| 3 | Type I IFN augments IL-27-dependent TRIM25 expression to inhibit HBV replication显示文摘Hepatitis B virus(HBV)can cause chronic hepatitis B,which may lead to cirrhosis and liver cancer.Type I interferon(IFN)is an approved drug for the treatment of chronic hepatitis B.However,the fundamental mechanisms of antiviral action by type I IFN and the downstream signaling pathway are unclear.TRIM25 is an IFN-stimulated gene(ISG)that has an important role in RIG-I ubiquitination and activation.Whether TRIM25 is induced in liver cells by type I IFN to mediate anti-HBV function remains unclear.Here we report that interleukin-27(IL-27)has a critical role in IFN-induced TRIM25 upregulation.TRIM25 induction requires both STAT1 and STAT3.In TRIM25 knockout HepG2 cells,type I IFN production was consistently attenuated and HBV replication was increased,whereas overexpression of TRIM25 in HepG2 cells resulted in elevated IFN production and reduced HBV replication.More interestingly,we found that TRIM25 expression was downregulated in HBV patients and the addition of serum samples from HBV patients could inhibit TRIM25 expression in HepG2 cells,suggesting that HBV might have involved a mechanism to inhibit antiviral ISG expression and induce IFN resistance.Collectively,our results demonstrate that type I IFN-induced TRIM25 is an important factor in inhibiting HBV replication,and the IFN-IL-27-TRIM25 axis may represent a new target for treating HBV infection. | Guangyun Tan Qingfei Xiao Hongxiao Song Feng Ma Fengchao Xu Di Peng Na Li Xiaosong Wang Junqi Niu Pujun Gao F Xiao-Feng Qin Genhong Cheng | 2018 | Cellular & Molecular Immunology2018,15,3: | 13 |
| 4 | Tectonic Constraints on the Transformation of Paleozoic Framework of Uplift and Depression in the Ordos Area显示文摘During the Paleozoic, the Ordos area in the western North China Plate was located at the intersecting position of microplates and controlled by their interaction. The structural framework in the Ordos area, which underwent transformations in the Ordovician, the Carboniferous and the Permian respectively, was dominated by the alternation of uplift and depression. The transformations of structural framework are utilized as the clues to investigate the microplates’ interacting type and its response in the Ordos area. According to the regional structural evolution, the Ordos area is simplified into an isopachous, isotropic and elastic shell model, and under proposed various boundary conditions, three series of numerical simulations corresponding to the three structural transformations are carried out to determine the detailed tectonic constraints. Numerical simulations reveal that the structure of the uplift and depression, which is similar to the actual pattern, develops only under one special boundary condition in each of the three series, indicating that the structural framework responds to the unique tectonic background. The simulation results show that in the Early Paleozoic, the L-shaped paleouplift formed nearby the southwestern corner of the Ordos area because the intensity of the compressions in the southern and western boundaries resulting from the ocean-continent collisions was similar. In the Late Paleozoic, it evolved into continent-continent (or arc-continent) interaction in the southern and northern boundaries; in the preliminary stage of the interaction, since the interface between the North China Plate and the plates on the south and north was narrow, the relative acting force was little and the regional western boundary immobile, and the structural framework in the basin was characterized by the N-S trending slender-waist-shaped uplift; as the interface between the plates expanded gradually, the extrusive force in the southern and northern boundaries of the North China Plate increased, resulting in the paleogeographic divisions showing E-W trending, and, the western boundary of the basin was extruded westward due to the intense compression inducing the local NE trending of paleogeographic division in the central area. | 5 | A novel heat-resistant Al–Si–Cu–Ni–Mg base material synergistically strengthened by Ni-rich intermetallics and nano-AlN_p microskeletons显示文摘In this work, nano-AlN particle(nano-AlN_p) microskeletons were introduced into an Al–Si–Cu–Ni–Mg alloy by Al–8 AlN master alloy in which the nano-AlN_p reinforcements connect with each other to form three-dimensional networks. It is found that these nano-AlN_p microskeletons mainly distribute in the binary Al–Si eutectic zones resulting in flaky eutectic Si phases being modified to particulates. Meanwhile,the microskeletons strengthen the matrix synergistically with semi-continuous Ni-rich intermetallics in three dimensions. The tensile mechanical properties, micro-hardness and thermal expansion properties of the alloy at different temperatures are significantly improved. | 6 | Type III interferon-induced CBFβinhibits HBV replication by hijacking HBx显示文摘Hepatitis B virus(HBV)and its associated chronic infection remain serious health threats worldwide.However,there is still no impactful approach for clinical treatment of hepatitis B patients.Therefore,developing a better understanding of the interactions between HBV and its host is particularly important.HBV infection has been reported to induce type-III but not type-I or type-II interferon(IFN).In this study,we identified CBFβ,an HIV enhancer,as an HBV restriction factor that is specifically induced by type-III IFN in the early stages of HBV infection.Type-III IFN-induced IL-10 played an important role in the production of CBFβ.Interestingly,the interaction between CBFβ-and HBV-encoded regulatory protein X(HBx)enhanced the stability of CBFβ,but notably blocked HBx-mediated promotion of HBV replication.CBFβexpression was lower in HBV patients than in healthy persons,and the addition of serum from HBV patients inhibited CBFβexpression in HepG2 cells.On the contrary,HBV via HBsAg inhibited type-III IFN-induced CBFβexpression and decreased the anti-HBV activity of type-III IFN,suggesting that HBV inhibits antiviral interferon-stimulated gene(ISG)expression and induces IFN resistance.Collectively,our results demonstrate that type-III IFN-triggered and IL-10-induced CBFβare crucial factors for inhibiting HBV replication,and the HBx–CBFβ–HBsAg axis reveals a new molecular mechanism of interaction between HBV and its hosts. | Fengchao Xu Hongxiao Song Qingfei Xiao Na Li Hong Zhang Genhong Cheng Guangyun Tan | 2019 | Cellular & Molecular Immunology2019,16,4: | 3 |

| 10 | Ample dietary fat reduced the risk of primary vesical calculi by inducing macrophages to engulf budding crystals in mice显示文摘Although primary vesical calculi is an ancient disease,the mechanism of calculi formation remains unclear.In this study,we established a novel primary vesical calculi model with D,L-choline tartrate in mice.Compared with commonly used melamine and ethylene glycol models,our model was the only approach that induced vesical calculi without causing kidney injury.Previous studies suggest that proteins in the daily diet are the main contributors to the prevention of vesical calculi,yet the effect of fat is overlooked.To assay the relationship of dietary fat with the formation of primary vesical calculi,D,L-choline tartrate-treated mice were fed a high-fat,low-fat,or normal-fat diet.Genetic changes in the mouse bladder were detected with transcriptome analysis.A high-fat diet remarkably reduced the morbidity of primary vesical calculi.Higher fatty acid levels in serum and urine were observed in the high-fat diet group,and more intact epithelia in bladder were observed in the same group compared with the normal-and low-fat diet groups,suggesting the protective effect of fatty acids on bladder epithelia to maintain its normal histological structure.Transcriptome analysis revealed that the macrophage differentiation-related gene C-X-C motif chemokine ligand 14(Cxcl14)was upregulated in the bladders of high-fat diet-fed mice compared with those of normal-or low-fat diet-fed mice,which was consistent with histological observations.The expression of CXCL14 significantly increased in the bladder in the high-fat diet group.CXCL14 enhanced the recruitment of macrophages to the crystal nucleus and induced the transformation of M2 macrophages,which led to phagocytosis of budding crystals and prevented accumulation of calculi.In human bladder epithelia(HCV-29)cells,high fatty acid supplementation significantly increased the expression of CXCL14.Dietary fat is essential for the maintenance of physiological functions of the bladder and for the prevention of primary vesical calculi,which provides new ideas for the reduction of morbidity of primary vesical calculi. | Huiling Chen Kaiqiang Hu Yaru Liang Yuqi Gao Chenye Zeng Kang Xu Xiaojin Shi Liyuan Li Yuemiao Yin Yi Qiao Ying Qiu Qingfei Liu Zhao Wang | 2022 | Acta Pharmaceutica Sinica B2022,12,2: | 1 |

| 13 | A metallogenic model for the supergiant gold system in Jiaodong province:Constraints from crustal velocity structure显示文摘We investigate the role of crustal architectures playing in controlling the genesis and nature of supergiant Jiaodong orogenic gold system via seismic imaging.We deployed an NWW-SEE-trending broadband seismic linear array traversing the province to construct a shear wave velocity(V_S)transect based on ambient noise tomography.Our crustal V_S transect reveals a prominent low velocity zone(LVZ)at 12–20 km depths across the whole province and high V_S anomalies at 8–12 km depths underneath the western part that hosts most of the ore tonnage.We interpret the former as a hydrothermal alteration zone related to mineralization and the latter as a crustal component containing large amounts of mafic rocks(e.g.,amphibolites).In addition,a listric fault system and a strike-slip fault system are imaged in the western and eastern parts of the province,respectively.Combining features of crustal velocities with regional geological observations that ores are tempo-spatially associated with mafic dikes,we propose a model of gold metallogenic processes.In this model,ponding and degassing of the mafic magmas led to the formation of a hydrothermal alteration LVZ in the middle crust prior to mineralization.Later,auriferous fluid was released from this intra-crustal hydrothermal alteration zone as it was heated by upwelling asthenosphere.The ore fluid ascending along the listric fault system possibly leached extra gold from the upper-crustal amphibolites.Then,the gold-rich fluid migrated up along different fault systems,leading to the disseminated-veinlet mineralization with great gold endowment in the western listric fault system and the quartz-vein type mineralization with relatively small ore tonnage in the eastern strike-slip fault system. | Jue HOU Tao XU Yinshuang AI Guiping YU Yingjie YANG Bo XU Qingfei WANG | 2023 | Science China Earth Sciences2023,66,12: | 0 |
