2Histological Subtypes of Lung Cancer in Chinese Males from 2000 to 2012显示文摘Objective To characterize the histological and epidemiological features of male lung cancer patients in China. Methods The demographic and histological information about male lung cancer patients identified from 2000-01-01 to 2012-12-31,was collected from the Cancer Hospital of the Chinese Academy of Medical Sciences. Relative frequencies(RF) were estimated for major histological subtypes and compared according to the years of diagnosis and birth. Results The RF of adenocarcinoma(ADC) increased from 21.96% to 43.36% and the RF of squamous cell carcinoma(SCC) decreased from 39.11% to 32.23% from 2000 to 2012 in the 15 427 male lung cancer patients included in this study(Z=17.909,P<0.0001; Z=-6.117,P<0.0001). The RF of ADC increased from 28.72% in 2000-2004,36.88% in 2005-2008 to 48.61% in 2009-2012 in patients born after 1960. The age-adjusted RF of ADC in 2007-2012 increased consistently in all the investigated areas. Conclusion The increased RF of ADC in male lung cancer patients highlights the need for further investigation of the etiologic factors of these tumors. 3Soil pH impact on microbial tetraether lipids and terrestrial input index (BIT) in China显示文摘The distributions of microbial tetraether lipids i.e. glycerol dialkyl glycerol tetraethers (GDGTs) were investigated in a variety of Chinese soils featured by a wide range of pH values in different climate zones. The bacterial branched GDGTs and archaeal isoprenoid GDGTs were found to be widely distributed in Chinese soils and their relative abundance was controlled primarily by soil pH. The bacterial bGDGTs usually dominate over archaeal iGDGTs in abundance when the soil pH is below 8.0. However archaeal iGDGTs become dominant and the abundance of halophilic archaea characterized by GDGT V without cyclopentyl ring increases dramatically in the alkaline soil with pH greater than 8.0. The relative abundance of archaeal iGDGTs to bacterial bGDGTs was defined here as RAI index. The RAI index decreases with elevated soil pH and a linear correlation is found between them indicating that the relative abundance of archaeal iGDGTs to bacterial bGDGTs increases with increased pH. The RAI index thus could be a novel proxy of the pH values of paleosol. The terrestrial input index BIT (Branched and Isoprenoid Tetraethers Index) decreases with increased soil pH suggesting that soil pH should be taken into consideration when the terrestrial contribution to marine sediments is estimated by BIT index. 4Human placenta-derived mesenchymal stem cells loaded on linear ordered collagen scaffold improves functional recovery after completely transected spinal cord injury in canine显示文摘Traumatic spinal cord injury(SCI) is a major challenge in the clinic. In this study, we sought to examine the synergistic effects of linear ordered collagen scaffold(LOCS) and human placenta-derived mesenchymal stem cells(hPMSCs) when transplanted into completely transected beagle dogs. After 36 weeks observation, we found that LOCS+hPMSCs implants promoted better hindlimb locomotor recovery than was observed in the non-treatment(control) group and LOCS group. Histological analysis showed that the regenerated tissue after treatment was well integrated with the host tissue, and dramatically reduced the volume of cystic and chondroitin sulfate proteoglycans(CSPGs) expression. Furthermore, the LOCS+hPMSCs group also showed more neuron-specific βIII-tubulin(Tuj-1)-and NeuN-positive neurons in the lesion area, as well as axonal regeneration, remyelination and synapse formation in the lesion site. Additionally, dogs in the LOCS+hPMSCs group experienced enhanced sprouting of both ascending(CGRP-positive) sensory fibers and descending(5-HT-and TH-positive) motor fibers at the lesion area. All these data together suggested that the combined treatment had beneficial effects on neuronal regeneration and functional improvement in a canine complete transection model. 5Identification of a Newly Isolated Getah Virus in the China-Laos Border,China显示文摘In this study, we isolated a virus strain(YN12031) from specimens of Armigeres subalbatus collected in the China-Laos border. BHK-21 cells infected with YN12031 exhibited an evident cytopathic effect(CPE) 32 h post-infection. The virus particles were spherical, 70 nm in diameter, and enveloped; they also featured surface fibers. Molecular genetic analysis revealed that YN12031 was closely related to alpha viruses such as Chikungunya virus and Sindbis virus, and located in the same clade as MM2021, the prototype of Getahvirus(GETV) isolated in Malaysia in 1955. Phylogenetic analysis of the E2 and capsid genes further revealed that YN12031 was located in the same clade as the Russian isolate LEIV/16275/Mag. Analysis of the homology of nucleotides and amino acids in the coding area and E2 gene demonstrated that the YN12031 isolated from the China-Laos border(tropical region) was related closest to the LEIV/16275/Mag isolate obtained in Russia(North frigid zone area) among other isolates studied. These results suggest that GETV can adapt to different geographical environments to propagate and evolve. 6Features of intestinal T-cell lymphomas in Chinese population without evidence of celiac disease and their close association with Epstein-Barr virus infection显示文摘Background Intestinal T-cell lymphoma (ITCL) is a heterogeneous lymphoid neoplastic group with variable clinical and pathological features. ITCL in oriental countries is different from enteropathy-type intestinal T-cell lymphoma (ETCL) in relation to celiac disease and Epstein-Barr virus (EBV). The objective of this study was to investigate the clinicopathological features, immunophenotype, expression of cytotoxic molecule (TIA-1), T-cell receptor (TCR)-γ gene rearrangement, and Epstein-Barr virus (EBV) latent infection in primary ITCL without celiac disease in Chinese.Methods The clinical data of 42 patients were analyzed, and the patients were followed up. Compared with human reactive lymphoid tissues, in situ hybridization for EBER1/2, polymerase chain reaction for TCR-γ gene rearrangement, and immunohistochemical staining for immunophenotypes, TIA-1 and EBV latent membrane proteins (LMP-1) were investigated. Survival curves of different clinicopathological features, immuno-phenotypes, expression of LMP1, TCR-γ gene rearrangement and therapy were analyzed.Results Three fourths of the patients suffered from ITCL in China were men with a peak age incidence in the 4th decade. Common presenting features included fever and hemotochezia. The prognosis was poor with a median survival of 3.0 months. The lesions were mostly localized in the ileocecum and colon. About 38/42 (90.5%) patients demonstrated pleomorphic medium-sized on large cells. Histological features of celiac disease were rarely seen. All 42 patients with ITCL revealed CD45RO positive. Neoplastic cells partially expressed T-cell differentiated antigens (CD3ε, CD4, CD8) and NK cell associated antigen (CD56). The positive frequency of CD3ε, CD4, CD8 and CD56 was 28/42 (66.7%), 7/42 (16.7%), 10/42 (23.8%) and 12/42 (28.6%) respectively. Thirty-nine cells (92.9%) expressed TIA-1, but none expressed CD20 and CD68. More than half of the patients (64.3%, 64.3% and 59.5%) revealed TCR-γ gene rearrangement by three different TCR-γ primers respectively. EBER1/2 was detected in 41 (97.6%) of the 42 patients. The expression frequency of LMP-1 was 38.1% (16/42).Conclusions Primary ITCL without celiac disease in Chinese is a special highly EBV-associated clinicopathological entity. There are few similarities in patients with celiac disease in western countries. 7Healing time of incision infection after hepatobiliary surgery treated by needle-free incision suture closure显示文摘AIM:To compare the effectiveness of needle-free incision suture closure with butterfly tape and traditional secondary suturing techniques in treating incision infection.METHODS:Two hundred and twenty-three patients with incision infection following hepatobiliary surgery at a tertiary hospital were randomly divided into three groups:90 patients were closed by needle-free incision suture closure,which gradually closed the incision wound when drainage from incision infection was visibly decreased and healthy granulation tissues had grown;79 patients were closed by butterfly bandage;another 54 patients were closed by traditional secondary suturing technique.Healing time of incision infection was calculated from the beginning of dressing change to the healing of the incision.RESULTS:Healing time in the needle-free incision suture closure group(24.2±7.2 d)was significantly shorter than that in the butterfly bandage group(33.3±11.2 d)and the traditional secondary suturing group(36.2±15.3 d)(P<0.05).Healing time in the butterfly bandage group appeared to be slightly shorter than that in the secondary suture group,but the difference was not statistically significant(P>0.05).CONCLUSION:Needle-free incision suture closure could gradually close the infection wound at the same time of drainage and dressing change,thereby shortening the healing time.Wen-Jie Ma Yong Zhou Hui Mao Rui-Hua Xu Anuj Shrestha Fu-Yu Li Alex Lorance Qin Yang Yong-Qiong Zhang Ting Jiang Huan Feng Wei Zhang Nan-Sheng Cheng 2014World Journal of Gastroenterology2014,20,42:8
8Energy-efficient power allocation for non-regenerative OFDM relay links显示文摘We consider a two-hop non-regenerative (amplify-and-forward) relay link with one source, one destination and one relay, in which orthogonal frequency division multiplexing (OFDM) is used. The power allocation (PA) to maximize the energy efficiency for the whole link is investigated under quality of service (Qos) require- ment and separate power constraints. An approximation method with low complexity is first proposed to convert the PA problem into a quasi-concave problem, which can obtain a tight lower bound in high SNR region. Furthermore, a more accurate approximation method with higher complexity is raised, which can approach the actual performance both in the low SNR and high SNR regions. We also introduce subcarrier pairing scheme to further enhance the energy efficiency for the link. 9Low WT1 transcript levels at diagnosis predicted poor outcomes of acute myeloid leukemia patients with t(8;21) who received chemotherapy or allogeneic hematopoietic stem cell transplantation显示文摘Background:Acute myeloid leukemia(AML) with t(8;21) is a heterogeneous disease.Identifying AML patients with t(8;21) who have a poor prognosis despite achieving remission is important for determining the best subsequent therapy.This study aimed to evaluate the impact of Wilm tumor gene-1(WT1) transcript levels and cellular homolog of the viral oncogene v-KIT receptor tyrosine kinase(C-KIT) mutations at diagnosis,and RUNXTRUNX1T1 transcript levels after the second consolidation chemotherapy cycle on outcomes.Methods:Eighty-eight AML patients with t(8;21) who received chemotherapy only or allogeneic hematopoietic stem cell transplantation(allo-HSCT) were included.Patients who achieved remission,received two or more cycles of consolidation chemotherapy,and had a positive measureable residual disease(MRD) test result(defined as <3-log reduction in RUNX1-RUNX1T1 transcript levels compared to baseline) after 2-8 cycles of consolidation chemotherapy were recommended to receive allo-HSCT.Patients who had a negative MRD test result were recommended to receive further chemotherapy up to only 8 cycles.WT1 transcript levels and C-KIT mutations at diagnosis,and RUNX1-RUNX1T1 transcript levels after the second consolidation chemotherapy cycle were tested.Results:Patients who had a C-KIT mutation had significantly lower WTl transcript levels than patients who did not have a C-KIT mutation(6.7%± 10.6%vs.19.5%± 19.9%,P < 0.001).Low WTl transcript levels(<5.0%) but not C-KIT mutation at diagnosis,a positive MRD test result after the second cycle of consolidation chemotherapy,and receiving only chemotherapy were independently associated with high cumulative incidence of relapse in all patients(hazard ratio[HR]= 3.53,2.30,and 11.49;95%confidence interval[CI]1.64-7.62,1.82-7.56,and 4.43-29.82;P = 0.002,0.034,and <0.001,respectively);these conditions were also independently associated with low leukemia-free survival(HR =3.71,2.33,and 5.85;95%CI 1.82-7.56,1.17-4.64,and 2.75-12.44;P < 0.001,0.016,and <0.001,respectively) and overall survival(HR = 3.50,2.32,and 4.34;95%CI 1.56-7.82,1.09-4.97,and 1.98-9.53;P = 0.002,0.030,and <0.001,respectively) in all patients.Conclusions:Testing for WTl transcript levels at diagnosis in patients with AML and t(8;21) may predict outcomes in those who achieve remission.A randomized study is warranted to determine whether allo-HSCT can improve prognosis in these patients.Ya-Zhen Qin Yu Wang Hong-Hu Zhu Robert Peter Gale Mei-Jie Zhang Qian Jiang Hao Jiang Lan-Ping Xu Huan Chen Xiao-Hui Zhang Yan-Rong Liu Yue-Yun Lai Bin Jiang Kai-Yan Liu Xiao-Jun Huang 2016Chinese Journal of Cancer2016,35,7:7

13Substrate stiffness modulates bone marrow-derived macrophage polarization through NF-κB signaling pathway显示文摘The stiffness of the extracellular matrix(ECM)plays an important role in regulating the cellular programming.However,the mechanical characteristics of ECM affecting cell differentiation are still under investigated.Herein,we aimed to study the effect of ECM substrate stiffness on macrophage polarization.We prepared polyacrylamide hydrogels with different substrate stiffness,respectively.After the hydrogels were confirmed to have a good biocompatibility,the bone marrow-derived macrophages(BMMs)from mice were incubated on the hydrogels.With simulated by the low substrate stiffness,BMMs displayed an enhanced expression of CD86 on the cell surface and production of reactive oxygen species(ROS)in cells,and secreted more IL-1βand TNF-αin the supernatant.On the contrary,stressed by the medium stiffness,BMMs expressed more CD206,produced less ROS,and secreted more IL-4 and TGF-β.In vivo study by delivered the hydrogels subcutaneously in mice,more CD68+CD86+cells around the hydrogels with the low substrate stiffness were observed while more CD68+CD206+cells near by the middle stiffness hydrogels.In addition,the expressions of NIK,phosphorylated p65(pi-p65)and phosphorylated IκB(pi-IκB)were significantly increased after stimulation with low stiffness in BMMs.Taken together,these findings demonstrated that substrate stiffness could affect macrophages polarization.Low substrate stiffness promoted BMMs to shift to classically activated macrophages(M1)and the middle one to alternatively activated macrophages(M2),through modulating ROS-initiated NF-κB pathway.Therefore,we anticipated ECM-based substrate stiffness with immune modulation would be under consideration in the clinical applications if necessary.Mimi Chen Yu Zhang Pinghui Zhou Xingzhi Liu Huan Zhao Xichao Zhou Qiaoli Gu Bin Li Xuesong Zhu Qin Shi 2020Bioactive Materials2020,5,4:7
141^H NMR-based metabolomics approach to investigating the renal protective effects of Genipin in diabetic rats显示文摘Diabetic nephropathy is one of the various complications of diabetes mellitus, affecting patients for lifetime. Earlier studies have revealed that genipin can not only improve diabetes, but also induce cytotoxicity. Therefore, it is not clear which effect of genipin on kidneys occurs, when it is used in the treatment of diabetes. In the present study, we performed nuclear magnetic resonance(NMR)-based metabolomics analysis of urine and kidney tissue samples obtained from diabetic rats to explore the change of endogenous metabolites associated with diabetes and concomitant kidney disease. Nine significant differential metabolites that were closely related to renal function were screened. They were mainly related to three metabolic pathways: synthesis and degradation of ketone bodies, glycine, serine and threonine metabolism, and butanoate metabolism, which are involved in methylamine metabolism, energy metabolism and amino acid metabolism. In addition, after the intervention of genipin, the metabolic levels of all the metabolites tended to be normal, indicating a protective effect of genipin on kidneys. Growing demand from the user community for a small beam and related experimental methods have motivated upgrades of the devices in the endstation.Minibeam modes have already been developed for operation. A self-integrated diffractometer reduces the sphere of confusion of the rotatory axis to 1μm. The new diffractometer is equipped with an upgraded on-axis viewing system that can improve the resolving power. Additionally,the area detector was also upgraded to the newest generation of detectors, the EIGER X 16 M, which can collect data at 133 Hz. After these upgrades, the endstation became virtually new. 18Pollen-based Holocene quantitative temperature reconstruction on the eastern Tibetan Plateau using a comprehensive method framework显示文摘Quantitative climate reconstruction on long timescales can provide important insights for understanding the climate variability and providing valuable data for simulations. Unfortunately, the credibility of some attempts was hampered by incomplete reconstruction procedures. We here establish a comprehensive framework resting on high-quality Chinese modern pollen database, including modern pollen data screening, calibration set selection, major climate factor analysis, appropriate model selection, strict statistical assessment of results and ecological interpretation. The application of this framework to three high-resolution pollen records from the eastern Tibetan Plateau allows accurate quantitative inferences of Holocene temperature changes, which is the major control of regional vegetation. The results show that the mean warmest month temperature(MTwa)during the early Holocene was ca. 10.4℃ and reached the highest value at 8.5–6 ka BP(ca. 11℃). The early and mid-Holocene(11–5 ka BP) warmth was followed by 1.2℃ temperature decrease, culminating in the coolest temperatures of the Holocene during the Neoglacial cooling. Superimposing on the general cooling trend, MTwareveals a significant 500-yr periodicity with varying intensities through time, showing that warm(cold) intervals are in phase with solar maxima(minima) periods. 20The VP2 protein of grass carp reovirus(GCRV) expressed in a baculovirus exhibits RNA polymerase activity显示文摘The double-shelled grass carp reovirus(GCRV) is capable of endogenous RNA transcription and processing. Genome sequence analysis has revealed that the protein VP2, encoded by gene seg-ment 2(S2), is the putative RNA-dependent RNA polymerase(RdRp). In previous work, we have ex-pressed the functional region of VP2 that is associated with RNA polymerase activity(denoted as rVP2390-900) in E. coli and have prepared a polyclonal antibody against VP2. To characterize the GCRV RNA polymerase, a recombinant full-length VP2(rVP2) was first constructed and expressed in a baculovirus system, as a fusion protein with an attached His-tag. Immunofluorescence(IF) assays, together with immunoblot(IB) analyses from both expressed cell extracts and purified His-tagged rVP2, showed that rVP2 was successfully expressed in Sf9 cells. Further characterization of the replicase activity showed that purified rVP2 and GCRV particles exhibited poly(C)-dependent poly(G) polymerase activity. The RNA enzymatic activity required the divalent cation Mg2+, and was optimal at 28 °C.
