| 9 | Expression and purification of Ctomp2aa167-aa434 and preparation of its mAb显示文摘Chlamydia trachomatis outer membrane protein 2 (Ctomp2) is a major immunogen in chlamydial infections and a highly genus-conserved structural protein of all Chlamydia species . To purify the protein and to prepare monoclonal antibodies (mAbs) against it, the recombinant protein was induced by IPTG, which was confirmed by SDS-PAGE and purified by means of a Ni2+ -charged resin column. The denatured protein was refolded in the GSH-GSSH buffer gradually and identified by Western blotting. Then the BALB/c mice were immunized with the recombinant protein to prepare the mAb against Ctomp2. The obtained mAbs were characterized. Genital specimens were tested with indirect ELISA mostly made of the mAb and cell culture in 84 patients with genital symptoms. The results showed that high-level expression of the recombinant protein was achieved, which existed as inclusion body and amounted to 38 % of total bacterium protein. A mAb against Ctomp2 was obtained. It belongs to IgG 2b. The titers were as high as 1:40 000. The Western blotting showed that the mAb only reacted with the recombinant protein. It had no crossing reactions against E. coli, N. gonorhoea, M. hominis, U. urealyticum and M. penetrans . It had high specifity. In comparison with gold standard test-cell culture, the sensitivities, specificities, positive predictive values and negative predictive values of indirect ELISA were 95.24%, 100%, 100% and 98.44%, respectively. | 11 | Morphologic evolution of bifurcated reaches in a macrotidal estuary with mountain streams显示文摘A macrotidal estuary with mountain streams(MEMS)is characterized by rapidly rising and falling flood peaks and large tidal ranges and is a typical estuary type with strong flow dynamics that is found worldwide.Understanding the morphodynamic evolution of MEMSs has great significance for river management.The roles of the mountain stream river flood process and macrotides on the morphology evolution still needs further quantitative study.Taking the Oujiang Estuary as an example,the evolution mechanisms of bifurcated reaches of a MEMS are studied by analyzing the hydrologic and topographic data and using a two-dimensional numerical model for flow-sediment transport.The results show that(1)under the combined actions of runoff and macrotides,sediment transport is active and morphological evolution is intensive,the main branch and subbranch historically switch frequently,and erosion generally occurs in wet years,while deposition occurs in dry years.(2)The river flood process of mountain streams causes rapid erosion,which rises and falls rapidly with a kurtosis index(K)of approximately 2.9.There is a logarithmic relation between the sediment transport load and K.The volume of sediment transport will be 40%larger than the river flood process with K=1.2 as for the Yangtze River.(3)Macrotides with large tidal ranges could enhance the discharge asymmetry(ψ).When the ψ is greater than 1,the side branches are dominated by flood tides.Although the current study is site specific,the results are expected to provide a valuable reference for sustainable management in similar estuaries. | Jing Liu Yongjun Lu Liqin Zuo Huaixiang Liu Qun Xu Qingzhi Hou | 2022 | International Journal of Sediment Research2022,37,6: | 0 |

| 13 | Intracortical screw insertion plus limited open reduction in treating type 31A3 irreducible intertrochanteric fractures in the elderly显示文摘BACKGROUND In most elderly patients with intertrochanteric fractures,satisfactory fracture reduction can be achieved by closed reduction using a traction table.However,intertrochanteric fractures cannot achieve satisfactory reduction in a few patients,which is called irreducible intertrochanteric fractures.Especially for type 31A3 irreducible intertrochanteric fractures,limited open reduction of the broken end with different intraoperative reduction methods is required to achieve satisfactory reduction and fixation.AIM To discuss clinical efficacy of intracortical screw insertion plus limited open reduction in type 31A3 irreducible intertrochanteric fractures in the elderly.METHODS A retrospective analysis was performed on 23 elderly patients with type 31A3 irreducible intertrochanteric fractures(12 males and 11 females,aged 65-89-yearsold)who received treatment at the orthopedics department.After type 31A3 irreducible intertrochanteric fractures were confirmed by intraoperative C-arm,all of these cases received intracortical screw insertion plus limited open reduction in the broken end with intramedullary screw internal fixation.The basic information of surgery,reduction effects,and functional recovery scores of the hip joint were assessed.RESULTS All patients were followed up for 13.8 mo on average.The operation time was 53.8±13.6 min(40-95 min).The intraoperative blood loss was 218.5±28.6 mL(170-320 mL).The average number of intraoperative X-rays was 22.8±4.6(18-33).The average time to fracture union was 4.8±0.7 mo.The reduction effect was assessed using Kim’s fracture reduction evaluation.Twenty cases achieved grade I fracture reduction and three cases grade II fracture reduction.All of them achieved excellent or good fracture reduction.Upon the last follow-up,the functional recovery scores score was 83.6±9.8,which was not significantly different from the functional recovery scores score(84.8±10.7)before the fracture(t=0.397,P=0.694).CONCLUSION With careful preoperative preparation,intracortical screw insertion plus limited open reduction contributed to high-quality fracture reduction and fixation.Good clinical outcomes were achieved without increasing operation time and intraoperative blood loss. | Xiao-Wen Huang Gu-Qi Hong Qiang Zuo Qun Chen | 2021 | World Journal of Clinical Cases2021,9,32: | 0 |

| 15 | Effect of Huoxue Huayu Detoxification on GSK-3β and β-catenin levels in OSF patients and its mechanism显示文摘Objective:To explore the effects of Huoxue Huayu detoxification method on the levels of GSK-3βandβ-catenin in OSF patients and its mechanism.Methods:A total of 120 patients with oral submucosal fibrosis(OSF)treated in our hospital from May 2018 to December 2019 were selected and divided into routine treatment group and Huoxue Huayu detoxification group of 60 cases according to the lottery method.The routine treatment group was treated with triamcinolone acetonide injection,and the Huoxue Huayu detoxification group was treated with Huoxue Huayu detoxification.The lesion area,mouth opening degree,SFRP1 level in gingival crevicular fluid and saliva,TCM symptom score,TGF-β1,IL-6,GSK-3β,β-catenin expression and clinical effect were observed in the two groups.Results:After treatment,the lesion area of oral mucosa in the Huoxue Huayu detoxification group was smaller than that in the routine treatment group(P<0.05),and the mouth opening degree in the Huoxue Huayu detoxification group was larger than that in the routine treatment group(P<0.05);After treatment,the level of SFRP1 in gingival crevicular fluid and saliva and clinical effect in the Huoxue Huayu detoxification group were higher than those in the routine treatment group(P<0.05);the TCM syndrome score,TGF-β1,IL-6,GSK-3β,β-catenin expression in the Huoxue Huayu detoxification group were lower than those in the routine treatment group(P<0.05).Conclusion:By acting on Wnt/β-catenin signaling pathway,Huoxue Huayu detoxification method can regulate the expression of TGF-β1,IL-6,GSK-3β,β-catenin,improve the clinical symptoms of patients with oral submucous fibrosis,alleviate the body inflammation,alleviate the pain,with remarkable effect. | Qiao-Juan Zuo Jin Tan Jie Yu Shi-Juan Chen Qun Li Xuan Zhu | 2021 | Journal of Hainan Medical University2021,27,5: | 0 |
| 16 | Highly Dynamic Polynuclear Metal Cluster Revealed in a Single Metallothionein Molecule显示文摘Human metallothionein(MT)is a small-size yet efficient metal-binding protein,playing an essential role in metal homeostasis and heavy metal detoxification.MT contains two domains,each forming a polynuclear metal cluster with an exquisite hexatomic ring structure.The apoprotein is intrinsically disordered,which may strongly influence the clusters and the metal-thiolate(M-S)bonds,leading to a highly dynamic structure.However,these features are challenging to identify due to the transient nature of these species.The individual signal from dynamic conformations with different states of the cluster and M-S bond will be averaged and blurred in classic ensemble measurement.To circumvent these problems,we combined a single-molecule approach and multiscale molecular simulations to investigate the rupture mechanism and chemical stability of the metal cluster by a single MT molecule,focusing on the Zn4S11 cluster in theαdomain upon unfolding.Unusual multiple unfolding pathways and intermediates are observed for both domains,corresponding to different combinations of M-S bond rupture.None of the pathways is clearly preferred suggesting that unfolding proceeds from the distribution of protein conformational substates with similar M-S bond strengths.Simulations indicate that the metal cluster may rearrange,forming and breaking metal-thiolate bonds even when MT is folded independently of large protein backbone reconfiguration.Thus,a highly dynamic polynuclear metal cluster with multiple conformational states is revealed in MT,responsible for the binding promiscuity and diverse cellular functions of this metal-carrier protein. | Guodong Yuan Felipe Curtolo Yibing Deng Tao Wu Fang Tian Qun Ma Yutong Liu Jinglin Zuo Guilherme Menegon Arantes Peng Zheng | 2021 | Research2021,,1: | 0 |
