvironment,and cultural interactions,and so on.Here stable carbon and nitrogen isotope analysis of human and pig bones from the Qinglongquan site was conducted,covering two cultural phases,namely the Qujialing Culture(3000 BC to 2600 BC) and the Shijiahe Culture(2600 BC to 2200 BC) .Based on this analysis,we further discussed the diets of ancient humans and pigs in the site,investigated the importance of rice-millet mixed agriculture to human and pig diets,and explored the relationship among the primitive rice agriculture and millet agriculture,cultural interactions,and paleoenvironment.The δ13C values of human bone collagen(-16.7‰ to-12.4‰,averaging-14.6‰±1.3‰,n=24) revealed that both C3 and C4 foods were consumed,probably from the contribution of rice(C3 plant) and millets(C4 plants) due to the coexistence of these crops at this site.In addition,the human mean δ13C value suggested that millet agriculture was only minor in human diets.The human δ15N values(6.6‰to 10.8‰,averaging 9.0‰±1.2‰,n=24) showed that animal resources played a significant role in human diets,and varied greatly.The mean δ13C value of the pigs(-14.3‰±2.5‰,n=13) was quite similar to that of the humans,but the mean δ15N value of the pigs was slightly less(1.3‰) .The similar δ13C and δ15N values between humans and pigs suggested that the pigs consumed a lot of humans'food remains.No correlations of the δ13C and δ15N values between humans and pigs showed that both human and pig diets were based mainly on plant foods,which might be related to highly developed rice-millet mixed agriculture at that time. In comparison with the human and pig diets between the two periods,millet agriculture contributed more than 10%in the Shijiahe Culture,if a simple mixing model was used.This apparent dietary shift matched the climatic variation and agricultural development through the time.In warm and humid climate with the expansion of the Qujialing Culture northwards,rice was widely cultivated.However,when the climate was cold and arid,northern culture was expanding southwards.Thus,millet agriculture became more important.GUO Yi HU YaoWu ZHU JunYing ZHOU Mi WANG ChangSui Michael P.
