1Hormones induce the formation of luminal-derived basal cells in the mammary gland显示文摘In the mammary gland, it is widely believed that the luminal cells are unipotent after birth, contributing only to the luminal compartment in normal development. Here, by lineage tracing, we uncovered an unexpected potential of luminal cells that can give rise to basal cells during pregnancy. These luminal-derived basal cells (LdBCs) persisted through mammary regression and gen erated more proge ny in successive rounds of preg nan cies. LdBCs express basal markers as well as estroge n receptor a (ERa). I n ovariectomized (OVX) mice, stimulation with estrogen and progesterone promoted the formation of LdBCs. In serial transplantation assays, LdBCs were able to rec on stitute new mammary glands in a hormone-depende nt manner. Tran scriptome an alysis and genetic experiments suggest that Wnt/p-catenin signaling is essential for the formation and maintenance of LdBCs. Our data uncover an unexpected bi-potency of luminal cells in a physiological context. 2Protein C receptor is a therapeutic stem cell target in a distinct group of breast cancers显示文摘Breast cancer is a heterogeneous disease.In particular,triple-negative breast cancer(TNBC)comprises various molecular subgroups with unclear identities and currently has few targeted treatment options.Our previous study identified protein C receptor(Procr)as a surface marker on mammary stem cells(MaSCs)located in the basal layer of the normal mammary gland.Given the possible connection of TNBC with basal layer stem cells,we conducted comparative analyses of Procr in breast cancers of mouse and human origin.In mouse mammary tumors,we showed that Procr+cells are enriched for cancer stem cells(CSCs)in Wnt1 basal-like tumors,but not in Brea basal-like tumors or PyVT luminal tumors.In human cancers,PROCR was robustly expressed in half of TNBC cases.Experiments with patient-derived xenografts(PDXs)revealed that PROCR marks CSCs in this discrete subgroup(referred to as PROCR+TNBC).Interfering with the function of PROCR using an inhibitory nanobody reduced the CSC numbers,arrested tumor growth and prevented rapid tumor recurrence.Our data suggest a key role of MaSC in breast tumorigenesis.Moreover,our work indicates that PROCR can be used as a biomarker to stratify TNBC into clinically relevant subgroups and may provide a novel targeted treatment strategy for this clinically important tumor subtype.Daisong Wang Xin Hu Chunye Liu Yingying Jia Yiqin Bai Cheguo Cai Jingqiang Wang Lanyue Bai Ruikai Yang ChangDong Lin Yi-Rong Liu Shan Li Feng Qiao Ling Yao Li Chen Gaoxiang Ge Hai Jiang Dianfan Li Lin Li JianFeng Chen Zhi-Ming Shao Yi Arial Zeng 2019Cell Research2019,29,10:2

4Flow Loss Mechanism in a Supercritical Carbon Dioxide Centrifugal Compressor at Low Flow Rate Conditions显示文摘With the advantages of high efficiency and compact structure,supercritical carbon dioxide(sC02)Brayton cycles have bright prospects for development in energy conversion field.As one of the core components of the power cycle,the centrifugal compressor tends to operate near the critical point(304.13 K,7.3773 MPa).Normally,the compressor efficiency increases as the inlet temperature decreases.When the inlet temperature is close to the critical point,the density increases sharply as the temperature decreases,which results in quickly decreasing of volume flow rate and efficiency reducing.The flow loss mechanism of the sCO_(2) compressor operating at low flow rate is studied in this paper.Computational fluid dynamics(CFD) simulations for sCO_(2)compressor were carried out at various inlet temperatures and various mass flow rates.When the sCO_(2)compressor operates at low volume flow rate,the flow loss is generated mainly on the suction side near the trailing edge of the blade.The flow loss is related to the counterclockwise vortexes generated on the suction side of the main blade.The vortexes are caused by the flow separation in the downstream region of the impeller passage,which is different from air compressors operating at low flow rates.The reason for this flow separation is that the effect of Coriolis force is especially severe for the sCO_(2) fluid,compared to the viscous force and inertial force.At lower flow rates,with the stronger effect of Coriolis force,the direction of relative flow velocity deviates from the direction of radius,resulting in its lower radial component.The lower radial relative flow velocity leads to severe flow separation on the suction side near the trailing edge of the main blade.YANG Zimu JIANG Hongsheng ZHUGE Weilin CAI Ruikai YANG Mingyang CHEN Haoxiang QIAN Yuping ZHANG Yangjun 2024Journal of Thermal Science2024,33,1:0
