Multiple genetic alterations and behavior of cellular biology in gastric cancer and other gastric mucosal lesions:H.pylori infection, histological types and staging显示文摘AIM To investigate the expression of multiplegenes and the behavior of cellular biology ingastric cancer(GC)and other gastric mucosallesions and their relations to Helicobacter pylori(H.pylori)infection, tumor staging andhistological subtypes.METHODS Three hundred and twenty-sevenspecimens of gastric mucosa obtained viaendoscopy or surgical resection, and ABCimmunohistochemical staining were used todetect the expression of p53,p16,Bcl-2 andCOX-2 proteins.H.pylori was determined byrapid urea test combined with pathologicalstaining or14C urea breath test.Cellular image analysis was performed in 66 patients withintestinal metaplasia(IM)and/or dysplasia(Dys). In 30 of them, both cancer and theparacancerous tissues were obtained at the timeof surgery.Histological pattern, tumor staging, lymph node metastasis, grading ofdifferentiation and other clinical data werestudied in the medical records.RESULTS p16 expression of IM or Dys wassignificantly lower in positive H.pylori chronicatrophic gastritis(CAG)than those withnegative H.pylori(CAG:54.8% vs 88.0%,IM:34.4% vs 69.6%,Dys:23.8% vs 53.6%,allP<0.05),Bcl-2 or COX-2 expression of IM orDys in positive H.pylori cases was significantlyhigher than that without H.pylori(Bcl-2:68.8%vs23.9%,90.5% vs 60.7%;COX-2:50.0% vs10.8%,61.8% vs 17.8%;all P<0.05).Themean number of most parameters of cellularimage analysis in positive H.pylori group wassignificantly higher than that in negative H.pylori group(Ellipser:53±14,40±12μm,Area1:748±572,302±202 μm2,Area2:3050±1661,1681±1990 μm2,all P<0.05;Ellipseb:79±23,58±15 μm,Ratio1:22%±5%,13%±4%,Ratio2:79%±17%,53%±20%,all P<0.01).There was significant correlation between Bcl-2and histologic pattern of gastric carcinoma, andbetween COX-2 and tumor staging or lymph nodemetastasis(Bcl-2:75.0% vs 16.7%;COX-2:76.0% vs 20.0%,79.2% vs 16.7%;allP<0.05).CONCLUSION p1l6, Bcl-2, and COX-2 but not p53 gene may play a role in the early genesis/ progression of gastric carcinoma and are associated with H. pylori infection. p53 gene is relatively late event in gastric tumorigenesis and mainly relates to its progression. There is more cellular-biological behavior of malignant tumor in gastric mucosal lesions with H. pylori infection. Aberrant Bcl-2 protein expression appears to be preferentially associated with the intestinal type cancer. COX-2 seems to be related to tumor staging and lymph node metastasis. | Heng Jun Gao Lian Zhen Yu Jian Feng Bai Yan Shen Peng Gu Sun Han Lin Zhao Kun Miu Xiu Zhen Lü Xiao Yong Zhang Zhi Quan Zhao | 2000 | World Journal of Gastroenterology2000,6,6: | 52

Gold nanorods together with HSP inhibitor-VER-155008 micelles for colon cancer mild-temperature photothermal therapy显示文摘Enhancing the heat-sensitivity of tumor cells provides an alternative solution to maintaining the therapeutic outcome of photothermal therapy(PTT). In this study, we constructed a therapeutic system, which was composed of methoxy-polyethylene-glycol-coated-gold-nanorods(MPEG-AuN R) and VER-155008-micelles, to evaluate the effect of VER-155008 on the sensitivity of tumor cells to heat, and further investigate the therapeutic outcome of MPEG-AuN R mediated PTT combined with VER-155008-micelles. VER-155008-micelles downregulate the expression of heat shock proteins and attenuate the heat-resistance of tumor cell. The survival of HCT116 cells treated with VER-155008-micelles under 45 ℃ is equal to that treated with high temperature hyperthermia(55 ℃) in vitro. Furthermore, we proved either the MPEG-AuN R or VER-155008-micelles can be accumulate in the tumor site by photoacoustic imaging and fluorescent imaging. In vivo anti-cancer evaluation showed that tumor size remarkably decreased(smaller than 100 mm^3 or vanished) when treated with combing 45℃ mild PTT system, which contrasted to the tumor size when treated with individual 45℃ mild PTT(around 500nm^3) or normal saline as control(larger than 2000 nm^3). These results proved that the VER-155008-micelles can attenuate the heat-resistance of tumor cells and enhance the therapeutic outcome of mild-temperature photothermal therapy. | Xichuan Tang Liwei Tan Kun Shi Jinrong Peng Yao Xiao Wenting Li Lijuan Chen Qian Yang Zhiyong Qian | 2018 | Acta Pharmaceutica Sinica B2018,8,4: | 12
Diagnostic value of elevated serum carbohydrate antigen 199 level in acute cholangitis secondary to choledocholithiasis显示文摘AIM To investigate the diagnostic value of abnormal serum carbohydrate antigen 199(CA199) level in acute cholangitis secondary to choledocholithiasis.METHODS In this retrospective cohort study, the clinical data of 727 patients with choledocholithiasis admitted to the Third Affiliated Hospital of Zunyi Medical College from June 2011 to June 2017 were collected. Among these patients, 258 patients had secondary acute cholangitis and served as observation group, and the remaining 569 choledocholithiasis patients served as the control group. Serum liver function indexes and tumor markers were detected in both groups, and the receiver operating characteristic(ROC) curves were constructed for markers showing statistical significances. The cutoff value, sensitivity, and specificity of each marker were calculated according to the ROC curves. RESULTS The results of liver function tests showed no significant differences between the two groups(P > 0.05). Tumor markers including serum CA125, CA153, carcinoembryonic antigen, and alpha fetoprotein levels were also not significantly different(P > 0.05); however, the serum CA199 level was significantly higher in the observation group than in the control group(P < 0.05). The ROC curve analysis showed that the area under the curve was 0.885(95%CI: 0.841-0.929) for CA199, and the cutoff value of 52.5 kU/L had the highest diagnostic accuracy, with a sensitivity of 86.8% and a specificity of 81.6%.CONCLUSION Abnormally elevated serum CA199 level has an important value in the diagnosis of acute cholangitis secondary to choledocholithiasis. It may be a specific inflammatory marker for acute cholangitis. | Yong Mei Li Chen Ci-Jun Peng Jun Wang Peng-Fei Zeng Guo-Xing Wang Wen-Ping Li Yan-Qing Luo Chao Du Kai Liu Kun Xiong Kai Leng Chun-Lin Feng Ji-Hu Jia | 2018 | World Journal of Clinical Cases2018,6,11: | 11
Role of macrophages in peripheral nerve injury and repair显示文摘Resident and inflammatory macrophages are essential effectors of the innate immune system. These cells provide innate immune defenses and regulate tissue and organ homeostasis. In addition to their roles in diseases such as cancer, obesity and osteoarthritis, they play vital roles in tissue repair and disease rehabilitation. Macrophages and other inflammatory cells are recruited to tissue injury sites where they promote changes in the microenvironment. Among the inflammatory cell types, only macrophages have both pro-inflammatory (Ml) and anti-inflammatory (M2) actions, and M2 macrophages have four subtypes. The co-action of Ml and M2 subtypes can create a favorable microenvironment, releasing cytokines for damaged tissue repair. In this review, we discuss the activation of macrophages and their roles in severe peripheral nerve injury. We also describe the therapeutic potential of macrophages in nerve tissue engineering treatment and highlight approaches for enhancing M2 cell-mediated nerve repair and regeneration. | Ping Liu Jiang Peng Gong-Hai Han Xiao Ding Shuai Wei Gang Gao Kun Huang Feng Chang Yu Wang | 2019 | Neural Regeneration Research2019,14,8: | 11
Brassinosteroid Enhances Jasmonate-Induced Anthocyanin Accumulation in Arabidopsis Seedlings显示文摘Jasmonate (JA) regulates plant development, mediates defense responses, and induces anthocyanin biosynthesis as well. Previously, we isolated the psc1 mutant that partially suppressed coi1 insensitivity to JA, and found that brassinosteroid (BR) was involved in JA signaling and negatively regulated JA inhibition of root growth in Arabidopsis. In this study it was shown that JA-induced anthocyanin accumulation was reduced in BR mutants or in wild type treated with brassinazole, an inhibitor of BR biosynthesis, whereas it was induced by an application of exogenous BR. It was also shown that the'late'anthocyanin biosynthesis genes including DFR, LDOX , and UF3GT, were induced slightly by JA in the BR mutants relative to wild type. Furthermore, the expression level of JA-induced Myb/bHLH transcription factors such as PAP1, PAP2, and GL3, which are components of the WD-repeat/Myb/bHLH transcriptional complexes that mediate the 'late' anthocyanin biosynthesis genes, was lower in the BR mutants than that in wild type. These results suggested that BR affects JA-induced anthocyanin accumulation by regulating the 'late' anthocyanin biosynthesis genes and this regulation might be mediated by the WD-repeat/Myb/bHLH transcriptional complexes. | Zhihong Peng Chengyun Han Libing Yuan Kun Zhang Hongman Huang Chunmei Ren | 2011 | Journal of Integrative Plant Biology2011,53,8: | 9
On the Key Dynamical Processes Supporting the 21.7 Zhengzhou Record-breaking Hourly Rainfall in China显示文摘An extremely heavy rainfall event occurred in Zhengzhou,China,on 20 July 2021 and produced an hourly rainfall rate of 201.9 mm,which broke the station record for China's Mainland.Based on radar observations and a convection-permitting simulation using the WRF-ARW model, this paper investigates the multiscale processes, especially those at the mesoscale, that support the extreme observed hourly rainfall. Results show that the extreme rainfall occurred in an environment characteristic of warm-sector heavy rainfall, with abundant warm moist air transported from the ocean by an abnormally northward-displaced western Pacific subtropical high and Typhoon In-Fa(2021).However,rather than through back building and echo training of convective cells often found in warm-sector heavy rainfall events, this extreme hourly rainfall event was caused by a single,quasi-stationary storm in Zhengzhou.Scale separation analysis reveals that the extreme-rainproducing storm was supported and maintained by the dynamic lifting of low-level converging flows from the north, south, and east of the storm. The low-level northerly flow originated from a mesoscale barrier jet on the eastern slope of the Taihang Mountain due to terrain blocking of large-scale easterly flows, which reached an overall balance with the southerly winds in association with a low-level meso-β-scale vortex located to the west of Zhengzhou. The large-scale easterly inflows that fed the deep convection via transport of thermodynamically unstable air into the storm prevented the eastward propagation of the weak, shallow cold pool.As a result, the convective storm was nearly stationary over Zhengzhou, resulting in record-breaking hourly precipitation. | Peng WEI Xin XU Ming XUE Chenyue ZHANG Yuan WANG Kun ZHAO Ang ZHOU Shushi ZHANG Kefeng ZHU | 2023 | Advances in Atmospheric Sciences2023,40,3: | 9
Effects of yeast and yeast cell wall polysaccharides supplementation on beef cattle growth performance, rumen microbial populations and lipopolysaccharides production显示文摘This experiment was conducted to investigate the effects of live yeast and yeast cell wall polysaccharides on growth performance, rumen function and plasma lipopolysaccharides(LPS) content and immunity parameters of beef cattle.Forty Qinchuan cattle were randomly assigned to one of four treatments with 10 replicates in each treatment. The dietary treatments were: control diet(CTR),CTR supplemented with 1 g live yeast(2×10^10 live cell g^-1 per cattle per day(YST1),CTR supplemented with 2 g live yeast per cattle per day(YST2) and CTR supplemented with 20 g of yeast cell wall polysaccharides(30.0%≤β-glucan≤35.0%,and 28.0%≤mannanoligosaccharide≤32.0%) per cattle per day(YCW).The average daily gain was higher(P=0.023) and feed conversion ratio was lower(P=0.042) for the YST2 than the CTR.The digestibility of neutral detergent fiber(P=0.039) and acid detergent fiber(P=0.016) were higher in yeast supplemented groups. The acetic acid:propionic acid of the YST2 was lower compared with the CTR(P=0.033).Plasma LPS(P=0.032),acute phase protein haptoglobin(P=0.033),plasma amyloid A(P=0.015) and histamine(P=0.038) were lower in the YST2 compared with the CTR.The copies of fibrolytic microbial populations such as Fibrobacter succinogenes S85,Ruminococcus albus 7 and Ruminococcus flavefaciens FD-1 of the YST2 were higher(P<0.001),while the copies of typical lactate producing bacteria Streptococcus bovis JB1 was lower(P<0.001) compared with the CTR.Little differences were observed between the CTR,YST1 and YCW in growth performance, ruminal fermentation characteristics, microbial populations, immunity indices and total tract nutrient digestibility. It is concluded that the YST2 could promote fibrolytic microbial populations, decrease starch-utilizing bacteria, reduce LPS production in the rumen and LPS absorption into plasma and decrease inflammatory parameters, which can lead to an improvement in growth performance in beef cattle. | PENG Quan-hui CHENG Long KANG Kun Tian Gang Mohammad Al-Mamun XUE Bai WANG Li-zhi ZOU Hua-wei Mathew Gitau Gicheha WANG Zhi-sheng | 2020 | Journal of Integrative Agriculture2020,19,3: | 9
Radiomics for the detection of microvascular invasion in hepatocellular carcinoma显示文摘Hepatocellular carcinoma(HCC)is the most common primary liver cancer, accounting for about 90%of liver cancer cases. It is currently the fifth most common cancer in the world and the third leading cause of cancer-related mortality. Moreover, recurrence of HCC is common.Microvascular invasion(MVI)is a major factor associated with recurrence in postoperative HCC.It is difficult to evaluate MVI using traditional imaging modalities.Currently,MVI is assessed primarily through pathological and immunohistochemical analyses of postoperative tissue samples.Needle biopsy is the primary method used to confirm MVI diagnosis before surgery. As the puncture specimens represent just a small part of the tumor, and given the heterogeneity of HCC,biopsy samples may yield false-negative results.Radiomics,an emerging, powerful, and non-invasive tool based on various imaging modalities, such as computed tomography, magnetic resonance imaging, ultrasound, and positron emission tomography, can predict the HCC-MVI status preoperatively by delineating the tumor and/or the regions at a certain distance from the surface of the tumor to extract the image features.Although positive results have been reported for radiomics, its drawbacks have limited its clinical translation. This article reviews the application of radiomics, based on various imaging modalities,in preoperative evaluation of HCC-MVI and explores future research directions that facilitate its clinical translation. | Kun Lv Xin Cao Peng Du Jun-Yan Fu Dao-Ying Geng Jun Zhang | 2022 | World Journal of Gastroenterology2022,28,20: | 9
Comparison of Two Methods to Assess the Size Structure of Phytoplankton Community Assemblages, in Liaodong Bay, China显示文摘Liaodong Bay is one of the largest semi-enclosed bays located in Bohai Sea,in northeast China. It is an important feeding and spawning place, even wintering and breeding migration base for many marine organisms. The size structure of phytoplankton communities is a key feature of marine ecosystems and can be used as an indicator for algae disaster, but the methods to evalute is still challenging. Here,we compared the size-fractionated chlorophyll-a method with a high-throughout sequencing method in an evaluation of the size structure of phytoplankton assemblages in Liaodong Bay,China,in 2014.The proportion of picophytoplankton was estimated at just 19%according to the results based on size-fractionated chlorophyll-a,which likely represents a severe underestimation. The sequencing method confirmed that many kinds of phytoplankton species appeared in more than one size-fractionated filter membrane, with some even dominating where the filter membrane pore size was greater than the individual phytoplankton size.According to the sequencing method, the phytoplankton assemblages in Liaodong Bay were dominated by picophytoplankton(average proportion=44%),followed by the nanophytoplankton(average 32%),a composition that is more consistent with other studies. The sequencing method provides a more robust way to assess the size structure and taxonomic diversity of marine phytoplankton communities. | SONG Lun WU Jing DU Jing LI Nan WANG Kun WANG Peng | 2019 | Journal of Ocean University of China2019,18,5: | 8
Combination of serum gamma-glutamyltransferase and alkaline phosphatase in predicting the diagnosis of asymptomatic choledocholithiasis secondary to cholecystolithiasis显示文摘BACKGROUND Gamma-glutamyltransferase(GGT) is one of the most important laboratory tests for the evaluation of liver damage. Through a long-term clinical observation of patients with secondary asymptomatic choledocholithiasis, we found that most patients had abnormal GGT serum levels.AIM To investigate the combination of serum GGT and alkaline phosphatase(ALP) in predicting the diagnosis of asymptomatic choledocholithiasis secondary to cholecystolithiasis.METHODS In this retrospective cohort study, the clinical data of 829 patients with cholecystolithiasis admitted to the Third Affiliated Hospital of Zunyi Medical College from August 2014 to August 2017 were collected. Among these patients,151 patients had secondary asymptomatic choledocholithiasis and served as the observation group, and the remaining 678 cholecystolithiasis patients served as the control group. Serum liver function indexes were detected in both groups, and the receiver operating characteristic(commonly known as ROC) curves were constructed for markers showing statistical significances. The cutoff value, sensitivity, and specificity of each marker were calculated according to the ROC curves.RESULTS The overall incidence of asymptomatic choledocholithiasis secondary to cholecystolithiasis was 18.2%. The results of liver function indexes including serum aspartate aminotransferase, alanine aminotransferase, direct bilirubin and total bilirubin levels showed no significant differences between the two groups(P> 0.05). However, the serum GGT and ALP levels were significantly higher in the observation group than in the control group(P < 0.05). The ROC curve analysis showed that the area under the curve was 0.881(95%CI: 0.830-0.932), 0.647(95%CI: 0.583-0.711) and 0.923(0.892-0.953) for GGT, ALP, and GGT + ALP,respectively. The corresponding cut-off values of GGT and ALP were 95.5 U/L and 151.5 U/L, sensitivity were 90.8% and 65.1%, and specificity were 83.6% and59.8%, respectively. The sensitivity and specificity of GGT + ALP were 93.5% and85.1%, respectively.CONCLUSION An abnormally elevated serum GGT level has an important value in the diagnosis of asymptomatic choledocholithiasis secondary to cholecystolithiasis. The combination of serum GGT and ALP has better diagnostic performance. As a convenient, rapid and inexpensive test, it should be applied in secondary asymptomatic choledocholithiasis routine screening. | Yong Mei Li Chen Peng-Fei Zeng Ci-Jun Peng Jun Wang Wen-Ping Li Chao Du Kun Xiong Kai Leng Chun-Lin Feng Ji-Hu Jia | 2019 | World Journal of Clinical Cases2019,7,2: | 8
Trial of small gateroad pillar in top coal caving longwall mining of large mining height显示文摘Coal seams in Tashan Mine of Datong Coal Group in China average 15 m thick and have been mined by the top coal caving longwall mining method of large mining height. Mining height was 3.8 m and the top coal caving height was 11.2 m. The gateroad pillar between panels was 38 m. During retreat mining, serious bumps occurred in the gateroads on both sides of the pillar affecting safety production. Therefore, pillarless mining was experimented. Using numerical modeling and comparative study of cases of similar mining condition,it was decided to employ a 6 m wide pillar, rather than the previous 38 m wide pillar.Support system for the gateroads was designed and implemented. During gateroad development, pillar failure conditions and entry deformation were monitored. Hydraulic fracturing method was employed to cut off the K3 sandstone along the entry rib so as to reduce the abutment pressure induced during retreat mining. Support reinforcement method combining grouting and advanced reinforcement methods was proposed to insure stable gateroad ahead of mining. Methane drainage and nitrogen injection were implemented to eliminate hazards associated with mine fire and spontaneous combustion. Since the development of gateroad has just completed, and retreat mining has not begun, the effectiveness of the proposed methods is unknown at this point. However, monitoring will continue until after mining. The results will be published in a separate paper. | Li Huamin Syd Peng Li Huigui Xu Yongxiang Yuan Ruifu Yue Shuaishuai Li Kun | 2016 | International Journal of Mining Science and Technology2016,26,1: | 8

Genome-wide variants of Eurasian facial shape differentiation and a prospective model of DNA based face prediction显示文摘It is a long-standing question as to which genes define the characteristic facial features among different ethnic groups. In this study, we use Uyghurs, an ancient admixed population to query the genetic bases why Europeans and Han Chinese look different. Facial traits were analyzed based on high-dense 3D facial images; numerous biometric spaces were examined for divergent facial features between European and Han Chinese, ranging from inter-landmark distances to dense shape geometrics. Genome-wide association studies(GWAS) were conducted on a discovery panel of Uyghurs. Six significant loci were identified, four of which, rs1868752, rs118078182, rs60159418 at or near UBASH3B, COL23A1, PCDH7 and rs17868256 were replicated in independent cohorts of Uyghurs or Southern Han Chinese. A prospective model was also developed to predict 3D faces based on top GWAS signals and tested in hypothetic forensic scenarios. | Lu Qiao Yajun Yang Pengcheng Fu Sile Hu Hang Zhou Shouneng Peng Jingze Tan Yan Lu Haiyi Lou Dongsheng Lu Sijie Wu Jing Guo Li Jin Yaqun Guan Sijia Wang Shuhua Xu Kun Tang | 2018 | Journal of Genetics and Genomics2018,45,8: | 7
