Association of Overlapped and Un-overlapped Comorbidities with COVID-19 Severity and Treatment Outcomes: A Retrospective Cohort Study from Nine Provinces in China显示文摘Objective Several COVID-19 patients have overlapping comorbidities. The independent role of each component contributing to the risk of COVID-19 is unknown, and how some non-cardiometabolic comorbidities affect the risk of COVID-19 remains unclear. Methods A retrospective follow-up design was adopted. A total of 1,160 laboratory-confirmed patients were enrolled from nine provinces in China. Data on comorbidities were obtained from the patients’ medical records. Multivariable logistic regression models were used to estimate the odds ratio(OR) and 95% confidence interval(95% CI) of the associations between comorbidities(cardiometabolic or non-cardiometabolic diseases), clinical severity, and treatment outcomes of COVID-19.Results Overall, 158(13.6%) patients were diagnosed with severe illness and 32(2.7%) had unfavorable outcomes. Hypertension(2.87, 1.30–6.32), type 2 diabetes(T2 DM)(3.57, 2.32–5.49),cardiovascular disease(CVD)(3.78, 1.81–7.89), fatty liver disease(7.53, 1.96–28.96), hyperlipidemia(2.15, 1.26–3.67), other lung diseases(6.00, 3.01–11.96), and electrolyte imbalance(10.40, 3.00–26.10)were independently linked to increased odds of being severely ill. T2 DM(6.07, 2.89–12.75), CVD(8.47,6.03–11.89), and electrolyte imbalance(19.44, 11.47–32.96) were also strong predictors of unfavorable outcomes. Women with comorbidities were more likely to have severe disease on admission(5.46,3.25–9.19), while men with comorbidities were more likely to have unfavorable treatment outcomes(6.58, 1.46–29.64) within two weeks. Conclusion Besides hypertension, diabetes, and CVD, fatty liver disease, hyperlipidemia, other lung diseases, and electrolyte imbalance were independent risk factors for COVID-19 severity and poor treatment outcome. Women with comorbidities were more likely to have severe disease, while men with comorbidities were more likely to have unfavorable treatment outcomes. | MA Yan ZHU Dong Shan CHEN Ren Bo SHI Nan Nan LIU Si Hong FAN Yi Pin WU Gui Hui YANG Pu Ye BAI Jiang Feng CHEN Hong CHEN Li Ying FENG Qiao GUO Tuan Mao HOU Yong HU Gui Fen HU Xiao Mei HU Yun Hong HUANG Jin HUANG Qiu Hua HUANG Shao Zhen JI Liang JIN Hai Hao LEI Xiao LI Chun Yan LI Min Qing LI Qun Tang LI Xian Yong LIU Hong De LIU Jin Ping LIU Zhang MA Yu Ting MAO Ya MO Liu Fen NA Hui WANG Jing Wei SONG Fang Li SUN Sheng WANG Dong Ting WANG Ming Xuan WANG Xiao Yan WANG Yin Zhen WANG Yu Dong WU Wei WU Lan Ping XIAO Yan Hua XIE Hai Jun XU Hong Ming XU Shou Fang XUE Rui Xia YANG Chun YANG Kai Jun YUAN Sheng Li ZHANG Gong Qi ZHANG Jin Bo ZHANG Lin Song ZHAO Shu Sen ZHAO Wan Ying ZHENG Kai ZHOU Ying Chun ZHU Jun Teng ZHU Tian Qing ZHANG Hua Min WANG Yan Ping WANG Yong Yan 无 | 2020 | Biomedical and Environmental Sciences2020,33,12: | 8
QTL Mapping for Important Agronomic Traits in Synthetic Hexaploid Wheat Derived from Aegiliops tauschii ssp. tauschii显示文摘Aegiliops tauschii is classified into two subspecies: Ae. tauschii ssp. tauschii and Ae. tauschii ssp. strangulata. Novel genetic variations exist in Ae. tauschii ssp. tauschii that can be utilized in wheat improvement. We synthesized a hexaploid wheat genotype(SHW-L1) by crossing an Ae. tauschii ssp. tauschii accession(AS60) with a tetraploid wheat genotype(AS2255). A population consisting of 171 F8 recombinant inbred lines was developed from SHW-L1 and Chuanmai 32 to identify QTLs associated with agronomic traits. A new genetic map with high density was constructed and used to detect the QTLs for heading date, kernel width, spike length, spikelet number, and thousand kernel weight. A total of 30 putative QTLs were identified for five investigated traits. Thirteen QTLs were located on D genomes of SHW-L1, six of them showed positive effect on agronomic traits. Chromosome region flanked by wPt-6133–wPt-8134 on 2D carried five environment-independent QTLs. Each QTL accounted for more than 10% phenotypic variance. These QTLs were highly consistent across environments and should be used in wheat breeding. | YU Ma CHEN Guo-yue ZHANG Lian-quan LIU Ya-xi LIU Deng-cai WANG Ji-rui PU Zhi-en ZHANG Li LAN Xiu-jin WEI Yu-ming LIU Chun-ji ZHENG You-liang | 2014 | Journal of Integrative Agriculture2014,13,9: | 6
Pseudomonas aeruginosa:pathogenesis, virulence factors, antibiotic resistance, interaction with host, technology advances and emerging therapeutics显示文摘Pseudomonas aeruginosa(P.aeruginosa)is a Gram-negative opportunistic pathogen that infects patients with cystic fibrosis, burn wounds, immunodeficiency, chronic obstructive pulmonary disorder(COPD),cancer, and severe infection requiring ventilation, such as COVID-19.P.aeruginosa is also a widely-used model bacterium for all biological areas. In addition to continued, intense efforts in understanding bacterial pathogenesis of P.aeruginosa including virulence factors(LPS,quorum sensing,two-component systems,6 type secretion systems, outer membrane vesicles(OMVs),CRISPR-Cas and their regulation),rapid progress has been made in further studying host-pathogen interaction, particularly host immune networks involving autophagy, inflammasome, noncoding RNAs,cGAS,etc.Furthermore, numerous technologic advances, such as bioinformatics,metabolomics,scRNA-seq,nanoparticles,drug screening, and phage therapy, have been used to improve our understanding of P.aeruginosa pathogenesis and host defense.Nevertheless, much remains to be uncovered about interactions between P.aeruginosa and host immune responses, including mechanisms of drug resistance by known or unannotated bacterial virulence factors as well as mammalian cell signaling pathways. The widespread use of antibiotics and the slow development of effective antimicrobials present daunting challenges and necessitate new theoretical and practical platforms to screen and develop mechanism-tested novel drugs to treat intractable infections, especially those caused by multi-drug resistance strains.Benefited from has advancing in research tools and technology, dissecting this pathogen’s feature has entered into molecular and mechanistic details as well as dynamic and holistic views.Herein,we comprehensively review the progress and discuss the current status of P.aeruginosa biophysical traits, behaviors, virulence factors, invasive regulators, and host defense patterns against its infection, which point out new directions for future investigation and add to the design of novel and/or alternative therapeutics to combat this clinically significant pathogen. | Shugang Qin Wen Xiao Chuanmin Zhou Qinqin Pu Xin Deng Lefu Lan Haihua Liang Xiangrong Song Min Wu | 2022 | Signal Transduction and Targeted Therapy2022,7,7: | 5
A highly potent and stable pan-coronavirus fusion inhibitor as a candidate prophylactic and therapeutic for COVID-19 and other coronavirus diseases显示文摘The development of broad-spectrum antivirals against human coronaviruses(HCoVs)is critical to combat the current coronavirus disease 2019(COVID-19)pandemic caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)and its variants,as well as future outbreaks of emerging CoVs.We have previously identified a polyethylene glycol-conjugated(PEGylated)lipopeptide,EK1C4,with potent pan-CoV fusion inhibitory activity. However,PEG linkers in peptide or protein drugs may reduce stability or induce anti-PEG antibodies in vivo. Therefore,we herein report the design and synthesis of a series of dePEGylated lipopeptide-based pan-CoV fusion inhibitors featuring the replacement of the PEG linker with amino acids in the heptad repeat 2 C-terminal fragment(HR2-CF)of HCoV-OC43.Among these lipopeptides,EKL1C showed the most potent inhibitory activity against infection by SARS-CoV-2 and its spike(S)mutants,as well as other HCoVs and some bat SARS-related coronaviruses(SARSr-CoVs)tested.The dePEGylated lipopeptide EKL1C exhibited significantly stronger resistance to proteolytic enzymes, better metabolic stability in mouse serum, higher thermostability than the PEGylated lipopeptide EK1C4,suggesting that EKL1C could be further developed as a candidate prophylactic and therapeutic for COVID-19 and other coronavirus diseases. | Jie Zhou Wei Xu Zezhong Liu Chao Wang Shuai Xia Qiaoshuai Lan Yanxing Cai Shan Su Jing Pu Lixiao Xing Youhua Xie Lu Lu Shibo Jiang Qian Wang | 2022 | Acta Pharmaceutica Sinica B2022,12,4: | 5
Detection of the Urinary Biomarkers PYD, CTX-Ⅱ, and DPD in Patients with Kashin-Beck Disease in the Qinghai Province of China显示文摘Kashin-Beck disease(KBD) is an endemic degenerative osteoarthropathy of uncertain etiology. The aim of our study was to identify changes in C-telopeptide of type Ⅱ collagen(CTX-Ⅱ), pyridinoline(PYD), and deoxypyridinoline(DPD) among KBD patients. 54 KBD patients and 78 healthy controls were included this study. Urinary samples were collected and measured by ELISA. The median quantities of PYD, CTX-Ⅱ, and DPD of KBD patients were 1107.73 ng/μmol.cre, 695.11 ng/μmol.cre, and 1342.34 pml/μmol.cre, while the median quantities of healthy controls were 805.59 ng/μmol.cre, 546.47 ng/μmol.cre, and 718.15 pml/μmol.cre, respectively. The differences between KBD patients and healthy controls were statistically significant(Z = 4.405, 3.653, and 3.724; P < 0.001). The higher levels of PYD, CTX-Ⅱ, and DPD detected in KBD patients indicate that they could be used as biomarkers of KBD. | ZHAO Zhi Jun PU Guang Lan ZHAN Pei Zhen LI Qiang WU Chun Ning WANG Li Hua | 2017 | Biomedical and Environmental Sciences2017,30,5: | 5
A fiber optoacoustic guide with augmented reality for precision breastconserving surgery显示文摘Lumpectomy, also called breast-conserving surgery, has become the standard surgical treatment for early-stage breast cancer. However, accurately locating the tumor during a lumpectomy, especially when the lesion is small and nonpalpable,is a challenge.Such difficulty can lead to either incomplete tumor removal or prolonged surgical time, which result in high re-operation rates(~25%)and increased surgical costs. Here,we report a fiber optoacoustic guide(FOG)with augmented reality(AR)for sub-millimeter tumor localization and intuitive surgical guidance with minimal interference. The FOG is preoperatively implanted in the tumor.Under external pulsed light excitation, the FOG omnidirectionally broadcasts acoustic waves through the optoacoustic effect by a specially designed nano-composite layer at its tip.By capturing the acoustic wave, three ultrasound sensors on the breast skin triangulate the FOG tip’s position with 0.25-mm accuracy. An AR system with a tablet measures the coordinates of the ultrasound sensors and transforms the FOG tip’s position into visual feedback with<1-mm accuracy, thus aiding surgeons in directly visualizing the tumor location and performing fast and accurate tumor removal. We further show the use of a head-mounted display to visualize the same information in the surgeons’first-person view and achieve hands-free guidance.Towards clinical application,a surgeon successfully deployed the FOG to excise a“pseudo tumor”in a female human cadaver. With the high-accuracy tumor localization by FOG and the intuitive surgical guidance by AR,the surgeon performed accurate and fast tumor removal, which will significantly reduce re-operation rates and shorten the surgery time. | Lu Lan Yan Xia Rui Li Kaiming Liu Jieying Mai Jennifer Anne Medley Samilia Obeng-Gyasi Linda K.Han Pu Wang Ji-Xin Cheng | 2018 | Light(Science & Applications)2018,7,1: | 4
Beta-and Novel Delta-Coronaviruses Are Identified from Wild Animals in the Qinghai-Tibetan Plateau, China显示文摘Outbreaks of severe virus infections with the potential to cause global pandemics are increasingly concerning. One type of those commonly emerging and re-emerging pathogens are coronaviruses(SARS-Co V, MERS-Co V and SARS-Co V-2).Wild animals are hosts of different coronaviruses with the potential risk of cross-species transmission. However, little is known about the reservoir and host of coronaviruses in wild animals in Qinghai Province, where has the greatest biodiversity among the world's high-altitude regions. Here, from the next-generation sequencing data, we obtained a known beta-coronavirus(beta-Co V) genome and a novel delta-coronavirus(delta-Co V) genome from faecal samples of 29 marmots, 50 rats and 25 birds in Yushu Tibetan Autonomous Prefecture, Qinghai Province, China in July 2019. According to the phylogenetic analysis, the beta-Co V shared high nucleotide identity with Coronavirus HKU24. Although the novel delta-Co V(Mt Co V) was closely related to Sparrow deltacoronavirus ISU42824, the protein spike of the novel delta-Co V showed highest amino acid identity to Sparrow coronavirus HKU17(73.1%). Interestingly, our results identified a novel host(Montifringilla taczanowskii) for the novel delta-Co V and the potential cross-species transmission. The most recent common ancestor(t MRCA) of Mt Co Vs along with other closest members of the species of Coronavirus HKU15 was estimated to be 289 years ago. Thus, this study increases our understanding of the genetic diversity of beta-Co Vs and deltaCo Vs, and also provides a new perspective of the coronavirus hosts. | Wentao Zhu Jing Yang Shan Lu Ruiting Lan Dong Jin Xue-lian Luo Ji Pu Shusheng Wu Jianguo Xu | 2021 | Virologica Sinica2021,36,3: | 3
Molecular characterization and functional expression of opioid receptor-libe_1 receptor显示文摘The opioid receptor-libel receptor (ORL), an orphan receptor whose human and murine complementary DNAs,has been characterized recently. ORL transcripts are particularly abundant in the central nervous system. We demonstrated that ORL expressed in human neuroblastoma SK-N-SH and SH-SY5Y cell lines by radioligand binding assay, reverse transcription polymerase chain reaction (RT-PCR) and Northern analysis in the present study. Stimulation with ORL1 specific agonist, nociceptin/orphanin Fo, increased [34S]GTPrγS binding to SK-N-SH cell membranes (EC50 = 14 ±0.45 nM), and attenuated forskolin-stimulated accumulation of cellular cAMP (EC50= 0.80 ±0.45 nM, indicative that activation of ORL1 activates G proteins and inhibits adenylyl cyclase. Activation of ORL1 receptor was also accessed using CHO:hORL1 cell line by microphysiometer. Treatment of nociceptin/orphanin FQ increased extracellular acidification rate significantly. | WU YA LAN LU PU KUN LING JIAN ZHAO ZHI JIE CHENG LAN MA GANG PEI(Shanghai Institute of Cell Biology, Chinese Academy of Sciences, Shanghai 200031, China)(Shanghai Medical University, Shanghai 200032, China) | 1997 | Cell Research1997,7,1: | 2
Impact of human leukocyte antigen mismatching on outcomes of liver transplantation:A meta-analysis显示文摘AIM:To assess the effect of human leukocyte antigen(HLA) mismatching on liver graft outcome and acute rejection from a meta-analysis of available cohort studies.METHODS:Articles in PubMed/MEDLINE,EMBASE and the Cochrane database from January 1970 to June 2009,including non-English literature identified in these databases, were searched.Only studies comparing HLA or sub-phenotype matching with mismatching were extracted. The percentage of graft survival was extracted by 'Engauge Digitizer' from survival curves if the raw data were not displayed.A meta-analysis was performed when at least 3 studies provided data.RESULTS:Sixteen studies met the inclusion criteria.A lower number of HLA mismatches(0-2 vs 3-6) did reduce the incidence of acute rejection(relative risk:0.77,P = 0.03).The degree of HLA mismatching(0-2 vs 3-6) had no significant effect on 1-year [hazard ratio(HR):1.04,P = 0.68] and 5-year(HR:1.09,P = 0.38) graft survival. In sub-phenotype analysis, the degree of HLA-A,B and DR mismatching(0 vs 1-2) had no significant effect on 1-year and 5-year graft survival, either. The HRs and P-values were 0.95,0.71(HLA-A,1-year);1.06,0.60(HLA-A,5-year);0.77,0.16(HLA-B,1-year);1.07,0.56(HLA-DR,1-year);1.18,0.23(HLADR,5-year),respectively.CONCLUSION:The results of this systematic review imply that good HLA compatibility can reduce the incidence of acute rejection in spite of having no influence on graft outcomes. To obtain a short recovery time and minimize rejection post transplantation,HLA matching studies should be considered before the operation. | Xiang Lan Ming-Man Zhang Cong-Lun Pu Chun-Bao Guo Quan Kang Ying-Chun Li Xiao-Ke Dai Yu-Hua Deng Qiang Xiong Zhi-Mei Ren | 2010 | World Journal of Gastroenterology2010,16,27: | 2

Si-Assisted Solidification Path and Microstructure Control of 7075 Aluminum Alloy with Improved Mechanical Properties by Selective Laser Melting显示文摘The construction and application of traditional high-strength 7075 aluminum alloy(Al7075) through selective laser melting(SLM) are currently restricted by the serious hot cracking phenomenon. To address this critical issue, in this study, Si is employed to assist the SLM printing of high-strength Al7075. The laser energy density during SLM is optimized, and the eff ects of Si element on solidification path, relative density, microstructure and mechanical properties of Al7075 alloy are studied systematically. With the modified solidification path, laser energy density, and the dense microstructure with refined grain size and semi-continuous precipitates network at grain boundaries, which consists of fine Si, β-MgSi, Q-phase and θ-AlCu, the hot cracking phenomenon and mechanical properties are eff ectively improved. As a result, the tensile strength of the SLM-processed Si-modified Al7075 can reach 486 ± 3 MPa, with a high relative density of ~ 99.4%, a yield strength of 291 ± 8 MPa, fracture elongation of(6.4 ± 0.4)% and hardness of 162 ± 2(HV) at the laser energy density of 112.5 J/mm~3. The main strengthening mechanism with Si modification is demonstrated to be the synergetic enhancement of grain refinement, solution strengthening, load transfer, and dislocation strengthening. | 19 | A Nomogram to Predict Patients with Obstructive Coronary Artery Disease: Development and Validation显示文摘Objective:To develop and validate clinical prediction models for the development of a nomogram to estimate the probability of patients having coronary artery disease(CAD).Methods and Results:A total of 1,025 patients referred for coronary angiography were included in a retrospective,single-center study.Randomly,720 patients(70%)were selected as the development group and the other patients were selected as the validation group.Multivariate logistic regression analysis showed that the seven risk factors age, sex, systolic blood pressure,lipoprotein-associated phospholipase A 2,type of angina, hypertension, and diabetes were signifi cant for diagnosis of CAD,from which we established model A.We established model B with the risk factors age, sex, height, systolic blood pressure,low-density lipoprotein cholesterol,lipoprotein-associated phospholipase A 2,type of angina, hypertension, and diabetes via the Akaike information criterion. The risk factors from the original Framingham Risk Score were used for model C.From comparison of the areas under the receiver operating characteristic curve, net reclassifi cation improvement, and integrated discrimination improvement of models A,B,and C,we chose model B to develop the nomogram because of its fi tness in discrimination, calibration, and clinical effi ciency. The nomogram for diagnosis of CAD could be used easily and conveniently. Conclusion:An individualized clinical prediction model for patients with CAD allowed an accurate estimation in Chinese populations. The Akaike information criterion is a better method in screening risk factors. The net reclassifi-cation improvement and integrated discrimination improvement are better than the area under the receiver operating characteristic curve in discrimination.Decision curve analysis can be used to evaluate the effi ciency of clinical prediction models. | Zesen Han Lihong Lai Zhaokun Pu Lan Yang | 2021 | Cardiovascular Innovations and Applications2021,,2: | 1
