1Characterization and enrichment of hepatic progenitor cells in adult rat liver显示文摘AIM: To detect the markers of oval cells in adult rat liver and to enrich them for further analysis of characterization in vitro.METHODS: Rat model for hepatic oval cell proliferation was established with 2-acetylaminofluorene and two third partial hepatectomy (2-AAF/PH). Paraffin embedded rat liver sections from model (11 d after hepatectomy) and control groups were stained with HE and OV6, cytokeratin19 (CK19),albumin, alpha fetoprotein (AFP), connexin43, and c-kit antibodies by immunohistochemistry. Oval cell proliferation was measured with BrdU incorporation test. C-kit positive oval cells were enriched by using magnetic activated cell sorting (MACS) .The sorted oval cells were cultured in a low density to observe colony formation and to examine their characterization in vitroby immunocytochemistry and RT-PCR.RESULTS: A 2-AAF/PH model was successfully established to activate the oval cell compartment in rat liver. BrdU incorporation test of oval cell was positive. The hepatic oval cells coexpressed oval cell specific marker OV6, hepatocytemarker albumin and cholangiocyte-marker CK19. They also expressed AFP and connexin 43. C-kit, one hematopoietic stem cell receptor, was expressed in hepatic oval cells at high levels. By using c-kit antibody in conjunction with MACS,we developed a rapid oval cell isolation protocol. The sorted cells formed colony when cultured in vitro. Cells in the colony expressed albumin or CK19 or coexpressed both and BrdU incorporation test was positive. RT-PCR on colony showed expression of albumin and CK19 gene.CONCLUSION: Hepatic oval cells in the 2-AAF/PH model had the properties of hepatic stem/progenitor cells. Using MACS, we established a method to isolate oval cells. 17High sensitivity of semimetal photodetection via Bose–Einstein condensation显示文摘The discovery of semiconductor has witnessed remarkable strides toward high performance of photodetectors attributed to its excellent carrier properties.However,semimetal,owning to the high carrier concentration and low carrier mobility compared to those of semiconductor,is generally considered unsuitable for photodetection.Herein,we demonstrate an outstanding photodetection in a layered semimetal titanium diselenide(TiSe_(2))in Bose-Einstein condensation(BEC)state.High sensitivity of semimetal photodetector is realized in the range of visible,infrared and terahertz bands.The noise equivalent power(NEP)has threefold improvement at the visible and infrared wavebands,and significant decrease by one order of magnitude in the terahertz frequencies via BEC phenomenon,attributed to the electrical parameter variation after condensation.The best NEP value in the terahertz frequency is comparable to that of commercial Si photodetector.Our results show another recipe to fabricate high performance of photodetection via semimetal except for semiconductor and pave the way to exploit macroscopic quantum phenomena for optoelectronics.Tuntan Wu Qinxi Qiu Yongzhen Li Qiangguo Zhou Wanli Ma Jingbo Li Lin Jiang Wei Zhou Zhiming Huang 2024InfoMat2024,6,2:0
18Earthquake Monitoring and Study in the Jingpohu Volcano Cluster Area显示文摘Seismicity in the Jingpohu volcanic area was investigated based on the seismic data recorded by the mobile seismic network consisting of 14 stations equipped with 24-bit broad-band 3-component seismographs around Crater Forest.Results show that there appears certain seismicity in Jingpohu and its adjacent areas with a low activity level and most of the recorded earthquakes are the volcanic-tectonic ones.The results of location indicate a dominant focal depth of 10km~30km,most of the earthquakes are smaller than M_L2.0,and are concentrated in the area of 'Crater Forest' and on the Dunhua-Mishan fault which runs through the volcanic area.At station No.2,which has better observation conditions,two types of events,likely associated to volcanism,were recorded;their waveform characteristics are somewhat similar to that of the long-period volcanic event and the volcanic tremor,but with different feature of frequencies.Liu Zhi Duan Yonghong Xu Zhaofan Yuan Qinxi Yang Jian Zhou Xuesong 2008Earthquake Research in China2008,22,2:0
19A new method for preparing a rat intracerebral hemorrhage model by combining focused ultrasound and microbubbles显示文摘Background:We aimed to prepare a non-invasive,reproducible,and controllable rat model of intracerebral hemorrhage with focused ultrasound(FUS).Methods:A rat intracerebral hemorrhage(ICH)model was established by combining FUS and microbubbles(μBs),and edaravone was used to verify whether the free radical scavenger had a protective effect on the model.The brain tissue of each group was sectioned to observe the gross histology,blood-brain barrier(BBB)permeability,cerebral infarction volume,and histopathological changes.Results:Compared with the FUS group,the BBB permeability was significantly increased in the FUS+μBs(F&B)group(p=0.0021).The second coronal slice in the F&B group had an obvious hemorrhage lesion,and the FUS+μBs+edaravone(F&B&E)group had smaller hemorrhage areas;however,ICH did not occur in the FUS group.The cerebral infarction volume in the F&B group was significantly larger than that in the FUS group(p=0.0030)and F&B&E group(p=0.0208).HE staining results showed that nerve fibrinolysis,neuronal necrosis,microglia production,and erythrocytes were found in both the F&B group and the F&B&E group,but the areas of the nerve fibrinolysis and neuronal necrosis in the F&B group were larger than the F&B&E group.Conclusions:A rat ICH model was successfully prepared using theμBs assisted FUS treatment,and edaravone had a therapeutic effect on this model.This model can be used to study the pathophysiological mechanism of ICH-related diseases and in preclinical research on related new drugs.Yao He Jie Yang Fengying Hu Min Liao Yuru Nie Xiaoxia Zhu Tao Zhang Keer Song Qinxi Li Xiaojie Li Chenghan Mei Zhe Wu Qiang Lu Zhihui Zhong 2023Animal Models and Experimental Medicine2023,6,2:0
20Analysis of pressure response at an observation well against pressure build-up by early stage of CO_(2)geological storage project显示文摘To ensure a safe and stable CO_(2)storage,pressure responses at an observation well are expected to be an important and useful field monitoring item to estimate the CO_(2)storage behaviors and the aquifer parameters during and after injecting CO_(2),because it can detect whether the injected CO_(2)leaks to the ground surface or the bottom of the sea.In this study,pressure responses were simulated to present design factors such as well location and pressure transmitter of the observation well.Numerical simulations on the pressure response and the time-delay from pressure build-up after CO_(2)injection were conducted by considering aquifer parameters and distance from the CO_(2)injection well to an observation well.The measurement resolution of a pressure transmitter installed in the observation well was presented based on numerical simulation results of the pressure response against pressure build-up at the injection well and CO_(2)plume front propagations.Furthermore,the pressure response at an observation well was estimated by comparing the numerical simulation results with the curve of CO_(2)saturation and relative permeability.It was also suggested that the analytical solution can be used for the analysis of the pressure response tendency using pressure build-up and dimensionless parameters of hydraulic diffusivity.Thus,a criterion was established for selecting a pressure transducer installed at an observation well to monitor the pressure responses with sufficient accuracy and resolution,considering the distance from the injection well and the pressure build-up at the injection well,for future carbon capture and storage(CCS)projects.Qiang Sun Kyuro Sasaki Qinxi Dong Zhenni Ye Hui Wang Huan Sun 2024Journal of Rock Mechanics and Geotechnical Engineering2024,16,2:0
