2Endocrine Disruption Activity of 30-day Dietary Exposure to Decabromodiphenyl Ethane in Balb/C Mouse显示文摘Objective This study aimed to evaluate the hepatotoxicity,metabolic disturbance activity and endocrine disrupting activity of mice treated by Decabromodiphenyl ethane(DBDPE).Methods In this study,Balb/C mice were treated orally by gavage with various doses of DBDPE.After 30 days of treatment,mice were sacrificed; blood,livers and thyroid glands were obtained,and hepatic microsomes were isolated.Biochemical parameters including 8 clinical chemistry parameters,blood glucose and hormone levels including insulin and thyroid hormone were assayed.The effects of DBDPE on hepatic cytochrome P450(CYP) levels and activities and uridinediphosphate-glucuronosyltransferase(UDPGT) activities were investigated.Liver and thyroid glands were observed.Results There were no obvious signs of toxicity and no significant treatment effect on body weight,or liver-to-body weight ratios between treatment groups.The levels of ALT and AST of higher dose treatment groups were markedly increased.Blood glucose levels of treatment groups were higher than those of control group.There was also an induction in TSH,T3,and f T3.UDPGT,PROD,and EROD activities were found to have been increased significantly in the high dose group.Histopathologic liver changes were characterized by hepatocyte hypertrophy and cytoplasmic vacuolization.Our findings suggest that DBDPE can cause a certain degree of mouse liver damage and insufficiency.Conclusion DBDPE has the activity of endocrine disruptors in Bal/C mice,which may induce drug-metabolizing enzymes including CYPs and UDPGT,and interfere with thyroid hormone levels mediated by Ah R and CAR signaling pathways.Endocrine disrupting activity of DBDPE could also affect the glucose metabolism homeostasis.SUN Ru Bao SHANG Shuai ZHANG Wei LIN Ben Cheng WANG Qiang SHI Yun XI Zhu Ge 2018Biomedical and Environmental Sciences2018,31,1:7
3Cytotoxicity and Apoptosis Induction in Human HepG2 Hepatoma Cells by Decabromodiphenyl Ethane显示文摘Objective To investigate the toxic effects of decabromodiphenyl ethane (DBDPE), used as an alternative to decabromodiphenyl ether in vitro. Methods HepG2 cells were cultured in the presence of DBDPE at various concentrations (3.125-100.0 mg/L) for 24, 48, and 72 h respectively and the toxic effect of DBDPE was studied. Results As evaluated by the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide and lactate dehydrogenase assays and nuclear morphological changes, DBDPE inhibited HepG2 viability in a time- and dose-dependent manner within a range of 12.5 mg/L to 100 mg/L and for 48 h and 72 h. Induction of apoptosis was detected at 12.5-100 mg/L at 48 h and 72 h by propidium iodide staining, accompanied with overproduction of reactive oxygen species (ROS). Furthermore, N-acetyl-L-cysteine, a widely used ROS scavenger, significantly reduced DBDPE-induced ROS levels and increased HepG2 cells viability. 4Subacute Effect of Decabromodiphenyl Ethane on Hepatotoxicity and Hepatic Enzyme Activity in Rats显示文摘Information regarding decabromodiphenyl ethane(DBDPE)effects on hepatotoxicity and metabolism is limited.In the present study,Wistar rats were given oral DBDPE at different doses.DBDPE induced oxidative stress,elevated blood glucose levels,increased CYP2B2 mRNA,CYP2B1/2protein,7-pentoxyresorufin O-depentylase(PROD)activity,and induced CYP3A2 mRNA,CYP3A2protein,and luciferin benzylether debenzylase(LBD)activity.UDPGT activity increased with its increasing exposure levels,suggesting that oral DBDPE exposure induces drug-metabolizing enzymes in rats via the CAR/PXR signaling pathway.The induction of CYPs and co-regulated enzymes of phase II biotransformation may affect the homeostasis of endogenous substrates,including thyroid hormones,which may,in turn,alter glucose metabolism.SUN Ru Bao XI Zhu Ge ZHANG Hua Shan ZHANG Wei 2014Biomedical and Environmental Sciences2014,27,2:4
5Crack status analysis for concrete dams based on measured entropy显示文摘The integrity and safety of concrete dams are seriously affected by the existing cracks in dam bodies, and some serious cracks may cause dam failure or disaster. The propagation of cracks in concrete dams is accompanied by changes in energy distribution, which can be represented by changes in the structure's system entropy. Therefore, the entropy theory can be used in analyzing the behavior of dam cracks. Due to the randomness and locality of crack propagation, it is difficult to predict the location of cracks by traditional monitoring methods. To solve this problem, the influence of spatial positions of monitoring points on inspection zones is represented by a weight index, and the weight index is determined by the distance measure method proposed in this paper. Through the weighted linear fusion method, the entropy of multiple monitoring points is obtained for analyzing the behavior of dam cracks in the selected zones. Meanwhile, the catastrophe theory is used as the variation criterion of an entropy sequence in order to predict the instability time of dam cracks. Case studies are put forward on a high arch dam, and the fusion entropy is calculated according to the monitoring data from strain gauges. 8Influence of Iron Supplementation on DMT1(IRE)-induced Transport of Lead by Brain Barrier Systems in vivo显示文摘Objective To investigate the potential involvement of DMT1(IRE) protein in the brain vascular system in vivo during Pb exposure. Methods Three groups of male Sprague-Dawley rats were exposed to Pb in drinking water, among which two groups were concurrently administered by oral gavage once every other day as the low and high Fe treatment group, respectively, for 6 weeks. At the same time, the group only supplied with high Fe was also set as a reference. The animals were decapitated, then brain capillary-rich fraction was isolate from cerebral cortex. Western blot method was used to identify protein expression, and RT-PCR to detect the change of the m RNA. Results Pb exposure significantly increased Pb concentrations in cerebral cortex. Low Fe dose significantly reduced the cortex Pb levels, However, high Fe dose increased the cortex Pb levels. Interestingly, changes of DMT1(IRE) protein in brain capillary-rich fraction were highly related to the Pb level, but those of DMT1(IRE) m RNA were not significantly different. Moreover, the consistent changes in the levels of p-ERK1/2 or IRP1 with the changes in the levels of DMT1(IRE). 11BRAF V600E/TERT promoter mutations and NIS/TSHR expression in differentiated thyroid carcinomaand their clinical significance显示文摘Objective Telomerase reverse transcriptase(TERT) promoter mutations have recently been described in thyroid carcinoma.The purpose of this study was to investigate the clinical significance of(v-raf murine sarcoma viral oncogene homolog B1) BRAF V600 E and TERT promoter mutations in differentiated thyroid carcinoma(DTC).The relationship between the two mutations and NIS/TSHR expression was also analyzed.Methods We have detected BRAF V600 E and TERT promoter mutations by direct sequencing and NIS/TSHR expression by immunohistochemistry in 229 cases of DTC,52 cases of benign nodular goiter,and 31 cases of normal thyroid tissue.Results The BRAF V600 E mutation was detected in 142(62.0%) of 229 cases of DTC [141 cases of papillary thyroid carcinoma(PTC) and 1 case of follicular thyroid carcinoma(FTC)].TERT promoter mutations were detected in 18(7.9%) of 229 cases of DTC(14 cases of PTC and 4 cases of FTC),including the mutations C228T(0.9%) and C250T(7.0%),which were mutually exclusive.Moreover,11(61.1%) cases also harbored the BRAF V600 E mutation,which was not associated with gender,age,tumor size,lymph node metastasis,and recurrence risk stratification(P >0.05).The rate of TERT promoter mutation was higher in males,age ≥45,and in the middle/high-risk group(P <0.05),and the rate of simultaneous BRAF V600 E and TERT promoter mutations were higher in the middle/high-risk group(P <0.05).In addition,NIS positive rate in the concurrent BRAF V600 E and TERT promoter mutation group(45.5 %) was lower than in other groups(that is,the DTC group with BRAF V600 E or TERT promoter mutations(55.1%),the DTC group with no BRAF V600 E or TERT promoter mutation(57.5%),the nodules and normal group(75.9%);| r | = 0.171,P = 0.002).Conclusion TERT promoter mutations were lower in patients with DTC,with the C250 T mutation being the most common.The detection of BRAF V600 E mutation combined with TERT promoter mutations was instructive for the prognosis assessment and treatment of DTC.Xiaoting Ru Qinjiang Liu Haihong Zhou Rong Yang Lie Bao 2017Oncology and Translational Medicine2017,3,2:1

14Efficacy of Moxifloxacin against Mycobacterium abscessus in Zebrafish Model in vivo显示文摘Objective Moxifloxacin(MFX)shows good in vitro activity against Mycobacterium abscessus and can be a possible antibiotic therapy to treat M.abscessus infection;however,other studies have shown a lower or no activity.We aimed to evaluate MFX activity against M.abscessus using zebrafish(ZF)model in vivo.Methods A formulation of M.abscessus labeled with CM-Dil was micro-injected into ZF.Survival curves were determined by recording dead ZF every day.ZF were lysed,and colony-forming units(CFUs)were enumerated.Bacteria dissemination and fluorescence intensity in ZF were analyzed.Inhibition rates of MFX and azithromycin(AZM,positive control)were determined and compared.Results Significantly increased survival rate was observed with different AZM concentrations.However,increasing MFX concentration did not result in a significant decrease in ZF survival curve.No significant differences in bacterial burdens by CFU loads were observed between AZM and MFX groups at various concentrations.Bacterial fluorescence intensity in ZF was significantly correlated with AZM concentration.However,with increasing MFX concentration,fluorescence intensity decreased slightly when observed under fluorescence microscope.Transferring rates at various concentrations were comparable between the MFX and AZM groups,with no significant difference.Conclusion MFX showed limited efficacy against M.abscessus in vivo using ZF model.Its activity in vivo needs to be confirmed.NIE Wen Juan XIE Zhong Yao GAO Shan TENG Tian Lu ZHOU Wen Qiang SHANG Yuan Yuan JING Wei SHI Wen Hui WANG Qing Feng HUANG Xue Rui CAI Bao Yun WANG Jun WANG Jing GUO Ru GE Qi Ping NIE Li Hui HAN Xi Qin DU Ya Dong CHU Nai Hui 2020Biomedical and Environmental Sciences2020,33,5:1
15Advancing intestinal organoid technology to decipher nano-intestine interactions and treat intestinal disease显示文摘With research burgeoning in nanoscience and nanotechnology,there is an urgent need to develop new biological models that can simulate native structure,function,and genetic properties of tissues to evaluate the adverse or beneficial effects of nanomaterials on a host.Among the current biological models,three-dimensional(3D)organoids have developed as powerful tools in the study of nanomaterial-biology(nano-bio)interactions,since these models can overcome many of the limitations of cell and animal models.A deep understanding of organoid techniques will facilitate the development of more efficient nanomedicines and further the fields of tissue engineering and personalized medicine.Herein,we summarize the recent progress in intestinal organoids culture systems with a focus on our understanding of the nature and influencing factors of intestinal organoid growth.We also discuss biomimetic extracellular matrices(ECMs)coupled with nanotechnology.In particular,we analyze the application prospects for intestinal organoids in investigating nano-intestine interactions.By integrating nanotechnology and organoid technology,this recently developed model will fill the gaps left due to the deficiencies of traditional cell and animal models,thus accelerating both our understanding of intestine-related nanotoxicity and the development of nanomedicines.Lin Bao Xuejing Cui Ru Bai Chunying Chen 2023Nano Research2023,16,3:0

18NMDAR-dependent somatic potentiation of synaptic inputs is correlated withβamyloid-mediated neuronal hyperactivity显示文摘Background:βAmyloid(Aβ)-mediated neuronal hyperactivity,a key feature of the early stage of Alzheimer’s disease(AD),is recently proposed to be initiated by the suppression of glutamate reuptake.Nevertheless,the underlying mechanism by which the impaired glutamate reuptake causes neuronal hyperactivity remains unclear.Chronic suppression of the glutamate reuptake causes accumulation of ambient glutamate that could difuse from synaptic sites at the dendrites to the soma to elevate the tonic activation of somatic N-methyl-D-aspartate receptors(NMDARs).However,less attention has been paid to the potential role of tonic activity change in extrasynaptic glutamate receptors(GluRs)located at the neuronal soma on generation of neuronal hyperactivity.Methods:Whole-cell patch-clamp recordings were performed on CA1 pyramidal neurons in acute hippocampal slices exposed to TFB-threo-β-benzyloxyaspartic acid(TBOA)or human Aβ_(1-42) peptide oligomer.A series of dendritic patch-clamp recordings were made at diferent distances from the soma to identify the location of the changes in synaptic inputs.Moreover,single-channel recording in the cell-attached mode was performed to investigate the activity changes of single NMDARs at the soma.Results:Blocking glutamate uptake with either TBOA or the human Aβ_(1-42) peptide oligomer elicited potentiation of synaptic inputs in CA1 hippocampal neurons.Strikingly,this potentiation specifcally occurred at the soma,depending on the activation of somatic GluN2B-containing NMDARs(GluN2B-NMDARs)and accompanied by a substantial and persistent increment in the open probability of somatic NMDARs.Blocking the activity of GluN2B-NMDARs at the soma completely reversed both the TBOA-induced or the Aβ_(1-42)-induced somatic potentiation and neuronal hyperactivity.Conclusions:The somatic potentiation of synaptic inputs may represent a novel amplifcation mechanism that elevates cell excitability and thus contributes to neuronal hyperactivity initiated by impaired glutamate reuptake in AD.Yifei Bao Xin Yang Yi Fu Zhengyan Li Ru Gong Wei Lu 2021Translational Neurodegeneration2021,10,3:0
19The m6A reader MhYTP2 regulates the stability of its target mRNAs contributing to low nitrogen tolerance in apple (Malus domestica)显示文摘Studies have shown that the m6A reader primarily affects genes expression by participating in the regulation of mRNA localization,splicing,degradation,translation,and other metabolic processes.Previously,we discovered that the apple(Malus domestica)m6A reader MhYTP2 bound with and destabilized m6A-modified MdMLO19 mRNA.In addition,it enhanced the translation efficiency of m6A-modified mRNA of MdGDH1L,encoding a glutamate dehydrogenase,which confers resistance to powdery mildew.In this study,we report the function of MhYTP2 in the regulation of resistance to low nitrogen(N).The overexpression of MhYTP2 enhances the resistance of apple to low N.We show that MhYTP2 binds with and stabilizes the mRNAs of MdALN,which participates in the allantoin catabolic process and cellular response to N starvation in apple;MdPIDL,which participates in root hair elongation;MdTTG1,which is involved in the differentiation process of trichomes;and MdATG8A,which is a core participant in the regulation of autophagy.In addition,MhYTP2 accelerates the degradation of MdRHD3 mRNA,which regulates root development.RNA immunoprecipitation-seq and electrophoretic mobility shift assays show that the mRNAs of MdALN,MdATG8A,MdPIDL,MdTTG1,and MdRHD3 are the direct targets of MhYTP2.Overexpressing or knocking down the above genes in MhYTP2 overexpressing plants dismisses the function of MhYTP2 under low N,suggesting the role of MhYTP2 is dependent on those genes.Together,these results demonstrate that MhYTP2 enhances the resistance of apple to N deficiency by affecting the stability of the bound mRNAs.Tianli Guo Zehua Yang Ru Bao Xiaomin Fu Na Wang Changhai Liu Fengwang Ma 2023Horticulture Research2023,10,6:0
20Investigation and mitigation of Mott neuronal oscillation fluctuation in spiking neural network显示文摘Mott devices,featuring low hardware cost and high energy efficiency,have been demonstrated as a key oscillatory element in artificial neurons to enable spiking neural networks(SNNs)such as conversionbased SNNs(CSNNs).However,there will be inevitably non-ideal fluctuation in the oscillation behavior,causing the accuracy degradation of networks.In this paper,we investigate the Mott neuronal oscillation fluctuation(NOF)through experiments and modeling.The results show that the NOF phenomenon conforms to Gaussian distribution and originates from thermal fluctuation induced switching voltage variations.We construct a two-layer CSNN for image recognition tasks to study the NOF effect and propose the activation function boundary(AFB)method to strengthen the stability of the network.The results indicate that AFB can improve the accuracy of CSNN by up to 15.5%by tightening output distribution.Lindong WU Zongwei WANG Lin BAO Linbo SHAN Zhizhen YU Yunfan YANG Shuangjie ZHANG Guandong BAI Cuimei WANG John ROBERTSON Yuan WANG Yimao CAI Ru HUANG 2024Science China(Information Sciences)2024,67,2:0
