| 3 | Uncompromised NK cell activation is essential for virus-specific CTL activity during acute influenza virus infection显示文摘Natural killer (NK) cells are indispensable components of both the innate and adaptive immune response. However,their precise roles in the cross-talk between innate and adaptive immunity during influenza virus infection remaincontroversial. By comparing NK cell dynamics and activity under a sub-lethal dose and high dose of influenza virusinfection, we showed that influenza virus PR8 directly infected NK cells during natural infection, which wasconsistent with our previous findings obtained from an in vitro investigation of human NK cells. The impairments incytotoxicity and IFN-γ production by spleen NK cells following high-dose infection were accompanied by decreasedvirus-specific killing mediated by cytotoxic T lymphocytes (CTLs). Importantly, the weakened CTL activity could bereversed by adoptive transfer of spleen NK cells harvested from low-dose-infected mice but not healthy donors.Taken together, our data provide direct evidence supporting the contribution of NK cells to antiviral T-cellresponses. | 4 | Secalonic acid D induces cell apoptosis in both sensitive and ABCG2-overexpressing multidrug resistant cancer cells through upregulating c-Jun expression显示文摘Secalonic acid D(SAD) could inhibit cell growth in not only sensitive cells but also multidrug resistant(MDR) cells. However, the molecular mechanisms need to be elucidated. Here, we identified that SAD possessed potent cytotoxicity in 3 pairs of MDR and their parental sensitive cells including S1-MI-80 and S1,H460/MX20 and H460, MCF-7/ADR and MCF-7 cells. Furthermore, SAD induced cell G2/M phase arrest via the downregulation of cyclin B1 and the increase of CDC2 phosphorylation. Importantly, JNK pathway upregulated the expression of c-Jun in protein level and increased c-Jun phosphorylation induced by SAD, which was linked to cell apoptosis via c-Jun/Src/STAT3 pathway. To investigate the mechanisms of upregulation of c-Jun protein by SAD, the mR NA expression level and degradation of c-Jun were examined. We found that SAD did not alter the mR NA level of c-Jun but inhibited its proteasome-dependent degradation. | 5 | Growth Inhibitory Effects of Garlic Polysaccharide on Human HepG2 Cells显示文摘[Objective] The growth inhibitory effects of garlic polysaccharide(GPS) on human Hep G2 cells were evaluated in this paper. [Method] Hep G2 cells were treated with GPS for 48 h for morphology assay by transition electron microscope. Anti-proliferative effects with the same treatment for 24 hand 48 h were assayed by MTT method.Cell cycle distribution and apoptosis assay of treated cells were performed in flow cytometry. [Result] The results showed that GPS enhanced growth inhibitory effect on Hep G2 cells in a time-and dose-dependent manner. PI(Propidium iodide)/Annexin V staining analyzed by FCM(flow cytometry) demonstrated that GPS has a cytotoxic effect on tumor cells. Cell cycle arrest of Hep G2 treated with GPS occurred in G2 phase. | 6 | Comparison of three remotely sensed drought indices for assessing the impact of drought on winter wheat yield显示文摘Agricultural drought threatens food security.Numerous remote-sensing drought indices have been developed,but their different principles,assumptions and physical quantities make it necessary to compare their suitability for drought monitoring over large areas.Here,we analyzed the performance of three typical remote sensing-based drought indices for monitoring agricultural drought in two major agricultural production regions in Shaanxi and Henan provinces,northern China(predominantly rain-fed and irrigated agriculture,respectively):vegetation health index(VHI),temperature vegetation dryness index(TVDI)and drought severity index(DSI).We compared the agreement between these indices and the standardized precipitation index(SPI),soil moisture,winter wheat yield and National Meteorological Drought Monitoring(NMDM)maps.On average,DSI outperformed the other indices,with stronger correlations with SPI and soil moisture.DSI also corresponded better with soil moisture and NMDM maps.The jointing and grain-filling stages of winter wheat are more sensitive to water stress,indicating that winter wheat required more water during these stages.Moreover,the correlations between the drought indices and SPI,soil moisture,and winter wheat yield were generally stronger in Shaanxi province than in Henan province,suggesting that remote-sensing drought indices provide more accurate predictions of the impacts of drought in predominantly rain-fed agricultural areas. | Jianxi Huang Wen Zhuo Ying Li Ran Huang Fernando Sedano Wei Su Jinwei Dong Liyan Tian Yanbo Huang Dehai Zhu Xiaodong Zhang | 2020 | International Journal of Digital Earth2020,13,4: | 4 |

[Method] SGC-7901 cells were treated with ligustrazine and DDP alone or combined for 48 h for morphology assay.Anti-proliferative effects with the same treatment for 24, 48 and 72 h were assayed by MTT method, respectively. Cell cycle distribution and apoptosis assay of treated cells were performed in flow cytometry. Zhengjun Jin's protocol was used to assay the effect of drug combination. [Result] Ligustrazine significantly increased the proliferation inhibition rate of DDP on SGC-7901 cells in combination with DDP, comparing with the effects of ligustrazine or DDP alone, and exhibited synergistic antitumor effect. [Methods] SGC-7901 cell lines were treated with ligustrazine and DDP alone or combined for 48 h for Western blot analysis, respectively. Western blot analysis was used to determine the expression of proteins involved in apoptosis including NF-κB p65, bax and caspase-3. [Results] The viability of SGC-7901 cells was inhibited after treated with ligustrazine and/or combined with DDP. The expression of NF-κB P65 protein decreased after treated with drugs, in which the protein decreased significantly in 1.2 mg/ml of TMP combined with 2 μg/ml of DDP group.Meanwhile, we investigated the protein expression of bax and caspase-3. The results showed that the expression of the two proteins increased following with the increasing concentration of TMP. | 17 | Vitamin B6 regulates IL-33 homeostasis to alleviate type 2 inflammation显示文摘Interleukin-33(IL-33)is a crucial nuclear cytokine that induces the type 2 immune response and maintains immune homeostasis.The fine-tuned regulation of IL-33 in tissue cells is critical to control of the type 2 immune response in airway inflammation,but the mechanism is still unclear.Here,we found that healthy individuals had higher phosphate-pyridoxal(PLP,an active form of vitamin B6)concentrations in the serum than asthma patients.Lower serum PLP concentrations in asthma patients were strongly associated with worse lung function and inflammation.In a mouse model of lung inflammation,we revealed that PLP alleviated the type 2 immune response and that this inhibitory effect relied on the activity of IL-33.A mechanistic study showed that in vivo,pyridoxal(PL)needed to be converted into PLP,which inhibited the type 2 response by regulating IL-33 stability.In mice heterozygous for pyridoxal kinase(PDXK),the conversion of PL to PLP was limited,and IL-33 levels were increased in the lungs,aggravating type 2 inflammation.Furthermore,we found that the mouse double minute 2 homolog(MDM2)protein,an E3 ubiquitin-protein ligase,could ubiquitinate the N-terminus of IL-33 and sustain IL-33 stability in epithelial cells.PLP reduced MDM2-mediated IL-33 polyubiquitination and decreased the level of IL-33 through the proteasome pathway.In addition,inhalation of PLP alleviated asthma-related effects in mouse models.In summary,our data indicate that vitamin B6 regulates MDM2-mediated IL-33 stability to constrain the type 2 response,which might help develop a potential preventive and therapeutic agent for allergy-related diseases. | Songling Zhu Shufen Zhong Kebin Cheng Li-Sha Zhang Jiu-wu Bai Zu Cao Su Wang Wen Chen Shipeng Cheng Liyan Ma Zhiyang Ling Yuying Huang Wangpeng Gu Xiaoyu Sun Chunyan Yi Meng Zhao Shuo Liang Jin-Fu Xu Bing Sun Yaguang Zhang | 2023 | Cellular & Molecular Immunology2023,20,7: | 1 |

Hu in LPS-stimulated RAW264.7 Macrophages显示文摘The juice leaked from broken phloem of Paulownia elongate S.Y.Hu has been used to cure acute inflammation resulted from stung injury by poisonous insects for a long time in China,but its potential mechanism remains unclear.The present study was designed to evaluate anti-inflammatory activity and mechanism of total flavonoids from the phloem of P.elongate S.Y.Hu in RAW264.7 cells.Lipopolysaccharide(LPS)-induced nitric oxide(NO)was measured by Griess and mRNA of pro-inflammatory mediators was analyzed by q-PCR.Cell viability was measured using cell counting kit(CCK)-8 assay.The protein level was analyzed by Western blot.The results showed that the total flavonoids of P.elongate significantly inhibited the production of the pro-inflammatory mediators such as NO,interleukin(IL)-1β and interleukin(IL)-6 in LPS-stimulated RAW264.7 cells.Total flavonoids of P.elongate exerted potential anti-inflammatory activity through the regulation of several signaling pathways.Total flavonoids of P.elongate inhibited JAK/STAT by blocking JAK2 and STAT3 phosphorylation levels.This is the first report on anti-inflammatory activity of total flavonoids from the phloem of P.elongate,which suggests that the total flavonoids of P.elongate may have great potential for the development of anti-inflammatory drug to treat inflammatory disorders. | Liyan LI Tao HUANG Chong LAN | 2021 | Agricultural Biotechnology2021,10,3: | 1 |
