5Natural killer cell-triggered vascular transformation:maternal care before birth?显示文摘Natural killer(NK)cells are found in lymphoid and non-lymphoid organs.In addition to important roles in immune surveillance,some NK cells contribute to angiogenesis and circulatory regulation.The uterus of early pregnancy is a non-lymphoid organ enriched in NK cells that are specifically recruited to placental attachment sites.In species with invasive hemochorial placentation,these uterine natural killer(uNK)cells,via secretion of cytokines,chemokines,mucins,enzymes and angiogenic growth factors,contribute to the physiological change of mesometrial endometrium into the unique stromal environment called decidua basalis.In humans,uNK cells have the phenotype CD56^(bright) CD16^(dim) and they appear in great abundance in the late secretory phase of the menstrual cycle and early pregnancy.Gene expression studies indicate that CD56^(bright) CD16^(dim) uterine and circulating cells are functionally distinct.In humans but not mice or other species with post-implantation decidualization,uNK cells may contribute to blastocyst implantation and are of interest as therapeutic targets in female infertility.Histological and genetic studies in mice first identified triggering of the process of gestation spiral arterial modification as a major uNK cell function,achieved via interferon(IFN)-c secretion.During spiral arterial modification,branches from the uterine artery that traverse the endometrium/decidua transiently lose their muscular coat and ability to vasoconstrict.The expression of vascular markers changes from arterial to venous as these vessels dilate and become low-resistance,high-volume channels.Full understanding of the vascular interactions of human uNK cells is difficult to obtain because endometrial time-course studies are not possible in pregnant women.Here we briefly review key information concerning uNK cell functions from studies in rodents,summarize highlights concerning human uNK cells and describe our preliminary studies on development of a humanized,pregnant mouse model for in vivo investigations of human uNK cell functions.Jianhong Zhang Zhilin Chen Graeme N Smith B.Anne Croy 2011Cellular & Molecular Immunology2011,8,1:19

8Water in the upper mantle and deep crust of eastern China: concentration, distribution and implications显示文摘Understanding the concentration and distribution of water in the Earth's mantle plays a substantial role in studying its chemical, physical and dynamic processes. After a decade of research, a comprehensive dataset of water content in upper-mantle samples has been built for eastern China, which is now the only place with water-content data from such diverse types of natural samples, and provides an integrated picture of the water content and its distribution in the upper mantle at a continental scale. The main findings include the following:(i) the temporal heterogeneity of the water content in the lithospheric mantle from early Cretaceous(~120 Ma) to Cenozoic(<40 Ma) was tightly connected with the stability of the North China Craton(from its destruction to its consolidation);(ii) the heterogeneous water content in the Cenozoic lithospheric mantle beneath different blocks of eastern China was not only inherited from tectonic settings from which they came, but was also affected later by geological processes they experienced;(iii) the distinct water content between the lowermost crust and lithospheric mantle of eastern China and its induced rheological contrast at the base of the crust indicate that the continental crust–mantle boundary could behave either in a coupled or decoupled manner beneath different areas and/or at different stages;(iv) the alkali basalts of eastern China demonstrate a heterogeneous distribution of water content in the mantle;local and regional comparisons of the water content between the lithospheric mantle and basalts' source indicate that the Cenozoic alkali basalts in eastern China were not sourced from the lithospheric mantle.Instead, the inferred high water contents in the mantle sources suggest that the Cenozoic eastern China basalts were likely sourced from the mantle transition zone(MTZ); and(v) both oceanic and continental crusts may carry a certain amount of water back into the deep mantle of eastern China by plate subduction.Such recycled crustal materials have not only created a local water-rich zone, but have also introduced crustal geochemical signatures into the mantle, both accounting for crustal geochemical imprints in the intra-plate magmatic rocks of eastern China.Qun-Ke Xia Jia Liu István Kovács Yan-Tao Hao Pei Li Xiao-Zhi Yang Huan Chen Ying-Ming Sheng 2019National Science Review2019,6,1:16

11Polymorphisms of MTHFD,plasma homocysteine levels, and risk of gastric cancer in ahigh-risk Chinese population显示文摘Purpose:Accumulative evidence suggests that folate has a protective effect on gastric cancer. The methylenetetrahyd-rofolate dlehydrogenase(MTHFD) plays an important role in folate and homocysteine metabolisms, and polymorphisms of MTHFD may result in disturbance of the folate-mediated homocysteine pathway. The aim of this study is to test the hypothesis that genetic variants of MTHFD and plasma homocysteine levels are associated with risk of gastric cancer and modulated by genotypes of methylenetetrahydrofolate reductase(MTHFR). Experimental Design: We genotyped G1958A and T401C in MTHFD and C677T in MTHFR and detected total plasma homocysteine(tHcy) levels in a case-control study of 589 gastric cancer cases and 635 cancer-free controls in a high-risk Chinese population. Results:The variant genotypes of MTHFD 1958AA and 401CC were associated with a significantly increased risk of gastric canceradjusted odds ratio(OR), 2.05; 95% confidence interval(95% CI),1.34-3.13 for 1958AA; adjusted OR,1.43; 95% CI,1.14-1.80 for 401CC compared with 1958GG/GA and 401TT/TC genotypes, respectively. Both of the effects were more evident in the subjects carrying MTHFR 677CT/TT genotypes. The average tHcy level was significantly higher in gastric cancer cases than in controls(P < 0.01), and the upper quartile of tHcy (> 13.6 mu mol/L) was associated with an 82% significantly increased risk of gastric cancer, compared with the lowest quartile of tHcy(<= 8.0 pmol/L;adjusted OR,1.82; 95% CI,1.20-2.75). Conclusions:The strong associations between MTHFD variants and the plasma tHcy levels and gastric cancer risk suggest, for the first time, a possible gene-environment interaction between genetic variants of folate-metabolizing genes and high tHcy levels in gastric carcinogenesis.Wang, L. 12A long-form α-neurotoxin from cobra venom produces potent opioid-independent analgesia显示文摘Aim:In light of the antinociceptive activity of the short-chain neurotoxin,cobrotoxin,and other acetylcholine antagonists,the antinociceptive activity andmechanisms of cobratoxin(CTX),a long-chain postsynaptic α-neurotoxin,wasinvestigated in rodent pain models.Methods:CTX was administered intraperito-neally(30,45,68 μg/kg),intra-cerebral ventricularly(4.5 μg/kg)or microinjectedinto periaqueductal gray(PAG;4.5 μg/kg).The antinociceptive action was testedusing the hot-plate and acetic acid writhing tests m mice and rats.The involve-ment of the cholinergic system and opioid system in CTX-induced analgesia wasexamined by pretreatment of animals with atropine(0.5 mg/kg,im;or 10 mg/kg,ip)or naloxone(1 and 5 mg/kg,ip).The effect of CTX on motor activity was testedusing the Animex test.Results:CTX exhibited a dose-dependent analgesic ac-tion in mice as determined by both the hot-plate and acetic acid writhing tests.The peak effect of analgesia was seen 3 h after administration.In the mouse aceticacid writhing test,the intra-cerebral ventricular administration of CTX at 4.5 μg/kg(1/12th of a systemic dose)produced marked analgesic effects.Microinjection ofCTX(4.5 μgkg)into the PAG region did not elicit an analgesic action in rats in thehot-plate test.Atropine at 0.5 mg/kg(im)and naloxone at 1 and 5 mg/kg(ip)bothfailed to block the analgesic effects of CTX,but atropine at 10 mg/kg(ip)didantagonize the analgesia mediated by CTX in the mouse acetic acid writhing test.Acetylsalicylic acid(300 mg/kg)did not enhance the analgesic effects of CTX.Atthe highest effective dose of 68 μg/kg the neurotoxin did not change the sponta-neous mobility of mice.Conclusion:CTX has analgesic effects,which are medi-ated in the central nervous system though not through the PAG.The centralcholinergic system but not opioid system appears to be involved in theantinociceptive action of CTX.Zhi-xin CHEN Hui-ling ZHANG Zhen-lun GU Bo-wen CHEN Rong HAN Paul F REID Laurence N RAYMOND Zheng-hong QIN 2006Acta Pharmacologica Sinica2006,27,4:7
13Micronuclei in Nasal Mucosa, Oral Mucosa and Lymphocytes in Students Exposed to Formaldehyde Vapor in Anatomy Class显示文摘The frequency of micronuclei (MN) in cells of the nasal mucosa, oral mucosa and in lym-phocytes was evaluated for 25 students in anatomy classes exposed to formaldehyde (FA) overan 8-week period. Each student served as his or her own control. The time-weighted averageconcentration (TWA) of fOrmaldehyde in anatomical laboratories and in students’ dormitorieswas 0. 508 ± 0. 299 mg/m3 and 0. 012 ± 0. 0025mg/m3, respectively. A higher frequency ofmicronuclei was observed in nasal and oral exfoliative cells after formaldehyde exposure (3. 85± 1. 48 vs 1 .20± 0. 676 and 0. 857 ± 0. 558 vs 0. 568 ± 0. 317, paired-t test: P< 0. 001and P < 0. 01, respectively). No significant increase in the frequency of lymphocyte micronu-clei was found after formaldehyde exposure (P >0. 05 ). 14Comparison of eastern paleo-Tethyan ophiolites and its geodynamic significance--Evidence from Dur'ngoi ophiolite显示文摘Nd-Pb isotopic geochemistry of metabasalt from Dur抧goi ophiolite, Qinghai Prov-ince, NW China reveals the presence of DMM mixing with EMII components in its upper mantle source, which is comparable with present Indian MORB. Comparisons indicate that the geo-chemistry, chronology and source characteristics of Dur抧goi ophiolite coincide with Pa-leo-tethyan ophiolites from 揝anjiang?area. 16Neuroprotective effects of INT-777 against Aβ1-42-induced cognitive impairment, neuroinflammation, apoptosis, and synaptic dysfunction in mice显示文摘OBJECTIVE To investigate the neuroprotective effects of TGR5 agonist INT-777 in the Aβ_(1-42)-treated mouse model of acute neurotoxicity.METHODS Cognitive impairment was induced by single intracerebroventricular injection of aggregated Aβ_(1-42)(410 pmol per mouse;5 μL)into the mouse brain in normal ICR mice.After3 d,INT-777(0.75,1.5 or 3.0 μg per mouse) was infused into the same site.Another 3 d later,the cognition function was evaluated by Morris water maze(MWM),novel objective recognition(NOR),and Y maze tests.Furthermore,the levels of TGR5,TNF-α,IL-1β,IL-6,NF-κB p65,Iba1,caspase 3,Bcl-2,Bax,PSD95,and synaptophysin in the hippocampus and frontal cortex were detected using Western blotting.Microglia activation in the hippocampal CA1,CA3 and DG regions and frontal cortex were detected by immunohistochemistry.TUNEL staining was used to detect the apoptotic cells.The number of spines in the hippocampal CA1,CA3 and DG regions and frontal cortex was detected by GolgiCox staining.RESULTS INT-777(1.5 or 3.0 μg per mouse) significantly ameliorated memory impairment in the Aβ_(1-42)-treated mice,evidenced by increased the time spent in the target quadrant(P<0.05),and the number of target crossings(P<0.05) in the MWM test,increased the discrimination index in the NOR test(P<0.05),and increased the number of correct choices(P<0.05,P<0.01) and decreased the latency(P<0.05) to enter the shock-free compartment in the Y-maze test.Importantly,this treatment reversed Aβ_(1-42)-induced TGR5 down-regulation(P<0.05,P<0.01),suppressed the increase of nuclear NF-κB p65(P<0.05,P<0.01) and mitigated neuroinflammation,evidenced by lower proinflammatory cytokines such as TNF-α(P<0.05),IL-1β(P<0.05),IL-6(P<0.05,P<0.01),and less Iba1-positive cells in the hippocampal CA1,CA3 and DG regions and frontal cortex(P<0.05,P<0.01) as well as the protein level of Iba1.INT-777(1.5 or3.0 μg per mouse) also pronouncedly suppressed apoptosis through the reduction of TUNEL-positive cells(P<0.05,P<0.01),decreased activation of caspase 3(P<0.05),increased ratio of Bcl-2/Bax(P<0.05,P<0.01),and ameliorated synaptic dysfunction via promoting dendritic spines generation(P<0.05,P<0.01) with the upregulation of PSD95(P<0.05) and synaptophysin proteins(P<0.05).CONCLUSION INT-777 has potent neuroprotective effects against Aβ_(1-42)-induced neurotoxicity,which suggests that the activation of TGR5 could be a novel and promising strategy for the treatment of AD.WU Xian LYU Yang-ge DU Yi-feng CHEN Fang Miranda N REED HU Mei Vishnu SUPPIRAMANIAM TANG Su-su HONG Hao 2018中国药理学与毒理学杂志2018,32,9:6
17Development of Soybean EST-SSR Markers and Their Use to Assess Genetic Diversity in the Subgenus Soja显示文摘Developing expressed sequence tag-derived SSR(EST-SSR)markers is imperative in genetic research.In this paper,we reported 37 EST-SSR markers which were developed from 286 unigenes obtained from soybean cDNA library.Among the 286 markers designed for the 4 accessions of Glycine max and 6 of its wild progenitor(G.soja)within the subgenus Soja,209 markers amplified DNA fragments,taking 73.1% and 37 markers appeared to be polymorphic,which was 12.9% of the total.The 37 loci detected a total of 142 alleles,while the PIC values varied from 0.194 to 0.794.Both the number of alleles per locus and PIC value were significantly related to the SSR motif.Six EST-SSR loci may be fixed for different alleles between G.max and G.soja since they were particularly polymorphic among the 6 G.soja accessions.A neighbor-joining tree placed the G.max accessions together as a group within the G.soja,though the average genetic distance among G.soja accessions was much higher.These new EST-SSRs markers will be useful for genetic diversity analysis,genetic mapping construction and gene discovery in Soja subgenus.LIU Yu-lin LI Ying-hui ZHOU Guo-an Uzokwe N CHANG Ru-zhen CHEN Shou-yi QIU Li-juan 2010Agricultural Sciences in China2010,9,10:6
18Discovering the roles of subsurface microorganisms: Progress and future of deep biosphere investigation显示文摘The discovery of the marine 'deep biosphere'-microorganisms living deep below the seafloor-is one of the most significant and exciting discoveries since the ocean drilling program began more than 40 years ago. Study of the deep biosphere has become a research frontier and a hot spot both for geological and biological sciences. Here, we introduce the history of the discovery of the deep biosphere, and then we describe the types of environments for life below the seafloor, the energy sources for the living creatures, the diversity of organisms within the deep biosphere, and the new tools and technologies used in this research field. We will highlight several recently completed Integrated Ocean Drilling Program Expeditions, which targeted the subseafloor deep biosphere within the crust and sediments. 19Air quality monitoring using mobile microscopy and machine learning显示文摘Rapid,accurate and high-throughput sizing and quantification of particulate matter(PM)in air is crucial for monitoring and improving air quality.In fact,particles in air with a diameter of≤2.5μm have been classified as carcinogenic by the World Health Organization.Here we present a field-portable cost-effective platform for high-throughput quantification of particulate matter using computational lens-free microscopy and machine-learning.This platform,termed c-Air,is also integrated with a smartphone application for device control and display of results.This mobile device rapidly screens 6.5 L of air in 30 s and generates microscopic images of the aerosols in air.It provides statistics of the particle size and density distribution with a sizing accuracy of~93%.We tested this mobile platform by measuring the air quality at different indoor and outdoor environments and measurement times,and compared our results to those of an Environmental Protection Agency–approved device based on beta-attenuation monitoring,which showed strong correlation to c-Air measurements.Furthermore,we used c-Air to map the air quality around Los Angeles International Airport(LAX)over 24 h to confirm that the impact of LAX on increased PM concentration was present even at 47 km away from the airport,especially along the direction of landing flights.With its machinelearning-based computational microscopy interface,c-Air can be adaptively tailored to detect specific particles in air,for example,various types of pollen and mold and provide a cost-effective mobile solution for highly accurate and distributed sensing of air quality.Yi-Chen Wu Ashutosh Shiledar Yi-Cheng Li Jeffrey Wong Steve Feng Xuan Chen Christine Chen Kevin Jin Saba Janamian Zhe Yang Zachary Scott Ballard Zoltán Göröcs Alborz Feizi Aydogan Ozcan 2017Light(Science & Applications)2017,6,1:5
