| 1 | RAB31 marks and controls an ESCRT-independent exosome pathway显示文摘Exosomes are generated within the multivesicular endosomes(MVEs)as intraluminal vesicles(ILVs)and secreted during the fusion of MVEs with the cell membrane.The mechanisms of exosome biogenesis remain poorly explored.Here we identify that RAB31 marks and controls an ESCRT-independent exosome pathway.Active RAB31,phosphorylated by epidermal growth factor receptor(EGFR),engages flotillin proteins in lipid raft microdomains to drive EGFR entry into MVEs to form ILVs,which is independent of the ESCRT(endosomal sorting complex required for transport)machinery.Active RAB31 interacts with the SPFH domain and drives ILV formation via the Flotillin domain of flotillin proteins.Meanwhile,RAB31 recruits GTPase-activating protein TBC1D2B to inactivate RAB7,thereby preventing the fusion of MVEs with lysosomes and enabling the secretion of ILVs as exosomes.These findings establish that RAB31 has dual functions in the biogenesis of exosomes:driving ILVs formation and suppressing MVEs degradation,providing an exquisite framework to better understand exosome biogenesis. | Denghui Wei Weixiang Zhan Ying Gao Liyan Huang Run Gong Wen Wang Ruhua Zhang Yuanzhong Wu Song Gao Tiebang Kang | 2021 | Cell Research2021,31,2: | 12 |

In the present study, we carried out a haplotype analysis to identify the exact bound-ary by using the combination of a set of microsatellite mark-ers and single nucleotide polymorphism (SNP) markers in two major NPC families. | 4 | Systematic screening for potential therapeutic targets in osteosarcoma through a kinome-wide CRISPR-Cas9 library显示文摘Objective:Osteosarcoma is the most common primary malignant bone tumor.However,the survival of patients with osteosarcoma has remained unchanged during the past 30 years,owing to a lack of efficient therapeutic targets.Methods:We constructed a kinome-targeting CRISPR-Cas9 library containing 507 kinases and 100 nontargeting controls and screened the potential kinase targets in osteosarcoma.The CRISPR screening sequencing data were analyzed with the Model-based Analysis of Genome-wide CRISPR/Cas9 Knockout(MAGeCK)Python package.The functional data were applied in the 143B cell line through lenti-CRISPR-mediated gene knockout.The clinical significance of kinases in the survival of patients with osteosarcoma was analyzed in the R2:Genomics Analysis and Visualization Platform.Results:We identified 53 potential kinase targets in osteosarcoma.Among these targets,we analyzed 3 kinases,TRRAP,PKMYT1,and TP53RK,to validate their oncogenic functions in osteosarcoma.PKMYT1 and TP53RK showed higher expression in osteosarcoma than in normal bone tissue,whereas TRRAP showed no significant difference.High expression of all 3 kinases was associated with relatively poor prognosis in patients with osteosarcoma.Conclusions:Our results not only offer potential therapeutic kinase targets in osteosarcoma but also provide a paradigm for functional genetic screening by using a CRISPR-Cas9 library,including target design,library construction,screening workflow,data analysis,and functional validation.This method may also be useful in potentially accelerating drug discovery for other cancer types. | Yuanzhong Wu Liwen Zhou Zifeng Wang Xin Wang Ruhua Zhang Lisi Zheng Tiebang Kang | 2020 | Cancer Biology & Medicine2020,17,3: | 2 |
