3BIG1 mediates sepsis-induced lung injury by modulating lipid raft-dependent macrophage inflammatory responses显示文摘Sepsis is a systemic inflammatory response syndrome with high mortality.It has been reported that brefeldin A-inhibited guanine nucleotide-exchange factor 1(BIG1)is involved in the pathogenesis of sepsis.However,the mechanism is not fully elucidated.In the present study,we explored the role of BIG1 in mediating lipid raft-dependent macrophage inflammatory response and its impact on lung injury in murine sepsis.In vitro studies revealed that BIG1 deficiency reduces the upregulation and secretion of tumor necrosis factor alpha(TNF-α),interleukin-6(IL-6),and IL-1β and inhibits the activation of the toll-like receptor 4(TLR4)/myeloid differentiation primary response 88-dependent nuclear factor kappa-B signaling pathway induced by the lipopolysaccha-ride(LPS)treatment.Further experiments revealed that the inhibitory effects of BIG1 deficiency on LPS-induced inflammation are due to the upregulation of adenosine triphosphate-binding cassette transporter A1.This promotes the free-cholesterol efflux from lipid rafts and results in the reduction of lipid raft TLR4 content.The decrease in TLR4 content in lipid raft thereby inhibits the LPS-induced inflammatory response.Furthermore,using the cecal ligation and puncture-induced polymicrobial sepsis mouse model,we found that conditional knockout(cKO)of the myeloid cell BIG1 significantly reduced the serum concentrations of TNF-α,IL-6,and IL-1β,and downregulated their mRNA expressions in the lungs.Pathological analysis confirmed that the BIG1 cKO alleviated the sepsis-induced lung injury.These results revealed the crucial new role of BIG1 in mediating lipid raft-dependent macrophage inflammatory response.Hence,BIG1 may be a potential promising therapeutic target for the treatment of septic lung injury.Minli Sun Xiaodan Han Di Zhou Jing Zhong Lixin Liu Yirui Wang Jiahui Ni Xiaoyan Shen Chao Liang Hao Fang 2021Acta Biochimica et Biophysica Sinica2021,53,8:1
4Analysis of wild vascular plant resources and diversity in Taibai Mountains of Qinling area,Shaanxi Province显示文摘Taibai Mountains,located at the northern foot of the Qinling Mountains,are composed of the Yuan Taibai Mountain(also known as East Taibai Mountain),Aoshan Mountain(also known as West Taibai Mountain)and their connection part.With its complex geographical and climatic conditions,Taibai Mountains are extremely rich in wild plant resources.Based on field investigation,literature review and specimen identification,wild vascular plants resources and their diversity in Taibai Mountains were studied by the sixth Traditional Chinese Medicine(TCM)Resources Scientific Expedition Team of Shenyang Pharmaceutical University.222 species of vascular plants belonging to 163 genera and 63 families were collected during July 2012 in this area,and most of the plants are angiosperms.The families with more species are Liliaceae,Ranunculaceae,Asteraceae,Rosaceae,and Saxifragaceae,and the dominant genus are Aconitum,Sedum,Eleutherococcus,Pedicularis,Polygonatum,and Patrinia.In terms of life form,perennial herbs are the main species,accounting for 72.97%of the total species,with others being some annual(or biennial)herbs,shrubs and lianas.Among all the collected vascular plants,170 species of them are medicinal vascular plants,accounting for 76.58%of the total.Most of the medicinal parts are roots and rhizomes,followed by the whole plants.Finally,on the basis of investigation and study,some suggestions are put forward to strengthen the protection and utilization of plant resources in Taibai Mountains.Bingjun Sun Qing Wang Minli Ruan Anhua Wang Jingming Jia 2023Asian Journal of Traditional Medicines2023,18,6:0
5The role of immune cells in obesogenic memory显示文摘Obesity is a serious chronic disease and worldwide public health problem.According to data from the World Health Organization(WHO),more than 650 million adults in the world were obese and three times more individuals were overweight in 2016.Obesity is associated with type II diabetes,hepatic steatosis,cardiovascular diseases,systemic chronic inflammation,and at least 13 types of cancer.Unfortunately,few effective remedies are available for obesity.The major problem is that most patients suffer from weight regain 2–3 years after obesity treatments,such as obesity drugs,lifestyle interventions(including exercise,diet restrictions,and diet modification),and bariatric surgery.Along with weight regain,most of the initial beneficial changes in metabolomic and transcriptomic profiles dissipate,including insulin sensitivity,the level of adipokines and blood cholesterol.This phenomenon has been defined as“obesogenic memory”.1,2 Obesogenic memory causes repeated weight loss and gain along with obesity therapy,which is called“weight cycling”,leading to a worse healthy status than would result from simple obesity.3,4.Minli Sun Shang Zheng Xiang Gao Zhaoyu Lin 2020Cellular & Molecular Immunology2020,17,8:0
6EVA2.0:Investigating Open-domain Chinese Dialogue Systems with Large-scale Pre-training显示文摘Large-scale pre-training has shown remarkable performance in building open-domain dialogue systems.However,previous works mainly focus on showing and evaluating the conversational performance of the released dialogue model,ignoring the discussion of some key factors towards a powerful human-like chatbot,especially in Chinese scenarios.In this paper,we conduct extensive experiments to investigate these under-explored factors,including data quality control,model architecture designs,training approaches,and decoding strategies.We propose EVA2.0,a large-scale pre-trained open-domain Chinese dialogue model with 2.8 billion parameters,and will make our models and codes publicly available.Automatic and human evaluations show that EVA2.0 significantly outperforms other open-source counterparts.We also discuss the limitations of this work by presenting some failure cases and pose some future research directions on large-scale Chinese open-domain dialogue systems.Yuxian Gu Jiaxin Wen Hao Sun Yi Song Pei Ke Chujie Zheng Zheng Zhang Jianzhu Yao Lei Liu Xiaoyan Zhu Minlie Huang 2023Machine Intelligence Research2023,20,2:0
7In vivo effects of neomycin sulfate on non-specific immunity,oxidative damage and replication of cyprinid herpesvirus 2 in crucian carp(Carassius auratus gibelio)显示文摘Neomycin belongs to the family of 2-deoxystreptamine-containing aminoglycoside antibiotics.It is widely used for bacterial infections,targeting most gram-negative and some gram-positive bacteria.Neomycin has also been reported to show antiviral activity.Here,we evaluated the toxicity of neomycin sulfate,and investigated its effect on non-specific immunity and viral infection in crucian carp(Carassius auratus gibelio).The safe concentration of neomycin sulfate for crucian carp was determined to be 102.9 mg/kg in vivo.In oxidative damage assays,neomycin sulfate increased superoxide dismutase and catalase activity and decreased malondialdehyde in the liver of crucian carp.In non-specific blood immune assays,the white blood cell count and complement 3 content significantly increased after neomycin sulfate treatment,while no significant changes were observed in antibacterial or lysozyme activity.In a challenge test,neomycin sulfate protected the crucian carp from cyprinid herpesvirus 2(CyHV-2)infection and inhibited CyHV-2 replication.In cytotoxicity assays,low concentrations of neomycin sulfate had no cytotoxicity on cells from the fins of crucian carp.The results of the present study indicate that oral administration of neomycin sulfate reduced oxidative damage,enhanced immunity and provided protection against CyHV-2 in crucian carp.Penghui Sun Fei Yu Jianfei Lu Minli Zhang Hao Wang Dan Xu Liqun Lu 2019Aquaculture and Fisheries2019,4,2:0
