1Comprehensive strategy of conduit guidance combined with VEGF producing Schwann cells accelerates peripheral nerve repair显示文摘Peripheral nerve regeneration requires stepwise and well-organized establishment of microenvironment.Since local delivery of VEGF-A in peripheral nerve repair is expected to promote angiogenesis in the microenvironment and Schwann cells(SCs)play critical role in nerve repair,combination of VEGF and Schwann cells may lead to efficient peripheral nerve regeneration.VEGF-A overexpressing Schwann cells were established and loaded into the inner wall of hydroxyethyl cellulose/soy protein isolate/polyaniline sponge(HSPS)conduits.When HSPS is mechanically distorted,it still has high durability of strain strength,thus,can accommodate unexpected strain of nerve tissues in motion.A 10 mm nerve defect rat model was used to test the repair performance of the HSPS-SC(VEGF)conduits,meanwhile the HSPS,HSPS-SC,HSPS-VEGF conduits and autografts were worked as controls.The immunofluorescent co-staining of GFP/VEGF-A,Ki67 and MBP showed that the VEGF-A overexpressing Schwann cells could promote the proliferation,migration and differentiation of Schwann cells as the VEGF-A was secreted from the VEGF-A overexpressing Schwann cells.The nerve repair performance of the multifunctional and flexible conduits was examined though rat behavioristics,electrophysiology,nerve innervation to gastrocnemius muscle(GM),toluidine blue(TB)staining,transmission electron microscopy(TEM)and NF200/S100 double staining in the regenerated nerve.The results displayed that the effects on the repair of peripheral nerves in HSPS-SC(VEGF)group was the best among the conduits groups and closed to autografts.HSPS-SC(VEGF)group exhibited notably increased CD31+endothelial cells and activation of VEGFR2/ERK signaling pathway in the regenerated nerve tissues,which probably contributed to the improved nerve regeneration.Altogether,the comprehensive strategy including VEGF overexpressing Schwann cells-mediated and HSPS conduit-guided peripheral nerve repair provides a new avenue for nerve tissue engineering.Ping Wu Zan Tong Lihua Luo Yanan Zhao Feixiang Chen Yinping Li Céline Huselstein Qifa Ye Qingsong Ye Yun Chen 2021Bioactive Materials2021,6,10:4

3Suppression of starch synthase I(SSI) by RNA interference alters starch biosynthesis and amylopectin chain distribution in rice plants subjected to high temperature显示文摘Based on known cDNAs of rice starch synthase isoforms,we constructed dsRNA interference vectors for starch synthase I(SSI)to produce transgenic plants containing starch with a moderately high amylose content.We investigated the effect of SSI suppression on grain quality traits,starch biosynthesis,and amylopectin chain distribution in rice plants exposed to two different temperature regimes.The activities and transcripts of BEs,DBEs,and other SS isoforms were further investigated to clarify the effect of SSI suppression on these key enzymes and their specific isoforms under different temperature treatments.Suppression of SSI by RNAi altered grain starch component and amylopectin chain distribution,but it exerted only a slight effect on total starch content(%)and accumulation amount(mg kernel?1)and on starch granule morphology and particle size distribution.Under normal temperature(NT),insignificant differences in kernel weight,chalky kernel proportion,chalky degree,and starch granule morphology between SSI-RNAi line and its wild type(WT)were observed.However,amylose content(AC)level and granule-bound starch synthase(GBSS)activity in rice endosperms were markedly increased by SSI-RNAi suppression.The chalky kernel proportion and chalky degree of SSIRNAi lines were significantly higher than those of WT under high temperature(HT)exposure at filling stage.Inhibition of SSI by RNAi affected amylopectin chain distribution and raised starch gelatinization temperature(GT)in two ways:directly from the SSI deficiency itself and indirectly by reducing BEIIb amounts in an SSI-deficient background.The deficiency of SSI expression led to an alteration in the susceptibility of grain chalkiness occurrence and starch gelatinization temperature to HT exposure,owing to a pleiotropic effect of SSI deficiency on the expression of other genes associated with starch biosynthesis.Qian Zhao Xiaoxia Du Zhanyu Han Yu Ye Gang Pan Muhammad-Asad-Ullah Asad Qifa Zhou Fangmin Cheng 2019The Crop Journal2019,7,5:2

7Wilson's Disease and Hepatic Transplantation显示文摘Summary: To investigate the changes in neurological symptoms and signs, as well as serum copper, serum ceruloplasmin after hepatic transplantation in patients with Wilson’s disease, neurological symptoms and signs, serum copper, serum ceruloplasmin before and after hepatic transplantation in 18 patients with Wilson’s disease were observed, and those changes were followed up in 20 non-operative controls treated with penicillamine. Our results showed that the neurological symptoms and signs, serum copper and serum ceruloplasmin were improved in the operative group but deteriorated in the non-operative control group. 8minimal genome design to maximally guarantee fertile inter-subspecific hybrid rice显示文摘Hybrid rice has made considerable contributions to achieve the ambitious goal of food security for the world's population.Hybrid rice from indica/xian and japonica/geng subspecies shows much higher heter-osis and is thereby an important innovation in promoting rice production in the next decade.However,such inter-subspecific hybrid rice has long suffered from serious hybrid sterility,which is a major challenge that needs to be addressed.In this study,we performed a genome design strategy to produce fertile inter-subspecific hybrid by creation of wide compatibility varieties that are able to overcome hybrid sterility.Based on combined genetic analyses in two indica-japonica crosses,we determined that four hybrid ste-rility loci,S5,f5,pf12 and Sc,are the major QTLs controlling inter-subspecific hybrid sterility and thus the minimal targets that can be manipulated for breeding sub-specific hybrid rice.We then cloned the pf12 lo-cus,one of the most effective loci for hybrid male sterility,by map-based cloning,and showed that artificial disruption of pf12A gene at this locus could successfully rescue hybrid fertility.We further dissected the genetic basis of wide compatibility using three pairwise crosses from a wide-compatibility variety Dular and representative indica and japonica varieties.On this basis,we constructed and assembled different combinations of naturally compatible alleles of four loci,S5,Sc,pf12,and f5,and found that the improved lines could fully recover pollen and embryo sac fertility in test-crossed F,s,thereby completely fulfilling the demands of inter-subspecific hybrid spikelet fertility in agricultural production.This breeding scheme would facilitate redesign of future inter-subspecific hybrid rice with a higher yield potential.Penghui Zhou Zhengji Wang Xingchen Zhu Yao Tang Liang Ye Huihui Yu Yating Li Ningke Zhang Ting Liu Tian Wang Yuying Wu Dengyun Cao Yuan Chen Xu Li Qinglu Zhang Jinghua Xiao Sibin Yu Qifa Zhang Jiaming Mi Yidan Ouyang 2023Molecular Plant2023,16,4:0

12PBLT AND THE CONCERNED PROBLEMS (WITH A REPORT OF 48 CASES)显示文摘Objective:To study the new surgical technique by piggyback of liver transplantation(PBLT) used to treat end-stage liver disease.Methods:Forty-eight cases of PBLT have been performed by author in 45 patients from September, 1995 to December,2000. The piggyback procedure was used in all cases, which included 4 retransplantation and 3 combined kidney-liver transplantation(CKLT), 3 Orthotopic inferior vena cava(IVC)-bridge-atrium LT and 1 paraorthotopic IVC-bridge to atrium LT, 1 auxiliary combined hepatovenal transplantation(ACHRT), and 1 auxiliary combined hepatointestinal transplantation (ACHIT).Result:All performances were successful and no technique-related complications occurred. The longest survival of liver malignancy that had PBLT was 1 year and 4 months, and the other 2 patients of benign end-stage liver disease who had PBLT have survived 4 years and 10 months and 5 years and 6 months respectively with a life of good quality. The survival rate of the benign end-stage liver disease that had PBLT since the end of 1998 is 67%. The 2 cases who had CKLT and 1 case ACHRT had been living more than 1 year and 2 year six months.Conclusion:As the new technique being used since the end of 1980s, the PBLT procedure is better than the classical one because no veno-venous bypass is needed and the hemodynamics are stable.
