1Oxidative stress, nutritional antioxidants and beyond显示文摘Free radical-induced oxidative stress contributes to the development of metabolic syndromes(Mets), including overweight,hyperglycemia, insulin resistance and pro-inflammatory state. Most free radicals are generated from the mitochondrial electron transport chain;under physiological conditions, their levels are maintained by efficient antioxidant systems. A variety of transcription factors have been identified and characterized that control gene expression in response to oxidative stress status.Natural antioxidant compounds have been largely studied for their strong antioxidant capacities. 2Fluorescent nanoparticles for chemical and biological sensing显示文摘Fluorescent nanoparticles (NPs), including quantum dots (QDs), dye-doped NPs, and rare earth-based NPs, etc., have been a major focus of research and development during the past decade. The impetus behind such endeavors can be attributed to their unique chemical and optical properties, such as bright fluorescence, high photostability, large Stocks shift and flexible processability. The introduction of fluorescent NPs into analytical chemistry has opened up new venues for fluorescent analysis. In this review, we focus on the developments and analytical applications of fluorescent NPs in the chemical and biological sensing of pH, ions, organic compounds, small biological molecules, nucleic acids, proteins, virus and bacteria. The review also points out the in vitro and in vivo imaging application of fluorescent NPs at the cell and body levels. 3Supplementation of branched-chain amino acids in protein-restricted diets modulates the expression levels of amino acid transporters and energy metabolism associated regulators in the adipose tissue of growing pigs显示文摘This experiment was conducted to investigate the effects of branched-chain amino acids(BCAA)supplemented in protein-restricted diets on the growth performance and the expression profile of amino acid transporters and energy metabolism related regulators in the white adipose tissue(WAT)of different regional depots including dorsal subcutaneous adipose(DSA) and abdominal subcutaneous adipose(ASA), A total of 24 crossbred barrows(7.40 ± 0.70 kg) were randomly divided into 4 groups and were fed the following isocaloric diets for 33 days: 1) a recommended adequate protein diet(AP, 20% CP, as a positive control); 2) a low protein diet(LP, 17% CP); 3) the LP diet supplemented with BCAA(LP + B, 17% CP) to reach the same level of the AP diet group; 4) the LP diet supplemented with 2 times the amount of BCAA(LP + 2B, 17% CP). The daily gain and daily feed intake of the LP diet group were the lowest among all the treatments(P < 0.01). The feed conversion was improved markedly in the group of LP + B compared with the LP diet group(P < 0.05). No significant difference was noted for the serum biochemical parameter concentrations of glucose, triglyceride, nonesterified fatty acid and insulin among the groups(P > 0.05). Moreover, BCAA supplementation down-regulated the expression levels of amino acid transporters including L-type amino acid transporter 1 and sodium-coupled neutral amino acid transporter 2 in DSA, but up-regulated the expression level of Ltype amino acid transporter 4 in ASA(P < 0.05), Meanwhile, the energy sensor AMP-activated protein kinase α was activated in the DSA of pigs fed LP diet and in the ASA of the pigs fed AP or LP + 2B diets(P < 0.05). The mRNA expression profile of the selected mitochondrial component and mitochondrial biogenesis associated regulators in DSA and ASA also responded differently to dietary BCAA supplementation. These results suggested that the growth performance of growing pigs fed protein restricted diets supplemented with BCAA could catch up to that of the pigs fed AP diets. 4The diagnostic rules of peripheral lung cancer preliminary study based on data mining technique显示文摘Objective: To discuss the clinical and imaging diagnostic rules of peripheral lung cancer by data mining technique, and to explore new ideas in the diagnosis of peripheral lung cancer, and to obtain early-stage technology and knowledge support of computer-aided detecting(CAD). Methods: 58 cases of peripheral lung cancer confirmed by clinical pathology were collected. The data were imported into the database after the standardization of the clinical and CT findings attributes were identified. The data was studied comparatively based on Association Rules (AR) of the knowledge discovery process and the Rough Set (RS) reduction algorithm and Genetic Algorithm(GA) of the generic data analysis tool (ROSETTA), respectively. Results: The genetic classification algorithm of ROSETTA generates 5 000 or so diagnosis rules. The RS reduction algorithm of Johnson`s Algorithm generates 51 diagnosis rules and the AR algorithm generates 123 diagnosis rules. Three data mining methods basically consider gender, age, cough, location, lobulation sign, shape, ground-glass density attributes as the main basis for the diagnosis of peripheral lung cancer. Conclusion: These diagnosis rules for peripheral lung cancer with three data mining technology is same as clinical diagnostic rules, and these rules also can be used to build the knowledge base of expert system. 5Protective effects of taurine against muscle damage induced by diquat in 35 days weaned piglets显示文摘Background: Oxidative stress is a key factor that influences piglets' health. Taurine plays an imperative role in keeping the biological system from damage. This study was conducted to investigate the protective effect of taurine against muscle injury due to the secondary effect of diquat toxicity.Results: Our study found that taurine effectively and dose-dependently alleviated the diquat toxicity induced rise of feed/gain, with a concurrent improvement of carcass lean percentage. The plasma content of taurine was considerably increased in a dose-dependent manner. Consequently, dietary taurine efficiently improved the activity of plasma antioxidant enzymes. Furthermore, taurine attenuated muscle damage by restoring mitochondrial micromorphology, suppressing protein degradation and reducing the percentage of apoptotic cells in the skeletal muscle. Taurine supplementation also suppressed the genes expression levels of the antioxidant-, mitochondrial biogenesis-, and muscle atrophy-related genes in the skeletal muscle of piglets with oxidative stress.Conclusions: These results showed that the dose of 0.60% taurine supplementation in the diet could attenuate skeletal muscle injury induced by diquat toxicity. 6Dietary proline supplementation alters colonic luminal microbiota and bacterial metabolite composition between days 45 and 70 of pregnancy in Huanjiang mini-pigs显示文摘Background: Pregnancy is associated with important changes in gut microbiota composition. Dietary factors may affect the diversity, composition, and metabolic activity of the intestinal microbiota. Among amino acids, proline is known to play important roles in protein metabolism and structure, cell differentiation, conceptus growth and development, and gut microbiota re-equilibration in case of dysbiosis.Results: Dietary supplementation with 1% proline decreased(P < 0.05) the amounts of Klebsiella pneumoniae,Peptostreptococcus productus, Pseudomonas, and Veillonella spp. in distal colonic contents than that in the control group. The colonic contents of Butyrivibrio fibrisolvens, Bifidobacterium sp., Clostridium coccoides, Clostridium coccoides-Eubacterium rectale, Clostridium leptum subgroup, Escherichia coli, Faecalibacterium prausnitzii,Fusobacterium prausnitzii, and Prevotella increased(P < 0.05) on d 70 of pregnancy as compared with those on d 45 of pregnancy. The colonic concentrations of acetate, total straight-chain fatty acid, and total short-chain fatty acids(SCFA) in the proline-supplemented group were lower(P < 0.05), and butyrate level(P = 0.06) decreased as compared with the control group. Almost all of the SCFA displayed higher(P < 0.05) concentrations in proximal colonic contents on d 70 of pregnancy than those on d 45 of pregnancy. The concentrations of 1,7-heptyl diamine(P = 0.09) and phenylethylamine(P < 0.05) in proximal colonic contents were higher, while those of spermidine(P = 0.05) and total bioamine(P = 0.06) tended to be lower in the proline-supplemented group than those in the control group. The concentrations of spermidine, spermine, and total bioamine in colonic contents were higher(P < 0.05) on d 70 of pregnancy than those measured on d 45 of pregnancy. In contrast, the concentration of phenylethylamine was lower(P < 0.05) on d 70 than on d 45 of pregnancy.(Continued on next page)(Continued from previous page)Conclusion: These findings indicate that L-proline supplementation modifies both the colonic microbiota composition and the luminal concentrations of several bacterial metabolites. Furthermore, our data show that both the microbiota composition and the concentrations of bacterial metabolites are evolving in the course of pregnancy. 7Ribozyme probe based on molecular beacon for real time monitoring of enzymatic cleavage process显示文摘Ribozyme probe based on molecular beacon (MBR) for monitoring enzymatic cleavage process in real time is designed and studied. The approach relies on ri-bozyme substrates modified at the two arms, with a fluores-cent moiety attached to the end of one arm and a non-fluorescent quenching moiety attached to the end of the other arm. MBR is employed to directly convert the cleavage information into fluorescence signal in real time. Compared with traditional approach, this method provides a no-radiolabeling, sensitive and effective way to research on the ribozyme activity, enzymatic dynamic process and ri-bozyme function during gene therapy. 8Novel protein detection method based on proximity- dependent polymerase reaction and aptamers显示文摘In recent years, specific detection of proteins is one of the hot issues about aptamers in proteomics. Here we reported a simple, sensitive and specific proximity-dependent protein assay with dual DNA aptamers. Thrombin was used as the model protein, and two aptamer probes with complementary se- quence at 3′-end were designed for the two distinct epitopes of the protein. Association of the two ap- tamers with thrombin resulted in stable hybrids due to the proximity of 3′-end, then polymerase reac- tion was induced. The amount of obtained dsDNA was indicated using the fluorescence dye Sybr Green I. The results showed that the initial velocity of polymerase reaction had a positive correlation with concentration of thrombin. The advantages of this dual-aptamer-based approach included simple and flexible design of aptamer probes, high selectivity and high sensitivity. 9Comparisons of carcass traits,meat quality,and serum metabolome between Shaziling and Yorkshire pigs显示文摘This study aims to compare the meat quality of Shaziling and Yorkshire pigs and to find the potential indicator in serum for superior meat quality.Six Shaziling and Yorkshire pigs at 30,60,90,150,210,and 300 d of age were selected to examine carcass traits,meat quality,and serum metabolome.The results showed that the body weight,carcass length,and loin eye area of Shaziling pigs at 150,210,and 300 d of age were significantly lower than those of Yorkshire pigs(P<0.05).Shaziling pigs at 150 and 300 d of age had significantly lower backfat thickness than Yorkshire pigs(P<0.05).Compared with Yorkshire pigs,Shaziling pigs at all 6 ages had a lower lean percentage and a higher fat percentage(P<0.05).At 60,90,and 150 d of age,the post-mortem pH-decline,b*value(yellowness),and drip loss of Shaziling pigs were significantly lower than those of Yorkshire pigs(P<0.05).Moreover,at 150 d of age,Shaziling pigs had significantly higher a^(*)value(redness)and intramuscular fat(IMF)content than Yorkshire pigs(P<0.05).Correlation analysis between the top 40 metabolites and phenotypes indicated that L-carnitine had positive correlations with fat percentage,pH24h,and IMF content,but had negative correlations with lean percentage,L^(*)value(lightness),and b^(*)value(P<0.05).Serum L-carnitine content,fat percentage,pH24h,and IMF content all decreased first and then increased as the pigs grew,which verified the positive correlations between L-carnitine and these phenotypes.In conclusion,Shaziling pigs have a slower growth rate but a better meat quality than Yorkshire pigs.The meat quality of Shaziling pigs is the best from 150 to 210 d of age.This study suggests that a higher serum L-carnitine content is a promising indicator for better meat quality.Bo Song Changbing Zheng Jie Zheng Shiyu Zhang Yinzhao Zhong Qiuping Guo Fengna Li Cimin Long Kang Xu Yehui Duan Yulong Yin 2022Animal Nutrition2022,,1:3
10Protein analysis based on molecular beacon probes and biofunctionalized nanoparticles显示文摘With the completion of the human genome-sequencing project, there has been a resulting change in the focus of studies from genomics to proteomics. By utilizing the inherent advantages of molecular beacon probes and biofunctionalized nanoparticles, a series of novel principles, methods and techniques have been exploited for bioanalytical and biomedical studies. This review mainly discusses the applications of molecular beacon probes and biofunctionalized nanoparticles-based technologies for realtime, in-situ, highly sensitive and highly selective protein analysis, including the nonspecific or specific protein detection and separation, protein/DNA interaction studies, cell surface protein recognition, and antigen-antibody binding process-based bacteria assays. 12Profiles of muscular amino acids,fatty acids,and metabolites in Shaziling pigs of different ages and relation to meat quality显示文摘Pork meat is closely related to physicochemical alterations during growth and development,resulting in differences in nutritional value and meat flavor.This study aimed to evaluate the composition of amino acids,fatty acids,and metabolic profiles in the longissimus thoracis muscle(LM)of Shaziling pigs aged 30,90,150,210,and 300 days.The results showed that the predominant fatty acids identified in the LM of Shaziling pigs were C16:0,C16:1,C18:0,C18:1n9c,and C18:2n6c.An opposite correlation was observed for C18:2n6c and n6/n3 polyunsaturated fatty acids(P<0.05).Alanine,aspartate,glutamate,Dglutamine,and D-glutamate metabolism were the main metabolic pathways for the Shaziling pig meat flavor(P<0.05).Moreover,the correlation coefficients revealed that the contents of anserine,C16:0,C16:1,and C18:1n9c were positively correlated with intramuscular fat and/or p H24hand were negatively correlated with the values of L^(*)(lightness)and b^(*)(yellowness)(P<0.05).In conclusion,age greatly affected the meat quality of Shaziling pigs,and the contents of muscular anserine,C16:0,C16:1,and C18:1n9c might be promising indicators for better meat quality.Yehui Duan Changbing Zheng Jie Zheng Lu Ma Xinran Ma Yinzhao Zhong Xichen Zhao Fengna Li Qiuping Guo Yulong Yin 2023Science China(Life Sciences)2023,66,6:2
13Identification of a major quantitative trait locus and its candidate underlying genetic variation for rice stigma exsertion rate显示文摘Stigma exsertion in male sterile lines of hybrid rice is important for seed yield.In the present study, ZS616 [Oryza sativa subsp.Xian(indica)], a male sterile line with a stigma exsertion rate(SER) as high as 94.5%, was crossed to DS552, a japonica line with almost no exserted stigmas.F3 plants with extremely low and high SER were sequenced to identify SER-associated quantitative trait loci(QTL).A major QTL for SER, qSER-3.1, was identified along with other QTL on chromosome 3 in a 3.9 Mb region.A total of 307 nonsynonymous single-nucleotide polymorphisms(SNPs) and 27 frame-shift insertion/deletions(InDels)differentiating ZS616 and DS552 were identified in the region containing qSER-3.1.Most SNPs(294) and InDels(25) were excluded after further analysis because they were shared by ZS616 and low(<2.0%) SER accessions in the Huazhong Agricultural University(HAU) core rice collection.Association analysis using the full HAU collection identified a 17-bp InDel in OS03 G0689400 as the most likely causal genetic variant underlying qSER-3.1.ZS616-type accessions(n = 54, with the 17-bp insertion) in the HAU collection had minimum(16.5%)and mean(39.6%) SERs significantly greater than those(n = 424) without the insertion(with minimum and mean SERs of 0.2% and 20.6%, respectively).Thus, this study identified a major QTL for stigma exsertion and revealed the mutation in a candidate gene for the QTL.Shouling Xu Yunchao Zheng Yang Liu Xiaohao Guo Yuanyuan Tan Qiuping Qian Qingyao Shu Jianzhong Huang 2019The Crop Journal2019,7,3:2
14Platinum-crosslinking polymeric nanoparticle for synergetic chemoradiotherapy of nasopharyngeal carcinoma显示文摘Despite extensive use of radiotherapy in nasopharyngeal carcinoma(NPC)treatment because of its high radiosensitivity,there have been huge challenges in further improving therapeutic effect,meanwhile obviously reducing radiation damage.To this end,synergistic chemoradiotherapy has emerged as a potential strategy for highly effective NPC therapy.Here,we developed RGD-targeted platinum-based nanoparticles(RGD-PtNPs,denoted as RPNs)to achieve targeted chemoradiotherapy for NPC.Such nanoparticles consist of an RGD-conjugated shell and a cis-platinum(CDDP)crosslinking core.Taking advantage of RGD,the RPNs may effectively accumulate in tumor,penetrate into tumor tissues and be taken by cancer cells,giving rise to a high delivery efficiency of CDDP.When they are fully enriched in tumor sites,the CDDP loaded RPNs can act as radiotherapy sensitizer and chemotherapy agents.By means of X-ray-promoted tumor cell uptake of nanoparticle and CDDP-induced cell cycle arrest in radiation-sensitive G2/M phases,RPNs may offer remarkable therapeutic outcome in the synergistic chemoradiotherapy for NPC.Yuxun Ding Xiaohui Xiao Lingli Zeng Qiuping Shang Wei Jiang Sha Xiong Xiaohui Duan Jun Shen Ruibing Wang Jinshan Guo Yue Pan 2021Bioactive Materials2021,6,12:1

18Research on consistency of identifying solitary pulmonary masses with CT显示文摘Objective:To research on consistency of identifying solitary pulmonary masses with CT. Methods:Three observers with different working backgrounds in imaging diagnosis individually interpreted the same group images of solitary pulmonary mass, by 12 indexes of objective signs. The differences in interpretation resulted in ante- and post-interpretations were assessed by the x2 test. The agreement of two interpretations from the same observer was confirmed with the kappa test. A double-blind method was adopted for analysis. Results:The agreement rates of ante- and post-interpreting from the three observers were respectively 82.65%(486/588) 80.27%(472/588) and 84.86% (499/588) while their interpreting results were generally accordant without significant difference(x2 = 4.975, df = 2, P = 0.083) however there was difference between the observer 2 and observer 3(x2 = 4.875, df = 1, P = 0.027). There were five indexes with k > 0.40 of ante- and post-interpreting results of the three observers, including clarity of nodule borderline, presence of sentus, uniformity of density, existence of cavity and calcification in pathological region, among them, the agreement rate of interpreting borderline and cavity was higher(k > 0.07); the blood vessel convergence poorer(0 < k ≤ 0.40); the other six CT signs of interpretation were slightly different.
