3Template-module assembly to prepare low-molecular-weight gene transport system with enhanced transmembrane capability显示文摘Based on specific host-guest interactions between amine-modified ?-cyclodextrin(CD-TAEA) and functional adamantane(AD) derivatives, a module-template strategy has been proposed for the construction of low-molecular-weight cationic assemblies for gene transport. This strategy offers great flexibility in terms of the introduction of mono- or multi-functionality by the inclusion of one or more adamantane-based modules with the desired functionalities. As proof of concept, phenylboronic acid(PB) containing adamantane(PB-AD) was used as a model module in the hope of offering enhanced cytosolic delivery in consideration of the special affinity of PB groups with cell membranes. The physicochemical properties of the complexes formed with plasmid DNA, such as particle size, zeta potential and morphology were investigated. Confocal laser scanning microscopy and flow cytometry experiments demonstrated the important contribution of the functional PB-AD module to the considerably enhanced intracellular internalization and uptake by cellular nuclei. Compared to the parent CD-TAEA, PB-AD/CDTAEA assemblies mediated higher transfection rates, which were even comparable to that of PEI25K. In addition, PB-AD/CDTAEA displayed much lower cytotoxicity than PEI25K in both 293T and HeLa cell lines. 4Research progress of magnetic resonance imaging contrast agents显示文摘Magnetic resonance imaging (MRI) is a clinical diagnostic modality, which has become popular in hospitals around the world. Approximately 30% of MRI exams include the use of contrast agents. The research progress of the paramagnetic resonance imaging contrast agents was described briefly. Three important approaches in the soluble paramagnetic resonance imaging contrast agents design including nonionic, tissue-specific and macromolecular contrast agents were investigated. 5Rates of oxygen consumption and tolerance of hypoxia and desiccation in Chinese black sleeper(Bostrichthys sinensis)and mudskipper(Boleophthalmus pectinirostris)embryos显示文摘The rates of oxygen consumption, tolerance of hypoxia and desiccation of the Chinese black sleeper (Bostrichthys sinensis) and mudskipper (Boleophthalmus pectinirostris) embryos were investigated. The pattern of oxygen consumption of the Chinese black sleeper embryos was similar to that of the mudskipper ones. The lowest rates of oxygen consumption[(1.65±0.66)nmol/ (ind.·h)] of the Chinese black sleeper embryos 16 h after fertilization and the lowest rates of oxygen consumption [(0.79± 0.08)nmol/(ind.·h)] of the mudskipper embryos 6 h after fertilization were recorded, respectively. Then the rates of oxygen consumption of these two species embryos increased gradually until hatching [(8.26±1.70)nmol/(ind.·h)in the Chinese black sleeper, (2.69±0.23)nmol/(ind.·h) in mudskipper]. After exposure to hypoxia water (0.16 mg/dm3), bradycardia of the embryos occurred in both the Chinese black sleeper and the mudskipper. However, the Chinese black sleeper embryos survived approximately 45 min longer than the mudskipper ones. 6Post-transcriptional regulator Rbm47 elevates IL-10 production and promotes the immunosuppression of B cells显示文摘Regulatory B cells(Bregs)are a functionally defined B cell subset,and IL-10 is crucial for the suppressive functions of Bregs.However,little is known regarding how IL-10 production is regulated in B cells.To explore the mechanisms by which IL-10 is regulated in B cells,we used mRNA microarrays to screen for molecules that are upregulated in IL-10-producing B cells and identified RNA-binding motif protein 47(Rbm47)as a post-transcriptional regulator.Rbm47 was found to promote IL-10 production in B cells.We found that Rbm47 promotes the stability of IL-10 mRNA by binding to AU-rich elements in the 3′untranslated region of Il10 mRNA.In addition,we demonstrated that the overexpression of Rbm47 enabled B cells to facilitate Foxp3+regulator T-cell induction and reduce the severity of DSS-induced ulcerative colitis.Taken together,these results suggest that Rbm47 plays an important role in regulating IL-10 at the post-transcriptional level,thus promoting the regulatory functions of B cells.The findings presented in this study not only increase our understanding of the post-translational regulation of IL-10 in B cells but also identify a novel strategy for the potential application of Bregs.Yinxiang Wei Fanghui Zhang Yu Zhang Xiaoqian Wang Chen Xing Jing Guo Hui Zhang Zhimin Suo Yan Li Jianli Wang Renxi Wang Zhijian Cai 2019Cellular & Molecular Immunology2019,16,6:2
7The Effect of CD3-Specific Monoclonal Antibody on Treating Experimental Autoimmune Myasthenia Gravis显示文摘CD3-specific monoclonal antibody was the first one used for clinical practice in field of transplantation. Recently,renewed interests have elicited in its capacity to prevent autoimmune diabetes by inducing immune tolerance. In this study, we tested whether this antibody can also be used to treat another kind of autoimmune disease myasthenia gravis (MG) and explored the possible mechanisms. MG is caused by an autoimmune damage mediated by antibody- and complement-mediated destruction of AChR at the neuromuscular junction. We found that administration of CD3-specific antibody (Fab)2 to an animal model with experimental autoimmune myasthenia gravis (EAMG) (B6 mice received 3 times of AChR/CFA immunization) could not significantly improve the clinical signs and clinical score. When the possible mechanisms were tested, we found that CD3 antibody treatment slightly down-regulated the T-cell response to AChR, modestly up-regulation the muscle strength. And no significant difference in the titers of IgG2b was found between CD3 antibody treated and control groups. These data indicated that CD3-specific antibody was not suitable for treating MG, an antibody- and complementmediated autoimmune disease, after this disease has been established. Wavelet transform has many advantages and has become a trend in data compression. 9Isolation and characterization of 49 polymorphic microsatellite loci for Decapterus maruadsi using SLAF-seq, and cross-amplifi cation to related species显示文摘Decapterus maruadsi is a commercially important species in China, but has been heavily exploited in some areas. There is a growing need to develop microsatellites promoting its genetic research for the adequate management of this ?shery resources. The recently developed speci?c-locus ampli?ed fragment sequencing(SLAF-seq) is an effcient and high-resolution method for genome-wide microsatellite markers discovery. In this study, 28 905 microsatellites(mono-to hexa-nucleotide repeats) were identi?ed using SLAF-seq technology, of which di-nucleotide was the most frequent(13 590, 47.02%), followed by mono-nucleotide(8 138, 28.15%), tri-nucleotide(5 727, 19.81%), tetra-nucleotide(1 104, 3.82%), pentanucleotide(234, 0.81%), and hexa-nucleotide(112, 0.39%). One hundred and thirty-two microsatellite loci(di-and tri-nucleotide) were randomly selected for ampli?cation and polymorphism, of which 49 were highly polymorphic and well-resolved. The average number of alleles per locus was 13.63, ranging from 4 to 25, and allele sizes varied between 110 bp and 309 bp. The observed heterozygosity( Ho) and expected heterozygosity( He) ranged from 0.233 to 1.000 and from 0.374 to 0.959, with mean values of 0.738 and0.836, respectively. The polymorphism information content(PIC) ranged from 0.341 to 0.941(mean=0.806).However, 12 loci deviated from Hardy-Weinberg equilibrium. Furthermore, transferability tests were also successful in validating the utility of the developed markers in ?ve phylogenetically related species of family Carangidae.
