1Helicobacter pylori upregulates prion protein expression in gastric mucosa: A possible link to prion disease显示文摘AIM: Pathological prion protein (PrPSC) is responsible for the development of transmissible spongiform encephalopathies (TSE). While PrPc enters the organism via the oral route, less data is available to know about its uptake and the role of gastrointestinal inflammation on the expression of prion precursor PrPc, which is constitutively expressed in the gastric mucosa.METHODS: We studied PrPc expression in the gastric mucosa of 10 Helicobacter pylori-positive patients before and after successful H pylori eradication compared to non-infected controls using RT-PCR and Western blotting.The effect of central mediators of gastric inflammation,i.e., gastrin, prostaglandin E2 (PGE2), tumor necrosis factor alpha (TNF-α) and interleukin 1 beta (IL-1β) on PrPc expression was analyzed in gastric cell lines.RESULTS: PrPc expression was increased in H pyloriinfection compared with non-infected controls and decreased to normal after successful eradication. Gastrin,PGE2, and IL-1β dose-dependently upregulated PrPc in gastric cells, while TNF-α had no effect.CONCLUSION: H pylori infection leads to the upregulation of gastric PrPc expression. 2Perioperative chemotherapy for advanced gastric cancer - results from a tertiary-care hospital in Germany显示文摘BACKGROUND Neoadjuvant/perioperative chemotherapy is the recommended treatment for advanced stages of gastric cancer(>T2,N+)before tumour resection in many European guidelines.However,there is no consensus as to whether perioperative chemotherapy is as effective in distal as in proximal tumours,in addition to a relevant uncertainty concerning appropriate treatment modalities for elderly patients.AIM To investigate the role of perioperative chemotherapy in advanced gastric cancer in patients from a German tertiary clinic with respect to efficacy,localisation,and age.METHODS We performed a retrospective analysis of 158 patients from our clinic with adenocarcinoma of the stomach or the gastroesophageal junction who underwent resection between 2008 and 2016.The data were evaluated particularly in relation to patient age,tumour site,and perioperative therapy.RESULTS Administration of perioperative chemotherapy did not lead to a significant survival advantage in our study population.The 5-year survival rates were 40%for patients who received perioperative chemotherapy and 29%for the group without perioperative chemotherapy(P=0.125).Our patients were on average distinctly older than patients in most of the published randomised controlled trials.Patients elder than 75 years received perioperative chemotherapy far less frequently.Patients with a proximal tumour received perioperative chemotherapy much more often.CONCLUSION This analysis reconfirms our previous data concerning the effectiveness of perioperative chemotherapy for advanced gastric cancer.There is reasonable doubt that the quality of the existing randomized controlled trials is sufficient to generally justify perioperative chemotherapy in patients with advanced gastric cancer independent of tumour localization or age.Katrin Bauer Giulia Manzini Doris Henne-Bruns Peter Buechler 2020World Journal of Gastrointestinal Oncology2020,12,5:4

7Highlights of the 2nd International Symposium on Tribbles and Diseases: tribbles tremble in therapeutics for immunity, metabolism, fundamental cell biology and cancer显示文摘The Tribbles(TRIB) family of pseudokinase proteins has been shown to play key roles in cell cycle, metabolic diseases, chronic inflammatory disease, and cancer development. A better understanding of the mechanisms of TRIB pseudokinases could provide new insights for disease development and help promote TRIB proteins as novel therapeutic targets for drug discovery. At the 2 nd International Symposium on Tribbles and Diseases held on May 7–9, 2018 in Beijing, China, a group of leading Tribbles scientists reported their findings and ongoing studies about the effects of the different TRIB proteins in the areas of immunity, metabolism, fundamental cell biology and cancer. Here, we summarize important and insightful overviews from 4 keynote lectures, 13 plenary lectures and 8 short talks that took place during this meeting. 12How Do Stomata Sense Reductions in Atmospheric Relative Humidity?显示文摘Dear Editor, It has been known since the work of Francis Darwin that,in response to a reduction in atmospheric relative humidity (rh),stomatal aperture decreases.Screening for Arabidopsis mutants compromised in stomatal responses to reduced rh resulted in the identification of two genes,OST1 and ABA2,that are involved in stomatal response to low rh conditions.Interestingly both encode proteins previously known to be involved in ABA signaling (Xie et al.,2006,and references therein).These findings strongly suggested that,at least in part,the stomatal response to low rh is mediated by ABA and the intracellular ABA signaling pathway.Our most recent data show that low rh-induced stomatal closure can proceed by guard cell autonomous ABA synthesis (Bauer et al.,2013),suggesting that there is significant overlap between the guard cell low air humidity-induced signaling pathway and the guard cell ABA signaling pathway.Such convergence has been noted before (Xie et al.,2006,and references therein) and the example of Open STomata 1 (OST1/SRK2E)that encodes a serine-threonine protein kinase and the SLow Anion Channel SLAC1 may represent a common mechanism for most other closure-inducing signals,too.Hubert Bauer Peter Ache Florian Wohlfart Khaled A.S.
