| 1 | Relationship between Fusobacterium nucleatum,inflammatory mediators and microRNAs in colorectal carcinogenesis显示文摘AIM To examine the effect of Fusobacterium nucleatum(F. nucleatum) on the microenvironment of colonic neoplasms and the expression of inflammatory mediators and microRNAs(miRNAs).METHODS Levels of F. nucleatum DNA, cytokine gene mRNA(TLR2, TLR4, NFKB1, TNF, IL1 B, IL6 and IL8), and potentially interacting miRNAs(miR-21-3p, miR-22-3p, mi R-28-5p, miR-34a-5p, miR-135b-5p) were measured by quantitative polymerase chain reaction(qPCR) TaqMan? assays in DNA and/or RNA extracted from the disease and adjacent normal fresh tissues of 27 colorectal adenoma(CRA) and 43 colorectal cancer(CRC) patients. KRAS mutations were detected by direct sequencing and microsatellite instability(MSI) status by multiplex PCR. Cytoscape v3.1.1 was used to construct the postulated miRNA:mRNA interaction network.RESULTS Overabundance of F. nucleatum in neoplastic tissue compared to matched normal tissue was detected in CRA(51.8%) and more markedly in CRC(72.1%). We observed significantly greater expression of TLR4, IL1 B, IL8, and miR-135 b in CRA lesions and TLR2, IL1 B, IL6, IL8, mi R-34 a and miR-135 b in CRC tumours compared to their respective normal tissues. Only two transcripts for miR-22 and miR-28 were exclusively downregulated in CRC tumour samples. The mRNA expression of IL1 B, IL6, IL8 and miR-22 was positively correlated with F. nucleatum quantification in CRC tumours. The mRNA expression of miR-135 b and TNF was inversely correlated. The miRNA:mRNA interaction network suggested that the upregulation of miR-34 a in CRC proceeds via a TLR2/TLR4-dependent response to F. nucleatum. | 3 | Serrated pathway in colorectal carcinogenesis显示文摘Serrated adenocarcinoma is a recently described subset of colorectal cancer(CRC),which account for about10%of all CRCs and follows an alternative pathway in which serrated polyps replace the traditional adenoma as the precursor lesion to CRC.Serrated polyps form a heterogeneous group of colorectal lesions that includes hyperplastic polyps(HPs),sessile serrated adenoma(SSA),traditional serrated adenoma(TSA)and mixed polyps.HPs are the most common serrated polyp followed by SSA and TSA.This distinct histogenesis is believed to have a major influence in prevention strategies,patient prognosis and therapeutic impact.Genetically,serrated polyps exhibited also a distinct pattern,with KRAS and BRAF having an important contribution to its development.Two other molecular changes that have been implicated in the serrated pathway include microsatellite instability and the CpG island methylator phenotype.In the present review we will address the current knowledge of serrated polyps,clinical pathological features and will update the most recent findings of its molecular pathways.The understanding of their biology and malignancy potential is imperative to implement a surveillance approach in order to prevent colorectal cancer development. | Letícia Yamane Cristovam Scapulatempo-Neto Rui Manuel Reis Denise Peixoto Guimares | 2014 | World Journal of Gastroenterology2014，20，10： | 8 |
| 4 | Mechanotransduction via the coordinated actions of integrins,PI3K signaling and Connexin hemichannels显示文摘Mechanical loading opens connexin 43(Cx43)hemichannels(HCs),leading to the release of bone anabolic molecules,such as prostaglandins,from mechanosensitive osteocytes,which is essential for bone formation and remodeling.However,the mechanotransduction mechanism that activates HCs remains elusive.Here,we report a unique pathway by which mechanical signals are effectively transferred between integrin molecules located in different regions of the cell,resulting in HC activation.Both integrinα5 andαV were activated upon mechanical stimulation via either fluid dropping or flow shear stress(FSS).Inhibition of integrinαV activation or ablation of integrinα5 prevented HC opening on the cell body when dendrites were mechanically stimulated,suggesting mechanical transmission from the dendritic integrinαV toα5 in the cell body during HC activation.In addition,HC function was compromised in vivo,as determined by utilizing an antibody blockingαV activation andα5-deficient osteocyte-specific knockout mice.Furthermore,inhibition of integrinαV activation,but not that ofα5,attenuated activation of the phosphoinositide 3-kinase(PI3K)-protein kinase B(AKT)signaling pathway upon mechanical loading,and the inhibition of PI3K/AKT activation blocked integrinα5 activation and HC opening.Moreover,HC opening was blocked only by an anti-integrinαV antibody at low but not high FSS levels,suggesting that dendriticαV is a more sensitive mechanosensor thanα5 for activating HCs.Together,these results reveal a new molecular mechanism of mechanotransduction involving the coordinated actions of integrins and PI3K/AKT in osteocytic dendritic processes and cell bodies that leads to HC opening and the release of key bone anabolic factors. | Manuel A.Riquelme Sumin Gu Rui Hua Jean X.Jiang | 2021 | Bone Research2021，9，1： | 7 |

| 18 | Biological oyster shell waste enhances polyphenylene sulfide composites and endows them with antibacterial properties显示文摘To date,there is no research that deals with biological waste as fillers in polyphenylene sulfide(PPS).In this study,oyster shells were recycled and treated to prepare thermally-treated oyster shells(TOS),which were used as PPS fillers to make new bio-based antibacterial composite materials.The effect of varying the content of TOS was studied by means of structure and performance characterization.PPS/TOS composites were demonstrated to have an antibacterial effect on the growth of E coli and S.aureus.Qualitative analysis showed that when the TOS content was≥30%and 40%,the composite materials had an apparent inhibition zone.Quantitative analysis showed that the antibacterial activity increased with the TOS content.Fourier transform infrared spectroscopy indicated the formation of hydrogen bonds between the molecular chains of TOS and PPS and the occurrence of a coordination reaction.At 10%TOS,the composite tensile strength reached a maximum value of 72.5 MPa,which is 9.65%higher than that of pure PPS.The trend of bending properties is the same as that of tensile properties,showing that the maximum property was reached for the composite with 10%TOS.At the same time,the crystallinity and contact angle were the highest,and the permeability coefficient was the lowest.The fatigue test results indicated that for the composite with 10%TOS,the tensile strength was 23%lower than static tensile strength,and the yield strength was 10%lower than the static yield strength.The results of the study showed that TOS not only could reduce the cost of PPS,but also could impart antibacterial properties and enhance the mechanical and,barrier properties,the thermostability,as well as the crystallinity. | Chi-Hui Tsou Rui Zeng Neng Wan Manuel Reyes De Guzman Xue-Fei Hu Tao Yang Chen Gao Xiaomei Wei Jia Yi Li Lan Rui-Tao Yang Ya-Li Sun | 2023 | Chinese Journal of Chemical Engineering2023，57，5： | 0 |

| 20 | 3D Resistivity Imaging of Buildings and Foundations in Urban and Protected Areas显示文摘The use of Geophysics in Civil Engineering is an established practice.Herein,it will be exploited the use of 3D resistivity imaging to investigate foundations and features under buildings in urban areas where no direct invasive methods are allowed.High-resolution 3D resistivity requires the deployment of a dense grid of electrodes and,in urban areas,these methods face problems such as space limitation and availability owing to buildings,pavements,asphalts,obstacles and infrastructure.Thus,the use of conventional electrode arrays and grids may not be possible and alternatives must be sought.Herein a non-conventional array,pole-pole,is explored to characterize foundations and features under existing constructions in delicate and protected environments.A simple but effective data check is also proposed and allows to estimate the resistivity distribution in the area.Results prove the adequacy and versatility of the pole-pole array to carry out 3D resistivity surveys in urban areas.Data quality checks also proved effective and provided early qualitative images of the resistivity distribution in the areas to be used as an aid in later interpretation and data modelling stages. | Manuel Matias Nuno Barraca Rui Moura | 2019 | Journal of Civil Engineering and Architecture2019，13，1： | 0 |
