1Potential one-step strategy for PET degradation and PHB biosynthesis through co-cultivation of two engineered microorganisms显示文摘The management and recycling of plastic waste is a challenging global issue.Polyethylene terephthalate(PET),one of the most widely used synthetic plastics,can be hydrolyzed by a series of enzymes.However,upcycling the resulting monomers is also a problem.In this study,we designed a co-cultivation system,in which PET degradation was coupled with polyhydroxybutyrate(PHB)production.First,PETase from Ideonalla sakaiensis was expressed in Yarrowia lipolytica Po1f with a signal peptide from lipase.The engineered PETase-producing Y.lipolytica was confirmed to hydrolyze bis(2-hydroxyethyl)terephthalate(BHET)and PET powder into the monomers terephthalate(TPA)and ethylene glycol(EG).Simultaneously,a TPA-degrading Pseudomonas stutzeri strain isolated from PET waste was transformed with a recombinant plasmid containing the phb CAB operon from Ralstonia eutropha,which encodes enzymes for the biosynthesis of PHB.The two co-cultivated engineered microbes could directly hydrolyze BHET to produce the bioplastic PHB in one fermentation step.During this process,5.16 g/L BHET was hydrolyzed in 12 h,and 3.66 wt%PHB(3.54 g/L cell dry weight)accumulated in 54 h.A total of 0.31g/L TPA was produced from the hydrolyzation of PET in 228 h.Although PHB could not be synthesized directly from PET because of the low hydrolyzing efficiency of PETase,this study provides a new strategy for the biodegradation and upcycling of PET waste by artificial microflora.Pan Liu Tong Zhang Yi Zheng Qingbin Li Tianyuan Su Qingsheng Qi 2021Engineering Microbiology2021,1,1:3
2Modulating the function of ABCB1: in vitro and in vivo characterization of sitravatinib, a tyrosine kinase inhibitor显示文摘Background:Overexpression of ATP-binding cassette(ABC)transporter is a major contributor to multidrug resistance(MDR),in which cancer cells acquire resistance to a wide spectrum of chemotherapeutic drugs.In this work,we evaluated the sensitizing effect of sitravatinib,a broad-spectrum tyrosine kinase inhibitor(TKI),on ATP-binding cassette subfamily B member 1(ABCB1)-and ATP-binding cassette subfamily C member 10(ABCC10)-mediated MDR.Methods:MTT assay was conducted to examine cytotoxicity and evaluate the sensitizing effect of sitravatinib at non-toxic concentrations.Tritium-labeled paclitaxel transportation,Western blotting,immunofluorescence analysis,and ATPase assay were carried out to elucidate the mechanism of sitravatinib-induced chemosensitization.The in vitro findings were translated into preclinical evaluation with the establishment of xenograft models.Results:Sitravatinib considerably reversed MDR mediated by ABCB1 and partially antagonized ABCC10-mediated MDR.Our in silico docking simulation analysis indicated that sitravatinib strongly and stably bound to the transmembrane domain of ABCB1 human-mouse chimeric model.Furthermore,sitravatinib inhibited hydrolysis of ATP and synchronously decreased the efflux function of ABCB1.Thus,sitravatinib could considerably enhance the intracellular concentration of anticancer drugs.Interestingly,no significant alterations of both expression level and localization of ABCB1 were observed.More importantly,sitravatinib could remarkably restore the antitumor activity of vincristine in ABCB1-mediated xenograft model without observable toxic effect.Conclusions:The findings in this study suggest that the combination of sitrvatinib and substrate antineoplastic drugs of ABCB1 could attenuate the MDR mediated by the overexpression of ABCB1.Yuqi Yang Ning Ji Chao-Yun Cai Jing-Quan Wang Zi-Ning Lei Qiu-Xu Teng Zhuo-Xun Wu Qingbin Cui Yihang Pan Zhe-Sheng Chen 2020Cancer Communications2020,40,7:2
3Responses of flood peaks to land use and landscape patterns under extreme rainstorms in small catchments-A case study of the rainstorm of Typhoon Lekima in Shandong,China显示文摘Investigations of the formation mechanisms of flood peaks in small catchments facilitate flood prediction and disaster prevention under extreme rainstorms.However,there have been few studies on the re-sponses of flood peaks to land use landscape patterns using field surveys during extreme rainstorm events.Based on field data from 17 small catchments near the rainstorm center of Typhoon Lekima,7 landscape indices were chosen.The flood peak and its sensitivity to the land use landscape were investigated by combining remote sensing interpretation and related analysis.The conclusions are as follows:(1)The peak discharge of the small catchment was 2.36e56.50 m3/s,the peak modulus was 8.00 e48.89 m3/(s$km2),and the flood index K ranged from 3.61 to 4.55.(2)Under similar rainfall conditions,the flood peak modulus,K and the proportion of sloping cropland had significantly positive correlations(p<0.05).The flood peak modulus was significantly negatively correlated with the proportion of forest-grassland and terrace(p<0.05),and K and the proportion of forest-grassland and terraced land exhibited a negative correlation.(3)The flood peak modulus and K were positively correlated with the landscape fragmentation.(4)The sensitivities of small catchments to floods were evaluated to be moderate compared to K values from other studies.The ability of small catchments to cope with extreme rain-storms can be improved by increasing the areas of forest-grassland,and terraces and reducing landscape fragmentation.Our results could be applied to provide a basis for land use planning and support for the response against disasters caused by extreme floods.Yuanhao Liu Jianqiao Han Juying Jiao Baoyuan Liu Wenyan Ge Qingbin Pan Fei Wang 2022International Soil and Water Conservation Research2022,10,2:1
4Prevalence of Metabolic Obese Normal Weight and Related Risk Factors in South China显示文摘[Objectives]Metabolic obese normal weight(MONW)is becoming one of the pubic problems which are threatening human health.Whereas,MONW was facing a great challenge for limited attention,especially for the female in China.The aim of this research was to estimate the prevalence of MONW and its related risk components in South China.[Methods]A community-based cross-sectional study was performed on 3349 residents aged 18-93 years in The First Affiliated Hospital of Jinan University,Guangzhou,China,in 2019.Data was collected by physical examination data which included physical measurements and laboratory examinations.[Results]In all subjects,55%were females(M/F=1509/1840).The prevalence of MONW was 16.09%(0.04%for male,16.05%for female,P<0.001).The prevalence increased significantly with increasing age in both genders(P<0.001).The binary logistic regression analysis shows that among the risk factors with MONW,age,BMI,gender,systolic pressure,hypertension[Male:ORs=2.56,95%CI(1.23,5.32);Female:ORs=2.88,95%CI(1.76,4.71)]and hypertriglyceridemia[Male:ORs=3.23,95%CI(1.67,6.19);Female:ORs=2.57,95%CI(1.64,4.03)]were found to be statistically significant.The level of ALT in MONW group was(27.88±15.85)in male and(24.33±15.75)in female,which were significantly higher than those in the non-MetS group.[Conclusions]The prevalence of MONW was pretty high.We considered MONW be significantly associated with the increase of ALT.Female gender,advanced age,and elevated ALT were independent risk factors for MONW.It was high time that the government should raise the public attention toward metabolic function in normal weight population.Effective prevention and treatment strategies for MONW and its risk factors should be developed targeting different ages and genders.Xuehua LUO Zien LIANG Yan LI Qingbin WU Zhiqiang PAN Yuqing LIANG Lingyu LI Juanjuan MAI Huijun XIE Li HAN 2021Medicinal Plant2021,12,2:0
