| 1 | Distribution and relationship between antimicrobial resistance genes and heavy metals in surface sediments of Taihu Lake,China显示文摘Heavy metals, pharmaceuticals, and other wastes released into the environment can significantly influence environmental antibiotic resistance. We investigated the occurrence of 22 antimicrobial resistance genes(ARGs) and 10 heavy metal concentrations, and the relationship between ARGs and heavy metals in surface sediment from seven sites of Lake Taihu. The results showed significant correlations(p < 0.05) between sediment ARG levels,especially for tetracycline and sulfonamides(e.g., tet(A), tet(D), tet(E), tet(O), sul1, sul2 and int-1) and specific heavy metals(Fe, Mn, Cr, Cu, Zn, among others) in the Lake. In the surface sediments, heavy metals showed an interaction with resistance genes, but the strength of interaction was diminished with increasing depth. For most of the heavy metals, the concentration of elements in the top sediments was higher than that in other depths.Tetracycline resistance genes(tet(A), tet(B), tet(D), tet(E) and tet(O), β-lactam resistance genes(SHV, TEM, CTX, OXA and OXY) and sulfonamide resistance genes(sulA, sul1, sul2, sul3 and int-1) were detected. They showed a trend which inferred a statistically significant increase followed by decreases in the relative abundance of these ARGs(normalized to 16 S rRNA genes) with increasing depth. | 3 | Retention mechanism of calcium ferrite and compositions of ash on selenium during chemical looping gasification显示文摘Selenium pollution by coal utilization is of increasing concern.Calcium-iron(Ca-Fe)oxygen carriers(OCs)and alkali metal ions have strong inhibitory effects on selenium,which can reduce the emissions of selenium vapor.The retention mechanisms of selenium by Fe_(2)O_(3),CaFe_(2)O_(4),Ca_(2)Fe_(2)O_(5) and bottom ash are investigated during chemical looping gasification(CLG).Iron-based OC can oxidize H_(2)Se(g)to SeO_(2)(g);furthermore,lattice oxygen is released by Fe_(2)O_(3),contributing to the formation of an Fe-O-Se structure to retain selenium and form selenite.Because calcium ferrite is poorly oxidizing,it cannot oxidize H_(2)Se(g),but the CaO produced when OCs are reduced can react with H_(2)Se(g)to form CaSe(s),and this process can be promoted by H_(2)S(g).The best retention rates reached 32.301%when Ca_(2)Fe_(2)O_(5) was used.In the cyclic experiment,the selenium retention of the bottom ash gradually increases.Alkali metal ions in bottom ash are the main factor in retaining selenium.Ca^(2+) and Mg^(2+) do not easily vaporize due to their high melting points;therefore,their selenium retention is significantly better than that of K^(+) and Na^(+).This research provided a new idea for the removal of selenium by using OCs and bottom ash par-ticles during CLG. | Ziheng Han Huifen Kang Nini Yuan Xintong Guo Jingjing Ma Qingjie Guo | 2023 | Particuology2023,,8: | 0 |
| 4 | A novel lytic phage potentially effective for phage therapy against Burkholderia pseudomallei in the tropics显示文摘Background:Burkholderia pseudomallei is a tropical pathogen that causes melioidosis.Its intrinsic drug-resistance is a leading cause of treatment failure,and the few available antibiotics require prolonged use to be effective.This study aimed to assess the clinical potential of B.pseudomallei phages isolated from Hainan,China.Methods:Burkholderia pseudomallei strain(HNBPoo1)was used as the isolation host,and phages were recovered from domestic environmental sources,which were submitted to the host range determination,lytic property assays,and stability tests.The best candidate was examined via the transmission electron microscope for classification.With its genome sequenced and analyzed,its protective efficacy against B.pseudomallei infection in A549 cells and Caenorhabditis elegans was evaluated,in which cell viability and survival rates were compared using the one-way ANOVA method and the log-rank test.Results:A phage able to lyse 24/25 clinical isolates was recovered.It was classified in the Podoviridae family and was found to be amenable to propagation.Under the optimal multiplicity of infection(MOl)of o.1,an eclipse period of around 20 min and a high titer(io12 PFU/ml)produced within 1 h were demonstrated.This phage was found stabile at a wide range of temperatures(24,37,40,50,and 60 C)and pH values(3-12).After being designated as vB_BpP_HN01,it was fully sequenced,and the 71,398 bp linear genome,containing 93 open reading frames and a tRNA-Asn,displayed a low sequence similarity with known viruses.Additionally,protective effects of applications of vB_BpP_HNo1(MOl=0.1 and MOl=1)alone or in combination with antibiotics were found to improve viability of infected cells(70.6±6.8%,85.8±5.7%,91.9±1.8%,and 96.8±1.8%,respectively).A significantly reduced mortality(10%)and a decreased pathogen load were demonstrated in infected C.elegans following the addition of this phage.Conclusions:As the frst B.pseudomallei phage was isolated in Hainan,China,phage vB_BpP_HNO1 was characterized by promising lytic property,stability,and effciency of bacterial elimination during the in vitro/vivo experiments.Therefore,we can conclude that it is a potential alternative agent for combating melioidosis. | Yanshuang Wang Xuemiao Li David A.B.Dance Han Xia Chen Chen Nini Luo Anyang Li Yanmei Li Qiao Zhu Qinghui Sun Xingyong Wu Yingfei Zeng Lin Chen Shen Tian Qianfeng Xia | 2022 | Infectious Diseases of Poverty2022,11,4: | 0 |
