| 5 | Machining characteristics of complex prism pattern on electroplated roll by copper显示文摘The BLU (back light unit) is the core component of the LCD for notebook, mobile-phone, navigation, as well as large sized TV, PID (public information display), etc. In order to enhance optical efficiency of LCD, optical films with the uniform prism patterns have been used for BLU by stacking two films up orthogonally. In this case, light interference-phenomenon occurred such as Morie, wet-out, u-turning, etc. It caused several problems such as low brightness, spots and stripes in LCD. Recently, the high-luminance micro complex prism patterns are actively studied to avoid the light interference-phenomenon and enhance the optical efficiency. In this study, the roll master to manufacture complex micro prism pattern film was machined by using the high precision lathe. The machined patterns on the roll master were 50, 45, 40, 35, 30, 25, 20, 15, 10 and 5 μm in the pitch with 25.0, 22.5, 20.0, 17.5, 15.0, 12.5, 10.0, 7.5, 5.0 and 2.5 μm in the peak height, respectively. The roll was 2 000 mm in length and 320 mm in diameter. The electroplated roll by copper and the natural single crystal diamond tool was used for machining the patterns. The cutting force was measured and analyzed for each cutting condition by using the dynamometer. | 6 | Improvement of machining quality of copper-plated roll mold by controlling temperature variation显示文摘Micro prism film used in LCD industry can be manufactured by roll to roll method with copper-plated roll mold. As copper-plated roll mold is getting larger, pitch error is getting severer. The pitch error drops the quality of micro prism film. The main cause of the pitch error was investigated during machining large roll mold whose machined length was 1 200 mm. The temperature of machining system was elevated during machining roll mold, and this elevation induced thermal expansion of the system. The temperature variation around the roll mold also made thermal expansion of the roll mold. The amount of thermal expansion had strong relationship to the amount of pitch error. Therefore, the roll mold was machined after warming-up of machining system and precise temperature controller around copper-plated roll mold was installed, which minimized the temperature variation. | 7 | Effects of exogenous recombinant human bone morphogenic protein-7 on the corneal epithelial mesenchymal transition and fibrosis显示文摘AIM:To evaluate the effect of exogenous recombinant human bone morphogenic protein-7(rhBMP-7)on transforming growth factor-β(TGF-β)-induced epithelial mesenchymal cell transition(EMT)and assessed its antifibrotic effect via topical application.METHODS:The cytotoxic effect of rhBMP-7 was evaluated and the EMT of human corneal epithelial cells(HECEs)was induced by TGF-β. HECEs were then cultured in the presence of rhBMP-7 and/or hyaluronic acid(HA). EMT markers,fibronectin,E-cadherin,α-smooth muscle actin(α-SMA),and matrix metaloproteinase-9(MMP-9),were evaluated. The level of corneal fibrosis and the reepithelization rate were evaluated using a rabbit keratectomy model. Expression of α-SMA in keratocytes were quantified following treatment with different concentrations of rhBMP-7.RESULTS:Treatment with rhBMP-7 attenuated TGF-β-induced EMT in HECEs. It significantly attenuated fibronectin secretion(31.6%; P<0.05),the α-SMA protein level(72.2%; P<0.01),and MMP-9 expression(23.6%,P<0.05)in HECEs compared with cells grown in the presence of TGF-β alone. E-cadherin expression was significantly enhanced(289.7%; P<0.01)in the presence of rhBMP-7. Topical application of rhBMP-7 combined with 0.1% HA significantly reduced the amount of α-SMA^+ cells by 43.18%(P<0.05)at a concentration of 2.5 μg/mL and by 47.73%(P<0.05)at 25 μg/mL,compared with the control group,without disturbing corneal reepithelization.CONCLUSION:rhBMP-7 attenuates TGF-β-induced EMT in vitro,and topical application of rhBMP-7 reduces keratocyte myodifferentiation during the early wound healing stages in vivo without hindering reepithelization. | 10 | Sequential disruption of ALV host receptor genes reveals no sharing of receptors between ALV subgroups A, B, and J显示文摘Background: Previously, we showed that targeted disruption of viral receptor genes in avian leukosis virus(ALV)subgroups using clustered regularly interspaced short palindromic repeats(CRISPR)/CRISPR-associated protein 9(Cas9))-based genome editing confers resistance to ALV subgroups B and J. Here, we used the same strategy to target the receptor expressed by ALV subgroup A(TVA) and generate chicken cells resistant to infection by this virus.Results: CRISPR/Cas9-based disruption of exon 2 within the tva gene of DF-1 fibroblasts conferred resistance to infection by ALV subgroup A regardless of whether frameshift mutations were introduced during editing. Conversely,overexpression of the wild-type TVA receptor(wtTVA) by tva-modified DF-1 clones restored susceptibility to ALV subgroup A. The results confirm that exon 2, which contains the low-density lipoprotein receptor class A domain of TVA, is critical for virus entry. Furthermore, we sequentially modified DF-1 cells by editing the tva, tvb, and Na^+/H^+ exchange 1(chNHE1) genes, which are the specific receptors for ALV subgroups A, B, and J, respectively.Conclusions: Simultaneous editing of multiple receptors to block infection by different subgroups of ALV confirmed that ALV subgroups A, B, and J do not share host receptors. | 11 | Room-temperature sputtered electrocatalyst WSe2 nanomaterials for hydrogen evolution reaction显示文摘The low-temperature physical vapor deposition process of atomically thin two-dimensional transition metal dichalcogenide(2D TMD) has been gaining attention owing to the cost-effective production of diverse electrochemical catalysts for hydrogen evolution reaction(HER) applications. We, herein, propose a simple route toward the cost-effective physical vapor deposition process of 2D WSe2 layered nanofilms as HER electrochemical catalysts using RF magnetron sputtering at room temperature(<27℃). By controlling the variable sputtering parameters, such as RF power and deposition time, the loading amount and electrochemical surface area(ECSA) of WSe2 films deposited on carbon paper can be carefully determined. The surface of the sputtered WSe2 films are partially oxidized, which may cause spherical-shaped particles. Regardless of the loading amount of WSe2, Tafel slopes of WSe2 electrodes in the HER test are narrowly distributed to be ~120–138 mV dec-1, which indicates the excellent reproducibility of intrinsic catalytic activity. By considering the trade-off between the loading amount and ECSA, the best HER performance is clearly observed in the 200 W-15 min sample with an overpotential of 220 mV at a current density of 10 mA cm-2. | 14 | Development of novel gene carrier using modified nano hydroxyapatite derived from equine bone for osteogenic differentiation of dental pulp stem cells显示文摘Hydroxyapatite(HA)is a representative substance that induces bone regeneration.Our research team extracted nanohydroxyapatite(EH)from natural resources,especially equine bones,and developed it as a molecular biological tool.Polyethylenimine(PEI)was used to coat the EH to develop a gene carrier.To verify that PEI is well coated in the EH,we first observed the morphology and dispersity of PEI-coated EH(pEH)by electron microscopy.The pEH particles were well distributed,while only the EH particles were not distributed and aggregated.Then,the existence of nitrogen elements of PEI on the surface of the pEH was confirmed by EDS,calcium concentration measurement and fourier transform infrared spectroscopy(FT-IR).Additionally,the pEH was confirmed to have a more positive charge than the 25 kD PEI by comparing the zeta potentials.As a result of pGL3 transfection,pEH was better able to transport genes to cells than 25 kD PEI.After verification as a gene carrier for pEH,we induced osteogenic differentiation of DPSCs by loading the BMP-2 gene in pEH(BMP-2/pEH)and delivering it to the cells.As a result,it was confirmed that osteogenic differentiation was promoted by showing that the expression of osteopontin(OPN),osteocalcin(OCN),and runt-related transcription factor 2(RUNX2)was significantly increased in the group treated with BMP-2/pEH.In conclusion,we have not only developed a novel nonviral gene carrier that is better performing and less toxic than 25 kD PEI by modifying natural HA(the agricultural byproduct)but also proved that bone differentiation can be effectively promoted by delivering BMP-2 with pEH to stem cells. | Myung Chul Lee Hoon Seonwoo Kyoung Je Jang Shambhavi Pandey Jaewoon Lim Sangbae Park Jae Eun Kim Yun-Hoon Choung Pankaj Garg Jong Hoon Chung | 2021 | Bioactive Materials2021,6,9: | 2 |
