| 4 | Terahertz wave generation from ring-Airy beam induced plasmas and remote detection by terahertz-radiation- enhanced-emission-of-fluorescence: a review显示文摘With the increasing demands for remote spectroscopy in many fields ranging from homeland security to environmental monitoring, terahertz (THz) spectroscopy has drawn a significant amount of attention because of its capability to acquire chemical spectral signatures non-invasively. However, advanced THz remote sensing techniques are obstructed by quite a few factors, such as THz waves being strongly absorbed by water vapor in the ambient air, difficulty to generate intense broadband coherent THz source remotely, and hard to transmit THz waveform information remotely without losing the signal to noise ratio, etc. In this review, after introducing different THz air-photonics techniques to overcome the difficulties of THz remote sensing, we focus mainly on theoretical and experimental methods to improve THz generation and detection performance for the purpose of remote sensing through tailoring the generation and detection media, air-plasma. For the THz generation part, auto-focusing ring-Airy beam was introduced to enhance the THz wave generation yield from two-color laser induced air plasma. By artificially modulated exotic wave packets, it is exhibited that abruptly auto-focusing beam induced air-plasma can give an up to 5.3-time-enhanced THz wave pulse energy compared to normal Gaussian beam induced plasma under the same conditions. At the same time, a red shift on the THz emission spectrum is also observed. A simulation using an interference model to qualitatively describe these behaviors has be developed. For the THz detection part, the results of THz remote sensing at 30 m using THz-radiation-enhanced-emission- of-fluorescence (THz-REEF) technique are demonstrated, which greatly improved from the 10 m demonstration last reported. The THz-REEF technique in the counter-propagation geometry was explored, which is proved to be more practical for stand-off detections than co-propagation geometry. We found that in the counter- propagating geometry the maximum amplitude of the REEF signal is comparable to that in the co-propagating case, whereas the time resolved REEF trace significantly changes. By performing the study with different plasmas, we observed that in the counter-propagating geometry the shape of the REEF trace depends strongly on the plasma length and electron density. A new theoretical model suggesting that the densest volume of the plasma does not contribute to the fluorescence enhancement is proposed to reproduce the experimental measurements. | 13 | IGF2-NR4A2 Signaling Regulates Macrophage Subtypes to Attenuate Liver Cirrhosis显示文摘Background and Aims:Liver cirrhosis can lead to liver failure and eventually death.Macrophages are the main contributors to cirrhosis and have a bidirectional role in regulating matrix deposition and degradation.Macrophage-based cell therapy has been developed as an alternative to liver transplantation.However,there is insufficient evidence regarding its safety and efficacy.In this study,we aimed to explore the effect of combining insulin-like growth factor 2(IGF2)with bone marrow-derived macrophages(BMDMs)to treat mice with liver cirrhosis.Methods:We assessed liver inflammation,fibrosis regression,liver function,and liver regeneration in mice with CCl4-induced cirrhosis and treated with BMDM only or IGF2+BMDM.We performed in vitro experiments in which activated hepatic stellate cells(HSCs)were co-cultured with macrophages in the presence or absence of IGF2.The polarity of macrophages and the degree of inhibition of HSCs were examined.The effect of IGF2 on macrophages was also verified by the overexpression of IGF2.Results:Combining IGF2 with BMDM reduced liver inflammation and fibrosis and increased hepatocyte proliferation.Combining IGF2 with BMDM was more effective than using BMDM alone.In vitro experiments demonstrated that IGF2 inhibited HSCs activation by upregulating NR4A2 to promote the anti-inflammatory macrophages phenotype.IGF2 also increased the synthesis of matrix metalloproteinases(MMPs)by macrophages,which may explain why administering IGF2 combined with BMDM was more effective than administering BMDM only.Conclusions:Our study provides a theoretical basis for the future use of BMDM-based cell therapy to treat liver cirrhosis. | Lichao Yao Xue Hu Mengqin Yuan Qiuling Zhang Pingji Liu Lian Yang Kai Dai Yingan Jiang | 2023 | Journal of Clinical and Translational Hepatology2023,11,4: | 0 |
| 14 | Gene-guided OX40L anchoring to tumor cells for synergetic tumor“self-killing”immunotherapy显示文摘The low objective response rates and severe side effects largely limit the clinical outcomes of immune checkpoint blockade(ICB)therapy.Here,a tumor“self-killing”therapy based on gene-guided OX40L anchoring to tumor cell membrane was reported to boost ICB therapy.We developed a highly efficient delivery system HA/PEI-KT(HKT)to co-deliver the OX40L plasmids and unmethylated CG-enriched oligodeoxynucleotide(CpG).On the one hand,CpG induced the expression of OX40 on T cells within tumors.On the other hand,OX40L plasmids achieved the OX40L anchoring on the tumor cell membrane to next promote T cells responses via OX40/OX40L axis.Such synergistic tumor“self-killing”strategy finally turned“cold”tumors to“hot”,to sensitize tumors to programmed cell death protein 1/programmed cell death ligand 1(PD-1/PD-L1)blockade therapy,and promoted an immune-mediated tumor regression in both B16F10 and 4T1 tumor models,with prevention of tumor recurrence and metastasis.To avoid the side effects,the gene-guided OX40L anchoring and PD-L1 silencing was proposed to replace the existing antibody therapy,which showed negligible toxicity in vivo.Our work provided a new possibility for tumor“self-killing”immunotherapy to treated various solid tumors. | Lin Lin Yingying Hu Zhaopei Guo Jie Chen Pingjie Sun Huayu Tian Xuesi Chen | 2023 | Bioactive Materials2023,,7: | 0 |
