| 1 | Current therapeutic strategies for advanced pancreatic cancer: A review for clinicians显示文摘Pancreatic cancer(PC) would become the second leading cause of cancer death in the near future, despite representing only 3% of new cancer diagnosis. Survival improvement will come from a better knowledge of risk factors, earlier diagnosis, better integration of locoregional and systemic therapies, as well as the development of more efficacious drugs rising from a deeper understanding of disease biology. For patients with unresectable, non-metastatic disease, combined strategies encompassing primary chemotherapy and radiation seems to be promising. In fit patients, new polychemotherapy regimens can lead to better outcomes in terms of slight but significant survival improvement associated with a positive impact on quality of life. The upfront use of these regimes can also increase the rate of radical resections in borderline resectable and locally advanced PC. Second line treatments showed to positively affect both overall survival and quality of life in fit patients affected by metastatic disease. At present, oxaliplatin-based regimens are the most extensively studied. Nonetheless, other promising drugs are currently under evaluation. Presently, in addition to surgery and conventional radiation therapy, new locoregional treatment techniques are emerging as alternative options in the multimodal approach to patients or diseases not suitable for radical surgery. As of today, in contrast with other types of cancer, targeted therapies failed to show relevant activity either alone or in combination with chemotherapy and, thus, current clinical practice does not include them. Up to now, despite the fact of extremely promising results in different tumors, also immunotherapy is not in the actual therapeutic armamentarium for PC. | 2 | Novel hepatocellular carcinoma molecules with prognostic and therapeutic potentials显示文摘Hepatocellular carcinoma(HCC), the predominant form of primary liver cancer, is the sixth most common cancer worldwide and the third leading cause of cancerrelated death. The difficulty to diagnose early cancer stages, the aggressive behaviors of HCC, and the poor effectiveness of therapeutic treatments, represent the reasons for the quite similar deaths per year and incidence number. Considering the fact that the diagnosis of HCC typically occurs in the advanced stages of the disease when the therapeutic options have only modest efficacy, the possibility to identify early diagnostic markers could be of significant benefit. So far, a large number of biomarkers have been associated to HCC progression and aggressiveness, but many of them turned out not to be of practical utility. This is the reason why active investigations are ongoing in this field. Given the huge amount of published works aimed at the identification of HCC biomarkers, in this review we mainly focused on the data published in the last year, with particular attention to the role of(1) molecular and biochemical cellular markers;(2) micro-interfering RNAs;(3) epigenetic variations; and(4) tumor stroma. It is worth mentioning that a significant number of the HCC markers described in the present review may be utilized also as targets for novel therapeutic approaches, indicating the tight relation between diagnosis and therapy. | 3 | A pilot study of antioxidant potential of endophytic fungi from some Sudanese medicinal plants显示文摘Objective: To evaluate the total phenolic content and total antioxidant capacity of ethyl acetate extracts of 21 endophytic fungi isolated from five Sudanese medicinal plants: Calotropis procera, Catharanthus roseus, Euphorbia prostrate, Vernonia amygdalina and Trigonella foenumgraecum. Methods: Crude extracts of endophytic fungi and their host plants were tested by classical Folin-Ciocalteu colorimetric method to determine the total phenolic content, also total antioxidant capacity was estimated using 1,1-diphenyl-2-picrylhydrazyl free radical scavenging in vitro method. Results: Among the endophytes, endophytic fungus Aspergillus sp. from Trigonella foenum-graecum seeds demonstrated the highest both total phenolic content in term of gallic acid equivalent(GAE) [(89.9±7.1 mg GAE/g)] and antioxidant activity for 1,1-diphenyl-2-picrylhydrazyl radical scavenging assay [IC50:(18.0±0.1 μg/m L)]. A high positive linear correlation(R2=0.999 1) was found between total antioxidant capacity and total phenolic content of endophytic fungi isolated from Vernonia amygdalina. | 4 | Gastric cancer:The times they are a-changin'显示文摘Gastric cancer is the third leading cause of cancer death worldwide.Even though during these last decades gastric cancer incidence decreased in Western countries,it remains endemic and with a high incidence in Eastern countries.The survival in advanced and metastatic stage of gastric cancer is still very poor.Recently the Cancer Genoma Atlas Research Network identified four subtypes with different molecular profiles to classify gastric cancer in order to offer the optimal targeted therapies for pre-selected patients.Indeed,the key point is still the selection of patients for the right treatment,on basis of molecular tumor characterization.Since chemotherapy reached a plateau of efficacy for gastric cancer,the combination between cytotoxic therapy and biological agents gets a better prognosis and decreases chemotherapeutic toxicity.Currently,Trastuzumab in combination with platinum and fluorouracil is the only approved targeted therapy in the first line for c-erb B2 positive patients,whereas Ramucirumab is the only approved targeted agent for patients with metastatic gastric cancer.New perspectives for an effective treatment derived from the immunotherapeutic strategies.Here,we report an overview on gastric cancer treatments,with particular attention to recent advances in targeted therapies and in immunotherapeutic approach. | Maria Antonietta Satolli Lucio Buffoni Rosella Spadi Ilaria Roato | 2015 | World Journal of Gastrointestinal Oncology2015,7,11: | 6 |

The enantioselective separation was accomplished on the immobilized-type Chiralpak ID-3 chiral stationary phase(CSP)under reversed-phase conditions. The results were evaluated and compared with those obtained by the of fi cial enantioselective method of European Pharmacopoeia used as the reference for checking the enantiomeric excess of(S)-OME. It has been established that the use of the Chiralpak ID-3 CSP allows the determination of the enantiomeric purity of(S)-OME without any interference coming from its chiral and achiral related substances. | 11 | Unusual retention behavior of omeprazole and its chiral impurities B and E on the amylose tris(3-chloro-5-methylphenylcarbamate)chiral stationary phase in polar organic mode显示文摘Recent reports have demonstrated that the new commercially available immobilized-type chiral stationary phases(CSPs) containing amylose tris(3-chloro-5-methylphenylcarbamate)(ACMPC) as a selector exhibit not only an exceptionally high enantioselectivity in high-performance liquid chromatography(HPLC) but they are also applicable to a wide range of chiral analytes. Herein, we report the results obtained in the HPLC analysis of omeprazole and its impurities B and E on the ACMPC-based Chiralpak IG-3 CSP(CSP) under polar organic conditions. A systematic evaluation of the retention characteristics of the selected benzimidazole chiral probes was carried out by changing the composition of the mobile phase and the column temperature. It is worth emphasizing that the high affinity of both enantiomers of all analytes recorded in pure methanol mode dramatically decreased incorporating small volumes of either basic or acid additives in the mobile phase.
