1A core-shell structured COYID-19 mRNA vaccine with favorable biodistribution pattern and promising immunity显示文摘Although inoculation of COVID-19 vaccines has rolled out globally,there is still a critical need for safe and effective vaccines to ensure fair and equitable supply for all countries.Here,we report on the development of a highly efficacious mRNA vaccine,SW0123 that is composed of sequence-modified mRNA encoding the full-length SARS-CoV-2 Spike protein packaged in core-shell structured lipopolyplex(LPP)nanoparticles.SWOT 23 is easy to produce using a large-scale microfluidics-based apparatus.The unique core-shell structured nanoparticle facilitates vaccine uptake and demonstrates a high colloidal stability,and a desirable biodistribution pattern with low liver targeting effect upon intramuscular administration.Extensive evaluations in mice and nonhuman primates revealed strong immunogenicity of SW0123,represented by induction of Th1-polarized T cell responses and high levels of antibodies that were capable of neutralizing not only the wild-type SARS-CoV-2,but also a panel of variants including D614G and N501Y variants.In addition,SW0123 conferred effective protection in both mice and non-human primates upon SARS-CoV-2 challenge.Taken together,SW0123 is a promising vaccine candidate that holds prospects for further evaluation in humans.Ren Yang Yao Deng Baoying Huang Lei Huang Ang Lin Yuhua Li Wenling Wang Jingjing Liu Shuaiyao Lu Zhenzhen Zhan Yufei Wang Ruhan A Wen Wang Peihua Niu Li Zhao Shiqiang Li Xiaopin Ma Luyao Zhang Yujian Zhang Weiguo Yao Xingjie Liang Jincun Zhao Zhongmin Liu Xiaozhong Peng Hangwen Li Wenjie Tan 2021Signal Transduction and Targeted Therapy2021,6,6:3
2Control measures during the COVID-19 outbreak reduced the transmission of hand,foot,and mouth disease显示文摘Control measures during the coronavirus disease 2019(COVID-19)outbreak may have limited the spread of infectious diseases.This study aimed to analyze the impact of COVID-19 on the spread of hand,foot,and mouth disease(HFMD)in China.A mathematical model was established to fit the reported data of HFMD in six selected cities in China's Mainland from 2015 to 2020.The absolute difference(AD)and relative difference(RD)between the reported incidence in 2020,and simulated maximum,minimum,or median incidence of HFMD in 2015-2019 were calculated.The incidence and R effof HFMD have decreased in six selected cities since the outbreak of COVID-19,and in the second half of 2020,the incidence and R effof HFMD have rebounded.The results show that the total attack rate(TAR)in 2020 was lower than the maximum,minimum,and median TAR fitted in previous years in six selected cities(except Changsha City).For the maximum,median,minimum fitted TAR,the range of RD(%)is 42·20-99·20%,36·35-98·41%48·35-96·23%(except Changsha City)respectively.The preventive and control measures of COVID-19 have significantly contributed to the containment of HFMD transmission.Yan Niu Li Luo Jia Rui Shiting Yang Bin Deng Zeyu Zhao Shengnan Lin Jingwen Xu Yuanzhao Zhu Yao Wang Meng Yang Xingchun Liu Tianlong Yang Weikang Liu Peihua Li Zhuoyang Li Chan Liu Jiefeng Huang Tianmu Chen 2021Journal of Safety Science and Resilience2021,2,2:3

4Increased intracellular calcium concentration causes electrical turbulence in guinea pig ventricular myocytes显示文摘Dysregulation of intracellular Ca2+ homeostasis is associated with various pathological conditions and arrhythmogenesis of the heart.The objective of this study was to investigate the effects of an acute increase in intracellular Ca2+ concentration ([Ca2+] i) on the electrophysiology of ventricular myocytes by mimicking intracellular Ca 2+ overload.The [Ca2+] i was clamped to either a controlled (65-100 nmol L-1) or increased (1 μmol L-1) level.The transmembrane action potentials and ionic currents were recorded using whole-cell patch clamp techniques.We found that the acute increase in [Ca2+] i shortened the action potential duration,reduced the action potential amplitude,maximum depolarization velocity and resting membrane potential,caused delayed after-depolarizations (DADs),and triggered activity--compared with these parameters in the control.The increased [Ca2+] i augmented late I Na in a time-dependent manner,reduced ICaL and IK1,and increased IKr but not IKs.The results of this study can be used to explain calcium overload-induced ventricular arrhythmias.FAN XinRong MA JiHua WAN Wei ZHANG PeiHua WANG Chao WU Lin 2011Science China(Life Sciences)2011,54,3:1

7A K^(+)-sensitive AND-gate dual-mode probe for simultaneous tumor imaging and malignancy identification显示文摘Although molecular imaging probes have the potential to non-invasively diagnose a tumor,imaging probes that can detect a tumor and simultaneously identify tumor malignancy remain elusive.Here,we demonstrate a potassium ion(K^(+))sensitive dual-mode nanoprobe(KDMN)for non-invasive tumor imaging and malignancy identification,which operates via a cascaded‘AND’logic gate controlled by inputs of magnetic resonance imaging(MRI)and fluorescence imaging(FI)signals.We encapsulate commercial K^(+)indicators into the hollow cavities of magnetic mesoporous silica nanoparticles,which are subsequently coated with a K^(+)-selective membrane that exclusively permits the passage of K^(+)while excluding other cations.The KDMN can readily accumulate in tumors and enhance the MRI contrast after systemic administration.Spatial information of the tumor lesion is thus accessible via MRI and forms the first layer of the‘AND’gate.Meanwhile,the KDMN selectively captures K^(+)and prevents interference from other cations,triggering a K^(+)-activated FI signal as the second layer of the‘AND’gate in the case of a malignant tumor with a high extracellular K^(+)level.This dual-mode imaging approach effectively eliminates false positive or negative diagnostic results and allows for non-invasive imaging of tumor malignancy with high sensitivity and accuracy.Qiyue Wang Fangyuan Li Zeyu Liang Hongwei Liao Bo Zhang Peihua Lin Xun Liu Shen Hu Jiyoung Lee Daishun Ling 2022National Science Review2022,9,7:0
8Modelling COVID-19 outbreak on the Diamond Princess ship using the public surveillance data显示文摘The novel coronavirus disease 2019(COVID-19)outbreak on the Diamond Princess(DP)ship has caused over 634 cases as of February 20,2020.We model the transmission process on DP ship as a stochastic branching process,and estimate the reproduction number at the innitial phase of 2.9(95%CrI:1.7e7.7).The epidemic doubling time is 3.4 days,and thus timely actions on COVID-19 control were crucial.We estimate the COVID-19 transmissibility reduced 34%after the quarantine program on the DP ship which was implemented on February 5.According to the model simulation,relocating the population at risk may sustainably decrease the epidemic size,postpone the timing of epidemic peak,and thus relieve the tensive demands in the healthcare.The lesson learnt on the ship should be considered in other similar settings.Shi Zhao Peihua Cao Daozhou Gao Zian Zhuang Weiming Wang Jinjun Ran Kai Wang Lin Yang Mohammad R.Einollahi Yijun Lou Daihai He Maggie H.Wang 2022Infectious Disease Modelling2022,7,2:0
9Experiment on microscale flames stabilized on surface of annular porous media显示文摘The characteristics of the surface-stabilized flame on a porous medium in an annular microscale channel were studied experimentally with methane/air mixture.In the pressure drop characteristic study,three porous media with the same pore diameter and different porosities were used and compared.The substitution of the air for the mixture was verified accurately at first.Then the relationship expression of the air pressure drop characteristic was acquired for each porous medium according to the Forcheimer correlation.The third porous medium was selected for the following research,as it had the expression closer to the Darcy's law.As for the ignition characteristic study of the surface-stabilized flame,a reasonable criterion was established for the flammability limit determination.Based on the criterion,the result showed that the limit reached the minimum value when the mixture equivalence ratio was 0.9.During the extinction characteristic study,through the well-designed operating procedure,the experiment was conducted to obtain the strict extinction limit of the surfacestabilized flame.It was indicated that the extinction limit at mixture equivalence ratio of 0.8 was the least.LIN Peihua LIN Yuzhen XUE Xin ZHANG Liang ZHANG Chi 2017航空动力学报2017,32,12:0
