1Chemical Characteristics and Environmental Records of a Snow-Pit at the Glacier No.12 in the Laohugou Valley, Qilian Mountains显示文摘In June 2006, samples were collected from a 90 cm snow-pit at an elevation of 5 040 m a.s.l. in the northern branch firn basin of the Glacier No. 12, Laohugou Valley, in the western part of Qilian Mountain, China. Oxygen isotopes(δ18O) and major soluble ions were measured. Results showed that Ca2+ is the dominant cation, and SO42- is the dominant anion. All major ions have close positive correlations with eachother except NO3- and NH4+. δ18O show positive correlation with air temperature. The method of correlation analysis, sea-salt ion tracer and trend analysis were used to determine the source of the chemical components. 2Identification and isolation of Mu-flanking fragments from maize显示文摘Transposable elements have been utilized as mutagens to create mutant libraries for functional genomics. Isolation of genomic seg-ments flanking the insertion Mutator (Mu) is a key step in insertion mutagenesis studies. Herein, we adopted a modified AFLP method to identify and isolate Mu-flanking fragments from maize. The method consists of the following steps: 1) double-digestion of genomic DNA with Bgl II/Msp I and ligation of digested fragments to the Bgl II-and Msp I-adaptors; 2) enrichment of a subset of Bgl II/Msp I fragments followed by selective amplification of the Mu-flanking fragments; 3) simultaneous display of AFLP bands derived from the flanking re-gions for both insert and native Mu transposons; 4) identification and isolation of AFLP bands resulting from Mu insertions by comparing the banding profiles between Mu-induced mutants and their parental lines; and 5) confirmation of flanking fragments related to these Mu insertions. Using this approach, we have isolated flanking fragment(s) resulting from Mu insertion for every Mu-induced mutant, and one such fragment, M196-FF, is found to contain a partial sequence of the DNA topoisomerase I gene Top1. Moreover, the modified AFLP method including all restriction enzymes, adaptors and primers has been optimized in this study. 4A mathematical modeling study of the HIV epidemics at two rural townships in the Liangshan Prefecture of the Sichuan Province of China显示文摘Background:As a response to a severe HIV epidemic in the Liangshan Prefecture,one of the worst in China,population based HIV interventions,including two population-wide HIV screening,have been carried out since 2005 at two townships in a remote mountainous region of Liangshan.The objective of our mathematical modeling study is to assess the temporal dynamics of the HIV epidemic in the two townships based on the data collected in the study area during the period 2005e2010.Methods:A mathematical model was set up to describe the population dynamics of HIV transmission in study area.The model was calibrated by fitting it to the HIV testing and treatment data from 2005 to 2008.Validation of the model was done by comparing its predicted value of HIV prevalence in 2010 to the prevalence data obtained in the 2010 population wide HIV testing.The validated model was used to produce estimation of HIV incidence,prevalence and death.Results:Our model estimations show that population-based HIV interventions have significantly slowed down the rise of the HIV epidemic in the two townships.Over the five-year period from 2005 to 2010,the year-over-year rate of increase in HIV incidence,prevalence,and death has declined by 91.5%,28.7%,and 52.3%,respectively.Conclusion:Mathematical models,when integrated with epidemiological and surveillance data,can be an effective tool for predicting the temporal dynamics of HIV and assessing the impacts of HIV interventions.Zhimin Su Caiting Dong Ping Li Hongxia Deng Yuhan Gong Shiyong Zhong Min Wu Yuhua Ruan Guangming Qin Wen Yang Yiming Shao Michael Li 2016Infectious Disease Modelling2016,1,1:2
